| 1 | Activation of STAT3 stimulates AHSP expression in K562 cells显示文摘Studies on the chaperone proteinα-hemoglobin stabilizing protein(AHSP)reveal that abundant AHSP in erythroid cells enhance the cells’tolerance to oxidative stress imposed by excessα-hemoglobin in pathological conditions.However,the potential intracellular modulation of AHSP expression itself in response to oxidative stress is still unknown.The present study examined the effect and molecular mechanism of STAT3,an oxidative regulator,on the expression of AHSP.AHSP expression increased in K562 cells upon cytokine IL-6-induced STAT3 activation and decreased in STAT3 knock-down K562 cells.Regulation of AHSP in oxidative circumstance was then examined inα-globin-overloaded K562 cells,and real-time PCR showed strengthened expression of both AHSP and STAT3.ChIP analysis showed binding of STAT3 to AHSP promoter and binding was significantly augmented with IL6 stimulation and uponα-globin overexpression.Dual luciferase reporter assays of the wildtype and mutated SB3 element,an IL-6RE site,in the AHSP promoter in K562 cells highlighted the direct regulatory effect of STAT3 on AHSP gene.Finally,direct binding of STAT3 to SB3 site of AHSP promoter was confirmed with EMSA assays.Our work reveals an adaptive AHSP regulation mediated by the redox-sensitive STAT3 signaling pathway,and provides clues to the therapeutic strategy for AHSP enhancement. | CAO Cong ZHAO GuoWei YU Wei XIE XueMin WANG WenTian YANG RuiFeng LV Xiang LIU DePei | 2014 | Science China(Life Sciences)2014,57,5: | 5 |
| 2 | A viral protein orchestrates rice ethylene signaling to coordinate viral infection and insect vector-mediated transmission显示文摘Arthropod-borne viruses cause serious threats to human health and global agriculture by rapidly spreading via insect vectors. Southern rice black-streaked dwarf virus (SRBSDV) is the most damaging rice-infecting virus that is frequently transmitted by planthoppers. However, the molecular mechanisms underlying its propagation in the host plants and epidemics in the field are largely unknown. Here, we showed that the SRBSDV-encoded P6 protein is a key effector that regulates rice ethylene signaling to coordinate viral infection and transmission. In early SRBSDV infection, P6 interacts with OsRTH2 in the cytoplasm to activate ethylene signaling and enhance SRBSDV proliferation;this also repels the insect vector to reduce infestation. In late infection, P6 enters the nucleus, where it interacts with OsEIL2, a key transcription factor of ethylene signaling. The P6-OsEIL2 interaction suppresses ethylene signaling by preventing the dimerization of OsEIL2, thereby facilitating viral transmission by attracting the insect vector. | 3 | Efficacy of inverso isomer of CendR peptide on tumor tissue penetration显示文摘The dense extracellular matrix and high interstitial fluid pressure of tumor tissues prevent the ability of anti-tumor agents to penetrate deep into the tumor parenchyma for treatment effects. C-end rule(CendR) peptides can enhance the permeability of tumor blood vessels and tumor tissues via binding to neuropilin-1(NRP-1), thus aiding in drug delivery. In this study, we selected one of the CendR peptides(sequence RGERPPR) as the parent L-peptide and substituted D-amino acids for the L-amino acids to synthesize its inverso peptide D(RGERPPR). We investigated the NRP-1 binding activity and tumorpenetrating ability of D(RGERPPR). We found that the binding affinity of D(RGERPPR) with NRP-1 and the cellular uptake was significantly higher than that of RGERPPR. Evans Blue tests revealed that D(RGERPPR) exhibited improved tumor-penetrating ability in C6, U87 and A549 tumor-bearing nude mice. Using nude mice bearing A549 xenograft tumors as a model, we found that the rate of tumor growth in the group co-administered with D(RGERPPR) and gemcitabine(Gem) was significantly lower than the gemcitabine-treated group with a tumor suppression rate(TSR%) of 55.4%. | 4 | Study on thermal wave based on the thermal mass theory显示文摘The conservation equations for heat conduction are established based on the concept of thermal mass.We obtain a general heat conduction law which takes into account the spatial and temporal inertia of thermal mass.The general law introduces a damped thermal wave equation.It reduces to the well-known CV model when the spatial inertia of heat flux and temperature and the temporal inertia of temperature are neglected,which indicates that the CV model only considers the temporal inertia of heat flux.Numerical simulations on the propagation and superposition of thermal waves show that for small thermal perturbation the CV model agrees with the thermal wave equation based on the thermal mass theory.For larger thermal perturbation,however,the physically impossible phenomenon pre-dicted by CV model,i.e.the negative temperature induced by the thermal wave superposition,is eliminated by the general heat conduction law,which demonstrates that the present heat conduction law based on the thermal mass theory is more reasonable. | HU RuiFeng CAO BingYang | 2009 | Science China(Technological Sciences)2009,52,6: | 3 |

