| 3 | Comparisons of carcass traits,meat quality,and serum metabolome between Shaziling and Yorkshire pigs显示文摘This study aims to compare the meat quality of Shaziling and Yorkshire pigs and to find the potential indicator in serum for superior meat quality.Six Shaziling and Yorkshire pigs at 30,60,90,150,210,and 300 d of age were selected to examine carcass traits,meat quality,and serum metabolome.The results showed that the body weight,carcass length,and loin eye area of Shaziling pigs at 150,210,and 300 d of age were significantly lower than those of Yorkshire pigs(P<0.05).Shaziling pigs at 150 and 300 d of age had significantly lower backfat thickness than Yorkshire pigs(P<0.05).Compared with Yorkshire pigs,Shaziling pigs at all 6 ages had a lower lean percentage and a higher fat percentage(P<0.05).At 60,90,and 150 d of age,the post-mortem pH-decline,b*value(yellowness),and drip loss of Shaziling pigs were significantly lower than those of Yorkshire pigs(P<0.05).Moreover,at 150 d of age,Shaziling pigs had significantly higher a^(*)value(redness)and intramuscular fat(IMF)content than Yorkshire pigs(P<0.05).Correlation analysis between the top 40 metabolites and phenotypes indicated that L-carnitine had positive correlations with fat percentage,pH24h,and IMF content,but had negative correlations with lean percentage,L^(*)value(lightness),and b^(*)value(P<0.05).Serum L-carnitine content,fat percentage,pH24h,and IMF content all decreased first and then increased as the pigs grew,which verified the positive correlations between L-carnitine and these phenotypes.In conclusion,Shaziling pigs have a slower growth rate but a better meat quality than Yorkshire pigs.The meat quality of Shaziling pigs is the best from 150 to 210 d of age.This study suggests that a higher serum L-carnitine content is a promising indicator for better meat quality. | Bo Song Changbing Zheng Jie Zheng Shiyu Zhang Yinzhao Zhong Qiuping Guo Fengna Li Cimin Long Kang Xu Yehui Duan Yulong Yin | 2022 | Animal Nutrition2022,,1: | 3 |
| 4 | Chidamide combined with cyclophosphamide,doxorubicin,vincristine and prednisone in previously untreated patients with peripheral T-cell lymphoma显示文摘Objective:Chidamide is an oral histone deacetylase subtype-selective inhibitor approved for relapsed or refractory peripheral T-cell lymphoma(PTCL).This phase 1 b study evaluated the safety,pharmacokinetics,and preliminary efficacy of chidamide in combination with cyclophosphamide,doxorubicin,vincristine and prednisone(CHOP)for treatment-na?ve PTCL patients.Methods:This study was an open-label,multicenter trial composed of dose escalation and dose expansion.Patients received CHOP for six 21-d cycles and chidamide on d 1,4,8 and 11 in each cycle.Four dose levels of chidamide(20,25,30 and 35 mg)were evaluated.The primary objective was to evaluate the safety and tolerability of the combination regimen.Results:A total of 30 patients were evaluated in this study:15 in the dose-escalation part and 15 in the doseexpansion part.In the dose-escalation study,three patients were enrolled in the 35 mg chidamide cohort.One had dose-limiting toxicity with grade 3 vascular access complications,and one had grade 2 neutropenia with a sustained temperature>38°C.Dose escalation was stopped at this chidamide dose level.The most common(≥10%)grade 3 or 4 adverse events(AEs)were leukopenia(90.0%),neutropenia(83.3%),vomiting(13.3%),thrombocytopenia(10.0%)and febrile neutropenia(10.0%).No significant changes in chidamide pharmacokinetic properties were observed before and after combination treatment.The objective response rate for the 28 patients evaluable for preliminary efficacy was 89.3%(25/28),with 16(57.1%)achieving complete response or unconfirmed complete response.The estimated median progression-free survival was 14.0 months.In summary,we chose chidamide 30 mg as the recommended dose for phase 2.Conclusions:The addition of chidamide to standard CHOP chemotherapy was tolerable with promising preliminary efficacy in previously untreated PTCL patients,which supports further clinical studies with this combination regimen for the frontline treatment of PTCL. | Lin Gui Junning Cao Dongmei Ji Huilai Zhang Qian Fan Jun Zhu Yuqin Song Shiyu Jiang Zhiqiang Ning Jia Yu Yuankai Shi | 2021 | Chinese Journal of Cancer Research2021,33,5: | 3 |

