Design and development of an oral remdesivir derivative W116 against SARS-CoV-2显示文摘Dear Editor,Since the declaration of COVID-19 as a global pandemic on March 11,2020,this pandemic has been circulating for 17 months throughout the world, leading to more than 200 million infections and nearly 4.4 million deaths as of August 15,2021.The pathogen of COVID-19 is a novel coronavirus named SARS-CoV-2,which shares-79%genome sequence identity with SARS-CoV.1 At the early stage of the COVID-19 outbreak,5ARS-CoV-2 caused great panic in the hardest-hit areas due to its high transmissibility and pathogenicity. To fight the COVID-19 crisis, drug repurposing was immediately pursued in order to find potential therapeutics. | Yuanchao Xie Wanchao Yin Yumin Zhang Weijuan Shang Zhen Wang Xiaodong Luan Guanghui Tian Haji A.Aisa Yechun Xu Gengfu Xiao Jia Li Hualiang Jiang Shuyang Zhanq Leike Zhang H.Eric Xu Jingshan Shen | 2021 | Cell Research2021,31,11: | 12
Group VB transition metal dichalcogenides for oxygen reduction reaction and strain-enhanced activity governed by p-orbital electrons of chalcogen显示文摘Developi ng alter native oxyge n reducti on reactio n (ORR) catalysts to replace precious Pt-based metals with abundant materials is the key challe nge of commercial application of fuel cells. Owing to their various compositi ons and tun able electronic properties, transition metal dichalcogenides (TMDs) have the great potential to realize high-efficiency catalysts for ORR. Here, various 3R-phase dichalcogenides of group VB and VIB transition metals (MX2, M = Nb, Ta, Mo, W;X = S, Se, Te) are investigated for ORR catalysts by using density functional theory calculations. The computed over-potentials of group VB TMDs are much less than those of group VIB TMDs. For group VB TMDs, a volcano-type plot of ORR catalytic activity is established on the adsorption energies of *OH, and NbS2 and TaTe2 exhibit best ORR activity with an oveepotential of 0.54 V. To achieve even better activity, strain engineering is performed to tune ORR catalytic activity, and the minimum over-potential of 0.43 V can be realized. We further dem on strate that the shift of p orbital center of surface chalcoge n elements under strain is responsible for tuning the catalytic activity of TMDs. | Shuyang Zhao Ke Wang Xiaolong Zou Lin Gan Hongda Du Chengjun Xu Feiyu Kang Wenhui Duan Jia Li | 2019 | Nano Research2019,12,4: | 4

Control Strategies of Large-scale Residential Air Conditioning Loads Participating in Demand Response Programs显示文摘Residential air conditioning(RAC)loads have great potential to be included in demand response(DR)programs. This paper studies large-scale RAC loads participating in DR programs, such as modeling, parameters identification,DR characteristics and control strategies.First,an aggregate model of large-scale RAC loads are established based on the buildings’performance with heat storage and insulation, avoiding the calculation of a single RAC model. Then, parameters of the aggregate model are identified based on the RACs’power and outdoor temperatures.Based on the aggregate model,DR characteristics of RAC loads are analyzed, including the dynamic relationship between power, outdoor and indoor temperature, and the potential of DR combined with the users’comfort.Next, the DR control strategies adapted for large-scale RAC loads are established by adjusting the temperature set-points.The DR strategies consider users’comfort and calculate the control signals of each RAC load according to the DR power, including adjustment temperature and adjustment time, which are sent to each RAC load for execution. In the DR process, the control center does not need to obtain the users’indoor temperature, which is conducive to protecting the users’privacy.DR strategies of RAC loads when the control degree within/beyond the DR potential are both proposed, and a load recovery control strategy is also introduced. Finally, the effectiveness and accuracy of the proposed model and DR control strategies are verified by simulation results. | Jixiang Wang Xingying Chen Jun Xie Shuyang Xu Kun Yu Lei Gan | 2022 | CSEE Journal of Power and Energy Systems2022,8,3: | 1

