Effects of ferrous and manganese ions on anammox process in sequencing batch biofilm reactors显示文摘Ferrous and manganese ions, as essential elements, significantly affect the synthesis of Haem-C, which participates in the energy metabolism and proliferation of anammox bacteria. In this study, two identical sequencing batch biofilm reactors were used to investigate the effects of ferrous and manganese ions on nitrogen removal efficiency and the potential of metal ions serving as electron donor/acceptors in the anammox process. Fluorescence in situ hybridization analysis was applied to investigate the microbial growth. Results showed that the nitrogen removal increased at high concentrations of Fe2+and Mn2+and the maximum removal efficiency was nearly 95% at Fe2+ 0.08 mmol/L and Mn2+0.05 mmol/L, which is nearly 15% and 8% higher than at the lowest Fe2+ and Mn2+concentrations(0.04 and 0.0125 mmol/L). The stabilities of the anammox reactor and the anammox bacterial growth were also enhanced with the elevated Fe2+and Mn2+concentrations. The Fe2+and Mn2+were consumed by anammox bacteria along with the removal of ammonia and nitrite. Stoichiometry analysis showed Fe2+could serve as an electron donor for NO-3-N in the anammox process. Nitrate could be reduced with Fe2+serving as the electron donor in the anammox system, which causes the value of NO-2-N/NH+ 4-N to decrease with the increasing of N-removal efficiency. | Xiaoli Huang Dawen Gao Sha Peng Yu Tao | 2014 | Journal of Environmental Sciences2014,26,5: | 13
Risk factors and outcomes of acute kidney injury after intracoronary stent implantation显示文摘BACKGROUND:Acute kidney injury following percutaneous coronary intervention(PCI) is associated with a worse outcome. However, the risk factors and outcomes of acute kidney injury(AKI) in patients after intracoronary stent implantation are still unknown.METHODS:A retrospective case control study was done in 325 patients who underwent intracoronary stent implantation from January 2010 to March 2011 at the Drum Tower Hospital of Nanjing University School of Medicine. Those were excluded from the study if they had incomplete clinical data. The patients were divided into a normal group and a AKI group according to the standard of post-operation day 7 to identify AKI.The parameters of the patients included:1) pre-operative ones:age, gender, hypertension, diabetes mellitus, cerebrovascular disease, left ventricular insufficiency, peripheral angiopathy, creatinine, urea nitrogen, estimated glomerular filtration rate(eGFR),hyperuricemia, proteinuria, emergency operation,hydration,medications(ACEI/ARBs,statins);2) intraoperative ones:dose of contrast media, operative time, hypotension;and 3) postoperative one:hypotension. The parameters were analyzed with univariate analysis and multivariate logistical regression analysis.RESULTS:Of the 325 patients,51(15.7%) developed AKI.Hospital day and in-hospital mortality were increased significantly in the AKI-group.Univariate analysis showed that age,pre-operative parameters(left ventricular insufficiency, peripheral angiopathy, creatinine, urea nitrogen, estimated glomerular filtration rate, hyperuricemia, proteinuria, hydration),emergency operation, intraoperative parameters(operative time, hypotension) and postoperative hypotension were significantly different. However, multivariate logistic regression analysis revealed that increased age(OR=0.253,95%CI=0.088-0.727),pre-operative proteinuria(OR=5.351,95%CI=2.128-13.459),pre-operative left ventricular insufficiency(OR=8.704,95%CI=3.170-23.898),eGFR<60 ml/min/1.73 m^2(OR=6.677,95%CI = 1.167-38.193),prolonged operative time, intraoperative hypotension(OR=25.245,95%CI=1.001-1.034) were independent risk factors of AKI.CONCLUSIONS:AKI is a common complication and associated with ominous outcome following intracoronary stent implantation.Increased age,pre-operative proteinuria,pre-operative left ventricular insufficiency,pre-operative low estimated glomerular filtration rate, prolonged operative time, intraoperative hypotension were the significant risk factors of AKI. | Fei He Jun Zhang Zhong-qiu Lu Qing-ling Gao Du-juan Sha Li-gang Pei Guo-feng Fan | 2012 | World Journal of Emergency Medicine2012,3,3: | 11
