1Cryptococcus neoformans,a global threat to human health显示文摘Background Emerging fungal pathogens pose important threats to global public health. The World Health Organization has responded to the rising threat of traditionally neglected fungal infections by developing a Fungal Priority Pathogens List (FPPL). Taking the highest-ranked fungal pathogen in the FPPL,Cryptococcus neoformans, as a paradigm, we review progress made over the past two decades on its global burden, its clinical manifestation and management of cryptococcal infection, and its antifungal resistance. The purpose of this review is to drive research efforts to improve future diagnoses, therapies, and interventions associated with fungal infections.Methods We first reviewed trends in the global burden of HIV-associated cryptococcal infection, mainly based on a series of systematic studies. We next conducted scoping reviews in accordance with the guidelines described in the Preferred Reporting Items for Systematic Reviews and Meta-analyses extension for Scoping Reviews using PubMed and ScienceDirect with the keywordCryptococcus neoformans to identify case reports of cryptococcal infections published since 2000. We then reviewed recent updates on the diagnosis and antifungal treatment of cryptococcal infections. Finally, we summarized knowledge regarding the resistance and tolerance ofC. neoformans to approved antifungal drugs.Results There has been a general reduction in the estimated global burden of HIV-associated cryptococcal meningitis since 2009, probably due to improvements in highly active antiretroviral therapies. However, cryptococcal meningitis still accounts for 19% of AIDS-related deaths annually. The incidences of CM in Europe and North America and the Latin America region have increased by approximately two-fold since 2009, while other regions showed either reduced or stable numbers of cases. Unfortunately, diagnostic and treatment options for cryptococcal infections are limited, and emerging antifungal resistance exacerbates the public health burden.Conclusions The rising threat ofC. neoformans is compounded by accumulating evidence for its ability to infect immunocompetent individuals and the emergence of antifungal-resistant variants. Emphasis should be placed on further understanding the mechanisms of pathogenicity and of antifungal resistance and tolerance. 2The effect of iron on the sound velocities ofδ-AlOOH up to 135 GPa显示文摘δ-(Al,Fe)OOH is considered to be one of the most important hydrous phases on Earth,remaining stable under the extreme conditions throughout the mantle.The behavior ofδ-(Al,Fe)OOH at high pressure is essential to understanding the deep water cycle.δ-(Al_(0.956)Fe_(0.044))OOH crystals synthesized at 21 GPa and 1473 K were investigated by high-pressure Brillouin light scattering spectroscopy and synchrotron X-ray diffraction up to 135.4 GPa in diamond anvil cells.The incorporation of 5 mol%FeOOH increases the unit-cell volume ofδ-AlOOH by~1%and decreases the shear-wave velocity(VS)by~5%at 20–135 GPa.In particular,the compressional(V_(P))and shear(VS)wave velocities ofδ-(Al_(0.956)Fe_(0.044))OOH are 7%–16%and 10%–24%greater than all the major minerals in the mantle transition zone including wadsleyite,ringwoodite,and majorite.The distinctly high sound velocities ofδ-(Al_(0.956)Fe_(0.044))OOH at 20–25 GPa may contribute to the seismic anomalies observed at~560–680 km depths in the cold and stagnant slab beneath Izu-Bonin and/or Korea.Furthermore,the VS ofδ-(Al_(0.956)Fe_(0.044))OOH is about 10%and 4%–12%lower than iron-bearing bridgmanite Mg_(0.96)Fe_(0.05)Si_(0.99O3)and ferropericlase(Mg_(0.92)Fe_(0.08))O,respectively,under the lowermost mantle conditions,which might partially contribute to the large low-shear-velocity provinces and ultralow velocity zones at the bottom of the lower mantle.Xiaowan Su Chaoshuai Zhao Chaojia Lv Yukai Zhuang Nilesh Salke Liangxu Xu Hu Tang Huiyang Gou Xiaohui Yu Qiang Sun Jin Liu 2021Geoscience Frontiers2021,12,2:1

5Photoresponse and carrier transport of protocrystalline silicon multilayer films显示文摘Alternating multilayer films of hydrogen diluted hydrogenated protocrystalline silicon (pc-Si:H) were prepared using a plasma-enhanced chemical vapor deposition technique.The microstructure of the deposited films and photoresponse characteristics of their Schottky diode structures were investigated by Raman scattering spectroscopy,Fourier transform infrared spectroscopy and photocurrent spectra.Microstructure and optical absorption analyses suggest that the prepared films were pc-Si:H multilayer films with a two-phase structure of silicon nanocrystals (NCs) and its amorphous counterpart and the band gap of the films showed a decreasing trend with increasing crystalline fraction.Photocurrent measurement revealed that silicon NCs facilitate the spatial separation of photo-generated carriers,effectively reduce the non-radiative recombination rate,and induce a photoresponse peak value shift towards the short-wavelength side with increasing crystallinity.However,the carrier traps near the surface defects of silicon NCs and their spatial carrier confinement result in a significant reduction of the diode photoresponse in the longwavelength region.An enhancement of the photoresponse from 350 to 1000 nm was observed when applying an increased bias voltage in the diode,showing a favorable carrier transport and an effective collection of photo-generated carriers was achieved.Both the spatial separation of the restricted electron-hole pairs in silicon NCs and the de-trapping of the carriers at their interface defects are responsible for the red-shift in photoresponse spectra and enhancement of external quantum efficiency.The results provide fundamental data for the carrier transport control of high-efficiency pc-Si:H solar cells.YU Wei ZHU HaiRong ZHAO Yi SUN YuKai LU HaiJiang FU GuangSheng 2012Chinese Science Bulletin2012,57,20:0
6One-step cell biomanufacturing platform:porous gelatin microcarrier beads promote human embryonic stem cell-derived midbrain dopaminergic progenitor cell differentiation in vitro and survival after transplantation in vivo显示文摘Numerous studies have shown that cell replacement therapy can replenish lost cells and rebuild neural circuitry in animal models of Parkinson’s disease.Transplantation of midbrain dopaminergic progenitor cells is a promising treatment for Parkinson’s disease.However,transplanted cells can be injured by mechanical damage during handling and by changes in the transplantation niche.Here,we developed a one-step biomanufacturing platform that uses small-aperture gelatin microcarriers to produce beads carrying midbrain dopaminergic progenitor cells.These beads allow midbrain dopaminergic progenitor cell differentiation and cryopreservation without digestion,effectively maintaining axonal integrity in vitro.Importantly,midbrain dopaminergic progenitor cell bead grafts showed increased survival and only mild immunoreactivity in vivo compared with suspended midbrain dopaminergic progenitor cell grafts.Overall,our findings show that these midbrain dopaminergic progenitor cell beads enhance the effectiveness of neuronal cell transplantation.Lin Feng Da Li Yao Tian Chengshun Zhao Yun Sun Xiaolong Kou Jun Wu Liu Wang Qi Gu Wei Li Jie Hao Baoyang Hu Yukai Wang 2024Neural Regeneration Research2024,19,2:0
