| 1 | Tubulin detyrosination promotes human trophoblast syncytium formation显示文摘Human trophoblast syncytialization is one of the most important yet least understood events during placental development.In this study,we found that detyrosinated a-tubulin(detyr-a-tub),which is negatively regulated by tubulin tyrosine Ugase(TTL),was elevated during human placental cytotrophoblast fusion.Correspondingly,relatively high expression of TTL protein was observed in first-trimester human placental cytotrophoblast cells,but fusing trophoblast cells exhibited much lower levels of T T L Notably,fusion of preeclamptic cytotrophoblast cells was compromised but could be partially rescued by knockdown of TTL levels.Mechanistically,chronic downregulation of TTL in trophoblast cells resulted in significantly elevated expression of detyra-tub.Restoration of detyr-a-tub thus contributed to the cell surface localization of the fusogenic protein Syncytin-2 and the gap junction protein Connexin 43(Cx43),which in turn promoted successful fusion between trophoblast cells.Taken together,the results suggest that tubulin detyrosination plays an essential role in human trophoblast fusogenic protein aggregation and syncytialization.Insufficient tubulin detyrosination leads to defects in syncytialization and potentially to the onset of preeclampsia. | Rui Wang Ruoxuan Yu Cheng Zhu Hai-Yan Lin Xiaoyin Lu Hongmei Wang | 2019 | Journal of Molecular Cell Biology2019,11,11: | 1 |
| 2 | LncRNA DACH1 protects against pulmonary fibrosis by binding to SRSF1 to suppress CTNNB1 accumulation显示文摘Idiopathic pulmonary fibrosis(IPF)is a progressive disease with unknown etiology and limited therapeutic options.Activation of fibroblasts is a prominent feature of pulmonary fibrosis.Here we report that lncRNA DACH1(dachshund homolog 1)is downregulated in the lungs of IPF patients and in an experimental mouse model of lung fibrosis.LncDACH1 knockout mice develop spontaneous pulmonary fibrosis,whereas overexpression of LncDACH1 attenuated TGF-β1-induced aberrant activation,collagen deposition and differentiation of mouse lung fibroblasts.Similarly,forced expression of LncDACH1 not only prevented bleomycin(BLM)-induced lung fibrosis,but also reversed established lung fibrosis in a BLM model.Mechanistically,LncDACH1 binding to the serine/arginine-rich splicing factor 1(SRSF1)protein decreases its activity and inhibits the accumulation of Ctnnb1.Enhanced expression of SRSF1 blocked the anti-fibrotic effect of LncDACH1 in lung fibroblasts.Furthermore,loss of LncDACH1 promoted proliferation,differentiation,and extracellular matrix(ECM)deposition in mouse lung fibroblasts,whereas such effects were abolished by silencing of Ctnnb1.In addition,a conserved fragment of LncDACH1 alleviated hyperproliferation,ECM deposition and differentiation of MRC-5 cells driven by TGF-β1.Collectively,LncDACH1 inhibits lung fibrosis by interacting with SRSF1 to suppress CTNNB1 accumulation,suggesting that LncDACH1 might be a potential therapeutic target for pulmonary fibrosis. | Jian Sun Tongzhu Jin Zhihui Niu Jiayu Guo Yingying Guo Ruoxuan Yang Qianqian Wang Huiying Gao Yuhan Zhang Tianyu Li Wenxin He Zhixin Li Wenchao Ma Wei Su Liangliang Li Xingxing Fan Hongli Shan Haihai Liang | 2022 | Acta Pharmaceutica Sinica B2022,12,9: | 1 |

| 7 | DLX5 promotes Col10a1 expression and chondrocyte hypertrophy and is involved in osteoarthritis progression显示文摘Osteoarthritis(OA)has been considered non-reversible as articular cartilage wears down with limited repair capacity.Enhanced chondrocyte hypertrophy and increased type X collagen gene(COL10A1)expression have been associated with OA.Therefore,regulators controlling collagen X expression and chondrocyte hypertrophy may play a role in OA intervention.Here,we investigated how Distal-less homeobox 5(DLX5),the distal-less homeobox family member,controls murine Col10a1 gene expression and chondrocyte hypertrophy in chondrogenic cell models and its role in a murine OA model.Through qRT-PCR and Western blot analyses,we detected significantly increased levels of COL10A1 and DLX5 in hypertrophic MCT and ATDC5 cells compared to their proliferative stage.Forced expression of Dlx5 further increases,while knockdown of Dlx5 decreases COL10A1 expression in hypertrophic MCT cells.We have performed dual-luciferase reporter and ChiP assays and demonstrated that DLX5 promotes reporter activity through direct interaction with Col10a1 cis-enhancer.We established a murine OA model and detected markedly increased COL10A1 and DLX5 in the articular cartilage and subchondral bone of the OA mice compared with the controls.Notably,forced overexpression of DLX5 in hypertrophic MCT cells up-regulates RUNX2,and adjacent DLX5 and RUNX2 binding sites have previously been found within the Col10a1 cis-enhancer.Together,our data suggest that DLX5 may cooperate with RUNX2 to control cell-specific Col10a1 expression and chondrocyte hypertrophy and is involved in OA pathogenesis. | Jinnan Chen Fangzhou Chen Xuan Wu Huiqin Bian Chen Chen Xiaojing Zhang Ruoxuan Hei Xiaotong Yang Haochun Yuan Qian Wang Yaojuan Lu Longwei Qiao Qiping Zheng | 2023 | Genes & Diseases2023,10,5: | 1 |