A new prognostic histopathologic classification of nasopharyngeal carcinoma显示文摘Background:The current World Health Organization(WHO) classification of nasopharyngeal carcinoma(NPC) con?veys little prognostic information. This study aimed to propose an NPC histopathologic classification that can poten?tially be used to predict prognosis and treatment response. Methods:We initially developed a histopathologic classification based on the morphologic traits and cell differentia?tion of tumors of 2716 NPC patients who were identified at Sun Yat?sen University Cancer Center(SYSUCC)(training cohort). Then, the proposed classification was applied to 1702 patients(retrospective validation cohort) from hospitals outside SYSUCC and 1613 patients(prospective validation cohort) from SYSUCC.The efficacy of radiochemotherapy and radiotherapy modalities was compared between the proposed subtypes. We used Cox proportional hazards models to estimate hazard ratios(HRs) with 95% confidence intervals(CI) for overall survival(OS).Results:The 5?year OS rates for all NPC patients who were diagnosed with epithelial carcinoma(EC;3708 patients),mixed sarcomatoid?epithelial carcinoma(MSEC;1247 patients),sarcomatoid carcinoma(SC;823 patients),and squamous cell carcinoma(SCC;253 patients) were 79.4%,70.5%,59.6%,and 42.6%,respectively(P < 0.001).In mul?tivariate models, patients with MSEC had a shorter OS than patients with EC(HR = 1.44,95% CI = 1.27–1.62),SC(HR = 2.00,95% CI = 1.76–2.28),or SCC(HR = 4.23,95% CI = 3.34–5.38).Radiochemotherapy significantly improved survival compared with radiotherapy alone for patients with EC(HR 49–0.75),and possibly for those with SCC(HR = 0.67,95% CI = 0.56–0.80),MSEC(HR = 0.58,95% CI = 0..74–1.28).= 0.63;95% CI = 0.40–0.98),but not for patients with SC(HR = 0.97,95% CI = 0Conclusions:The proposed classification offers more information for the prediction of NPC prognosis compared with the WHO classification and might be a valuable tool to guide treatment decisions for subtypes that are associ?ated with a poor prognosis. | Hai-Yun Wang Yih-Leong Chang Ka-Fai To Jacqueline S.G.Hwang Hai-Qiang Mai Yan-Fen Feng Ellen T.Chang Chen-Ping Wang Michael Koon Ming Kam Shie-Lee Cheah Ming Lee Li Gao Hui-Zhong Zhang Jie-Hua He Hao Jiang Pei-Qing Ma Xiao-Dong Zhu Liang Zeng Chun-Yan Chen Gang Chen Ma-Yan Huang Sha Fu Qiong Shao An-Jia Han Hai-Gang Li Chun-Kui Shao Pei-Yu Huang Chao-Nan Qian Tai-Xiang Lu Jin-Tian Li Weimin Ye Ingemar Ernberg Ho Keung Ng Joseph T.S.Wee Yi-Xin Zeng Hans-Olov Adami Anthony T.C.Chan Jian-Yong Shao | 2016 | Chinese Journal of Cancer2016,35,6: | 10
Effects of Primary Annealing Condition on Recrystallization Texture in a Grain Oriented Silicon Steel显示文摘The recrystallization texture in grain oriented silicon steel sheets, which were annealed at different primary annealing temperatures with and without an electric field, was investigated. An automated electron backscattered diffraction (EBSD) technique was used to analyze the recrystallization texture. It was found that recovery and application of electric field in primary annealing lead to an increase of {001} component and a decrease of {111} component after annealing at 900℃. The development of recrystallization texture can be explained in terms of the effects of electric field and primary annealing temperature on recovery. | Yubui SHA Fang ZHANG Song LI Xaoyu GAO Jiazhen XU Liang ZUO School of Materials and Metallurgy,Northeastern University,Shenyang 110004,China | 2004 | Journal of Materials Science & Technology2004,20,3: | 9