| 8 | Electronic tattoos based on large-area Mo2C grown by chemical vapor deposition for electrophysiology显示文摘Tattoo electronics has attracted intensive interest in recent years due to its comfortable wearing and imperceivable sensing,and has been broadly applied in wearable healthcare and human-machine interface.However,the tattoo electrodes are mostly composed of metal films and conductive polymers.Two-dimensional(2D)materials,which are superior in conductivity and stability,are barely studied for electronic tattoos.Herein,we reported a novel electronic tattoo based on large-area Mo_(2)C film grown by chemical vapor deposition(CVD),and applied it to accurately and imperceivably acquire on-body electrophysiological signals and interface with robotics.High-quality Mo_(2)C film was obtained via optimizing the distribution of gas flow during CVD growth.According to the finite element simulation(FES),bottom surface of Cu foil covers more stable gas flow than the top surface,thus leading to more uniform Mo_(2)C film.The resulting Mo_(2)C film was transferred onto tattoo paper,showing a total thickness of~3μm,sheet resistance of 60-150Ω/sq,and skin-electrode impedance of~5×10^(5)Ω.Such thin Mo_(2)C electronic tattoo(MCET in short)can form conformal contact with skin and accurately record electrophysiological signals,including electromyography(EMG),electrocardiogram(ECG),and electrooculogram(EOG).These body signals collected by MCET can not only reflect the health status but also be transformed to control the robotics for human-machine interface. | Shiyu Wang Xin Wang Weifeng Zhang Xiaohu Shi Dekui Song Yan Zhang Yan Zhao Zihan Zhao Nan Liu | 2023 | Nano Research2023,16,3: | 1 |
| 9 | Effects of V and Al on Microstructure and Room Temperature Fracture Toughness of Nb-Si In-situ Composites显示文摘The effect of alloying elements V and Al on microstructure and room temperature fracture toughness of an experimental Nb-Si in-situ composite was investigated. The Nb-Si alloys with different amount of V and Al were prepared by non-consumable arc-melting furnace. The experimental results showed that with the addition of V and Al, the microstructure of the Nb-Si materials transformed from Nbss+Nb3Si to Nbss+Nb5Si3 and the lattice parameters of Nbss phase decreased, and the alloying element V promoted the formation of the finer Nbss structure. It was observed that the room temperature fracture toughness was improved by the addition of V and Al, and the optimum room-temperature fracture toughness of the material appeared in the alloy with 2.6at.%V and 3.6at.%Al. | 12 | Understanding Miocene Climate Evolution in Northeastern Tibet: Stable Carbon and Oxygen Isotope Records from the Western Tianshui Basin, China显示文摘To investigate climate evolution during the Miocene, especially during the Middle Miocene climate transition on the northeastern Tibetan Plateau, stable oxygen and carbon isotopes of carbonates from a 288-m-thick lacustrine-fluvial sediment sequence covering the period from 17.1 to 6.1 Ma from Tianshui Basin, China, were analyzed. The relatively low stable oxygen isotope values indicate the prevalence of wet climate conditions during the period of 17.1–13.6 Ma, an interval corresponding to the well-known Middle Miocene Climate Optimum. The interval between 13.6 and 11.0 Ma(i.e., the late Middle Miocene) is marked by a progressive increase in the δ18O values, indicative of a decrease in precipitation, probably linked to the expansion of the East Antarctic Ice Sheet and global cooling since about 14 Ma. The climate in the study area continued to get drier as shown by the enrichment of the heavy oxygen isotope from 11 Ma. | 15 | Tumor-microenvironment activated duplex genome-editing nanoprodrug for sensitized near-infrared titania phototherapy显示文摘Near-infrared(NIR)-light-triggered nanomedicine, including photodynamic therapy(PDT)and photothermal therapy(PTT), is growing an attractive approach for cancer therapy due to its high spatiotemporal controllability and minimal invasion, but the tumor eradication is limited by the intrinsic anti-stress response of tumor cells. Herein, we fabricate a tumor-microenvironment responsive CRISPR nanoplatform based on oxygen-deficient titania(TiO_(2-x)) for mild NIR-phototherapy. In tumor microenvironment, the overexpressed hyaluronidase(HAase) and glutathione(GSH) can readily destroy hyaluronic acid(HA) and disulfide bond and releases the Cas9/sgRNA from TiO_(2-x) to target the stress alleviating regulators, i.e., nuclear factor E2-related factor 2(NRF2) and heat shock protein 90a(HSP90a), thereby reducing the stress tolerance of tumor cells. Under subsequent NIR light illumination, the TiO_(2-x) demonstrates a higher anticancer effect both in vitro and in vivo. | 16 | Investigating association between gut microbiota and sarcopenia-related traits:a Mendelian randomization study显示文摘Background Observational studies have indicated a potential link between gut microbiota and sarcopenia.However,the underlying mechanisms and a causal relationship have not been established.Thus,the objective of this study is to examine the possible causal association between gut microbiota and sarcopenia-related traits,including low hand-grip strength and appendicular lean mass(ALM),to shed light on the gut–muscle axis.Methods To investigate the potential impact of gut microbiota on low hand-grip strength and ALM,we utilized a two-sample Mendelian randomization(MR)approach.Summary statistics were obtained from genome-wide association studies of gut microbiota,low hand-grip strength,and ALM.The primary MR analysis employed the random-effects inverse-variance weighted(IVW)method.To assess the robustness,we conducted