| 1 | Tomato SlAN11 regulates flavonoid biosynthesis and seed dormancy by interaction with bHLH proteins but not with MYB proteins显示文摘The flavonoid compounds are important secondary metabolites with versatile human nutritive benefits and fulfill a multitude of functions during plant growth and development.The abundance of different flavonoid compounds are finely tuned with species-specific pattern by a ternary MBW complex,which consists of a MYB,a bHLH,and a WD40 protein,but the essential role of SlAN11,which is a WD40 protein,is not fully understood in tomato until now.In this study,a tomato WD40 protein named as SlAN11 was characterized as an effective transcription regulator to promote plant anthocyanin and seed proanthocyanidin(PA)contents,with late flavonoid biosynthetic genes activated in 35S::SlAN11 transgenic lines,while the dihydroflavonol flow to the accumulation of flavonols or their glycosylated derivatives was reduced by repressing the expression of SlFLS in this SlAN11-overexpressed lines.The above changes were reversed in 35S::SlAN11-RNAi transgenic lines except remained levels of flavonol compounds and SlFLS expression.Interestingly,our data revealed that SlAN11 gene could affect seed dormancy by regulating the expressions of abscisic acid(ABA)signaling-related genes SlABI3 and SlABI5,and the sensitivity to ABA treatment in seed germination is conversely changed by SlAN11-overexpressed or-downregulated lines.Yeast two-hybrid assays demonstrated that SlAN11 interacted with bHLH but not with MYB proteins in the ternary MBW complex,whereas bHLH interacted with MYB in tomato.Our results indicated that low level of anthocyanins in tomato fruits,with low expression of bHLH(SlTT8)and MYB(SlANT1 and SlAN2)genes,remain unchanged upon modification of SlAN11 gene alone in the transgenic lines.These results suggest that the tomato WD40 protein SlAN11,coordinating with bHLH and MYB proteins,plays a crucial role in the fine adjustment of the flavonoid biosynthesis and seed dormancy in tomato. | Yongfeng Gao Jikai Liu Yongfu Chen Hai Tang Yang Wang Yongmei He Yongbin Ou Xiaochun Sun Songhu Wang Yinan Yao | 2018 | Horticulture Research2018,5,1: | 13 |
| 2 | Construction and external validation of a nomogram that predicts lymph node metastasis in early gastric cancer patients using preoperative parameters显示文摘Objective: To create a nomogram to predict the incidence of lymph node metastasis(LNM) in early gastric cancer(EGC) patients and to externally validate the nomogram.Methods: To construct the nomogram,we retrospectively analyzed a primary cohort of 272 EGC patients.Univariate analysis and a binary logistic regression were performed.A nomogram predicting the incidence of LNM in EGC patients was created.The discrimination ability of the nomogram was measured using the concordance index(c-index),and the nomogram was also calibrated.Then,another prospective cohort of 81 patients was analyzed to validate the nomogram.Results: In the primary cohort,LNM was pathologically confirmed in 37(13.6%) patients.In multivariate analysis,the presence of an ulcer,the maximum lesion diameter observed via gastroscopy,the thickness of the lesion observed via endoscopic ultrasonography,and the presence of enlarged lymph nodes on computed tomography(CT) were independent risk factors for LNM.A nomogram was then created based on the regression model with the c-index of 0.905,and the calibration curve of the nomogram fell approximately on the ideal 45-degree line.The cut-off score of the nomogram was 110,and the sensitivity,specificity,positive predictive and negative predictive values of the nomogram in the primary cohort were 81.1%,86.0%,47.6% and 96.7%,respectively,and in the prospective validation cohort were 75.0%,91.0%,60.0% and 95.5%,respectively.The calibration curve of the external validation cohort was almost on the 45-degree line.Conclusions: We developed an effective nomogram predicting the incidence of LNM for EGC patients. | Yinan Zhang Yiqiang Liu Ji Zhang Xiaojiang Wu Xin Ji Tao Fu Ziyu Li Qi Wu Zhaode Bu Jiafu Ji | 2018 | Chinese Journal of Cancer Research2018,30,6: | 12 |