Effect of Tangweian Jianji on upper gastrointestinal remodeling in streptozotocin-induced diabetic rats显示文摘AIM:To investigate the effect of Tangweian Jianji(TWAJJ) on the biomechanical and morphometrical remodeling of the upper gastrointestinal tract in diabetic rats.METHODS:Diabetes was induced in 27 rats by injecting streptozotocin(40 mg/kg body weight),the animals were then divided into three groups(n = 9 in each group),i.e.,diabetic control(DM);high dose(10 g/kg,T1) and low dose(5 g/kg,T2).Another 10 rats acted as normal controls(Control).TWAJJ was administered by gavage once daily.Blood glucose and serum insulin levels were measured.Circumferential length,wall thickness and opening angle were measured from esophageal,duodenal,jejunal and ileal ring segments.The residual strain was calculated from the morphometric data.Step-wise distension was carried out on esophageal and jejunal segments.The obtained data on the length,diameter and pressure changes were then used to calculate the circumferential and longitudinal stresses and strains.Real-time reverse transcription polymerase chain reaction was used to detect the receptor of advanced glycation end-products(RAGE) mRNA level in jejunal tissues.RESULTS:At the end of the experiment,the blood glucose level was significantly higher and the serum insulin level was significantly lower in DM,T1 and T2 groups than in the control group(Glucose:30.23 ± 0.41 mmol/L,27.48 ± 0.27 mmol/L and 27.84 ± 0.29 mmol/L vs 5.05 ± 0.04 mmol/L,P = 1.65 × 10-16,P = 5.89 × 10-19 and P = 1.63 × 10-18,respectively;Insulin:1.47 ± 0.32 g/L,2.66 ± 0.44 g/L,2.03 ± 0.29 g/L and 4.17 ± 0.54 g/L,P = 0.0001,P = 0.029 and P = 0.025,respectively).However,these levels did not differ among the DM,T1 and T2 groups.The wet weight per unit length,wall thickness and opening angle of esophageal and intestinal segments in the DM group were significantly higher than those in the control group(from P = 0.009 to P = 0.004).These parameters in the T1 group were significantly lower than those in the DM group(wet weight,duodenum:0.147 ± 0.003 g/cm vs 0.158 ± 0.001 g/cm,P = 0.047;jejunum,0.127 ± 0.003 g/cm vs 0.151 ± 0.002 g/cm,P = 0.017;ileum,0.127 ± 0.004 g/cm vs 0.139 ± 0.003 g/cm,P = 0.046;wall thickness,esophagus:0.84 ± 0.03 mm vs 0.94 ± 0.02 mm,P = 0.014;duodenum:1.27 ± 0.06 mm vs 1.39 ± 0.05 mm,P = 0.031;jejunum:1.19 ± 0.07mm vs 1.34 ± 0.04 mm,P = 0.047;ileum:1.09 ± 0.04 mm vs 1.15 ± 0.03 mm,P = 0.049;opening angle,esophagus:112.2 ± 13.2 vs 134.7 ± 14.7,P = 0.027;duodenum:105.9 ± 12.3 vs 123.1 ± 13.1,P = 0.046;jejunum:90.1 ± 15.4 vs 115.5 ± 13.3,P = 0.044;ileum:112.9 ± 13.4 vs 136.1 ± 17.1,P = 0.035).In the esophageal and jejunal segments,the inner residual stain was significantly smaller and the outer residual strain was larger in the DM group than in the control group(P = 0.022 and P = 0.035).T1 treatment significantly restored this biomechanical alteration(P = 0.011 and P = 0.019),but T2 treatment did not.Furthermore,the circumferential and longitudinal stiffness of the esophageal and jejunal wall increased in the DM group compared with those in the control group.T1,but not T2 treatment,significantly decreased the circumferential wall stiffness in the jejunal segment(P = 0.012) and longitudinal wall stiffness in the esophageal segment(P = 0.023).The mRNA level of RAGE was significantly decreased in the T1 group compared to that in the DM group(P = 0.0069).CONCLUSION:TWAJJ(high dose) treatment partly restored the morphometric and biomechanical remodeling of the upper gastrointestinal tract in diabetic rats. | Gui-Fang Liu Jing-Bo Zhao Zhong Zhen Hong Sha Peng-Min Chen Min Li Jia-Cheng Zhang Ming-Ze Yuan Wen Gao Hans Gregersen Xiao-Lin Tong | 2012 | World Journal of Gastroenterology2012,18,35: | 8