| 13 | Autumn nitrogen enrichment destabilizes ecosystem biomass production in a semiarid grassland显示文摘Nitrogen(N)deposition decreases the temporal stability of ecosystem aboveground biomass production(ecosystem stability).However,little is known about how the responses of ecosystem stability differ based on seasonal N enrichment.By adding N in autumn,winter,or growing season,from October 2014 to May 2020,in a temperate grassland in northern China,we found that only N addition in autumn resulted in a significantly positive correlation between ecosystem mean aboveground net primary productivity(ANPP)and its standard deviation and significantly reduced ecosystem stability.Autumn N-induced reduction in ecosystem stability was associated with the vanished negative effect of community-wide species asynchrony(asynchronous dynamics among populations to environmental perturbations)on the standard deviation of ecosystem ANPP in combination with the emerged positive effect of dominance(Simpson's dominance index that indicates the relative weight of dominant species in a community).Our findings indicate that autumn N addition might overestimate the negative effect of annual atmospheric N deposition on ecosystem stability,suggesting that to better evaluate the influence of N deposition in temperate grasslands,both field experiments and global modeling should consider not only the annual N load but also its seasonal dynamics.Moreover,further studies should pay more attention to the alteration in the ecosystem temporal deviations,which might be more sensitive to human-induced environmental changes. | Yuqiu Zhang Zhengru Ren Haining Lu Xu Chen Ruoxuan Liu Yunhai Zhang | 2023 | Fundamental Research2023,3,2: | 0 |

| 17 | Tailoring the multiscale mechanics of tunable decellularized extracellular matrix (dECM) for wound healing through immunomodulation显示文摘With the discovery of the pivotal role of macrophages in tissue regeneration through shaping the tissue immune microenvironment, various immunomodulatory strategies have been proposed to modify traditional biomaterials. Decellularized extracellular matrix (dECM) has been extensively used in the clinical treatment of tissue injury due to its favorable biocompatibility and similarity to the native tissue environment. However, most reported decellularization protocols may cause damage to the native structure of dECM, which undermines its inherent advantages and potential clinical applications. Here, we introduce a mechanically tunable dECM prepared by optimizing the freeze-thaw cycles. We demonstrated that the alteration in micromechanical properties of dECM resulting from the cyclic freeze-thaw process contributes to distinct macrophage-mediated host immune responses to the materials, which are recently recognized to play a pivotal role in determining the outcome of tissue regeneration. Our sequencing data further revealed that the immunomodulatory effect of dECM was induced via the mechnotrasduction pathways in macrophages. Next, we tested the dECM in a rat skin injury model and found an enhanced micromechanical property of dECM achieved with three freeze-thaw cycles significantly promoted the M2 polarization of macrophages, leading to superior wound healing. These findings suggest that the immunomodulatory property of dECM can be efficiently manipulated by tailoring its inherent micromechanical properties during the decellularization process. | 18 | Two‑Level Biomimetic Designs Enable Intelligent Stress Dispersion for Super‑Foldable C/NiS Nanofiber Free‑Standing Electrode显示文摘Due to the lack of in-depth understanding about the folding issues of the electronic materials,it is a huge challenge to pre-pare a super-foldable and highly electrochemical faradic electrode.Here,inspired from from the fully nimble structures of cuit cocoons and cockscomb petals,with two-level biomimetic design,for the first time we prepared a super-foldable and electrochemically functional freestanding cathode,made of C-fiber@NiS-cockscomb(SFCNi).In virtue of its nimble biomi-metic structures,SFCNi can remarkably sustain over 100,000 times,repeated true-folding without composite fibers fracture,functional matters detachment,conductivity degradation,or electrochemical performance change.The main mechanism behind these behaviors was disclosed by Real-time scanning electron microscopy and mechanical simulations,on the folding process.Results unveil that the cockscomb-like NiS with atomic thickness can deform freely due to the need of bending,and the cuit-cocoon-like SFCNi can generate an“ε-shape”folding structure at the crease.Such a smart self-adaptive deforma-tion capability can effectively reduce the effect of stresses and local excessive deformations,so that the chemical bonds can preserve their interaction,and the material won’t fracture.This subtle and exceptional mechanical behavior realizes a super-foldable property.The two-level biomimetic design strategy is a novel method for fabrication of super-foldable composite electrodes and integrated multi-functional super-foldable devices. | Guangtao Zan Tong Wu Wenya Dong Junchen Zhou Teng Tu Ruoxuan Xu Yun Chen Ying Wang Qingsheng Wu | 2022 | Advanced Fiber Materials2022,4,5: | 0 |