Profiles of metabolic gene expression in the white adipose tissue, liver and hypothalamus in leptin knockout(Lep^(△I14/△I14)) rats显示文摘Leptin deficiency is principally linked to metabolic disorders. Leptin knockout(Lep^(△I14/△I14) Sprague Dawley rats created by CRISPR/Cas9 is a new model to study metabolic disorders. We used a whole rat genome oligonucleotide microarray to obtain tissue-specific gene expression profiles of the white adipose tissue, liver and hypothalamus in Lep^(△I14/△I14))and wild-type(WT) rats. We found 1,651 differentially expressed(enriched) genes in white adipose tissue,916 in the liver, and 306 in the hypothalamus in the Lep^(△I14/△I14) rats compared to WT. Gene ontology category and KEGG pathway analysis of the relationships among differentially expressed genes showed that these genes were represented in a variety of functional categories, including fatty acid metabolism, molecular transducers and cellular processes. The reliability of the data obtained from microarray was verified by quantitative real-time PCR on 14 representative genes. These data will contribute to a greater understanding of different metabolic disorders, such as obesity and diabetes. | Leijian Guan Kaixuan Xu Shuyang Xu Ningning Li Xinru Wang Yankai Xia Di Wu | 2017 | The Journal of Biomedical Research2017,31,6: | 1
Bimetallic MOF derived nickel nanoclusters supported by nitrogendoped carbon for efficient electrocatalytic CO_(2)reduction显示文摘Utilizing electrocatalytic CO_(2)reduction(ECR)to decrease the carbon footprint has been regarded as a promising pathway.Herein,we report the synthesis of Ni nanoclusters(NCs)of below 2 nm highly dispersed on N-doped carbon using a Ni/Zn bimetallic metal-organic framework(MOF)precursor. The size and the content of the Ni catalyst can be effectively controlled by varying the Ni:Zn ratio in MOF precursors.The-NH2 group in MOF ligand critically influences the size of Ni catalyst,as well as the property of the carbon substrate. At the optimum ratio of 1:150,Ni NCs with an average size of 1.9 nm anchored on pyridinic N-rich carbon were obtained after MOF pyrolysis. The resultant catalyst exhibits a high Faradaic efficiency for CO(FECO,98.7%)and considerable partial current density for CO(JCO,−40.4 mA·cm^(−2))at−0.88 V versus reversible hydrogen electrode(RHE).Benefiting from the synergistic effect of small Ni clusters and their optimal interaction with the carbon support, the catalyst displays exceptional long-term stability.Density functional theory(DFT)calculations carried out for the three model structures confirm that Ni NCs anchored on N-doped carbon facilitate the easier formation of*COOH intermediate and faster electron transfer rate compared with the large-sized Ni particles represented by Ni(111)and the N-doped carbon without Ni. | Haojing Wang Xiaodong Wu Guanyu Liu Shuyang Wu Rong Xu | 2023 | Nano Research2023,16,4: | 1

Agent-based Modeling and Simulation for the Electricity Market with Residential Demand Response显示文摘Currently, critical peak load caused by residential customers has attracted utility companies and policymakers to pay more attention to residential demand response(RDR)programs. In typical RDR programs, residential customers react to the price or incentive-based signals, but the actions can fall behind flexible market situations. For those residential customers equipped with smart meters, they may contribute more DR loads if they can participate in DR events in a proactive way.In this paper,we propose a comprehensive market framework in which residential customers can provide proactive RDR actions in a day-ahead market(DAM).We model and evaluate the interactions between generation companies(GenCos),retailers, residential customers, and the independent system operator(ISO)via an agent-based modeling and simulation(ABMS)approach. The simulation framework contains two main procedures—the bottom-up modeling procedure and the reinforcement learning(RL)procedure. The bottom-up modeling procedure models the residential load profiles separately by household types to capture the RDR potential differences in advance so that residential customers may rationally provide automatic DR actions.Retailers and GenCos