Urodynamic assessment of bladder storage function after radical hysterectomy for cervical cancer显示文摘Background:After radical hysterectomy for cervical cancer, the most common complication is lower urinary tract symptoms.Post-operatively,bladder capacity can alter bladder function for a prolonged period. This study aimed to identify factors affecting bladder storage function. Methods:A multicenter, retrospective cohort study was conducted.Information of patients with stages IA2 to IIB cervical cancer with urodynamic study results were retrospectively collected from nine hospitals between June 2013 and June 2018 according to the inclusion criteria.Demographic, surgical, and oncological data were collected. The univariate and multivariate logistic regression was used to identify clinical factors associated with bladder storage function. Results:Two hundred and three patients with cervical cancer had urodynamic testing post-operatively.Ninety-five(46.8%)patients were diagnosed with stress urinary incontinence(SUI).The incidence of low bladder compliance(LBC)was 23.2%.Twenty-seven(13.3%)patients showed detrusor overactivity(DO).Fifty-seven patients(28.1%)presented with a decreased maximum cystometric capacity(DMCC).The probability of composite bladder storage dysfunction was 68.0%.Multivariate analysis confirmed that laparoscopy represents a protective factor for SUI with an odds ratio of 0.498(P=0.034).Patients who underwent a nerve-sparing procedure were less odds to experience SUI(P=0.014).A significant positive correlation between LBC and DO was observed(P<0.001).A greater length of the resected vagina and chemoradiotherapy were common risk factors for LBC and DO,while radiotherapy exerted a stronger effect than chemotherapy.Additionally, patients who received chemoradiotherapy frequently developed a DMCC.The follow-up time was not correlated with bladder storage function. Conclusion:A nerve-sparing procedure without longer resected vagina is recommended for protecting the bladder storage function. | Ting-Ting Cao Hong-Wu Wen Yu-Nong Gao Qiu-Bo Lyu Hui-Xin Liu Sha Wang Shi-Yan Wang Hua-Xin Sun Na Yu Hai-Bo Wang Yi Li Zhi-Qi Wang Olivia H.Chang Xiu-Li Sun Jian-Liu Wang | 2020 | Chinese Medical Journal2020,,19: | 7
CXCL10/CXCR3 Signaling in the DRG Exacerbates Neuropathic Pain in Mice显示文摘Chemokines and receptors have been implicated in the pathogenesis of chronic pain. Here,we report that spinal nerve ligation(SNL)increased CXCR3 expression in dorsal root ganglion(DRG)neurons, and intra-DRG injection of Cxcr3 shRNA attenuated the SNL-induced mechanical allodynia and heat hyperalgesia.SNL also increased the m RNA levels of CXCL9,CXCL10,and CXCL11,whereas only CXCL10 increased the number of action potentials(APs)in DRG neurons.Furthermore,in Cxcr3^(-/-)mice,CXCL10 did not increase the number of APs,and the SNL-induced increase of the numbers of APs in DRG neurons was reduced.Finally,CXCL10 induced the activation of p38 and ERK in ND7-23 neuronal cells and DRG neurons.Pretreatment of DRG neurons with the P38 inhibitor SB203580 decreased the number of APs induced by CXCL10.Our data indicate that CXCR3,activated by CXCL10,mediates p38 and ERK activation in DRG neurons and enhances neuronal excitability, which contributes to the maintenance of neuropathic pain. | Yan-Fang Kong Wei-Lin Sha Xiao-Bo Wu Lin-Xia Zhao Ling-Jie Ma Yong-Jing Gao | 2021 | Neuroscience Bulletin2021,37,3: | 6
Lipoprotein(a) as a Marker for Predicting the Presence and Severity of Coronary Artery Disease in Untreated Chinese Patients Undergoing Coronary Angiography显示文摘Objective Low-density lipoprotein cholesterol(LDL-C) has been well known as the risk factor of coronary artery disease(CAD). However, the role of lipoprotein(a) [Lp(a)] in the development of CAD is of great interest but still controversial. Thus, we aim to explore the effect of Lp(a) on predicting the presence and severity of CAD in Chinese untreated patients, especially in combination with LDL-C. Methods We consecutively recruited 1,980 non-treated patients undergoing coronary angiography, among which 1,162 patients were diagnosed with CAD. Gensini score(GS) was used to assess the severity of CAD. Lp(a) was measured by immunoturbidimetric method. Results Patients with CAD had higher level of LDL-C and Lp(a) compared with non-CAD(P < 0.05). Multivariable logistic regression