1Inhibiting Hv1 channel in peripheral sensory neurons attenuates chronic inflammatory pain and opioid side effects显示文摘Both opioids and nonsteroidal anti-inflammatory drugs(NSAIDS)produce deleterious side effects and fail to provide sustained relief in patients with chronic inflammatory pain.Peripheral neuroinflammation(PN)is critical for initiation and development of inflammatory pain.A better understanding of molecular mechanisms underlying PN would facilitate the discovery of new analgesic targets and the development of new therapeutics.Emerging evidence suggests that peripheral sensory neurons are not only responders to painful stimuli,but are also actively engaged in inflammation and immunity,whereas the intrinsic regulatory mechanism is poorly understood.Here we report the expression of proton-selective ion channel Hv1 in peripheral sensory neurons in rodents and humans,which was previously shown as selectively expressed in microglia in mammalian central nervous system.Neuronal Hv1 was up-regulated by PN or depolarizing stimulation,which in turn aggravates inflammation and nociception.Inhibiting neuronal Hv1 genetically or by a newly discovered selective inhibitor YHV98-4 reduced intracellular alkalization and ROS production in inflammatory pain,mitigated the imbalance in downstream SHP-1-pAKT signaling,and also diminished pro-inflammatory chemokine release to alleviate nociception and morphine-induced hyperalgesia and tolerance.Thus,our data reveal neuronal Hv1 as a novel target in analgesia strategy and managing opioids-related side effects.Qiansen Zhang Yimin Ren Yiqing Mo Peipei Guo Ping Liao Yuncheng Luo Jie Mu Zhuo Chen Yang Zhang Ya Li Linghui Yang Daqing Liao Jie Fu Juwen Shen Wei Huang Xuewen Xu Yanyan Guo Lianghe Mei Yunxia Zuo Jin Liu Huaiyu Yang Ruotian Jiang 2022Cell Research2022,32,5:3
2The increasing expression of GPX7 related to the malignant clinical features leading to poor prognosis of glioma patients显示文摘Background:Glioma is the most common malignant brain tumor in adults.The standard treatment scheme of glioma is surgical resection combined alternative radio-and chemotherapy.However,the outcome of glioma patients was unsatisfied.Here,we aimed to explore the molecular and biological function characteristics of GPX7 in glioma.Methods:The multidimensional data of glioma samples were downloaded from Chinese Glioma Genome Atlas(CGGA).RT-qPCR method was used to identify the expression status of GPX7.Kaplan–Meier curves and Cox regression analysis were used to explore the prognostic value of GPX7.Gene Set Enrichment Analysis(GSEA)was applied to investigate the GPX7-related functions in glioma.Results:The results indicated that the expression of GPX7 in glioma was higher compared to that in normal brain tissue.Univariate and multivariate Cox regression analyses confirmed that the expression value of GPX7 was an independent prognostic factor in glioma.The GSEA analysis showed that GPX7 was significantly enriched in the cell cycle pathway,ECM pathway,focal adhesion pathway,and toll-like receptor pathway.Conclusions:The GPX7 was recommended as an independent risk factor for patients diagnosed with glioma for the first time and GPX7 could be potentially used as the therapy target in future.Furthermore,we attempted to explore a potential biomarker for improving the diagnosis and prognosis of patients with glioma.Jiawei Yao Xin Chen Zhendong Liu Ruotian Zhang Cheng Zhang Quan Yang Penglei Yao Qiuyi Jiang Jianing Wu Shiguang Zhao 2021Chinese Neurosurgical Journal2021,7,2:1
3Probing of coupling effect induced plasmonic charge accumulation for water oxidation显示文摘A key issue for redox reactions in plasmon-induced photocatalysis, particularly for water oxidation, is the concentration of surface-accumulating charges(electrons or holes) at a reaction site for artificial photosynthesis. However, where plasmonic charge accumulated at a catalyst’s surface, and how to improve local charge density at active sites, remains unknown because it is difficult to identify the exact spatial location and local density of the plasmon-induced charge, particularly with regard to holes. Herein, we show that at the single particle level, plasmon-coupling-induced holes can be greatly accumulated at the plasmonic Au nanoparticle dimer/TiO_(2) interface in the nanogap region, as directly evidenced by the locally enhanced surface photovoltage. Such an accumulation of plasmonic holes can significantly accelerate the water oxidation reaction(multi-holes involved) at the interfacial reaction site, with nearly one order of magnitude enhancement in photocatalytic activities compared to those of highly dispersed Au nanoparticles on TiO_(2). Combining Kelvin probe force microscopy and theoretical simulation, we further clarified that the local accumulated hole density is proportional to the square of the local near-field enhancement. 