| 9 | Direct Effects of Activin A on the Activation of Mouse Macrophage RAW264.7 Cells显示文摘Macrophages play critical roles in innate immune and acquired immune via secreting pro-inflammatory mediators, phagocytosing microorganisms and presenting antigens. Activin A, a member of transforming growth factor β (TGF-β) superfamily, is produced by macrophages and microglia cells. In this study, we reported a direct effect of activin A as a pro-inflammatory factor on mouse macrophage cell line RAW264.7 cells. Our data revealed that activin A could not only increase IL-1β and IL-6 production from RAW264.7 cells, but also promote pinocytic and phagocytic activities of RAW264.7 cells. In addition, activin A obviously up-regulated MHC II expression on the surface of RAW264.7 cells, whereas did not influence MHC I expression. Activin A also enhanced CD80 expression, which is a marker of activated macrophages, but did not influence RAW264.7 cell proliferation. | 10 | Effect of direct current pulse stimulating acupoints of JiaJi (T10-L3) and Ciliao (BL 32) with Han's Acupoint Nerve Stimulator on labour pain in women: a randomized controlled clinical study显示文摘OBJECTIVE: To assess the clinical effect and safety of direct current(DC) pulse produced by Han's Acupoint Nerve Stimulator in reduction(HANS) of labor pain.METHODS: Totally 120 participants were enrolled in this clinical trial, and were randomly divided into4 groups including: HANS group, patient controlled intravenous analgesia(PCIA) group, patient-controlled epidural analgesia(PCEA) group and control group. The HANS group was treated by stimulating the acupoints of Jia Ji(T10-L3) and Ciliao(BL 32)with DC pulse of 100 Hz and 15-30 m A produced by a portable battery-powered Han's Acupoint Nerve Stimulator for 30 min. The PCIA group was intravenously infused Ondansetron(8 mg) for 5 min,then tramadol injection(1.5 mg/kg) was slowly dripped by using Baxter APⅡ electronic pump with50 m L tramadol(0.70%) + ondansetron(8 mg),background infusion 2 m L/h, PCA dose of 2 m L,lockout interval of 10 min. In PCEA group, women received intrathecal injection ropivacaine(3 mg) in L2-3, and epidural catheter was connected to Baxter APⅡ electronic pump, with 100 m L Ropivacaine(0.1%) and Sufentanil(50 ug), background infusion5 m L, Patient controlled analgesia(PCA) dose of 5m L, lockout interval of 10 min. The control group was not received analgesia. The visual analogue scale(VAS), stage and manner of labor, Apgar score of newborn, neonatal weights, oxytocin dosage,postpartum hemorrhage and side effects were monitored in all groups.RESULTS: The vital signs were all stable in the four analgesic groups. After analgesia, there was statistical difference in VAS score between HANS group and control group, between PCEA group and the control group, between PCIA group and control group. The analgesic effect in the PCEA group was significantly better than that of other two groups.The second stage of labor in the PCEA group was longer than the other three groups, showing significant difference between them. The Apgar score of newborn 1min after birth in the PCIA group was slightly lower than that of the other two groups,showing significant difference between them. The neonatal weights between four groups were not significantly different. The rate of cesarean sectionin the control group was significantly higher than that of the labor analgesia group, there was statistically difference in four groups. The number of PCIA group that used oxytocin was lower than that of other three groups. There was no significant difference in postpartum hemorrhage between four groups. The side effects of the PCEA group were itching, uroschesis and neonatal asphyxia and PCIA group were nausea and vomiting and neonatal asphyxia. | 12 | Crustal P-wave velocity structure in the northeastern margin of the Qinghai-Tibetan Plateau and insights into crustal deformation显示文摘The transitional area between the northeastern margin of the Qinghai-Tibetan Plateau, Ordos Block and Alxa Block,also being the northern segment of the North-South Seismic Belt, is characterized by considerably high seismicity level and high risk of strong earthquakes. In view of the special tectonic environment and deep tectonic setting in this area, this study used two seismic wide-angle reflection/refraction cross profiles for double constraining, so as to more reliably obtain the fine-scale velocity structure characteristics in both the shallow and deep crust of individual blocks and their boundaries in the study