revealed that Lp(a) > 205 mg/L(highest tertile) predicted 1.437-fold risk for CAD(95% CI: 1.108-1.865, P = 0.006) and 1.480-fold risk for high GS(95% CI: 1.090-2.009, P = 0.012) respectively. Interestingly, concomitant elevated level of Lp(a) and LDL-C conferred the highest risk for both presence [OR = 1.845, 95% CI: 1.339-2.541, P < 0.001] and severity [OR = 1.736, 95% CI: 1.188-2.538, P = 0.004] of CAD. Conclusion Lipoprotein(a) is a useful marker for predicting the presence and severity of CAD, especially combined with LDL-C. | SUN Di ZHAO Xi LI Sha ZHANG Yan WU Na Qiong ZHU Cheng Gang GUO Yuan Lin GAO Ying QING Ping LIU Geng DONG Qian LI Jian Jun | 2018 | Biomedical and Environmental Sciences2018,31,4: | 6
Biofilm-inspired adhesive and antibacterial hydrogel with tough tissue integration performance for sealing hemostasis and wound healing显示文摘Uncontrolled bleeding and infection can cause significant increases in mortalities.Hydrogel sealants have attracted extensive attention for their ability to control bleeding. However, because interfacial water is a formidable barrier to strong surface bonding,a challenge remains in finding a product that offers robust tissue adhesion combined with anti-infection properties.Inspired by the strong adhesive mechanism of biofilm and mussels,we report a novel dual bionic adhesive hydrogel(DBAH)based on chitosan grafted with methacrylate(CS-MA),dopamine(DA),and N-hydroxymethyl acrylamide(NMA)via a facile radical polymerization process.CS-MA and DA were simultaneously included in the adhesive polymer for imitating the two key adhesive components:polysaccharide intercellular adhesin(PIA)of staphylococci biofilm and 3,4-dihydroxy-L-phenylalanine(Dopa)of mussel foot protein, respectively.DBAH presented strong adhesion at 34 kPa even upon three cycles of full immersion in water and was able to withstand up to 168 mm Hg blood pressure, which is significantly higher than the 60–160 mm Hg measured in most clinical settings. Most importantly, these hydrogels presented outstanding hemostatic capability under wet and dynamic in vivo movements while displaying excellent antibacterial properties and biocompatibility. Therefore,DBAH represents a promising class of biomaterials for high-efficiency hemostasis and wound healing. | Wei Han Bo Zhou Kai Yang Xin Xiong Shifang Luan Yu Wang Zheng Xu Peng Lei Zhengshan Luo Jian Gao Yijing Zhan Guopu Chen Lei Liang Rui Wang Sha Li Hong Xu | 2020 | Bioactive Materials2020,5,4: | 6
Electrochemical behavior of microbiologically influenced corrosion on Fe_3Al in marine environment显示文摘In this article, microbiologically influenced corrosion behavior of Fe3Al intermetal-lic compound in microorganism culture medium has been investigated by using weight loss methods, electrochemical techniques, and electron microscopy.Polar-ization curves showed that a sharp electrical current peak caused by surface pitting could be observed after Fe3Al electrodes were immersed in culture medium for 15 days when the polarization potential was about-790 mV vs SCE.Based on the electrochemical impedance spectroscopy(EIS)and the equivalent circuit parameters of the associated system, the corrosion products were found to exhibit a two-layer structured feature and the microorganisms could induce pitting and erosion corrosion of the inner layer. In addition, the passivating film of the inner layer was absolutely destroyed by microbial metabolic products. | Xueting CHANG Shougang CHEN Guanhui GAO Yansheng YIN Sha CHENG Tao LIU | 2009 | Acta Metallurgica Sinica(English Letters)2009,22,4: | 5
