1Establishment of apparent quantum yield and maximum ecosystem assimilation on Tibetan Plateau alpine meadow ecosystem显示文摘The alpine meadow is widely distributed on the Tibetan Plateau with an area of about 1.2×106kn2. Damxung County, located in the hinterland of the Tibetan Plateau, is the place covered with this typical vegetation. An open-path eddy covariance system was set up in Damxung rangeland station to measure the carbon flux of alpine meadow from July to October,2003. The continuous carbon flux data were used to analyze the relationship between net ecosystem carbon dioxide exchange (NEE) and photosynthetically active radiation (PAR), as well as the seasonal patterns of apparent quantum yield (α) and maximum ecosystem assimilation (Pmax).Results showed that the daytime NEE fitted fairly well with the PAR in a rectangular hyperbola function, with α declining in the order of peak growth period (0.0244 μmolCO2 · μmol-1pAR) >early growth period > seed maturing period > withering period (0.0098 μmolCO2 · μmol-1pAR).The Pmax did not change greatly during the first three periods, with an average of 0.433mgCO2· m-2· s-1, i.e. 9.829 μmolCO2· m-2· s-1. However, during the withering period, Pmax was only 0.35 mgCO2 · m-2 · s-1, i.e. 7.945 μmolCO2 · m-2 · s-1. 2Toxicological effects involved in risk assessment of rare earth lanthanum on roots of Vicia faba L.seedlings显示文摘Combined chemical analyses and biological measurements were utilized to investigate potential toxicological effects and possible mechanisms involved in risk assessment of rare earth elements (REEs) on Vicia faba L.seedlings,which were hydroponically cultivated and exposed to various concentrations of lanthanum (La) for 15 days.The results showed that La contents in both the solution and roots increased with the increase of extraneous La,contributing to hormetic dose responses of superoxide dismutase (SOD),catalase (CAT),guaiacol peroxidase (GPX),ascorbate peroxidase (APX) and endoprotease (EP) isozymes activities,and HSP 70 production enhanced at low doses but suppressed at higher doses of La.These physiological responses constituted antioxidant and detoxification systems against La-induced oxidative stress.The elevated La levels also contributed to oxidatively modified proteins,which were most responsible for subsequent cell death and growth retardation of the roots.By combination of hormetic and traditional threshold dose levels,the threshold dose range was deduced to be 108-195 μg La/g dry weight in the roots,corresponding to 0.90-3.12 mg/L of soluble La in the culture solution.It suggests that persistent applications of REEs may lead to potential ecological risk in the environment.Chengrun Wang Mei He Wen Shi Jessie Wong Tao Cheng Xiaorong Wang Lingling Hu Fenfen Chen 2011Journal of Environmental Sciences2011,23,10:9
3Sequential fate-switches in stem-like cells drive the tumorigenic trajectory from human neural stem cells to malignant glioma显示文摘Glioblastoma(GBM)is an incurable and highly heterogeneous brain tumor,originating from human neural stem/progenitor cells(hNSCs/hNPCs)years ahead of diagnosis.Despite extensive efforts to characterize hNSCs and end-stage GBM at bulk and single-cell levels,the de novo gliomagenic path from hNSCs is largely unknown due to technical difficulties in early-stage sampling and preclinical modeling.Here,we established two highly penetrant hNSC-derived malignant glioma models,which resemble the histopathology and transcriptional heterogeneity of human GBM.Integrating time-series analyses of whole-exome sequencing,bulk and single-cell RNA-seq,we reconstructed gliomagenic trajectories,and identified a persistent NSC-like population at all stages of tumorigenesis.Through trajectory analyses and lineage tracing,we showed that tumor progression is primarily driven by multi-step transcriptional reprogramming and fate-switches in the NSC-like cells,which sequentially generate malignant heterogeneity and induce tumor phenotype transitions.We further uncovered stage-specific oncogenic cascades,and among the candidate genes we functionally validated C1QL1 as a new glioma-promoting factor.Importantly,the neurogenic-to-gliogenic switch in NSC-like cells marks an early stage characterized by a burst of oncogenic alterations,during which transient AP-1 inhibition is sufficient to inhibit gliomagenesis.Together,our results reveal previously undercharacterized molecular dynamics and fate choices driving de novo gliomagenesis from hNSCs,and provide a blueprint for potential early-stage treatment/diagnosis for GBM.Xiaofei Wang Ran Zhou Yanzhen Xiong Lingling Zhou Xiang Yan Manli Wang Fan Li Chuanxing Xie Yiming Zhang Zongyao Huang Chaoqiong Ding Kaidou Shi Weida Li Yu Liu Zhongwei Cao Zhen-Ning Zhang Shengtao Zhou Chong Chen Yan Zhang Lu Chen Yuan Wang 2021Cell Research2021,31,6:8
