Joint utilization of genetic analysis and semi-cloning technology reveals a digenic etiology of Müllerian anomalies显示文摘Dear Editor,Identifying pathogenic gene mutations and their combination is critical but challenging in dissecting the etiology of complex diseases when more than one gene is involved.1 The digenic/oligogenic/omnigenic models, holding that more than one gene could act synergistically, appeal to a wide range of genes responsible for complex phenotypes.1,2,3,4 These genetic models advanced our understanding of genetic factors underlying complex phenotypes, yet an accordingly rapid and efficient experimental assay for identifying pathogenic combinations of genetic variants at animal model level is lacking and urgently needed. | Lingbo Wang Ying Zhang Xiaoyi Fu Shuangshuang Dong Shuyan Tang Ning Zhang Chengcheng Song Nan Yang Lin Zhang Hongyan Wang Huijuan Shi Li Jin Feng Zhang Jinsong Li Keqin Hua | 2020 | Cell Research2020,30,1: | 8
Inducible regulation of GDNF expression in human neural stem cells显示文摘Glial cell derived neurotrophic factor (GDNF) holds promises for treating neurodegenerative diseases such as Parkinson's disease. Human neural stem cells (hNSCs) have proved to be a suitable cell delivery vehicle for the safe and efficient introduction of GDNF into the brain. In this study, we used hNSCs-infected with a lentivirus encoding GDNF and the hygromycin resistance gene as such vehicles. A modified tetracycline operator 7 (tetO7) was inserted into a region upstream of the EF1- promoter to drive GDNF expression. After hygromycin selection, hNSCs were infected with a lentivirus encoding a KRAB-tetracycline repressor fusion protein (TTS). TTS bound to tetO7 and suppressed the expression of GDNF in hNSCs. Upon administration of doxycycline (Dox) the TTS-tetO7 complex separated and the expression of GDNF resumed. The hNSCs infected with GDNF expressed the neural stem cell specific markers, nestin and sox2, and exhibited no significant change in proliferation rate. However, the rate of apoptosis in hNSCs expressing GDNF was lower compared with normal NSCs in response to actinomycin treatment. Furthermore, a higher percentage of Tuj-1 positive cells were obtained from GDNF-producing NSCs under conditions that induced differentiation compared to control NSCs. The inducible expression of GDNF in hNSCs may provide a system for the controllable delivery of GDNF in patients with neurodegenerative diseases. | WANG ShuYan REN Ping GUAN YunQian ZOU ChunLin FU LinLin ZHANG Yu | 2013 | Science China(Life Sciences)2013,56,1: | 5

Ultra-long room temperature phosphorescence of indium-based organic inorganic metal halides for naked-eye-visible afterglow显示文摘Solid-state molecules based on room-temperature phosphorescent(RTP)emission have received extensive attention due to their special optical properties of triplet excitons. However, there are still few solid molecular systems with naked-eye-visible afterglow characteristics.Herein,we introduce 4-phenylbenzylamine(namely PBA)with a long conjugated system into common non-toxic In^(3+)to form an indium-based organic inorganic halide, whose chemical formula is PBA_(3)[InCl_6]·H_(2)O.Interestingly,this hybrid halide generates a RTP emission at 617 nm with a lifetime decay as long as 290.4 ms,expressing a naked-eye-visible afterglow for more than 7 s.The mechanism study shows that the long lifetime RTP originated from the specific lamellar stacking of organic molecules and metal halide units, facilitating the interaction between the inorganic layers and organic layers. Therefore, the material can be potentially used in emergency lighting, information security, and other fields.Meanwhile, this work provides a reference for the design and implementation of a more efficient organic-inorganic hybrid system with the ultralong RTP emission. | Heng Yu Hao Gong Zhaorui Hua Yang Zhang Wenming Sun Shuyan Gong Ganglong Cui Yang Tian Hongbing Fu | 2023 | Science China Chemistry2023,66,9: | 0