optimize their bidding strategies via the RL procedure. The modified optimization approach in this procedure can prevent the training results from falling into local optimum solutions. The ISO clears the DAM to maximize social welfare via Karush-Kuhn-Tucker(KKT)conditions.Based on realistic residential data in China, the proposed models and methods are verified and compared in a large multi-scenario test case with 30,000 residential households. Results show that proactive RDR programs and interactions between market entities may yield significant benefits for both the supply and demand sides. The models and methods in this paper may be used by utility companies, electricity retailers, market operators, and policy makers to evaluate the consequences of a proactive RDR and the interactions among multi-entities. | Shuyang Xu Xingying Chen Jun Xie Saifur Rahman Jixiang Wang Hongxun Hui Tao Chen | 2021 | CSEE Journal of Power and Energy Systems2021,7,2: | 1
Non-Ischemic, Non-Hypoxic Myocardial Injury, and Long-Term Mortality in Patients with Coronavirus Disease 2019: A Retrospective Cohort Study显示文摘Objective:Cardiac damage is commonly reported in patients with coronavirus disease 2019(COVID-19)but its prevalence and impact on the long-term survival of patients remain uncertain. This study aimed to explore the prevalence of myocardial injury and assess its prognostic value in patients with COVID-19.Methods:A single-center,retrospective cohort study was performed at the Affiliated Hospital of Jianghan University.Data from 766 patients with confirmed COVID-19 who were hospitalized from December 27,2019 to April 25,2020 were collected.Demographic, clinical, laboratory, electrocardiogram, treatment data and all-cause mortality during follow-up were collected and analyzed. Results:Of the 766 patients with moderate to critically ill COVID-19,86(11.2%)died after a mean follow-up of 72.8 days.Myocardial injury occurred in 94(12.3%)patients.The mortality rate was 64.9%(61/94)and 3.7%(25/672)in patients with and without myocardial injury, respectively.Cox regression showed that myocardial injury was an independent risk factor for mortality(hazard ratio:8.76,95%confidence interval:4.76–16.11,P<0.001).Of the 90 patients with myocardial injury with electrocardiogram results, sinus tachycardia was present in 29,bundle branch block in 26,low voltage in 10,and abnormal T-wave in 53.Conclusions:COVID-19 not only involves pneumonia but also cardiac damage.Myocardial injury is a common complication and an independent risk factor for mortality in COVID-19 patients. | Fajiu Li Xijie Zhu Ziyang Zhu Yinjian Yang Zhuang Tian Duolao Wang Shi Chen Xiaoyan Gao Yalin Xu Bo Zhang Wei Yu Min Liu Xiqi Xu Chenghong Li Shuyang Zhang | 2022 | Cardiology Discovery2022,2,2: | 0
甜高粱青贮菌剂研究进展显示文摘Sweet sorghum has the advantages of water saving, high biomass, high temperature tolerance, salt and alkali tolerance. It is suitable for planting as high quality feed in arid,semi-arid and high salinity areas[1].Lactic acid bacteria can use high content of water-soluble carbohydrates in sweet sorghum to grow rapidly, improve its adaptability as ruminant silage, and better preserve the nutrients of silage[2].Cellulase can degrade the high content of cellulose in sweet sorghum silage and improve its palatability and digestibility. At present, there is no special silage inoculum of sweet sorghum in the market. | Wang Shuyang Dong Miaoyin Xu Fuqiang Li Qiaoqiao Chen Jihong Li Wenjian | 2018 | IMP & HIRFL Annual Report2018,,1: | 0

Tumor microenvironment-responsive artesunate loaded Z-scheme heterostructures for synergistic photo-chemodynamic therapy of hypoxic tumor显示文摘Tumor microenvironment(TME)with the particular features of severe hypoxia, insufficient endogenous H2O2,and overexpression of glutathione(GSH)markedly reduced the antitumor efficacy of monotherapy.Herein,a TME-responsive multifunctional nanoplatform(Bi2S3@Bi@PDA-HA/Art NRs)was presented for synergistic photothermal therapy(PTT),chemodynamic therapy(CDT),and photodynamic therapy(PDT)to achieve better therapeutic outcomes. The Z-scheme heterostructured bismuth sulfide@bismuth nanorods(Bi2S3@Bi NRs)guaranteed excellent