A fully automatic registration approach based on contour and SIFT for HJ-1 images显示文摘To achieve a fully automatic registration between HJ-1 CCD images and HJ-1 infrared images is a difficult task as it must deal with the varying illuminations and resolutions of the images, different perspectives, and the local deformations within the images. In this paper, aimed at those registration issues,a fully automatic registration approach based on contour and SIFT is proposed. The registration technique performs a pre-registration process using contour feature matching algorithm that decides the overlapping region between a reference image and an input image.Once the coarse regions are obtained,it performs a fine registration process based on SIFT detector and a local adaptive matching strategy. In the fine registration process, image blocking theory is used, which not only speeds up the features extraction and matching, but also makes the matching point pairs distributed uniformly in images, and further improves the accuracy of input image rectification.Experiments with visible images and infrared images from HJ-1A/B demonstrate the efficiency and the accuracy of the proposed technique for multisource remote sensing images registration. | NI XiLiang CAO ChunXiang DING Lin JIANG Tao ZHANG Hao JIA HuiCong LI GuangHe ZHAO Jian CHEN Wei JI Wei XU Min GAO MengXu ZHENG Sheng TIAN Rong LIU Cheng LI Sha | 2012 | Science China Earth Sciences2012,55,10: | 5
Circulating MicroRNAs as Novel Diagnostic Biomarkers for Very Early-onset(≤40 years) Coronary Artery Disease显示文摘Objective Very early-onset coronary artery disease(CAD) is a great challenge in cardiovascular medicine throughout the world, especially regarding its early diagnosis. This study explored whether circulating micro RNAs(mi RNAs) could be used as potential biomarkers for patients with very early-onset CAD.Methods We performed an initial screening of mi RNA expression using RNA isolated from 20 patients with angiographically documented very early-onset CAD and 20 age-and sex-matched normal controls. For further confirmation,we prospectively examined the mi RNAs selected from 40 patients with very early-onset CAD and 40 angiography-normal controls. Results A total of 22 overexpressed mi RNAs and 22 underexpressed mi RNAs were detected in the initial screening.RT-q PCR analysis of the mi RNAs obtained from the initial screening revealed that four mi RNAs including mi R-196-5p,mi R-3163-3p,mi R-145-3p,and mi R-190a-5p exhibited significantly decreased expression in patients compared with that in controls(P<0.05).The areas under the receiver operating characteristic curve for these mi RNAs were 0.824(95% CI,0.731-0.917;P<0.001),0.758(95% CI,0.651-0.864;P<0.001),0.753(95% CI,0.643-0.863;P<0.001),and 0.782(95% CI,0.680-0.884;P<0.001),respectively,in the validation set. Conclusion To our knowledge, this is an advanced study to report about four serum mi RNAs(mi R-196-5p,mi R-3163-3p,mi R-145-3p,and mi R-190a-5p) that could be used as novel biomarkers for the diagnosis of very early-onset CAD. | DU Ying YANG Sheng Hua LI Sha CUI Chuan Jue ZHANG Yan ZHU Cheng Gang GUO Yuan Lin WU Na Qiong GAO Ying SUN Jing DONG Qian LIU Geng LI Jian Jun | 2016 | Biomedical and Environmental Sciences2016,29,8: | 5
The role of pineal microRNA-325 in regulating circadian rhythms after neonatal hypoxic-ischemic brain damage显示文摘Circadian rhythm disorder is a common, but often neglected, consequence of neonatal hypoxic-ischemic brain damage(HIBD).However, the underlying molecular mechanisms remain largely unknown. We previously showed that,in a rat model of HIBD,up-regulation of microRNA-325(miR-325)in the pineal gland is responsible for the suppression of Aanat,a key enzyme involved in melatonin synthesis and circadian rhythm regulation. To better understand the mechanism by which miR-325 affects circadian rhythms in neonates with HIBD,we compared clinical samples from neonates with HIBD and samples from healthy neonates recruited from the First Affiliated Hospital of Soochow University(Dushuhu Branch)in 2019.We found that circulating miR-325 levels correlated positively with the severity of sleep and circadian rhythm disorders in neonates with HIBD.Furthermore,a luciferase reporter gene assay revealed that LIM homeobox 3(LHX3)is a novel downstream target of miR-325.In addition,in miR-325 knock-down