The Effect of Xu’s Influenza Decoction Combined with Oseltamivir on Influenza A:A Propensity Score Matching Study显示文摘Background:The resurgence of seasonal influenza virus circulation has been seen in 2021-2022 after the tempo-rary suppression in 2020-2021.Neuraminidase inhibitors(NAIs)are widely applied in the clinical treatment of influenza A despite several limitations. Objective:To access the efficacy of Xu’s influenza decoction(XID)in combination therapy with oseltamivir for the treatment of influenza A.Methods:In this retrospective cohort study, the eligible participants were diagnosed with influenza A between June 1,2018,and May 30,2022,in the First Affiliated Hospital of Zhejiang Chinese Medical University.According to whether Xu’s influenza decoction was applied, patients were divided into two groups:treated with or without XID.Propensity score matching(PSM)was used to further adjust the covariates between groups. The primary outcome was to compare time to defervescence via K-M curves,Breslow tests, and Cox regression analysis. In Cox proportional hazards model,a univariate analysis was performed to obtain preliminary results, while a further multivariate analysis was conducted to study the independent factors that influence defervescence.Subgroup analysis was conducted according to body temperature and time from onset to admission. The secondary outcome consisted of routine blood and C-reactive protein(CRP),length of stay, and medical costs. Results:A total of 336 patients with influenza A were enrolled in this study(i.e.,163 patients in the XID+oseltamivir group;173 patients in the oseltamivir group). After 1:1 matching via PSM,230 patients meeting the criteria were included in the analysis, with 115 in each arm.The XID+oseltamivir group had shorter time to defervescence(36 h vs 44 h,P=0.011),shorter length of stay(3 days vs 4 days,P=0.018),and higher defervescence possibility(HR=1.384,95%CI:1.054-1.818).Subgroup analysis indicated that for patients during non-window period(≥48 h)with medium-grade fever(38.1℃-39℃),the XID+oseltamivir combination therapy reduced time to defervescence(P=0.04995/0.004)with a higher defervescence possibility(HR=1.524/1.683).Meanwhile,there’s no statistical significance but observable trends of the XID+oseltamivir group in the lower medical costs(3068.07 yuan vs 3120.68 yuan),the lower neutrophils%(48.53%vs 51.00%)and the higher lymphocyte%(39.83%vs 37.72%).Conclusion:The combination of XID and oseltamivir can shorten the time to defervescence and length of stay in influenza A.Its antipyretic effect is mainly reflected in the medium-grade and non-window periods. | Tianxi Chen Shuyan Fu Fengyuan Tian Qiushuang Li Hongyu Ling Yijie Lou Jun Tang Hong Zheng | 2024 | Clinical Complementary Medicine and Pharmacology2024,4,1: | 0

Functional polymer passivatingFA_(0.85)PEA_(0.15)SnI_(3) for efficient and stable lead-free perovskite solar cells显示文摘Due to their excellent advantages such as low toxicity, superior optoelectronic properties,low-temperature fabrication, and costeffectiveness,Sn-based perovskites have become the most promising alternatives for high performance lead-free perovskite solar cells. However, the character of Sn ^(2+)is easily oxidized toSn ^(4+),causing unnecessary p-type self-doping and high leakage current.More seriously,trap-induced non-radiative recombination from rapid crystallization causes into large energy loss with a low open circuit voltage. Therefore, the Sn-based solar cells have efficiency far behind the Pb-based solar cells.Herein, the polymer poly(ethylene glycol)diacrylate(PEGDA)is used to control crystal growth and passivate the defects in FA_(0.85)PEA_(0.15)SnI_(3) thin film. This Sn-perovskite layer shows compact crystal with large grain size and reduced defects.Optimized perovskite thin film is further processed to fabricate the inverted solar cell with device structure of ITO(indium tin oxide)/PEDOT:PSS(Poly(3,4-ethylenedioxythiophene)/poly(styrenesulfonate))/FA_(0.85)PEA_(0.15)SnI_(3)/ICBA(indene-C60 bisadduct)/BCP(bathocuproine)/Ag,which shows the power conversion efficiency(PCE)of 11.45%with voltage of 0.82 V.Moreover, corresponding perovskite solar cells exhibit an enhanced stability due to PEGDA induced compressive strain in perovskite. This work could shed light on one of successful attempts to improve Sn-based solar cell efficiency for sustainable energy conversion. | Wenzhan Xu Yu Gao Miao He Shuyan Chen Hongyan Fu Guodan Wei | 2023 | Nano Research2023,16,1: | 0
