| 2 | Regulation of the inflammatory cycle by a controllable release hydrogel for eliminating postoperative inflammation after discectomy显示文摘Surgery is the final choice for most patients with intervertebral disc degeneration(IDD).Operation-caused trauma will cause inflammation in the intervertebral disc.Serious inflammation will cause tissue defects and induce tissue degeneration,IDD recurrence and the occurrence of other diseases.Therefore,we proposed a scheme to treat recurrence after discectomy by inhibiting inflammation with an aspirin(ASP)-loaded hydrogel to restore the mechanical stability of the spine and relieve local inflammation.ASP-liposomes(ASP-Lips)were incorporated into a photocrosslinkable gelatin-methacryloyl(GelMA)via mixing.This material can effectively alleviate inflammation by inhibiting the release of high mobility group box 1(HMGB1)from the nucleus to the cytoplasm.We further assessed the expression of inflammatory cytokines,such as interleukin 6(IL-6)and tumor necrosis factor-α(TNF-α),and degeneration-related factors,such as type II collagen(COL-2),Aggrecan,matrix metallopeptidases-3(MMP-3),MMP-13,a disintegrin and metalloproteinase with thrombospondin motifs-4(ADAMTS-4)and ADAMTS-5 in rat nucleus pulpous cells.The level of IDD was analyzed through H&E,safranin-O staining and immunohistochemistry in rabbit samples.In vitro,we found that ASP-Lip@GelMA treatment significantly decreased inflammatory cytokines,MMP-3 and-13,and ADAMTS-4 and-5 and up-regulated COL-2 and Aggrecan via the inhibited release of HMGB-1 from the nucleus.In vivo,ASP-Lip@GelMA can effectively inhibit inflammation of local tissue after disc surgery and fill local tissue defects.This composite hydrogel system is a promising way to treat the recurrence of IDD after surgery without persistent complications. | Yu Liu Jiacheng Du Peng Peng Ruoyu Cheng Jiayi Lin Congxin Xu Huilin Yang Wenguo Cui Haiqing Mao Yuling Li Dechun Geng | 2021 | Bioactive Materials2021,6,1: | 6 |
| 3 | FluxExplorer: A general platform for modeling and analyses of metabolic net-works based on stoichiometry显示文摘Stoichiometry-based analyses of meta- bolic networks have aroused significant interest of systems biology researchers in recent years. It is necessary to develop a more convenient modeling platform on which users can reconstruct their network models using completely graphical operations, and explore them with powerful analyzing modules to get a better understanding of the properties of metabolic systems. Herein, an in silico platform, FluxExplorer, for metabolic modeling and analyses based on stoichiometry has been developed as a publicly available tool for systems biology research. This platform integrates various analytic approaches, in- cluding flux balance analysis, minimization of meta- bolic adjustment, extreme pathways analysis, shadow prices analysis, and singular value decom- position, providing a thorough characterization of the metabolic system. Using a graphic modeling process, metabolic networks can be reconstructed and modi- fied intuitively and conveniently. The inconsistencies of a model with respect to the FBA principles can be proved automatically. In addition, this platform sup- ports systems biology markup language (SBML). FluxExplorer has been applied to rebuild a metabolic network in mammalian mitochondria, producing meaningful results. | 6 | Comparison of schemes for preparing magnetic Fe_3O_4 nanoparticles显示文摘Magnetic Fe3O4 nanoparticles were prepared by means of coprecipitation using NH3·H2O in water and in alcohol, and using NaOH in water. A series of instruments such as SEM, TEM, HRTEM, FT-IR, XRD and VSM were used to characterize the properties of the magnetic nanoparticles. The results indicated that the magnetism of Fe3O4 nanoparticles synthesized using NH3·H2O in water was the highest, although the reaction time was the longest. The process using NaOH in water was the simplest and the reaction time was the shortest, but the particle characteristics were inferior to those of the other two methods. The mean size of magnetic Fe3O4 nanoparticles prepared by coprecipitation in alcohol was the smallest among the three, but the nanoparticles aggregated severely. The magnetic Fe3O4 nanoparticles were coated with oleic acid using saturated sodium oleate, and the polarity of the surface-modified nanoparticles was measured. | 8 | A CORRELATION EQUATION FOR CALCULATING INCLINED JET PENETRATION LENGTH IN A GAS-SOLID FLUIDIZED BED显示文摘Numerical simulation of gas-solid flow in a two-dimensional fluidized bed with an inclined jet was performed. The numerical model is based on the two-fluid model of gas and solids phase in which the solids constitutive equations are based on the kinetic theory of granular flow. The improved ICE algorithm, which can be used for both low and high-velocity fluid flow, were used to solve the model equations. The mechanism of jet formation was analyzed using both numerical simulations and experiments. The emergence and movement of gas bubbles were captured numerically and experimentally. The influences of jet velocity, nozzle diameter, nozzle inclination and jet position on jet penetration length were obtained. A semi-empirical expression was derived and the parameters were correlated from experimental data. | 9 | N, F?Codoped Microporous Carbon Nanofibers as Efficient Metal?Free Electrocatalysts for ORR显示文摘Currently, the oxygen reduction reaction(ORR) mainly depends on precious metal platinum(Pt) catalysts. However, Pt-based catalysts have several shortcomings, such as high cost, scarcity, and poor long-term stability. Therefore, development of e cient metal-free electrocatalysts to replace Pt-based electrocatalysts is important. In this study, we successfully prepared nitrogen-and fluorinecodoped microporous carbon nanofibers(N, F-MCFs) via electrospinning polyacrylonitrile/polyvinylidene