| 1 | CPSF30-L-mediated recognition of mRNA m^(6)A modification controls alternative polyadenylation of nitrate signaling-related gene transcripts in Arabidopsis显示文摘N6-methyladenosine(m^(6)A),a ubiquitous internal modification of eukaryotic mRNAs,plays a vital role in almost every aspect of mRNA metabolism.However,there is little evidence documenting the role of m^(6)A in regulating alternative polyadenylation(APA)in plants.APA is controlled by a large protein-RNA complex with many components,including CLEAVAGE AND POLYADENYLATION SPECIFICITY FACTOR30(CPSF30).In Arabidopsis,CPSF30 has two isoforms and the longer isoform(CPSF30-L)contains a YT512-B Homology(YTH)domain,which is unique to plants.In this study,we showed that CPSF30-L YTH domain binds to m^(6)A in v itro.In the cpsf30-2 mutant,the transcripts of many genes including several important nitrate signaling-related genes had shifts in polyadenylation sites that were correlated with m^(6)A peaks,indicating that these gene transcripts carrying m^(6)A tend to be regulated by APA.Wild-type CPSF30-L could rescue the defects in APA and nitrate metabolism in cpsf30-2,but m^(6)A-binding-defective mutants of CPSF30-L could not.Taken together,our results demonstrated that m^(6)A modification regulates APA in Arabidops is and revealed that the m^(6)A reader CPSF30-L affects nitrate signaling by controlling APA,shedding new light on the roles of the m^(6)A modification during RNA 3-end processing in nitrate metabolism. | Yiteng Hou Jing Sun Baixing Wu Yangyang Gao Hongbo Nie Zhentian Nie Shuxuan Quan Yong Wang Xiaofeng Cao Sisi Li | 2021 | Molecular Plant2021,14,4: | 12 |
| 2 | Primary structure of helix C to helix G of a new retinal protein in H.sp.xz515显示文摘The gene fragment encoding the retinal protein from helix C to helix G in a new strain of halobacteria, H.sp.xz515 has been amplified by PCR method. The nucleotide sequence of this fragment has been determined. The deduced amino acid sequence has been compared with halobium br and other two br-like proteins, ar-1 and ar-2. Results show that those amino acid residues in br, essential for proton pumping and binding to retinal, are conserved. | 3 | Peripheral Lymphocyte Subsets as a Marker of Parkinson's Disease in a Chinese Population显示文摘In this study, we conducted a clinical analysis of lymphocyte subtypes in 268 patients with Parkinson's disease(PD) to assess their clinical impact as a potential marker of advanced PD in Chinese patients. The participants comprised 268 sporadic PD patients and 268 healthy controls. The numbers of natural killer(NK) cells and CD3+, CD3+CD4+, CD3+CD8+, and CD19+ lymphocytes from peripheral blood were determined by immunostaining and flow cytometric analysis and the percentages of these CD+ T cells were calculated. The ratio of regulatory T(Treg)/helper T 17(Th17) lymphocytes from 64 PD patients and 46 controls was determined by flow cytometric analysis.The results showed that the percentage of NK cells was higher in advanced PD patients than in controls(22.92% ±10.08% versus 19.76% ± 10.09%, P = 0.006), while CD3+ T cells are decreased(62.93% ± 9.27% versus65.75% ± 9.13%, P = 0.005). The percentage of CD19+B cells in male patients was lower(P = 0.021) than in female patients, whereas NK cells were increased(P \ 0.0001). The scores on the Unified Parkinson's Disease Rating Scale(UPDRS) and the Non-Motor Symptoms Scale in late-onset PD patients were significantly higher than those in earlyonset patients(P = 0.024 and P = 0.007, respectively). The percentage of CD19+ B cells in patients with UPDRS scores[24 was lower than in those with scores \24(10.17% ±4.19% versus 12.22% ± 5.39%, P = 0.009). In addition, the Treg/Th17 ratio in female patients was higher than that in female controls(13.88 ± 6.32 versus 9.94 ± 4.06, P =0.042). | 4 | Multi-functional vesicles improve Helicobacter pylori eradication by a comprehensive strategy based on complex pathological microenvironment显示文摘Helicobacter pylori(H.pylori),creating a global infection rate over 50%,presents great challenges in clinical therapies due to its complex pathological microenvironment