1Scientific design and preliminary results of three-dimensional variational data assimilation system of GRAPES显示文摘The scientific design and preliminary results of the data assimilation component of the Global-Regional Prediction and Assimilation System (GRAPES) recently developed in China Meteorological Administration (CMA) are presented in this paper. This is a three-dimensional variational (3DVar) assimilation system set up on global and regional grid meshes favorable for direct assimilation of the space-based remote sensing data and matching the frame work of the prediction model GRAPES. The state variables are assumed to decompose balanced and unbalanced components. By introducing a simple transformation from the state variables to the control variables with a recursive or spectral filter, the convergence rate of iteration for minimization of the cost function in 3DVar is greatly accelerated. The definition of dynamical balance depends on the characteristic scale of the circulation considered. The ratio of the balanced to the unbalanced parts is controlled by the prescribed statistics of background errors. Idealized trials produce the same results as the analytic solution. The results of real data case studies show the capability of the system to improve analysis compared to the traditional schemes. 2Celastrus orbiculatus extract induces mitochondrial-mediated apoptosis in human hepatocellular carcinoma cells显示文摘OBJECTIVE:To investigate the apoptotic effects and underlying molecular mechanisms of Celastrus orbiculatus(C.orbiculatus) extract in human hepatocellular carcinoma cells.METHODS:Human hepatocellular carcinoma cells(HCCLM6) were treated with C.orbiculatus extract(COE) at different nontoxic concentrations(10,20,40,80,and 160 μg/mL).The effect of COE on HC-CLM6 viability was examined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assays.Cellular apoptosis following COE treatment was assessed by flow cytometry and western blot analysis.RESULTS:COE significantly inhibited cell viability and induced apoptosis of HCCLM6 cells in a dose-dependent manner.Apoptosis was accompanied by increased Bax expression and decreased Bcl-2 expression.In addition,COE treatment led to the release of cytochrome c,activation of caspase-3,and cleavage of poly(ADP-ribose) polymerase(PARP).Furthermore,activation of extracellular signal-regulated kinase(ERK),p38 kinase,and c-Jun N-terminal kinase(JNK) phosphorylation,and down-regulation of Akt phosphorylation was observed.CONCLUSION:COE induces mitochondrial-mediated,caspase-dependent apoptosis in HCCLM6 cells,which might be attributed to the activation of mitogen-activated protein kinase(MAPK) and inhibition of Akt signaling pathways.These data suggest that COE may be a potential treatment for human hepatocellular carcinoma.Hua Zhang Yayun Qian Yanqing Liu Guoqing Li Pingfang Cui Yaodong Zhu Hui Ma Xue Ji Shiyu Guo Hisamits Tadashi 2012Journal of Traditional Chinese Medicine2012,32,4:18
3Direct assimilation of satellite radiance data in GRAPES variational assimilation system显示文摘Variational method is capable of dealing with observations that have a complicated nonlinear relation with model variables representative of the atmospheric state, and so make it possible to directly assimilate such measured variables as satellite radiance, which have a nonlinear relation with the model variables. Assimilation of any type of observations requires a corresponding observation operator, which establishes a specific mapping from the space of the model state to the space of observation. This paper presents in detail how the direct assimilation of real satellite radiance data is implemented in the GRAPES-3DVar analysis system. It focuses on all the components of the observation operator for direct assimilation of real satellite radiance data, including a spatial interpolation operator that transforms variables from model grid points to observation locations, a physical transformation from model variables to observed elements with different choices of model variables, and a data quality control. Assimilation experiments, using satellite radiances such as NOAA17 AMSU-A and AMSU-B (Advanced Microwave Sounding Unit), are carried out with two different schemes. 