| 8 | Engineered platelets-based drug delivery platform for targeted thrombolysis显示文摘Thrombolytic agents have thus far yielded limited therapeutic benefits in the treatment of thrombotic disease due to their short half-life,low targeting ability,and association with serious adverse reactions,such as bleeding complications.Inspired by the natural roles of platelets during thrombus formation,we fabricated a platelet-based delivery system(NO@uPA/PLTs)comprising urokinase(uPA)and arginine(Arg)for targeted thrombolysis and inhibition of re-embolism.The anchoring of uPA to the platelet surface by lipid insertion increased the thrombotic targeting and in vivo circulation duration of uPA without disturbing platelet functions.Nitric oxide(NO)generated by the loaded Arg inhibited platelet aggregation and activation at the damaged blood vessel,thereby inhibiting re-embolism.NO@uPA/PLTs effectively accumulated at the thrombi in pulmonary embolism and carotid artery thrombosis model mice and exerted superior thrombolytic efficacy.In addition,the platelet delivery system showed excellent thrombus recurrence prevention ability in a mouse model of secondary carotid artery injury.The coagulation indicators in vivo showed that the platelet-based uPA and NO co-delivery system possessed a low hemorrhagic risk,providing a promising tool for rapid thrombolysis and efficient inhibition of posttreatment re-embolism. | Songli Wang Ruifeng Wang Nana Meng Linwei Lu Jun Wang Jianfen Zhou Jiasheng Lu Qianzhu Xu Cao Xie Changyou Zhan Yao Li Yang Yu Weiyue Lu Min Liu | 2022 | Acta Pharmaceutica Sinica B2022,12,4: | 1 |

| 14 | The prediction of exit choice using cumulative prospect value显示文摘Exit choice is essential for pedestrian safety and evacuation efficiency during the context of an emergency. Cumulative prospect theory is a widespread realistic decision-making theory that can transform choice outcomes and probabilities into subjective terms and integrate them as a parameter of cumulative prospect value (CPV), which determines the decision. The main contribution of this paper is the use of the CPV to predict exit choice. Furthermore, the different decision-making rules including Max, exponential(Expo), and Ratio (i.e., the three variable choice functions) were summarized and examined. This study used a confusion matrix to compare the predicted results with experimental data. | 15 | Study on single-group dynamics in evacuation considering pre-defined and undeclared leadership显示文摘Grouping is a common phenomenon that occurs everywhere.The leader-follower relationship inside groups has often been qualitatively characterized in previous models using simple heuristics.However,a general method is lacking to quantitatively explain leadership in an evacuating group.To understand the evolution of single-group dynamics throughout an evacuation,we developed an extended social force model integrated with a group force.A series of single-group evacuations from a room were simulated.An information-theoretic method,transfer entropy(TE),was applied to detect predefined and undeclared leadership among evacuees.The results showed that the predefined leader was correctly detected by TE,suggesting its capability in measuring leadership based on time series of evacuees’movement information(e.g.,velocity and acceleration).When evacuees were grouped together,TE was higher than when they were alone.Leaders presented a monotonically increasing cumulative influence curve over the investigated period,whereas followers showed a diminishing tendency.We found that leadership emergence correlated with evacuees’spatial positions.The individual located in the foremost part of the group was most likely to become a leader of those in the rear,which concurred with the experimental observations.We observed how a large group split into smaller ones with undeclared leadership during evacuation.These observations were quantitatively verified by TE results.This study provides novel insights into quantifying leadership and understanding single-group dynamics during evacuations. | Wei Xie Dongli Gao Ruifeng Cao Eric Wai Ming Lee Richard Kwok Kit Yuen Jingwen Weng | 2023 | Journal of Safety Science and Resilience2023,4,1: | 0 |
| 16 | Quality variations and controlling factors of deepwater submarine-fan reservoirs,Rovuma Basin,East Africa显示文摘The mechanisms causing quality variations and key control factors of submarine-fan reservoirs in the gas field X of the Rovuma Basin,East Africa are analyzed based on core and well-log data in this paper.Depositional fabric,lithofacies difference and characteristics of genetic units are the fundamental reasons of reservoir quality variations.In the case of weak cementation,porosity and permeability of submarine-fan reservoirs are controlled by grain sorting and clay content,respectively.Reservoir quality variations for 5 main lithofacies are related to variable depositional fabrics and calcite cementation.Among them,massive medium-coarse sandstones with weak cementation have the highest porosity and permeability,and coarser or finer sandstones have poorer reservoir quality.The existence of bottom current can develop laminated sandstones,improving the pore structure and physical properties greatly.Lithofacies vary among different types,locations and stages of genetic units,and they control the distribution patterns of submarine-fan reservoir quality:the physical properties of channel shaft or lobe main body are better than those of the channel or lobe edge.The sandstone sorting and physical properties are gradually improved from near-source to far-source.When multi-stage sand bodies are superimposed,the sand-mud ratio in the later stage is higher than that in the earlier stage,making the physical properties get better in the later stage. | WANG Min ZHANG Jiajia WANG Ruifeng XU Qingyan WEN Siying CAO Quanbin YU Jitao WANG Li | 2022 | Petroleum Exploration and Development2022,49,3: | 0 |