area,and further discuss the seismogenic environment in seismic zones with strong historical earthquakes. In this paper, the P-wave data from the two profiles are processed and interpreted, and two-dimensional crustal velocity structure models along the two profiles are constructed by travel time forward modeling. The results show that there are great differences in velocity structure,shape of intra-crustal interfaces and crustal thickness among different blocks sampled by the two seismic profiles. The crustal thickness along the Lanzhou-Huianbu-Yulin seismic sounding profile(L1) increases from ~43 km in the western margin of Ordos Block to ~56 km in the Qilian Block to the west. In the Ordos Block, the velocity contours vary gently, and the average velocity of the crust is about 6.30 km s-1; On the other hand, the velocity structures in the crust of the Qilian Block and the arclike tectonic zone vary dramatically, and the average crustal velocities in these areas are about 0.10 km s-1 lower than that of the Ordos Block. In addition, discontinuous low-velocity bodies(LVZ1 and LVZ2) are identified in the crust of the Qilian Block and the arc-like tectonic zone, the velocity of which is 0.10–0.20 km s-1 lower than that of the surroundings. The average crustal thickness of the Ordos Block is consistently estimated to be around 43 km along both Profile L2(Tongchuan-Huianbu-Alashan left banner seismic sounding profile) and Profile L1. In contrast to the gently varying intra-crustal interfaces and velocity contours in the Ordos Block along Profile L1, which is a typical structure characteristic of stable cratons, the crustal structure in the Ordos Block along Profile L2 exhibits rather complex variations. This indicates the presence of significant structural differences in the crust within the Ordos Block. The crustal structure of the Helan Mountain Qilian Block and the Yinchuan Basin is featured by 'uplift and depression' undulations, showing the characteristics of localized compressional deformation.Moreover, there are low-velocity zones with alternative high and low velocities in the middle and lower crust beneath the Helan Mountain, where the velocity is about 0.15–0.25 km s-1 lower than that of the surrounding areas. The crustal thickness of the Alxa Block is about 49 km, and the velocity contours in the upper and middle-lower crust of the block vary significantly. The complex crustal velocity structure images along the two seismic sounding profiles L1 and L2 reveal considerable structural differences among different tectonic blocks, their coupling relationships and velocity structural features in the seismic zones where strong historical earthquakes occurred. | 13 | Single-cell transcriptomics reveals gene signatures and alterations associated with aging in distinct neural stem/progenitor cell subpopulations显示文摘Aging associated cognitive decline has been linked to dampened neural stem/progenitor cells (NSC/NPCs) activities manifested by decreased proliferation, reduced propensity to produce neurons, and increased differentiation into astrocytes. While gene transcription changes objectively reveal molecular alterations of cells undergoing various biological processes, the search for molecular mechanisms underlying aging of NSC/NPCs has been confronted by the enormous heterogeneity in cellular compositions of the brain and the complex cellular microenvironment where NSC/NPCs reside. Moreover, brain NSC/NPCs themselves are not a homogenous population, making it even more difficult to uncover NSC/NPC sub-type specific aging mechanisms. Here, using both population-based and single cell transcriptome analyses of young and aged mouse forebrain ependymal and subependymal regions and comprehensive “big-data” processing, we report that NSC/NPCs reside in a rather inflammatory environment in aged brain, which likely contributes to the differentiation bias towards astrocytes versus neurons. Moreover, single cell transcriptome analyses revealed that different aged NSC/NPC subpopulations, while all have reduced cell proliferation, use different gene transcription programs to regulate age-dependent decline in cell cycle. Interestingly, changes in cell proliferation capacity are not influenced by inflammatory cytokines, but likely result from cell intrinsic mechanisms. The Erk/Mapk pathway