sensitivity analyses using the MR pleiotropy residual sum and outlier(MR-PRESSO)test to detect and correct for horizontal pleiotropy,as well as the MR-Egger intercept test and leave-one-out analysis.Results Alcaligenaceae,Family XIII,and Paraprevotella were positively associated with the risk of low hand-grip strength(P-values<0.05).Streptococcaceae were negatively associated with low hand-grip strength(P-values<0.05).Eight bacterial taxa(Actinomycetales,Actinomycetaceae,Bacteroidaceae,Porphyromonadaceae,Prevotellaceae,Bacteroides,Marvinbryantia,and Phascolarctobacterium)were associated with a higher risk of ALM(P-values<0.05).Eubacterium fissicatena group was negatively associated with ALM(P-values<0.05).Conclusion We found several gut microbiota components causally associated with sarcopenia-related traits.Our findings provided insights into novel strategies for the prevention and treatment of sarcopenia through the regulation of the gut microbiota,contributing to a better understanding of the gut–muscle axis. | Jiaxi Zhao Rui Liang Quhong Song Shiyu Song Jirong Yue Chenkai Wu | 2023 | Precision Clinical Medicine2023,6,2: | 0 |
| 17 | Myeloid peroxisome proliferator-activated receptor α deficiency accelerates liver regeneration via IL-6/STAT3 pathway after 2/3 partial hepatectomy in mice显示文摘Background:Liver regeneration is a fundamental process for sustained body homeostasis and liver function recovery after injury.Emerging evidence demonstrates that myeloid cells play a critical role in liver regeneration by secreting cytokines and growth factors.Peroxisome proliferator-activated receptorα(PPARα),the target of clinical lipid-lowering fibrate drugs,regulates cell metabolism,proliferation,and survival.However,the role of myeloid PPARαin partial hepatectomy(PHx)-induced liver regeneration remains unknown.Methods:Myeloid-specific PPARa-deficient(Ppara^(Mye−/−))mice and the littermate controls(Ppara^(fl/fl))were subjected to sham or 2/3 PHx to induce liver regeneration.Hepatocyte proliferation and mitosis were assessed by immunohistochemical(IHC)staining for 5-bromo-2'-deoxyuridine(BrdU)and Ki67 as well as hematoxylin and eosin(H&E)staining.Macrophage and neutrophil infiltration into livers were reflected by IHC staining for galectin-3 and myeloperoxidase(MPO)as well as flow cytometry analysis.Macrophage migration ability was evaluated by transwell assay.The mRNA levels for cell cycle or inflammation-related genes were measured by quantitative real-time RT-PCR(qPCR).The protein levels of cell proliferation related protein and phosphorylated signal transducer and activator of transcription 3(STAT3)were detected by Western blotting.Results:Ppara^(Mye−/−)mice showed enhanced hepatocyte proliferation and mitosis at 32 h after PHx compared with Ppara^(fl/fl)mice,which was consistent with increased proliferating cell nuclear antigen(Pcna)mRNA and cyclinD1(CYCD1)protein levels in Ppara^(Mye−/−)mice at 32 h after PHx,indicating an accelerated liver regeneration in Ppara^(Mye−/−)mice.IHC staining showed that macrophages and neutrophils were increased in Ppara^(Mye−/−)liver at 32 h after PHx.Livers of Ppara^(Mye−/−)mice also showed an enhanced infiltration of M1 macrophages at 32 h after PHx.In vitro,Ppara-deficient bone marrow-derived macrophages(BMDMs)exhibited markedly enhanced migratory capacity and upregulated M1 genes Il6 and Tnfa but downregulated M2 gene Arg1 expressions.Furthermore,the phosphorylation of STAT3,a key transcript factor mediating IL6-promoted hepatocyte survival and proliferation,was reinforced in the liver of Ppara^(Mye−/−)mice after PHx.Conclusions:This study provides evidence that myeloid PPARαdeficiency accelerates PHx-induced liver regeneration via macrophage polarization and consequent IL-6/STAT3 activation,thus providing a potential target for manipulating liver regeneration. | Guomin Xie Yanting Song Na Li Zhenzhen Zhang Xia Wang Ye Liu Shiyu Jiao Ming Wei Baoqi Yu Yan Wang Hua Wang Aijuan Qu | 2022 | Hepatobiliary Surgery and Nutrition2022,11,2: | 0 |

| 19 | Functional characterization of novel NPRL3 mutations identified in three families with focal epilepsy显示文摘Focal epilepsy accounts for 60% of all forms of epilepsy, but the pathogenic mechanism is not well understood. In this study,three novel mutations in NPRL3(nitrogen permease regulator-like 3), c.937_945del, c.1514dup C and 6,706-bp genomic DNA(g DNA) deletion, were identified in three families with focal epilepsy by linkage analysis, whole exome sequencing(WES) and Sanger sequencing. NPRL3 protein is a component of the GATOR1 complex, a major inhibitor of m TOR signaling. These mutations led to truncation of the NPRL3 protein and hampered the binding between NPRL3 and DEPDC5, which is another component of the GATOR1 complex. Consequently, the mutant proteins enhanced m TOR signaling in cultured cells, possibly due to impaired inhibition of m TORC1 by GATOR1. Knockdown of nprl3 in Drosophila resulted in epilepsy-like behavior and abnormal synaptic development.