5Visualizing the role of applied voltage in non-metal electrocatalysts显示文摘Understanding how applied voltage drives the electrocatalytic reaction at the nanoscale is a fundamental scientific problem,particularly in non-metallic electrocatalysts,due to their low intrinsic carrier concentration.Herein,using monolayer molybdenum disulfide(MoS_(2))as a model system of non-metallic catalyst,the potential drops across the basal plane of MoS_(2)(△V_(sem))and the electric double layer(△V_(edl))are decoupled quantitatively as a function of applied voltage through in-situ surface potential microscopy.We visualize the evolution of the band structure under liquid conditions and clarify the process of EF keeping moving deep into Ec,revealing the formation process of the electrolyte gating effect.Additionally,electron transfer(ET)imaging reveals that the basal plane exhibits high ET activity,consistent with the results of surface potential measurements.The potential-dependent behavior of kf and ns in the ET reaction are further decoupled based on the measurements of△V_(sem)and△V_(edl).Comparing the ET and hydrogen evolution reaction imaging results suggests that the low electrocatalytic activity of the basal plane is mainly due to the absence of active sites,rather than its electron transfer ability.This study fills an experimental gap in exploring driving forces for electrocatalysis at the nanoscale and addresses the long-standing issue of the inability to decouple charge transfer from catalytic processes.Ziyuan Wang Jun Chen Chenwei Ni Wei Nie Dongfeng Li Na Ta Deyun Zhang Yimeng Sun Fusai Sun Qian Li Yuran Li Ruotian Chen Tiankai Bu Fengtao Fan Can Li 2023National Science Review2023,10,9:0
6Bladder microenvironment actuated proteomotors with ammonia amplification for enhanced cancer treatment显示文摘Enzyme-driven micro/nanomotors consuming in situ chemical fuels have attracted lots of attention for biomedical applications.However,motor systems composed by organism-derived organics that maximize the therapeutic efficacy of enzymatic products remain challenging.Herein,swimming proteomotors based on biocompatible urease and human serum albumin are constructed for enhanced antitumor therapy via active motion and ammonia amplification.By decomposing urea into carbon dioxide and ammonia,the designed proteomotors are endowed with self-propulsive capability,which leads to improved internalization and enhanced penetration in vitro.As a glutamine synthetase inhibitor,the loaded L-methionine sulfoximine further prevents the conversion of toxic ammonia into non-toxic glutamine in both tumor and stromal cells,resulting in local ammonia amplification.After intravesical instillation,the proteomotors achieve longer bladder retention and thus significantly inhibit the growth of orthotopic bladder tumor in vivo without adverse effects.We envision that the as-developed swimming proteomotors with amplification of the product toxicity may be a potential platform for active cancer treatment.Hao Tian Juanfeng Ou Yong Wang Jia Sun Junbin Gao Yicheng Ye Ruotian Zhang Bin Chen Fei Wang Weichang Huang Huaan Li Lu Liu Chuxiao Shao Zhili Xu Fei Peng Yingfeng Tu 2023Acta Pharmaceutica Sinica B2023,13,9:0
7Tailoring morphology symmetry of bismuth vanadate photocatalysts for efficient charge separation显示文摘Although spatial charge separation between different facets of semiconductor crystals has been recognized as a general strategy in photocatalysis, the vital role of crystal morphology symmetry in charge separation properties still remains elusive. Herein,taking monoclinic bismuth vanadate(BiVO_(4)) as a platform, we found distinct charge separation difference via rationally tailoring the morphology symmetry from octahedral to truncated octahedral crystals. For octahedral BiVO_(4), photogenerated electrons and holes can be separated between edges and quasi-equivalent facets. However, as for truncated octahedral crystals,photogenerated electrons tend to transfer to {010} facets while photogenerated holes prefer to accumulate on {120} facets, thus realizing the spatial separation of photogenerated charge between different facets. Morphology tailoring of BiVO_(4) crystals leads to a significantly improved photogenerated charge separation efficiency and photocatalytic water oxidation activity. The built-in electric field for driving the separation of photogenerated electrons and holes is considered to be modulated by tuning the morphology symmetry of BiVO_(4) crystals. 