| 1 | Long-chain acyl-CoA synthetase in fatty acid metabolism involved in liver and other diseases:An update显示文摘Long-chain acyl-Co A synthetase(ACSL) family members include five different ACSL isoforms, each encoded by a separate gene and have multiple spliced variants. ACSLs on endoplasmic reticulum and mitochondrial outer membrance catalyze fatty acids with chain lengths from 12 to 20 carbon atoms to form acyl-Co As, which are lipid metabolic intermediates and involved in fatty acid metabolism, membrane modifications and various physiological processes. Gain- or lossof-function studies have shown that the expression of individual ACSL isoforms can alter the distribution and amount of intracellular fatty acids. Changes in the types and amounts of fatty acids, in turn, can alter the expression of intracellular ACSLs. ACSL family members affect not only the proliferation of normal cells, but the proliferation of malignant tumor cells. They also regulate cell apoptosis through different signaling pathways and molecular mechanisms. ACSL members have individual functions in fatty acid metabolism in different types of cells depending on substrate preferences, subcellular location and tissue specificity, thus contributing to liver diseases and metabolic diseases, such as fatty liver disease, obesity, atherosclerosis and diabetes. They are also linked to neurological disorders and other diseases. However, the mechanisms are unclear. | 4 | Classification of four distinct osteoarthritis subtypes with a knee joint tissue transcriptome atlas显示文摘The limited molecular classifications and disease signatures of osteoarthritis(OA)impede the development of prediagnosis and targeted therapeutics for OA patients.To classify and understand the subtypes of OA,we collected three types of tissue including cartilage,subchondral bone,and synovium from multiple clinical centers and constructed an extensive transcriptome atlas of OA patients.By applying unsupervised clustering analysis to the cartilage transcriptome,OA patients were classified into four subtypes with distinct molecular signatures:a glycosaminoglycan metabolic disorder subtype(C1),a collagen metabolic disorder subtype(C2),an activated sensory neuron subtype(C3),and an inflammation subtype(C4).Through ligand-receptor crosstalk analysis of the three knee tissue types,we linked molecular functions with the clinical symptoms of different OA subtypes.For example,the Gene Ontology functional term of vasculature development was enriched in the subchondral bone-cartilage crosstalk of C2 and the cartilage-subchondral bone crosstalk of C4,which might lead to severe osteophytes in C2 patients and apparent joint space narrowing in C4 patients.Based on the marker genes of the four OA subtypes identified in this study,we modeled OA subtypes with two independent published RNA-seq datasets through random forest classification.The findings of this work contradicted traditional OA diagnosis by medical imaging and revealed distinct molecular subtypes in knee OA patients,which may allow for precise diagnosis and treatment of OA. | Chunhui Yuan Zongyou Pan Kun Zhao Jun Li Zixuan Sheng Xudong Yao Hua Liu Xiaolei Zhang Yang Yang Dongsheng Yu Yu Zhang Yuzi Xu Zhi-Yong Zhang Tianlong Huang Wanlu Liu Hongwei Ouyang | 2020 | Bone Research2020,8,4: | 4 |

| 6 | Ameliorating end-product inhibition to improve cadaverine production in engineered Escherichia coli and its application in the synthesis of bio-based diisocyanates显示文摘Cadaverine is an important C5 platform chemical with a wide range of industrial applications.However,the cadaverine inhibition on the fermenting strain limited its industrial efficiency of the strain.In this study,we report an engineered Escherichia coli strain with high cadaverine productivity that was generated by developing a robust host coupled with metabolic engineering to mitigate cadaverine inhibition.First,a lysine producing E.coli was treated with a combination of radiation(ultraviolet and visible spectrum)and ARTP(atmospheric and room temperature plasma)mutagenesis to obtain a robust host with high cadaverine tolerance.Three mutant targets including HokD,PhnI and PuuR are identified for improved