5Aerodynamic optimization of an adaptive flap for next-generation green aircraft显示文摘Adaptive,morphing flaps are taking ever-increasing attention in civil aviation thanks to the expected benefits this technology can bring at the aircraft level in terms of high-lift performance improvement and related fuel burnt reduction per flight.Relying upon morphing capabilities,it is possible to fix a unique setting for the flap and adapt the flap shape to match the aerodynamic requirements for take-off or landing.The proper morphed shapes can assure better high-lift performances than those achievable by referring to a conventional flap.Moreover,standing the unique flap setting for take-off and landing,a dramatic simplification of the flap deployment systems may be achieved.As a consequence of this simplification,the deployment system can be fully hosted in the wing,thus avoiding under-wing nacelles with significantly better aerodynamics and fuel consumption.The first step for a rational design of an adaptive flap consists in defining the target morphed shapes and the unique optimal flap setting in the take-off and landing phases.In this work,aerodynamic optimization analyses are carried out to determine the best flap setting and related morphed shapes in compliance with the take-off and landing requirements of a reference civil transport aircraft.Four different initial conditions are adopted to avoid the optimization falling into local optima,thus obtaining four groups of optimal candidate configurations.After comparing each candidate’s performance through 2D and 3D simulations,the optimal configuration has been selected.2D simulations show that the optimal configuration is characterized by a maximum lift increase of 31.92%in take-off and 9.04%in landing.According to 3D simulations,the rise in maximum lift equals 22.26%in take-off and 3.50%in landing.Numerical results are finally verified through wind tunnel tests,and the aerodynamic mechanism behind the obtained improvements is explained by carefully analyzing the flow field around the flap.Tianlong LIN Rosario PECORA Danilo CILIBERTI Wei XIA Shuling HU 2024Chinese Journal of Aeronautics2024,37,2:0
6Comparative Analysis of Social Forces Participating in Emergency Management of Public Health Events in China, Australia, Germany and the United States显示文摘Objective To provide a reference and suggestions for Chinese social forces to participate in the emergency management of public health events.Methods Through literature research method,comparative research method and SWOT matrix analysis,four aspects of prevention,early warning,response and recovery of the social forces in China,Germany,Australia and the United States participating in emergency management were studied comparatively.Results and Conclusion In the emergency management of public health events,China’s social forces play greater role in the response and recovery phase than that in the prevention and response phase.As to its shortcomings,the following suggestions are made,such as incorporating social forces into the national emergency management system,strengthening the construction and training of social forces,popularizing public health knowledge and awareness,clarifying property rights and using incentives and punishment together.Wang Shuling Dou Lele Lin Xiangpeng Liu Liang Chen Hui 2022Asian Journal of Social Pharmacy2022,17,2:0
7Structural insights into dimerization and activation of the mGlu2-mGlu3 and mGlu2-mGlu4 heterodimers显示文摘Nicotinamide adenine dinucleotide(NAD+)is a central metabolite in cellular processes.Depletion of NAD+has been demonstrated to be a prevalent theme in both prokaryotic and eukaryotic immune responses.Short prokaryotic Argonaute proteins(Agos)are associated with NADase domain-containing proteins(TIR-APAZ or SIR2-APAZ)encoded in the same operon.They confer immunity against mobile genetic elements,such as bacteriophages and plasmids,by inducing NAD+depletion upon recognition of target nucleic acids.However,the molecular mechanisms underlying the activation of such prokaryotic NADase/Ago immune systems remain unknown.Here,we report multiple cryo-EM structures of NADase/Ago complexes from two distinct systems(TIR-APAZ/Ago and SIR2-APAZ/Ago).Target DNA binding triggers tetramerization of the TIR-APAZ/Ago complex by a cooperative self-assembly mechanism,while the heterodimeric SIR2-APAZ/Ago complex does not assemble into higher-order oligomers upon target DNA binding.However,the NADase activities of these two systems are unleashed via a similar closed-to-open transition of the catalytic pocket,albeit by different mechanisms.Furthermore,a functionally conserved sensor loop is employed to inspect the guide RNA–target DNA base pairing and facilitate the conformational rearrangement of Ago proteins required for the activation of these two systems.Overall,our study reveals the mechanistic diversity and similarity of Ago protein-associated NADase systems in prokaryotic immune response.Xinwei Wang Mu Wang Tuo Xu Ye Feng Qiang Shao Shuo Han Xiaojing Chu Yechun Xu Shuling Lin Qiang Zhao Beili Wu 2023Cell Research2023,33,10:0
