| 1 | PGE2 activates EP4 in subchondral bone osteoclasts to regulate osteoarthritis显示文摘Prostaglandin E2(PGE2), a major cyclooxygenase-2(COX-2) product, is highly secreted by the osteoblast lineage in the subchondral bone tissue of osteoarthritis(OA) patients. However, NSAIDs, including COX-2 inhibitors, have severe side effects during OA treatment. Therefore, the identification of novel drug targets of PGE2 signaling in OA progression is urgently needed. Osteoclasts play a critical role in subchondral bone homeostasis and OA-related pain. However, the mechanisms by which PGE2 regulates osteoclast function and subsequently subchondral bone homeostasis are largely unknown. Here, we show that PGE2 acts via EP4 receptors on osteoclasts during the progression of OA and OA-related pain. Our data show that while PGE2 mediates migration and osteoclastogenesis via its EP2 and EP4 receptors, tissue-specific knockout of only the EP4 receptor in osteoclasts(EP4 Lys M) reduced disease progression and osteophyte formation in a murine model of OA. Furthermore, OA-related pain was alleviated in the EP4 Lys M mice, with reduced Netrin-1 secretion and CGRP-positive sensory innervation of the subchondral bone. The expression of plateletderived growth factor-BB(PDGF-BB) was also lower in the EP4 Lys Mmice, which resulted in reduced type H blood vessel formation in subchondral bone. Importantly, we identified a novel potent EP4 antagonist, HL-43, which showed in vitro and in vivo effects consistent with those observed in the EP4 Lys Mmice. Finally, we showed that the Gαs/PI3 K/AKT/MAPK signaling pathway is downstream of EP4 activation via PGE2 in osteoclasts. | 2 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 5 | The Effects of Chronic Hypoxia on Thermoregulation and Metabolism in Phrynocephalus vlangalii显示文摘Phrynocephalus vlangalii are widely distributed on Tibetan plateau spanning diverse altitudes and habitats. In the present study, P.vlangalii were exposed to 8% oxygen concentration in a hypoxic chamber for 6 weeks. Then the body temperature(T_b), standard metabolic rate(SMR), heart rate and metabolic enzyme activities of the lizards were measured at 20℃ and 30℃. The results indicated that hypoxia exposure decreased T_b, SMR and heart rate. Lactate dehydrogenase(LDH) activity of 8% O_2 group became significant elevated in liver and skeletal muscle compared with control group at 20℃, but descended significantly in heart. Using electrophoresis we found that LDH contains five isozymes(LDH1, LDH2, LDH3, LDH4 and LDH5) and are expressed specifically in liver, skeletal muscle and heart. Citrate synthase(CS) activity in the liver also decreased at 20℃ and 30℃. | 7 | Increased Expression of Colonic Mucosal Melatonin in Patients with Irritable Bowel Syndrome Correlated with Gut Dysbiosis显示文摘Dysregulation of the gut microbiota/gut hormone axis contributes to the pathogenesis of irritable bowel syndrome(IBS).Melatonin plays a beneficial role in gut motility and immunity.However,altered expression of local mucosal melatonin in IBS and its relationship with the gut microbiota remain unclear.Therefore,we aimed to detect the colonic melatonin levels and microbiota profiles in patients with diarrhea-predominant IBS(IBS-D)and explore their relationship in germ-free(GF)rats and BON-1 cells.Thirty-two IBS-D patients and twenty-eight healthy controls(HCs)were recruited.Fecal specimens from IBS-D patients and HCs were separately transplanted into GF rats by gavage.The levels of colon mucosal melatonin were assessed by immunohistochemical methods,and fecal microbiota communities were analyzed using 16S rDNA sequencing.The effect of butyrate on melatonin synthesis in BON-1 cells was evaluated by ELISA.Melatonin levels were significantly increased and negatively correlated with visceral hypersensitivity in IBS-D patients.GF rats inoculated with fecal microbiota from IBS-D patients had high colonic melatonin levels.Butyrate-producing Clostridium cluster XIVa species,such as Roseburia species and Lachnospira species,were positively related to colonic mucosal melatonin expression.Butyrate significantly increased melatonin secretion in BON-1 cells.Increased melatonin expression may be an adaptive protective mechanism in the development of IBS-D.Moreover,some Clostridium cluster XIVa species could increase melatonin expression via butyrate production.Modulation of the gut hormone/gut microbiota axis offers a promising target of interest for IBS in the future. | Ben Wang Shiwei Zhu Zuojing Liu Hui Wei Lu Zhang Meibo He Fei Pei Jindong Zhang Qinghua Sun Liping Duan | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 2 |

| 10 | A promising new class of plasticine: Metallic plasticine显示文摘Soft, malleable, and non-dry on exposure in air are the typical features for plain plasticine, which lead plasticine to be widely used in many industrial fields and our daily life. As a kind of clay, poorly electric conductivity and thermal conductivity of plain plasticine seriously limit its applications. Therefore,synthesizing a kind of plasticine having metallic bond is of importance for extending its applications in some special cases, such as thermal-cooling medium, anti-static electricity, electromagnetic shielding,etc. Here, we report a novel GaInSnCdZn_2 alloy, which exhibits similar behavior as compared to those of plasticine at near room temperature(30–40℃), and a good electrical conductivity due to its nature of metal. This new GaInSnCdZn_2 alloy can be called as metallic plasticine that contains the near-eutectic structure with low melting point and the other relatively high melting point phases. In this metallic plasticine, the near-eutectic structure with low melting point plays the same role as the oily ingredient in plain plasticine, dominating the plastic deformation, while the other relatively high melting point phases act as the stuffing like the CaCO3 in plain plasticine. | 17 | Extraction of Alpha transmitter signals from single-station observations using the direction-finding method显示文摘For the applications of Alpha very-low-frequency(VLF) systems consisting of multiple transmitters, determining the origin transmitter station of received signals is a crucial problem. Based on single-station observations, this study develops a directionfinding method to extract Russian Alpha transmitter signals. First, the amplitudes of Alpha signals received in Suizhou City,Hubei Province(31.57°N, 113.32°E) in the east-west(EW) and north-south(NS) directions are obtained by the power spectrum method. The amplitude ratios of signals in the two directions are subsequently adopted to estimate the propagation angles of the signals with respect to the NS direction of the VLF receiver station. Phase ambiguity in our system is eliminated by comparing Alpha signal waveform with that from the VTX transmitter in India(8.39°N, 77.75°E) as a reference station. Finally, we can determine the quadrant where the incoming wave signals are located relative to the receiver and eventually distinguish the exact Alpha transmitters. Based on the direction-finding results, the amplitudes of each Alpha signal are extracted, and their diurnal variation features are analyzed to verify the performance of our method.