mice, the transcription factor LHX3 exhibited an miR-325-dependent circadian pattern of expression in the pineal gland. We established a neonatal mouse model of HIBD by performing doublelayer ligation of the left common carotid artery and exposing the pups to a low-oxygen environment for 2 hours.Lhx3 mRNA expression was significantly down-regulated in these mice and partially rescued in miR-325 knockout mice subjected to the same conditions. Finally,we showed that improvement in circadian rhythm-related behaviors in animals with HIBD was dependent on both miR-325 and LHX3.Taken together, our findings suggest that the miR-325-LHX3 axis is responsible for regulating circadian rhythms and provide novel insights into the identification of potential therapeutic targets for circadian rhythm disorders in patients with neonatal HIBD.The clinical trial was approved by Institutional Review Board of Children’s Hospital of Soochow University(approval No.2015028)on July 20,2015.Animal experiments were approved by Animal Care and Use Committee,School of Medicine,Soochow University,China(approval No.XD-2016-1)on January 15,2016. | Ning Sha Hua-Wei Wang Bin Sun Min Gong Po Miao Xiao-Lu Jiang Xiao-Feng Yang Mei Li Li-Xiao Xu Chen-Xi Feng Yuan-Yuan Yang Jie Zhang Wen-Jing Zhu Yuan-Yuan Gao Xing Feng Xin Ding | 2021 | Neural Regeneration Research2021,16,10: | 4
Generating broad-spectrum tolerance to ALS-inhibiting herbicides in rice by base editing显示文摘Herbicide-tolerant rice varieties generated by genome editing are highly desirable for weed control. We have used a cytosine base editor to create a series of missense mutations in the P171 and/or G628 codons of the acetolactate synthase(ALS)gene to confer herbicide tolerance in rice. The four different missense mutations in the P171 codon,P171S,P171A,P171Yand P171F,exhibited different patterns of tolerance towards five representative herbicides from five chemical families of ALS inhibitors. For example,P171S and P171A had lower levels of tolerance than P171Y and P171F to bispyribac but not to the other herbicides.Interestingly,a novel triple mutant(P171F/G628E/G629S)had the highest tolerance to all five tested herbicides.Field trials showed that both P171F and P171F/G628E/G629S could potentially be used with nicosulfuron. Our work illustrates an effective way of using base editing to generate herbicide tolerance in elite rice varieties. | Rui Zhang Sha Chen Xiangbing Meng Zhuangzhuang Chai Delin Wang Yuge Yuan Kunling Chen Linjian Jiang Jiayang Li Caixia Gao | 2021 | Science China(Life Sciences)2021,64,10: | 4
Long-term adenosine A1 receptor activation-induced sortilin expression promotes α-synuclein upregulation in dopaminergic neurons显示文摘Prolonged activation of adenosine A1 receptor likely leads to damage of dopaminergic neurons and subsequent development of neurodegenerative diseases. However, the pathogenesis underlying long-term adenosine A1 receptor activation-induced neurodegeneration remains unclear. In this study, rats were intraperitoneally injected with 5 mg/kg of the adenosine A1 receptor agonist N6-cyclopentyladenosine(CPA)for five weeks. The mobility of rats was evaluated by forced swimming test, while their cognitive capabilities were evaluated by Y-maze test.Expression of sortilin,α-synuclein,p-JUN,and c-JUN proteins in the substantia nigra were detected by western blot analysis. In addition, immunofluorescence staining of sortilin andα-synuclein was performed to detect expression in the substantia nigra. The results showed that, compared with adenosine A1 receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine(5 mg/kg)+CPA co-treated rats, motor and memory abilities were reduced, surface expression of sortin andα-synuclein in dopaminergic neurons was reduced, and total sortilin and totalα-synuclein were increased in CPA-treated rats.MN9D cells were incubated with 500 nM CPA alone or in combination with 10μM SP600125(JNK inhibitor)for 48 hours.Quantitative real-time