4The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 5Histogram method for reliable thickness measurements of graphene films using atomic force microscopy(AFM)显示文摘Atomic force microscopy(AFM) is a commonly used technique for graphene thickness measurement.However, due to surface roughness caused by graphene itself and variation introduced in AFM measurement, graphene thickness is difficult to be accurately determined by AFM. In this paper, a histogram method was used for reliable measurements of graphene thickness using AFM. The influences of various measurement parameters in AFM analysis were investigated. The experimental results indicate that significant deviation can be introduced using various order of flatten and improperly selected measurement parameters including amplitude setpoint and drive amplitude. At amplitude setpoint of 100 mV and drive amplitude of 100 m V, thickness of 1 layer(1L), 2 layers(2L) and 4 layers(4L) graphene were measured.The height differences for 1L, 2L and 4L were 1.51 ± 0.16 nm, 1.92 ± 0.13 nm and 2.73 ± 0.10 nm, respectively. 7Fabric changes induced by super-absorbent polymer on cementelime stabilized excavated clayey soil显示文摘This paper studies the microstructure variation induced by super-absorbent polymer(SAP)to understand the mechanism of macroscopic strength improvement of stabilized soil.The fabric changes of cement elime stabilized soil were analyzed with respect to the variation of SAP content,water content,lime content and curing time,using mercury intrusion porosimetry(MIP)tests.It can be observed that the delimitation pore diameter between inter-and intra-aggregate pores was 0.2 mm for the studied soil,determined through the intrusion/extrusion cycles.Experimental results showed that fabric in both inter-and intra-aggregate pores varied significantly with SAP content,lime content,water content and curing time.Two main changes in fabric due to SAP are identified as:(1)an increase in intra-aggregate pores(<0.2 mm)due to the closer soilecementelime cluster space at higher SAP content;and(2)a decrease in inter-aggregate pores represented by a reduction in small-pores(0.2e2 mm)due to the lower pore volume of soil mixture after water absorption by SAP,and a slight increase in large-pores(>2 mm)due to the shrinkage of SAP particle during the freezeedry process of MIP test.Accordingly,the strength gain due to SAP for cementelime stabilized soil was mainly due to a denser fabric with less interaggregate pores.The cementitious products gradually developed over time,leading to an increase in intra-aggregate pores with an increasing proportion of micro-pores(0.006e0.2 mm).Meanwhile,the inter-aggregate pores were filled by cementitious products,resulting in a decrease in total void ratio.Hence,the strength development over time is attributable to the enhancement of cementation bonding and the refinement of fabric due to the increasing cementitious compounds.Xia Bian Lingling Zeng Xiaozhao Li Xiusong Shi Shuming Zhou Fuqing Li 2021Journal of Rock Mechanics and Geotechnical Engineering2021,13,5:3
8DVL mutations identified from human neural tube defects and Dandy-Walker malformation obstruct the Wnt signaling pathway显示文摘Wnt signaling pathways,including the canonical Wnt/β-catenin pathway,planar cell polarity pathway,and Wnt/Ca2+signaling pathway,play important roles in neural development during embryonic stages.The DVL genes encode the hub proteins for Wnt signaling pathways.The mutations in DVL2 and DVL3 were identified from patients with neural tube defects(NTDs),but their functions in the pathogenesis of human neural diseases remain elusive.Here,we sequenced the coding regions of three DVL genes in 176 stillborn or miscarried fetuses with NTDs or Dandy-Walker malformation(DWM)and 480 adult controls from a Han Chinese population.Four rare mutations were identified:DVL1 p.R558 H,DVL1 p.R606 C,DVL2 p.R633 W,and DVL3 p.R222 Q.To assess the effect of these mutations on NTDs and DWM,various functional analyses such as luciferase reporter assay,stress fiber formation,and in vivo teratogenic assay were performed.The results showed that the DVL2 p.R633 W mutation destabilized DVL2 protein and upregulated activities for all three Wnt signalings(Wnt/β-catenin signaling,Wnt/planar cell polarity signaling,and Wnt/Ca2+signaling)in mammalian cells.In contrast,DVL1 mutants(DVL1 p.R558 H and DVL1 p.R606 C)decreased canonical Wnt/β-catenin signaling but increased the activity of Wnt/Ca2+signaling,and DVL3 p.R222 Q only decreased the activity of Wnt/Ca2+signaling.We also found that only the DVL2 p.R633 W mutant displayed more severe teratogenicity in zebrafish embryos than wild-type DVL2.Our study demonstrates that these four rare DVL mutations,especially DVL2 p.R633 W,may contribute to human neural diseases such as NTDs and DWM by obstructing Wnt signaling pathways.Lingling Liu Weiqi Liu Yan Shi Ling Li Yunqian Gao Yunping Lei Richard Finnell Ting Zhang Feng Zhang Li Jin Huili Li Wufan Tao Hongyan Wang 2020Journal of Genetics and Genomics2020,47,6:2