8Formation of multifaceted nano-groove structure on rutile TiO_(2) photoanode for efficient electron-hole separation and water splitting显示文摘Photoelectrochemical(PEC)water-splitting using solar energy holds great promise for the renewable energy future,and a key challenge in the development of industry viable PEC devices is the unavailability of high-efficient photoanodes.Herein,we designed a TiO_(2) model photocatalyst with nano-groove pattern and different surface orientation using low-energy Ar+irradiation and photoetching of TiO_(2),and significantly improved the intrinsic activity for PEC water oxidation.High-resolution transmission electron microscopy directly manifests that the grooves consist of highly stepped surface with<110>steps and well-crystallized.Transient absorption spectroscopy reveals the groove surface that allows for increased recovery lifetime,which ensures promoted electron-hole separation efficiency.Surface photovoltage directly shows the carrier separation and transportation behaviors,verified by selective photodeposition,demonstrating the groove surface on TiO_(2) contributes to electron-hole separation.This work proposes an efficient and scalable photoanode strategy,which potentially can open new opportunities for achieving efficient PEC water oxidation performance.Xiaoyi Zhan Yaling Luo Ziyu Wang Yao Xiang Zheng Peng Yong Han Hui Zhang Ruotian Chen Qin Zhou Hongru Peng Hao Huang Weimin Liu Xin Ou Guijun Ma Fengtao Fan Fan Yang Can Li Zhi Liu 2022Journal of Energy Chemistry2022,31,2:0
9Light-driven directional ion transport for enhanced osmotic energy harvesting显示文摘Light-driven ion(proton)transport is a crucial process both for photosynthesis of green plants and solar energy harvesting of some archaea.Here,we describe use of a TiO_(2)/C_(3)N_(4)semiconductor heterojunction nanotube membrane to realize similar light-driven directional ion transport performance to that of biological systems.This heterojunction system can be fabricated by two simple deposition steps.Under unilateral illumination,the TiO_(2)/C_(3)N_(4)heterojunction nanotube membrane can generate a photocurrent of about 9μA/cm^(2),corresponding to a pumping stream of~5500 ions per second per nanotube.By changing the position of TiO_(2)and C_(3)N_(4),a reverse equivalent ionic current can also be realized.Directional transport of photogenerated electrons and holes results in a transmembrane potential,which is the basis of the light-driven ion transport phenomenon.As a proof of concept,we also show that this system can be used for enhanced osmotic energy generation.The artificial light-driven ion transport system proposed here offers a further step forward on the roadmap for development of ionic photoelectric conversion and integration into other applications,for example water desalination.Kai Xiao Paolo Giusto Fengxiang Chen Ruotian Chen Tobias Heil Shaowen Cao Lu Chen Fengtao Fan Lei Jiang 2021National Science Review2021,8,8:0
10Tip-induced directional charge separation on one-dimensional BiVO4 nanocones for asymmetric light absorption显示文摘One dimensional(1D)semiconductor is a class of extensively attractive materials for many emerging solar energy conversion technologies.However,it is still of shortage to assess the impact of 1D structural symmetry on spatial charge separation and understand its underlying mechanism.Here we take controllably-synthesized 1D BiVO_(4)nanocones and nanorods as prototypes to study the influence of 1D symmetry on charge separation.It is found that the asymmetric BiVO_(4)nanocones enable more effective charge separation compared with the symmetric nanorods.The unexpected spatial charge separation on the nanocones is mainly ascribed to uneven light absorption induced diffusion-controllable charge separation due to symmetry breaking of 1D nanostructure,as evidenced by spatial and temporal resolved spectroscopy.Moreover,the promotion effect of charge separation on the nanocones was quantitatively evaluated to be over 20 times higher than that in BiVO_(4)nanorods.This work gives the first demonstration of the influence of 1D structural symmetry on the charge separation behavior,providing new insights to design and fabricate semiconductor materials for efficient solar energy conversion.Nengcong Yang Ruotian Chen Chenwei Ni Dongfeng Li Qi Sun Lifang Liu Yu Qi Shengye Jin Xiuli Wang Fengtao Fan Can Li Fuxiang Zhang 2022Journal of Energy Chemistry2022,31,9:0
