| 1 | Decrease in Serum Amyloid a Protein Levels Following Three-month Stays in Negatively Charged Particle-dominant Indoor Air Conditions显示文摘Objective The changes in serum adipokines and cytokines related to oxidative stress were examined during 3 months ‘Off to On' and ‘On to Off' periods using negatively charged particle-dominant indoor air conditions(NCPDIAC).Methods Seven volunteers participated in the study,which included ‘OFF to 3 months ON' periods(ON trials) for a total of 16 times,and ‘ON to 3 months OFF'(OFF trials) periods for a total of 13 times.Results With the exception of one case,serum amyloid A(SAA) levels decreased significantly during the ON trials.Conclusion Considering that SAA is an acute phase reactive protein such as C reactive protein(CRP),this observed decrease might indicate the prevention of cardiovascular and atherosclerotic changes,since an increase in high-sensitive CRP is associated with the subsequent detection of these events. | Suni Lee Shoko Yamamoto Yasumitsu Nishimura Hidenori Matsuzaki Kei Yoshitome Tamayo Hatayama Miho Ikeda YU Min Nagisa Sada Naoko Kumagai-Takei Takemi Otsuki | 2018 | Biomedical and Environmental Sciences2018，31，5： | 5 |

| 6 | Relationships of early esophageal cancer with human papillomavirus and alcohol metabolism显示文摘BACKGROUND It is well known that an alcohol consumption habit together with inactive heterozygous aldehyde dehydrogenase-2(ALDH2)is an important risk factor for the development of esophageal squamous cell carcinoma(ESCC).It remains controversial whether human papillomavirus(HPV)infection contributes to the occurrence/development of ESCC.There has been no study in which the relationship between ESCC and HPV in addition to alcohol dehydrogenase-1B(ADH1B)and ALDH2 genotypes was evaluated.AIM To evaluate relationships between HPV infection and development of esophageal cancer,particularly early esophageal cancer,based on ADH1B/ALDH2 polymorphisms.METHODS We conducted an exploratory retrospective study using new specimens,and we enrolled 145 patients who underwent endoscopic resection for superficial ESCC and had been observed for more than two years by both physical examination and endoscopic examination in Hokkaido University Hospital.Saliva was collected to analyze genetic polymorphisms of ADH1B/ALDH2.We performed in situ hybridization for resected specimens to detect HPV by using an HPV type 16/18 probe.RESULTS HPV was detected in 15(10.3%)of the 145 patients with ESCC.HPV-positive rates in inactive ALDH2*1/*2 and ALDH2*1/*1+*2/*2 were 10.8%and 9.8%,respectively(P=1.00).HPV-positive rates in slow-metabolizing ADH1B*1/*1 and ADH1B*1/*2+*2/*2 were 12.0%and 10.0%,respectively(P=0.72).HPV-positive rates in the heavy or moderate alcohol consumption group and the light or rare consumption group were 11.1%and 8.7%,respectively(P=0.68).HPV-positive rates in the heavy smoking group and the light or no smoking group were 11.8%and 8.3%,respectively(P=0.59).The 3-year incidence rates of secondary ESCC or head and neck cancer after initial treatment in the HPV-positive and HPVnegative groups were 14.4%and 21.4%(P=0.22),respectively.CONCLUSION In the present situation,HPV status is considered to be less important than other risk factors,such as alcohol consumption,smoking habit,ADH1B/ALDH2 polymorphisms,and HPV status would therefore have no effect on ESCC risk management. | Masaki Inoue Yuichi Shimizu Marin Ishikawa Satoshi Abiko Yoshihiko Shimoda Ikko Tanaka Sayoko Kinowaki Masayoshi Ono Keiko Yamamoto Shoko Ono Naoya Sakamoto | 2020 | World Journal of Gastroenterology2020，26，39： | 2 |

| 8 | ERK phosphorylation functions in invadopodia formation in tongue cancer cells in a novel silicate fibre-based 3D cell culture system显示文摘To screen for additional treatment targets against tongue cancer, we evaluated the contributions of extracellular signal-related kinase(ERK), AKT and ezrin in cancer development. Immunohistochemical staining showed that ERK and ezrin expressions were significantly higher in invasive squamous cell carcinoma than in carcinoma in situ. To investigate the roles of ERK and ezrin in cancer development, we used the non-woven silica fibre sheet Cellbed^(TM)with a structure resembling the loose connective tissue morphology in a novel 3 D culture system. We confirmed that the 3 D system using CellbedTMaccurately mimicked cancer cell morphology in vivo. Furthermore, cell projections were much more apparent in 3 D-cultured tongue cancer cell lines than in 2 D cultures. Typically, under conventional 2 D culture conditions, F-actin and cortactin are colocalized in the form of puncta within cells.However, in the 3 D-cultured cells, colocalization was mainly observed at the cell margins, including the projections. Projections containing F-actin and cortactin colocalization were predicted to be invadopodia. Although suppressing ezrin expression with small interfering RNA transfection caused no marked changes in morphology, cell projection formation was decreased, and the tumour thickness in vertical sections after 3 D culture was markedly decreased after suppressing ERK activity because both the invasion ability and proliferation were inhibited. An association between cortactin activation as well as ERK activity and invadopodia formation was detected. Our novel 3 D culture systems using Cellbed? are simple and useful for in vitro studies before conducting animal experiments. | 14 | Biological Effects of Cloth Containing Specific Ore Powder in Patients with Pollen Allergy显示文摘Objective The custom-homebuilding company,Cosmic Garden Co.Ltd.,located in Okayama City,Japan was established in 1997 and uses specific natural ore powder(SNOP) in wall materials and surveys customers in order to improve allergic symptoms.Methods To investigate the biological effects of SNOP,patients with a pollen allergy were recruited to stay in a room surrounded by cloth containing SNOP(CCSNOP),and their symptoms and various biological parameters were compared with those of individuals staying in a room surrounded by control non-woven cloth(NWC).Each stay lasted 60 min.Before and immediately after the stay,a questionnaire regarding allergic symptoms,as well as POMS(Profile of Mood Status) and blood sampling,was performed.Post-stay minus pre-stay values were calculated and compared between CCSNOP and NWC groups.Results Results indicated that some symptoms,such as nasal obstruction and lacrimation,improved,and POMS evaluation showed that patients were calmer following a stay in CCSNOP.Relative eosinophils,non-specific Ig E,epidermal growth factor,monocyte chemotactic protein-1,and tumor necrosis factor-α increased following a stay in CCSNOP.Conclusion This ore powder improved allergic symptoms,and long-term monitoring involving 1 to 2 months may be necessary to fully explore the biological and physical effects of SNOP on allergic patients. | Suni LEE Hitoshi OKAMOTO Shoko YAMAMOTO Tamayo HATAYAMA Hidenori MATSUZAKI Naoko KUMAGAI-TAKEI Kei YOSHI-TOME Yasumitsu N1SHIMURA Toshiaki SATO Yasuzo KIRITA Yoshio FUJII Takemi OTSUKI | 2016 | Biomedical and Environmental Sciences2016，29，8： | 0 |