9Unusual coastal ocean cooling in the northern South China Sea by a katabatic cold jet associated with Typhoon Mujigea(2015)显示文摘This study deals with a unusual cooling event after Typhoon Mujigea passed over the northern South China Sea(SCS) in October 2015. We analyze the satellite sea surface temperature(SST) time series from October 3 to 18,2015 and find that the cooling process in the coastal ocean had two different stages. The first stage occurred immediately after typhoon passage on October 3, and reached a maximum SST drop of –2℃ on October 7 as the usual cold wake after typhoon. The second stage or the unusual extended cooling event occurred after 7d of the typhoon passage, and lasted for 5d from October 10 to 15. The maximum SST cooling was –4℃ and occurred after 12d of typhoon passage. The mechanism analysis results indicate that after landing and moving northwestward to the Yunnan-Guizhou Plateau(YGP), Typhoon Mujigea(2015) met the westerly wind front on October 5. The lowpressure and positive-vorticity disturbances to the front triggered meridional air flow and low-pressure trough,thus induced a katabatic cold jet downward from the Qinghai-Tibet Plateau(QTP) passing through the YGP to the northwestern SCS. The second cooling reached the maximum SST drop 4d later after the maximum air temperature drop of –9℃ on October 11. 10RNA modifications:importance in immune cell biology and related diseases显示文摘RNA modifications have become hot topics recently.By influencing RNA processes,including generation,transportation,function,and metabolization,they act as critical regulators of cell biology.The immune cell abnormality in human diseases is also a research focus and progressing rapidly these years.Studies have demonstrated that RNA modifications participate in the multiple biological processes of immune cells,including development,differentiation,activation,migration,and polarization,thereby modulating the immune responses and are involved in some immune related diseases.In this review,we present existing knowledge of the biological functions and underlying mechanisms of RNA modifications,including N6-methyladenosine(m^(6)A),5-methylcytosine(m^(5)C),N1-methyladenosine(m1 A),N7-methylguanosine(m^(7)G),N4-acetylcytosine(ac^(4)C),pseudouridine(Ψ),uridylation,and adenosine-to-inosine(A-to-I)RNA editing,and summarize their critical roles in immune cell biology.Via regulating the biological processes of immune cells,RNA modifications can participate in the pathogenesis of immune related diseases,such as cancers,infection,inflammatory and autoimmune diseases.We further highlight the challenges and future directions based on the existing knowledge.All in all,this review will provide helpful knowledge as well as novel ideas for the researchers in this area.Lian Cui Rui Ma Jiangluyi Cai Chunyuan Guo Zeyu Chen Lingling Yao Yuanyuan Wang Rui Fan Xin Wang Yuling Shi 2022Signal Transduction and Targeted Therapy2022,7,10:2

12Facilitated transport separation of CO_(2) and H_(2)S by supported liquid membrane based on task-specific protic ionic liquids显示文摘Selective separation of CO_(2) and H_(2)S from CH_(4) is of importance to the natural gas upgrading because of its cor-rosivity. As is well known, amine group can recognize reversibly CO_(2) and H_(2)S with a moderate interaction. Withthis in mind, this work prepared a series of amine-functionalized supported-protic-ionic-liquid membranes(SPILMs) for the selective separation of CO_(2) and H_(2)S from CH_(4) . For comparison, aprotic ionic liquid membranewas also prepared for natural gas sweetening. The effect of temperature and transmembrane pressure differenceon the permeability of single gas and ideal selectivity of CO_(2)/CH_(4) and H_(2)S/CO_(2) were studied. Facilitated transportmechanism was studied by NMR spectra. The permeability of CO_(2) in primary amine functionalized [DMPDAH][Tf_(2)N]-based membrane decreases with the increasing transmembrane pressure, indicating a typical feature offacilitated transport membrane. The permeability of CO_(2) and the ideal selective of CO_(2)/CH_(4) are as high as 2000 barrers and 53.2 at 313.2 K in [DMPDAH][Tf_(2)N] membrane, significantly higher than those in conventional ILsmembrane. 