6Angiogenesis in a 3D model containing adipose tissue stem cells and endothelial cells is mediated by canonical Wnt signaling显示文摘Adipose-derived stromal cells(ASCs) have gained great attention in regenerative medicine. Progress in our understanding of adult neovascularization further suggests the potential of ASCs in promoting vascular regeneration, although the specific cues that stimulate their angiogenic behavior remain controversial. In this study, we established a three-dimensional(3D) angiogenesis model by co-culturing ASCs and endothelial cells(ECs) in collagen gel and found that ASC-EC-instructed angiogenesis was regulated by the canonical Wnt pathway. Furthermore, the angiogenesis that occurred in implants collected after injections of our collagen gelbased 3D angiogenesis model into nude mice was confirmed to be functional and also regulated by the canonical Wnt pathway. Wnt regulation of angiogenesis involving changes in vessel length, vessel density,vessel sprout, and connection numbers occurred in our system. Wnt signaling was then shown to regulate ASCmediated paracrine signaling during angiogenesis through the nuclear translocation of β-catenin after its cytoplasmic accumulation in both ASCs and ECs. This translocation enhanced the expression of nuclear cofactor Lef-1 and cyclin D1 and activated the angiogenic transcription of vascular endothelial growth factor A(VEGFA), basic fibroblast growth factor(bF GF), and insulin-like growth factor 1(IGF-1). 7Micro-droplet Digital Polymerase Chain Reaction and Real-Time Quantitative Polymerase Chain Reaction Technologies Provide Highly Sensitive and Accurate Detection of Zika Virus显示文摘The establishment of highly sensitive diagnostic methods is critical in the early diagnosis and control of Zika virus(ZIKV)and in preventing serious neurological complications of ZIKV infection. In this study, we established micro-droplet digital polymerase chain reaction(ddPCR) and real-time quantitative PCR(RT-qPCR) protocols for the detection of ZIKV based on the amplification of the NS5 gene. For the ZIKV standard plasmid, the RT-qPCR results showed that the cycle threshold(Ct) value was linear from 10~1 to 10~8 copy/l L, with a standard curve R^2 of 0.999 and amplification efficiency of 92.203%;however, a concentration as low as 1 copy/l L could not be detected. In comparison with RT-qPCR, the dd PCR method resulted in a linear range of 10~1–10~4 copy/l L and was able to detect concentrations as low as 1 copy/l L. Thus, for detecting ZIKV from clinical samples, RT-qPCR is a better choice for high-concentration samples(above 10~1 copy/l L),while ddPCR has excellent accuracy and sensitivity for low-concentration samples. 10Chidamide combined with cyclophosphamide,doxorubicin,vincristine and prednisone in previously untreated patients with peripheral T-cell lymphoma显示文摘Objective:Chidamide is an oral histone deacetylase subtype-selective inhibitor approved for relapsed or refractory peripheral T-cell lymphoma(PTCL).This phase 1 b study evaluated the safety,pharmacokinetics,and preliminary efficacy of chidamide in combination with cyclophosphamide,doxorubicin,vincristine and prednisone(CHOP)for treatment-na?ve PTCL patients.Methods:This study was an open-label,multicenter trial composed of dose escalation and dose expansion.Patients received CHOP for six 21-d cycles and chidamide on d 1,4,8 and 11 in each cycle.Four dose levels of chidamide(20,25,30 and 35 mg)were evaluated.The primary objective was to evaluate the safety and tolerability of the combination regimen.Results:A total of 30 patients were evaluated in this study:15 in the dose-escalation part and 15 in the doseexpansion part.In the dose-escalation study,three patients were enrolled in the 35 mg chidamide cohort.One had dose-limiting toxicity with grade 3 vascular access complications,and one had grade 2 neutropenia with a sustained temperature>38°C.Dose escalation was stopped at this chidamide dose level.The most common(≥10%)grade 3 or 4 adverse events(AEs)were leukopenia(90.0%),neutropenia(83.3%),vomiting(13.3%),thrombocytopenia(10.0%)and febrile neutropenia(10.0%).No significant changes in chidamide pharmacokinetic properties were observed before and after combination treatment.The objective response rate for the 28 patients evaluable for preliminary efficacy was 89.3%(25/28),with 16(57.1%)achieving complete response or unconfirmed complete response.The estimated median progression-free survival was 14.0 months.In summary,we chose chidamide 30 mg as the recommended dose for phase 2.Conclusions:The addition of chidamide to standard CHOP chemotherapy was tolerable with promising preliminary efficacy in previously untreated PTCL patients,which supports further clinical studies with this combination regimen for the frontline treatment of PTCL.Lin Gui Junning Cao Dongmei Ji Huilai Zhang Qian Fan Jun Zhu Yuqin Song Shiyu Jiang Zhiqiang Ning Jia Yu Yuankai Shi 2021Chinese Journal of Cancer Research2021,33,5:3
11Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. 12The quality attribute of watershed ecosystem is more important than the landscape attribute in controlling erosion of red soil in southern China显示文摘Landscape and quality attributes are major ecosystem characteristics closely associated with soil conservation service(SCS).However,the intrinsic mechanisms by which these two attributes influence SCS are still unclear.Therefore,this study quantitatively analyzed the landscape pattern,ecological quality,and SCS in the Lianshui River watershed(a typical soil and water loss area of red soil in southern China)and its sub-watersheds in 2019.The boosted regression tree model was used to explore the influence of 15 factors(i.e.,landscape and quality attributes)on SCS at the sub-watershed scale.According to the results,compared with the landscape attribute,the quality attribute of the watershed ecosystem could better explain the spatial heterogeneity of SCS across 66 sub-watersheds.The overall degree of influence of five quality factors on SCS reached 57.81%,with the highest being the normalized differential build-up and bare soil index(NDBSI),at 25.11%.Among 10 landscape factors,aggregation had the greatest influence on SCS,at 28.64%.The relationships between key influencing factors and SCS were nonmonotonic and non-linear,with threshold effects.For example,NDBSI values of 0.18e0.41 had a positive influence on SCS,while NDBSI values of 0.41e0.65 had a negative influence on SCS.The findings broaden our understanding of the response of SCS to changes in landscape and quality attributes at the sub-watershed scale,and could offer comprehensive support for soil erosion management in the watershed ecosystem.Qing Zhu Xi Guo Jiaxin Guo Jun Wu Yingcong Ye Wenbo Cai Shiyu Liu 2022International Soil and Water Conservation Research2022,10,3:2
13Formation mechanisms,potentials and exploration practices of large lithologic gas reservoirs in and around an intracratonic rift:Taking the Sinian-Cambrian of Sichuan Basin as an example显示文摘By examining structures,sediments,reservoirs and accumulation assemblages in the Deyang-Anyue rift and its surrounding area,four new understandings are obtained.First,during the initiation period of Deyang-Anyue rift,multiple groups of faults developed in the rift due to the effect of tensile force,bringing about multiple mound and shoal belts controlled by horsts in the second member of the Sinian Dengying Formation;in the development stage of the rift,the boundary faults of the rift controlled the development of mound and shoal belts at the platform margin in the fourth member of Dengying Formation;during the shrinkage period of the rift,platform margin grain shoals of the Cambrian Canglangpu Formation developed in the rift margin.Second,four sets of large-scale mound and shoal reservoirs in the second member of Dengying Formation,the fourth member of Dengying Formation,Canglangpu Formation and Longwangmiao Formation overlap with several sets of source rocks such as Qiongzhusi Formation source rocks and Dengying Formation argillaceous limestone or dolomite developed inside and outside the rift,forming good source-reservoir-cap rock combinations;the sealing of tight rock layers in the lateral and updip direction results in the formation model of large lithologic gas reservoirs of oil pool before gas,continuous charging and independent preservation of each gas reservoir.Third,six favorable exploration zones of large-scale lithologic gas reservoirs have been sorted out through comprehensive evaluation,namely,mound and shoal complex controlled by horsts in the northern part of the rift in the second member of Dengying Formation,isolated karst mound and shoal complex of the fourth member of Dengying Formation in the south of the rift,the superimposed area of multi-stage platform margin mounds and shoals of the second and fourth members of Dengying Formation and Canglangpu Formation in the north slope area,the platform margin mounds and shoals of the second and fourth members of Dengying Formation in the west side of the rift,the platform margin mound and shoal bodies of the fourth member of Dengying Formation in the south slope area,etc.Fourth,Well Pengtan-1 drilled on the mound and shoal complex controlled by horsts of the second member of Dengying Formation in the rift and Well Jiaotan-1 drilled on the platform margin mound and shoal complex of the North Slope have obtained high-yield gas flows in multiple target layers,marking the discovery of a new gas province with reserves of(2-3)×10^(12) m^(3).This has proved the huge exploration potential of large lithologic gas reservoir group related to intracratonic rift.WEI Guoqi YANG Wei XIE Wuren SU NanX IE Zengye ZENG Fuying MA Shiyu JIN Hui WANG Zhihong ZHU Qiuying HAO Cuiguo WANG Xiaodan 2022Petroleum Exploration and Development2022,49,3:2