8Intratumoral CD103+CD8+T cells predict response to neoadjuvant chemoimmunotherapy in advanced head and neck squamous cell carcinoma显示文摘Background Immune cell heterogenicity is known to determine the therapeutic response to cancer progression.Neoadjuvant chemoimmunotherapy(NACI)has shown clinical benefits in some patients with advanced head and neck squamous cell carcinoma(HNSCC),but the underlying mechanism behind this clinical response is unknown.The efficacy of NACI needs to be potentiated by identifying accurate biomarkers to predict clinical responses.Here,we attempted to identify molecules predicting NACI response in advanced HNSCC.Methods We performed combined single-cell RNA sequencing(scRNA-seq)and multiplex immunofluorescence(mIHC)staining with tumor samples derived from NACI-treated HNSCC patients to identify a new tumor-infiltrating cell(TIL)subtype,CD103+CD8+TILs,associated with clinical response,while both in vitro and in vivo assays were carried out to determine its antitumor efficiency.The regulatory mechanism of the CD103+CD8+TILs population was examined by performing cell-cell interaction analysis of the scRNA-seq data and spatial analysis of the mIHC images.Results We established intratumoral CD103+CD8+TILs density as a determinant of NACI efficacy in cancers.Our scRNA-seq results indicated that the population of CD103+CD8+TILs was dramatically increased in the responders of NACI-treated HNSCC patients,while mIHC analysis confirmed the correlation between intratumoral CD103+CD8+TILs density and NACI efficacy in HNSCC patients.Further receiver operating characteristic curve analysis defined this TIL subset as a potent marker to predict patient response to NACI.Functional assays showed that CD103+CD8+TILs were tumor-reactive T cells,while programmed cell death protein-1(PD-1)blockade enhanced CD103+CD8+TILs cytotoxicity against tumor growth in vivo.Mechanistically,targeting the triggering receptor expressed on myeloid cells 2-positive(TREM2+)macrophages might enhance the population of CD103+CD8+TILs and facilitate antitumor immunity during NACI treatment.Conclusions Our study highlights the impact of intratumoral CD103+CD8+TILs density on NACI efficacy in different cancers,while the efforts to elevate its population warrant further clinical investigation.Siqi Ren Tianjun Lan Fan Wu Suling Chen Xue Jiang Chuying Huo Zitian Li Shule Xie Donghui Wu Ruixin Wang Yanyan Li Lin Qiu Guoxin Huang Shurui Li Xiaojuan Wang Meifeng Cen Tingting Cai Zhaoyu Lin Jinsong Li Bowen Li 2023Cancer Communications2023,43,10:0
9Reflections on the Participation of Social Organizations in the Prevention of COVID-19显示文摘Objective To study the problems of Chinese social organizations in participating in the prevention and control of the COVID-19 pandemic so that we can standardize the participation mechanism,improve the collaborative management mechanism,and promote the development of social organizations.Methods Through the analysis of CNKI,news reports,industry conferences and other data,combined with the current situation,the participation of social organizations in public health emergencies were investigated.Results and Conclusion The coordination between government and social organizations is not good.Social organizations in the field of public health are unable to stand out in a timely manner.The public do not have self-management ability.Some of the media spread rumors.Besides,most social organizations are highly dependent on the government and they are ack supervision.Social organizations should be incorporated into the emergency management system and national hub-based social organizations must be established.In addition,supervision should be strengthened to increase the credibility of social organizations.The responsibilities of community and emergency management at the grassroots level should be enhanced.The public opinion response and disposal system should be improved.Lastly,the responsibility of social organizations in the field of public health must be fulfilled.Liao Wenjing Lin Xiangpeng Liu Liang Dou Lele Wang Shuling 2022Asian Journal of Social Pharmacy2022,17,4:0