fluoride/polyvinylpyrrolidone(PAN/PVDF/PVP) tricomponent polymers followed by a hydrothermal process and thermal treatment, which was achieved for the first time in the literature. The results indicated that N, F-MCFs exhibit a high catalytic activity(E_(onset): 0.94 V vs. RHE, E_(1/2): 0.81 V vs. RHE, and electron transfer number: 4.0) and considerably better stability and methanol tolerance for ORR in alkaline solutions as compared to commercial Pt/carbon(Pt/C, 20 wt%) catalysts. Furthermore, in acidic media, N, F-MCFs showed a four-electron transfer pathway for ORR. | 10 | Application of Bronsted acid ionic liquids as green catalyst in the synthesis of 2-propanol with reactive distillation显示文摘Five Br?nsted acidic ionic liquids(ILs) were prepared and characterized by FT-IR,~1H NMR and ^(13)C NMR. Their catalytic activities for the synthesis of 2-propanol(IPOH) via transesterification of isopropyl acetate(IPAc) with methanol(Me OH) were investigated. Among all the tested ILs, [Ps-mim]HSO_4 performed best and was used as catalyst for further studies. The reaction kinetics were carried out to correlate the parameters in a homogeneous second order kinetic model. It has been found that there is close agreement between the calculated and experimental values. The high-pressure batch reactive distillation experimental apparatus was set up in order to enhance the conversion of IPAc. A high conversion of IPAc of 99.4% was obtained under the optimal reaction conditions. The catalyst [Ps-mim]HSO_4 can be recycled easily by a rotary evaporator and reused without any further treatment. | 12 | Synergistic osteogenic and angiogenic effects of KP and QK peptides incorporated with an injectable and self-healing hydrogel for efficient bone regeneration显示文摘Irregular defects generated by trauma or surgery in orthopaedics practice were usually difficult to be fitted by the preformed traditional bone graft substitute. Therefore, the injectable hydrogels have attracted an increasing interest for bone repair because of their fittability and mini-invasivity. However, the uncontrollable spreading or mechanical failures during its manipulation remain a problem to be solved. Moreover, in order to achieve vascularized bone regeneration, alternatives of osteogenic and angiogenic growth factors should be adopted to avoid the problem of immunogenicity and high cost. In this study, a novel injectable self-healing hydrogel system (GMO hydrogel) loaded with KP and QK peptides had been developed for enhancing vascularized regeneration of small irregular bone defect. The dynamic imine bonds between gelatin methacryloyl and oxidized dextran provided the GMO hydrogel with self-healing and shear-thinning abilities, which led to an excellent injectability and fittability. By photopolymerization of the enclosed GelMA, GMO hydrogel was further strengthened and thus more suitable for bone regeneration. Besides, the osteogenic peptide KP and angiogenic peptide QK were tethered to GMO hydrogel by Schiff base reaction, leading to desired releasing profiles. In vitro, this composite hydrogel could significantly improve the osteogenic differentiation of BMSCs and angiogenesis ability of HUVECs. In vivo, KP and QK in the GMO hydrogel demonstrated a significant synergistic effect in promoting new bone formation in rat calvaria. | 15 | Prevalent fecal contamination in drinking water resources and potential health risks in Swat, Pakistan显示文摘Fecal bacteria contaminate water resources and result in associated waterborne diseases.This study assessed drinking water quality and evaluated their potential health risks in Swat, Pakistan. Ground and surface drinking water were randomly collected from upstream to downstream in the River Swat watershed and analyzed for fecal contamination using fecal indicator bacteria(Escherichia coli) and physiochemical parameters(potential of hydrogen, turbidity, temperature, electrical conductivity, total dissolved solid, color, odor and taste). The physiochemical parameters were within their safe limits except in a few locations, whereas, the fecal contaminations in drinking water resources exceeded the drinking water quality standards of Pakistan Environmental Protection Agency(Pak-EPA),2008 and World Health Organization(WHO), 2011. Multivariate and univariate analyses revealed that downstream urbanization trend, minimum distance between water sources and pit latrines/sewerage systems, raw sewage deep well injection and amplified urban,pastures and agricultural runoffs having human and animal excreta were the possible sources of contamination. | 18 | An overview of the switching parameter variation of RRAM显示文摘Resistive random access memory(RRAM) has been considered as one of the most promising candidates for next-generation nonvolatile memory, due to its advantages of simple device structure, excellent scalability, fast operation speed and low power consumption. Deeply understanding the physical mechanism and effectively controlling the statistical variation of switching parameters are the basis of fostering RRAM into commercial application. In this paper, based on the deep understanding on the mechanism of the formation and rupture of conductive filament, we summarize the methods of analyzing and modeling the statistics of switching parameters such as SET/RESET voltage, current, speed or time. Then, we analyze the distributions of switching parameters and the influencing factors. Additionally, we also sum up the analytical model of resistive switching statistics composed of the cell-based percolation model and SET/RESET switching dynamics. The results of the model can successfully explain the experimental distributions of switching parameters of the Ni O- and Hf O2-based RRAM devices. The model also provides theoretical guide on how to improve the uniformity and reliability such as disturb immunity.