appears to be critically involved in regulating age-dependent changes in the capacity for NSC/NPCs to undergo clonal expansion. | 15 | Inhibitory Effect of Short-Term Palpebral Margin Cleaning with Antibiotic Eye Drops on Ocular Surface Flora before Cataract Extraction:A New Preoperative Antibacterial Method显示文摘Objective To explore the clinical significance of the combined application of palpebral margin cleaning and antibiotic eye drops in inhibiting bacterial growth in the palpebral margin and conjunctival sacs before cataract extraction.Methods In this study,61 patients(97 eyes)with age-related cataract who underwent phacoemulsification and intraocular lens implantation were selected,and randomly grouped.In the experimental group,the combined application of palpebral margin cleaning with cotton pads and levofloxacin eye drops was given for three days before the surgery.In the control group,levofloxacin eye drops alone were applied for three consecutive days.Bacteria samples from the conjunctival sac and eyelid margins were cultivated and identified before and three days after taking antimicrobial measures,respectively.Results In the experimental group,the positive rates of the two bacteria samples were 100%(50/50)and 40%(20/50)before and 10%(5/50)and 0%(0/50)after the treatment.In the control group,the positive rates of the two bacteria samples were 97.9%(46/47)and 29.8%(14/47)before and 40.4%(19/47)and 10.6%(5/47)after the treatment.The positive rates between the two groups were not significantly different before taking antimicrobial measures(P=0.485 and 0.395),while they were significantly different after taking antimicrobial measures(P=0.001 and 0.024).Conclusion Combined application of eyelid and palpebral margin cleaning with cotton pads and antibiotic eye drops before cataract extraction imparted excellent antibacterial effects. | Xiaoyong Chen Hongyuan Cai Yinan Liu Jing Hong | 2022 | Chinese Medical Sciences Journal2022,37,2: | 2 |
| 16 | Nomogram for predicting lymph node metastasis rate of submucosal gastric cancer by analyzing clinicopathological characteristics associated with lymph node metastasis显示文摘Background:To combine clinicopathological characteristics associated with lymph node metastasis for submucosal gastric cancer into a nomogram.Methods:We retrospectively analyzed 262 patients with submucosal gastric cancer who underwent D2 gastrectomy between 1996 and 2012.The relationship between lymph node metastasis and clinicopathological features was statistically analyzed.With multivariate logistic regression analysis,we made a nomogram to predict the possibility of lymph node metastasis.Receiver operating characteristic(ROC) analysis was also performed to assess the predictive value of the model.Discrimination and calibration were performed using internal validation.Results:A total number of 48(18.3%) patients with submucosal gastric cancer have pathologically lymph node metastasis.For submucosal gastric carcinoma,lymph node metastasis was associated with age,tumor location,macroscopic type,size,differentiation,histology,the existence of ulcer and lymphovascular invasion in univariate analysis(all P<0.05).The multivariate logistic regression analysis identified that age ≤50 years old,macroscopic type III or mixed,undifferentiated type,and presence of lymphovascular invasion were independent risk factors of lymph node metastasis in submucosal gastric cancer(all P<0.05).We constructed a predicting nomogram with all these factors for lymph node metastasis in submucosal gastric cancer with good discrimination [area under the curve(AUC)=0.844].Internal validation demonstrated a good discrimination power that the actual probability corresponds closely with the predicted probability.Conclusions:We developed a nomogram to predict the rate of lymph node metastasis for submucosal gastric cancer.With good discrimination and internal validation,the nomogram improved individualized predictions for assisting clinicians to make appropriated treatment decision for submucosal gastric cancer patients. | Zhixue Zheng Yinan Zhang Lianhai Zhang Ziyu Li Aiwen Wu Xiaojiang Wu Yiqiang Liu Zhaode Bu Jiafu Ji | 2015 | Chinese Journal of Cancer Research2015,27,6: | 2 |

This review aims at contributing a comprehensive overview on the operation performance of the integrated process.