in vivo.To improve the eradication efficacy,herein we fabricated a pharmaceutical vesicle RHL/Cl-Ch-cal where cholesterol-PEG,calcitriol and first-line antibiotic clarithromycin were co-loaded in the rhamnolipid-composed outer lipid layer.RHL/Cl-Ch-cal could quickly penetrate through gastric mucus layer to reach H.pylori infection sites,and then effectively destroyed the architecture of H.pylori biofilms,killed dispersed H.pylori and inhibited the re-adhesion of residual bacteria(called biofilms eradication tetralogy).Moreover,RHL/Cl-Ch-cal activated the host immune response to H.pylori by replenishing cholesterol to repair lipid raft on the cell membrane of host epithelial cells.Finally,RHL/Cl-Ch-cal killed the intracellular H.pylori through recovering the lysosomal acidification and assisting degradation.In experiments,RHL/Cl-Ch-cal demonstrated prominent anti-H.pylori efficacy in the classical H.pylori-infected mice model.Therefore,the study provides a“comprehensive attack”strategy for anti-H.pylori therapies including biofilms eradication tetralogy,immune activation and intracellular bacteria killing. | Xiaonan Chen Yiqing Zou Shuqi Zhang Pengchao Fang Shuxuan Li Pengyu Li Yingying Sun Gang Yuan Haiyan Hu | 2022 | Acta Pharmaceutica Sinica B2022,12,9: | 2 |
| 5 | Multi-scale variation characteristics of polar vortex at 10 hPa in the Southern Hemisphere显示文摘By use of 1948-2007 NCEP/NCAR reanalysis monthly geopotential data, a set of circulation indices are defined to characterize the polar vortex at 10 hPa in the Southern Hemisphere, including area-(S), intensity-(P) and centre position-(λc , φc) indices. Sea-sonal variation, interannual anomalies and their possible causes of 10 hPa polar vortex in the Southern Hemisphere are analyzed by using these indices, the relationship between 10 hPa polar vortex strength and the Antarctic Oscillation are analyzed as well. The results show that: (1) the polar region at 10 hPa in the Southern Hemisphere is controlled by anticyclone (cyclone) from Dec. to Jan. (from Mar. to Oct.), Feb. and Nov. are circulation transition seasons. (2) Intensity index (P) and area index (S) of anticy-clone (cyclone) in Jan. (Jul.) show a significant spike in the late 1970s, the anticyclone (cyclone) enhances (weakens) from ex-tremely weak (strong) oscillation to near the climatic mean before a spike, anticyclone tends to the mean state from very strong oscillation and cyclone oscillates in the weaker state after the spike. (3) There is significant interdecadal change for the anticyclone center in Jan., while markedly interannual variation for cyclone center in July. (4) The ozone anomalies can cause the interannual anomaly of the polar anticyclone at 10 hPa in the Southern Hemisphere in Jan. (positive correlation between them), but it is not related to the polar cyclone anomalies. | 8 | Regulating ethane and ethylene synthesis by proton corridor microenvironment for CO_(2) electrolysis显示文摘Electrocatalytic reduction of carbon dioxide is one of the most effective strategies to achieve carbon neutrality and energy sustainability.Although high-value multi-carbon products have been widely studied,limited electrocatalysts have been reported for the selective conversion of ethane.More importantly,the factors tuning the selectivity between ethane and ethylene have not been clarified.Here,Zn@Cu nanowire arrays(Zn@Cu-NWAs) catalyst is proposed to stimulate the maintenance of efficient CO_(2)-to-C_(2)H_(6) conversion at high current densities.Meanwhile,in order to investigate the factors affecting the interconversion between ethane and ethylene,the counterpart catalyst that facilitates C–C coupling to ethylene was also synthesized.Time-of-flight secondary-ion mass spectroscopy(TOF-SIMS),in-situ Raman spectroscopy,and simulation results show that Zn@Cu-NWAs can provide a localized proton corridor environment for the formation of ethane,accelerating the further proton-coupled CO_(2) reduction reaction(CO_(2)RR)kinetics.Hence,this catalyst delivered an ethane Faraday efficiency of over 65% at-1.14 V vs.RHE with a total current density of 142.3 mA/cm^(2).This work provides a new perspective on regulating the local microenvironment to modify the selectivity of multi-carbon products. | Xiaowen Zhang Bohua Ren Hao Li Shuxuan Liu Haoyang Xiong Silong Dong Yifan Li Dan Luo Yi Cui Guobin Wen Xin Wang | 2023 | Journal of Energy Chemistry2023,,12: | 0 |