photothermal performance of the nanoplatform. Moreover, its ability to produce O2 and reactive oxygen species(ROS)synchronously could relieve tumor hypoxia and improve PDT outcomes. The densely coated polydopamine/ammonium bicarbonate(PDA/ABC)and hyaluronic acid(HA)layers on the surface of the nanoplatform enhanced the cancer-targeting capacity and induced the acidic TME-triggered in situ“bomb-like”release of Art.The CDT treatment was achieved by activating the released Art through intracellular Fe2+ions in an H2O2-independent manner.Furthermore, decreasing the glutathione peroxidase 4(GPX4)levels by Art could also increase the PDT efficiency of Bi2S3@Bi NRs.Owing to the synergistic effect, this nanoplatform displayed improved antitumor efficacy with minimal toxicity both in vitro and in vivo. Our design sheds light on the application of phototherapy combined with the traditional Chinese medicine monomer-artesunate in treating the hypoxic tumor. | Jie Lv Xiaoyu Wang Xue Zhang Runpei Xu Shuyang Hu Shuangling Wang Meng Li | 2023 | Asian Journal of Pharmaceutical Sciences2023,18,3: | 0
Debris Flow Susceptibility Evaluation in Meizoseismal Region:A Case Study in Jiuzhaigou,China显示文摘Jiuzhaigou is situated on a mountain-canyon region and is famous for frequent tectonic activities. An abundance of loose co-seismic landslides and collapses were produced on gullies after the Jiuzhaigou Earthquake on August 8,2017,which was served as material source for debris flow in later years.Debris flow appears frequently which are seriously endangering the safety of people's lives and properties.Even the earliest debris flow appeared in areas where no case ever reported before. The debris flow susceptibility evaluation(DFSE)is used for predicting the areas prone to debris flow, which is urgently required to avoid hazards and help to guide the strategy of preventive measures. Therefore, this work employs debris flow in Jiuzhaigou to reveal the characteristics of disaster-pregnant environment and to explore the application of machine learning in DFSE.Some new viewpoints are suggested:(i)Material density factor of debris flow is first adopted in this work, and it is proved to be a critical factor for triggering debris flows by sensitivity analysis method.(ii)Deep neural network and convolutional neural network(CNN)achieve relatively good area under the curve(AUC)values and are 0.021-0.024 higher than traditional machine learning methods.(iii)Watershed units combined with CNNbased model can achieve more accurate, reliable and practical susceptibility map. This work provides an idea for prevention of debris flow in mountainous lands. | Yongwei Li Linrong Xu Yonghui Shang Shuyang Chen | 2024 | Journal of Earth Science2024,35,1: | 0

Efficient expansion of rare human circulating hematopoietic stem/progenitor cells in steady-state blood using a polypeptide-forming 3D culture显示文摘Although widely applied in treating hematopoietic malignancies, transplantation of hematopoietic stem/progenitor cells(HSPCs)is impeded by HSPC shortage.Whether circulating HSPCs(cHSPCs)in steady-state blood could be used as an alternative source remains largely elusive. Here we develop a three-dimensional culture system(3DCS)including arginine, glycine, aspartate, and a series of factors.Fourteen-day culture of peripheral blood mononuclear cells(PBMNCs)in 3DCS led to 125-and 70-fold increase of the frequency and number of CD34+cells.Further,3DCS-expanded cHSPCs exhibited the similar reconstitution rate com-pared to CD34+HSPCs in bone marrow.Mechanistically,3DCS fabricated an immunomodulatory niche, secreting cytokines as TNF to support cHSPC survival and proliferation.Finally,3DCS could also promote the expansion of cHSPCs in patients who failed in HSPC mobilization. Our 3DCS successfully expands rare cHSPCs,providing an alternative source for the HSPC therapy, particularly for the patients/donors who have failed in HSPC mobilization. | Yulin Xu Xiangjun Zeng Mingming Zhang Binsheng Wang Xin Guo Wei Shan Shuyang Cai Qian Luo Honghu Li Xia Li Xue Li Hao Zhang Limengmeng Wang Yu Lin Lizhen Liu Yanwei Li Meng Zhang Xiaohong Yu Pengxu Qian He Huang | 2022 | Protein & Cell2022,13,11: | 0