14Human-animal interactions and bat coronavirus spillover potential among rural residents in Southern China显示文摘Human interaction with animals has been implicated as a primary risk factor for several high impact zoonoses,including many bat-origin viral diseases.However the animal-to-human spillover events that lead to emerging diseases are rarely observed or clinically examined,and the link between specific interactions and spillover risk is poorly understood.To investigate this phenomenon,we conducted biological-behavioral surveillance among rural residents in Yunnan,Guangxi,and Guangdong districts of Southern China,where we have identified a number of SARS-related coronaviruses in bats.Serum samples were tested for four bat-borne coronaviruses using newly developed enzyme-linked immunosorbent assays(ELISA).Survey data were used to characterize associations between human-animal contact and bat coronavirus spillover risk.A total of 1,596 residents were enrolled in the study from 2015 to 2017.Nine participants(0.6%)tested positive for bat coronaviruses.265(17%)participants reported severe acute respiratory infections(SARI)and/or influenza-like illness(ILI)symptoms in the past year,which were associated with poultry,carnivore,rodent/shrew,or bat contact,with variability by family income and district of residence.This study provides serological evidence of bat coronavirus spillover in rural communities in Southern China.The low seroprevalence observed in this study suggests that bat coronavirus spillover is a rare event.Nonetheless,this study highlights associations between human-animal interaction and zoonotic spillover risk.These findings can be used to support targeted biological behavioral surveillance in high-risk geographic areas in order to reduce the risk of zoonotic disease emergence.Li Hongying Mendelsohn Emma Zong Chen Zhang Wei Hagan Emily Wang Ning Li Shiyue Yan Hong Huang Huimin Zhu Guangjian Ross Noam Chmura Aleksei Terry Philip Fielder Mark Miller Maureen Shi Zhengli Daszak Peter 2019Biosafety and Health2019,1,2:2

16Coaxing Polysaccharide Granules into Supramolecular Biocolloidal Additives for Papermaking显示文摘In light of developments in polysaccharide-based sustainable processes involving supramolecular interactions,we herein present our findings pertaining to coaxing polysaccharide granules into functional supramolecular biocolloids.Translucent biocolloidal dispersions containing various forms of starch are facilely designable,essentially built upon complexation between disassembled native cornstarch granules and amphiphilic ligands.Oily moieties of guest molecules are dynamically attractable into cavities of helical structures,with cationic groups pointing toward the bulk phase.This noncovalent attraction can generate core-shell biocolloidal particles.The significantly higher gelatinizability of freeze-dried biocolloids in contrast to native cornstarch granules is attributable to complex formation,and a homogenous dispersion is readily formable at room temperature.Our results also show biocolloids'ligand-related antibacterial activity.The use of biocolloids as wet-end additives for biofiber assemblies(cellulosic paper)can enhance mechanical strength,fines retention,and filler bondability.Supramolecular biocolloids with positively charged,translucent,easily gelatinizable,antibacterial,and polysaccharide-bondable functionalities would find tailorable use in the paper industry.Yongsheng Wang Wenyuan Zhu Xueren Qian Meiyun Zhang Shunxi Song Chuyan Jing Zhengren Meng Shiyu Xu Xianhui An Xiujie Huang Jing Shen 2020Paper And Biomaterials2020,5,2:1