cadaverine tolerance.Further transcriptome analysis suggested that cadaverine suppressed the synthesis of ATP and lysine precursor.Accordingly,the related genes involved in glycolysis and lysine precursor,as well as cadaverine exporter was engineered to release the cadaverine inhibition.The final engineered strain was fed-batch cultured and a titer of 58.7 g/L cadaverine was achieved with a yield of 0.396 g/g,both of which were the highest level reported to date in E.coli.The bio-based cadaverine was purified to>99.6%purity,and successfully used for the synthesis of polyurethane precursor 1,5-pentamethylene diisocyanate(PDI)through the approach of carbamate decomposition. | Xin Wang Xing Guo Jing Wang Hui Li Feng He Sheng Xu Kequan Chen Pingkai Ouyang | 2021 | Synthetic and Systems Biotechnology2021,6,4: | 3 |
| 7 | Targeting a novel inducible GPX4 alternative isoform to alleviate ferroptosis and treat metabolic-associated fatty liver disease显示文摘Metabolic-associated fatty liver disease(MAFLD),which is previously known as non-alcoholic fatty liver disease(NAFLD),represents a major health concern worldwide with limited therapy.Here,we provide evidence that ferroptosis,a novel form of regulated cell death characterized by iron-driven lipid peroxidation,was comprehensively activated in liver tissues from MAFLD patients.The canonical-GPX4(cGPX4),which is the most important negative controller of ferroptosis,is downregulated at protein but not mRNA level.Interestingly,a non-canonical GPX4 transcript-variant is induced(inducible-GPX4,iGPX4)in MAFLD condition.The high fat-fructose/sucrose diet(HFFD)and methionine/choline-deficient diet(MCD)-induced MAFLD pathologies,including hepatocellular ballooning,steatohepatitis andfibrosis,were attenuated and aggravated,respectively,in cGPX4-and iGPX4-knockin mice.cGPX4 and iGPX4 isoforms also displayed opposing effects on oxidative stress and ferroptosis in hepatocytes.Knockdown of iGPX4 by siRNA alleviated lipid stress,ferroptosis and cell injury.Mechanistically,the triggered iGPX4 interacts with cGPX4 to facilitate the transformation of cGPX4 from enzymatic-active monomer to enzymatic-inactive oligomers upon lipid stress,and thus promotes ferroptosis.Co-immunoprecipitation and nano LC–MS/MS analyses confirmed the interaction between iGPX4 and cGPX4.Our results reveal a detrimental role of non-canonical GPX4 isoform in ferroptosis,and indicate selectively targeting iGPX4 may be a promising therapeutic strategy for MAFLD. | Jie Tong Dongjie Li Hongbo Meng Diyang Sun Xiuting Lan Min Ni Jiawei Ma Feiyan Zeng Sijia Sun Jiangtao Fu Guoqiang Li Qingxin Ji Guoyan Zhang Qirui Shen Yuanyuan Wang Jiahui Zhu Yi Zhao Xujie Wang Yi Liu Shenxi Ouyang Chunquan Sheng Fuming Shen Pei Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,9: | 3 |

| 12 | Preparation and Properties of Bio-Based Flame Retardant Polyvinyl Alcohol显示文摘Polyvinyl alcohol (PVA) has been widely used in the fields of medical, food and packaging due to its excellentbiocompatibility, good fiber-forming and film-forming properties. However, the high flammability of PVA hasgreatly limited its wider applications. The flame-retardant PVA was prepared by melt blending of a bio-basedflame retardant (prepared from lignin, phosphoric acid and carbamide) with thermoplastic PVA (TPVA). Thechemical structure, morphology, thermal properties, mechanical properties, fire property and fluidity of thisflame retardant PVA were investigated by Fourier transform infrared spectrometer(FTIR), field emission scanning electron microscope(SEM), thermogravimetric analyzer(TGA), impact tester, universal testing machine,horizontal-vertical burning tester, limiting oxygen indexer(LOI) and melt flow rate meter(MFR). The resultsshowed that the prepared flame retardant had good compatibility with the PVA substrate;The impact strength,melt flow rate, fire property and char residue of this PVA material increased with the content of bio-based flameretardant. When the content of flame retardant was of 20%, the five indices including impact strength, meltflow rate, UL-94 level, LOI and char residual were 11.3 KJ/m^(2), 21.2 g/10 min, V-0 UL-94 level, 33.1%, and19.2%, respectively.