14Purification and molecular weight distribution of a key exopolysaccharide component of Bacillus megaterium TF10显示文摘Extracellular polymeric substances(EPS) are organic metabolic compounds excreted by microorganisms. They largely impact microbial aggregate structures and functions.Extracellular polysaccharides(EP) in EPS are responsible for the formation of microbial aggregates. In this work, we successfully separated and characterized EP from EPS of the bacterium Bacillus megaterium TF10. Extraction of EP from EPS was optimized using Sevag's reagent. Chemical characteristics, functional groups, and molecular weight(MW) distribution of EP were compared with the harvested EPS and soluble microbial products(SMP). We found that the polymers of lower MW and free proteins were successfully removed by Sevag's reagent. The higher MW components of EPS were predominantly polysaccharides,while the polymers of lower MW tended to secrete to the supernatant and were described as SMP. A part of the proteins in the EP was polysaccharide-bonded. 15Association of myostatin Variants with Growth Traits of Zhikong Scallop(Chlamys farreri)显示文摘Scallop is a popular sea food and an important aquaculture shellfish.Identification of genes and genetic variants relating to scallop growth could benefit high-yielding scallop breeding.Myostatin(MSTN) is a conservative regulator of muscle growth,and has become one of the most important target genes for genetic improvement of the production of farmed animals.In this study,four single nucleotide polymorphisms(SNPs) were identified in the 5' flanking region of MSTN gene(Cf MSTN) in Zhikong scallop(Chlamys farreri).The association of these SNPs with scallop growth traits,including shell length,shell height,body weight and striated muscle weight was analyzed.The SNP g-1162G>T was found to associate with shell length,shell height,and striated muscle weight.The TT type scallops showed significantly higher trait values than those of GT type,and the GG type individuals exhibited median values.On the contrary,significantly more Cf MSTN transcripts were detected in the striated muscle of GT type scallops than in those of TT and GG type ones.Our results suggested that Cf MSTN might regulate the scallop muscle growth negatively,and SNP g-1162G>T can be used as a candidate marker for the selective breeding of high-yielding scallop.FU Qiang GUO Huihui FENG Liying LI Xue ZHANG Lingling WANG Shi HU Xiaoli BAO Zhenmin 2016Journal of Ocean University of China2016,15,1:2
16CLDASSD:Reconstructing Fine Textures of the Temperature Field Using Super-Resolution Technology显示文摘Before 2008,the number of surface observation stations in China was small.Thus,the surface observation data were too sparse to effectively support the High-resolution China Meteorological Administration’s Land Assimilation System(HRCLDAS)which ultimately inhibited the output of high-resolution and high-quality gridded products.This paper proposes a statistical downscaling model based on a deep learning algorithm in super-resolution to research the above problem.Specifically,we take temperature as an example.The model is used to downscale the 0.0625°×0.0625°,2-m temperature data from the China Meteorological Administration’s Land Data Assimilation System(CLDAS)to 0.01°×0.01°,named CLDASSD.We performed quality control on the paired data from CLDAS and HRCLDAS,using data from 2018 and 2019.CLDASSD was trained on the data from 31 March 2018 to 28 February 2019,and then tested with the remaining data.Finally,extensive experiments were conducted in the Beijing-Tianjin-Hebei region which features complex and diverse geomorphology.Taking the HRCLDAS product and surface observation data as the'true values'and comparing them with the results of bilinear interpolation,especially in complex terrain such as mountains,the root mean square error(RMSE)of the CLDASSD output can be reduced by approximately 0.1℃,and its structural similarity(SSIM)was approximately 0.2 higher.CLDASSD can estimate detailed textures,in terms of spatial distribution,with greater accuracy than bilinear interpolation and other sub-models and can perform the expected downscaling tasks.Ruian TIE Chunxiang SHI Gang WAN Xingjie HU Lihua KANG Lingling GE 2022Advances in Atmospheric Sciences2022,39,1:2