polymerase chain reaction analysis of sortilin andα-synuclein mRNA levels in MN9D cells revealed upregulated sortilin expression in MN9D cells cultured with CPA alone, but the combination of CPA and SP600125 could inhibit this expression.Predictions made using Jasper,PROMO,and Alibaba online databases identified a highly conserved sequence in the sortilin promoter that was predicted to bind JUN in both humans and rodents.A luciferase reporter assay of sortilin promoter plasmid-transfected HEK293T cells confirmed this prediction. After sortilin expression was inhibited by sh-SORT1,expression of p-JUN and c-JUN was detected by western blot analysis.Long-term adenosine A1 receptor activation levels upregulatedα-synuclein expression at the post-transcriptional level by affecting sortilin expression. The online tool Raptor-X-Binding and Discovery Studio 4.5 prediction software predicted that sortilin can bind toα-synuclein.Co-immunoprecipitation revealed an interaction between sortilin andα-synuclein in MN9D cells. Our findings indicate that suppression of prolonged adenosine A1 receptor activation potently inhibited sortilin expression andα-synuclein accumulation, and dramatically improved host cognition and kineticism. This study was approved by the University Committee of Animal Care and Supply at the University of Saskatchewan(approval No.AUP#20070090)in March 2007 and the Animals Ethics Committee of University of South China(approval No.LL0387-USC)in June 2017. | Yun-Cheng Lv An-Bo Gao Jing Yang Li-Yuan Zhong Bo Jia Shu-Hui Ouyang Le Gui Tian-Hong Peng Sha Sun Francisco S.Cayabyab | 2020 | Neural Regeneration Research2020,15,4: | 4
Sortilin promotes macrophage cholesterol accumulation and aortic atherosclerosis through lysosomal degradation of ATP-binding cassette transporter A1 protein显示文摘Sortilin is closely associated with hyperlipidemia and the risk of atherosclerosis(AS).The role of sortilin and the underlying mechanism in peripheral macrophage are not fully understood. In this study,we investigated the effect of macrophage sortilin on ATP-binding cassette transporter A1(ABCA1)expression,ABCA1-mediated cholesterol efflux, and aortic AS.Macrophage sortilin expression was upregulated by oxidized low-density lipoproteins(ox-LDLs)in both concentration-and time-dependent manners. Its expression reached the peak level when cells were incubated with 50μg/ml ox-LDL for 24 h.Overexpression of sortilin in macrophage reduced cholesterol efflux, leading to an increase in intracellular total cholesterol, free cholesterol, and cholesterol ester.Sortilin was found to bind with ABCA1 protein and suppress macrophage ABCA1 expression, resulting in a decrease in cholesterol efflux from macrophages. The inhibitory effect of sortilin in cholesterol efflux was partially reversed by treatment with chloroquine,a lysosomal inhibitor. On the contrary, the ABCA1 protein level and ABCA1-mediated cholesterol efflux is increased by sortilin short hairpin RNA transfection. The fecal and biliary cholesterol 3H-sterol from cholesterol-laden mouse peritoneal macrophage was reduced by sortilin overexpression through lentivirus vector(LV)-sortilin in low-density lipoprotein receptor knockout mice, which was prevented by co-treatment with chloroquine.Treatment with LV-sortilin reduced plasma high-density lipoprotein and increased plasma ox-LDL levels.Accordingly, aortic lipid deposition and plaque area were exacerbated, and ABCA1 expression was reduced in mice in response to infection with LV-sortilin alone. These effects of LV-sortilin were partially reversed by chloroquine.Sortilin enhances lysosomal degradation of ABCA1 protein and suppresses ABCA1-mediated cholesterol efflux from macrophages, leading to foam cell formation and AS development. | Yuncheng Lv Jing Yang Anbo Gao Sha Sun Xilong Zheng Xi Chen Wei Wan Chaoke Tang Wei Xie Suyun Li Dongming Guo Tianhong Peng Guojun Zhao Liyuan Zhong | 2019 | Acta Biochimica et Biophysica Sinica2019,51,5: | 4