17Evaluation of the Mechanisms Underlying Amino Acid and Microbiota Interactions in Intestinal Infections Using Germ-Free Animals显示文摘Intestinal infectious diseases refer to the inflammatory changes in the intestinal tract caused by pathogens(including bacteria,viruses,fungi,protozoa,or parasites)or their toxic products.A large number of microorganisms colonize the intestinal tract of healthy people,which together with the intestinal epithelium constitute the biological barrier of the intestinal tract to resist infectious diseases.As an“invisible organ,”the intestinal flora is closely related to human nutrition metabolism and intestinal infections.A variety of intestinal flora participates in the nutritional metabolism of amino acids,and the small molecular substances produced by the amino acid metabolism through the intestinal flora can enhance intestinal immunity and resist bacterial infections.In turn,amino acids can also regulate the composition of the intestinal flora,maintain the steady-state of the intestinal flora,protect the intestinal barrier,and inhibit colonization by pathogenic bacteria.As a model animal with a clear microbial background,germ-free(GF)animals can clarify the mechanisms of interactions between intestinal microbes and amino acid metabolism in intestinal infections by combining genetic engineering technology and multi-omics studies.This article reviews related researches on the involvement of intestinal microbes in host amino acid metabolism and resistance to intestinal infections and discusses the advantages of GF animal models for studying the underlying mechanisms.The GF animal model is helpful to further study the intervention effects of amino acid metabolism of targeted intestinal flora on intestinal infections.Yapeng Yang Peng Bin Shiyu Tao Guoqiang Zhu Zhifeng Wu Wei Cheng Wenkai Ren Hong Wei 2021Infectious Microbes & Diseases2021,3,2:1

19Perivascular localized cells commit erythropoiesis in PDGF-B-expressing solid tumors显示文摘Background:Tumors possess incessant growth features,and expansion of their masses demands sufficient oxygen supply by red blood cells(RBCs).In adult mammals,the bone marrow(BM)is the main organ regulating hematopoiesis with dedicated manners.Other than BM,extramedullary hematopoiesis is discovered in various pathophysiological settings.However,whether tumors can contribute to hematopoiesis is completely unknown.Accumulating evidence shows that,in the tumor microenvironment(TME),perivascular localized cells retain progenitor cell properties and can differentiate into other cells.Here,we sought to better understand whether and how perivascular localized pericytes in tumors manipulate hematopoiesis.Methods:To test if vascular cells can differentiate into RBCs,genome-wide expression profiling was performed using mouse-derived pericytes.Genetic tracing of perivascular localized cells employing NG2-CreERT2:R26R-tdTomato mouse strain was used to validate the findings in vivo.Fluorescence-activated cell sorting(FACS),single-cell sequencing,and colony formation assays were applied for biological studies.The production of erythroid differentiationspecific cytokine,erythropoietin(EPO),in TME was checked using quantitative polymerase chain reaction(qPCR),enzyme-linked immunosorbent assay(ELISA,magnetic-activated cell sorting and immunohistochemistry.To investigate BM function in tumor erythropoiesis,BM transplantation mouse models were employed.Results:Genome-wide expression profiling showed that in response to plateletderived growth factor subunit B(PDGF-B),neural/glial antigen 2(NG2)+perivascular localized cells exhibited hematopoietic stem and progenitor-like features and underwent differentiation towards the erythroid lineage.PDGF-B simultaneously targeted cancer-associated fibroblasts to produce high levels of EPO,a crucial hormone that necessitates erythropoiesis.FACS analysis using genetic tracing of NG2+cells in tumors defined the perivascular localized cell-derived subpopulation of hematopoietic cells.Single-cell sequencing and colony formation assays validated the fact that,upon PDGF-B stimulation,NG2+cells isolated from tumors acted as erythroblast progenitor cells,which were distinctive from the canonical BM hematopoietic stem cells.Conclusions:Our data provide a new concept of hematopoiesis within tumor tissues and novel mechanistic insights into perivascular localized cell-derived erythroid cells within TME.Targeting tumor hematopoiesis is a novel therapeutic concept for treating various cancers that may have profound impacts on cancer therapy.Kayoko Hosaka Chenchen Wang Shiyue Zhang Xue Lv Takahiro Seki Yin Zhang Xu Jing Jieyu Wu Qiqiao Du Xingkang He Yulong Fan Xuan Li Makoto Kondo Masahito Yoshihara Hong Qian Lihong Shi Ping Zhu Yuanfu Xu Yunlong Yang Tao Cheng Yihai Cao 2023Cancer Communications2023,43,6:1