| 11 | A multifunctional cholesterol-free liposomal platform based on protopanaxadiol for alopecia therapy显示文摘Liposome could form a long-term drug reservoir in the skin for sustained drug release,which is beneficial to improving efficacy and alleviating adverse effects.Thus,it has become a better option for anti-alopecia drugs delivery.However,cholesterol used as the fluidity buffer in conventional liposomes is a precursor for testosterone biosynthesis,which could convert to dihydrotestosterone,resulting in hair follicle damage and potentiating hair loss.To overcome the limitations,in this study we prepared a cholesterol-free liposome(PPD-Lip)using protopanaxadiol(PPD)instead of cholesterol to avoid the biosynthesis of testosterone which is adverse to alopecia therapy.PPD-Lip also worked as an active ingredient to facilitate hair growth by promoting dermal papilla cells proliferation and migration,upregulating mRNA levels of hair growth-related positive regulators,and accelerating angiogenesis in vitro.Meanwhile,it promoted hair regrowth in telogen and androgenetic alopecia mice models in vivo.In addition,our study showed that as the liposomal vehicle,PPD-Lip loaded with dutasteride exerted a stronger efficacy in the treatment of androgenetic alopecia and such a strategy could extend to other anti-alopecia agents.To the best of our knowledge,being easy for clinical transformation,the PPD-based liposomal delivery system provides a promising and multifunctional alternative platform for the delivery of alopecia treatment agents. | Xuefei Zhang Shuxuan Li Yating Dong Hehui Rong Junke Zhao Haiyan Hu | 2022 | Nano Research2022,15,10: | 0 |

| 14 | Expression and analysis of zinc fi nger family gene in Lenzites gibbosa显示文摘Zinc finger transcription factors play significant roles in the growth and development of plant and animal,but their function remains obscure in fungi.Lenzites gibbosa mycelia were extracted and sequenced by transcriptome analysis after growing on sawdust at different times to support mycelial growth of L.gibbosa in a nutrient matrix.Data bases used for analysis were the Kyoto encyclopedia of genes and genomes(KEGG)annotation,the cluster of orthologous groups of proteins(COG)and gene ontology(GO)annotation.Zinc finger class genes related to the growth and development of L.gibbosa were screened.GO annotation and enrichment analysis of diff erentially expressed genes were carried out.A total of 114.55 Gb Clean Data were obtained from the L.gibbosa transcriptome.The average Clean Data in each sample was 6.16 Gb.The relative efficiency of reads between each sample and the reference genome was 88.5%to 91.4%.The COG analysis showed that most zinc finger protein genes were related to replication,recombination and repair function.GO enrichment analysis showed that the expressed genes involved in cellular process,cell part and binding.We identifi ed seventy-two expressed genes including seven up-regulated genes and sixty-five down-regulated genes by applying DESeq2 data analysis software.By comparing the significantly expressed genes with KEGG database,66 annotated sequences were obtained,and 35 primary metabolic pathways were annotated.Pathway enrichment analysis showed that differentially expressed genes were signifi cantly enriched in protein processing in endoplasmic reticulum and ubiquitin-mediated proteolysis pathways.Gene_11750 and gene_5266 are highly correlated with the growth and development of L.gibbosa and are closely related to protein processing in endoplasmic reticulum and ubiquitin-mediated proteolysis pathway.According to gene functional analysis,seven important differentially expressed genes related to the growth and development of L.gibbosa were identified. | Jun Zhang Yujie Chi Shuxuan Li Jian Zhang Jie Chen | 2020 | Journal of Forestry Research2020,31,5: | 0 |
