| 1 | Application of platelet-rich plasma with stem cells in bone and periodontal tissue engineering显示文摘Presently, there is a high paucity of bone grafts in the United States and worldwide. Regenerating bone is of prime concern due to the current demand of bone grafts and the increasing number of diseases causing bone loss. Autogenous bone is the present gold standard of bone regeneration. However, disadvantages like donor site morbidity and its decreased availability limit its use. Even allografts and synthetic grafting materials have their own limitations. As certain specific stem cells can be directed to differentiate into an osteoblastic lineage in the presence of growth factors(GFs), it makes stem cells the ideal agents for bone regeneration.Furthermore, platelet-rich plasma(PRP), which can be easily isolated from whole blood, is often used for bone regeneration, wound healing and bone defect repair. When stem cells are combined with PRP in the presence of GFs, they are able to promote osteogenesis. | 2 | Advanced landfill leachate treatment using a two-stage UASB-SBR system at low temperature显示文摘A two-stage upflow anaerobic sludge blanket(UASB) and sequencing batch reactor(SBR) system was introduced to treat landfill leachate for advanced removal of COD and nitrogen at low temperature.In order to improve the total nitrogen(TN) removal efficiency and to reduce the COD requirement for denitrification,the raw leachate with recycled SBR nitrification supernatant was pumped into the first-stage UASB(UASB1) to achieve simultaneous denitrification and methanogenesis.The results showed that UASB1 played an important role in COD removal and UASB2 and SBR further enhanced the nutrient removal efficiency.When the organic loading rates of UASB1,UASB2 and SBR were 11.95,1.63 and 1.29 kg COD/(m3·day),respectively,the total COD removal efficiency of the whole system reached 96.7%.The SBR acted as the real undertaker for NH4+-N removal due to aerobic nitrification.The system obtained about 99.7% of NH4+-N removal efficiency at relatively low temperature(14.9-10.9°C).More than 98.3% TN was removed through complete denitrification in UASB1 and SBR.In addition,temperature had a significant effiect on the rates of nitrification and denitrification rather than the removal of TN and NH4+-N once the complete nitrification and denitrification were achieved. | Hongwei Sun,Qing Yang,Yongzhen Peng,Xiaoning Shi,Shuying Wang,Shujun Zhang Key Laboratory of Beijing Water Quality Science and Water Envirocnnment Recovery Engineering,Beijing University of Technology,Beijing 100124,China | 2010 | Journal of Environmental Sciences2010,22,4: | 26 |
| 3 | A structural view of the antibiotic degradation enzyme NDM-1 from a superbug显示文摘Gram-negative Enterobacteriaceae with resistance to carbapenem conferred by New Delhi metallo-β-lactamase 1(NDM-1)are a type of newly discovered antibioticresistant bacteria.The rapid pandemic spread of NDM-1 bacteria worldwide(spreading to India,Pakistan,Europe,America,and Chinese Taiwan)in less than 2 months characterizes these microbes as a potentially major global health problem.The drug resistance of NDM-1 bacteria is largely due to plasmids containing the blaNDM-1 gene shuttling through bacterial populations.The NDM-1 enzyme encoded by the blaNDM-1 gene hydrolyzes β-lactam antibiotics,allowing the bacteria to escape the action of antibiotics.Although the biological functions and structural features of NDM-1 have been proposed according to results from functional and structural investigation of its homologues,the precise molecular characteristics and mechanism of action of NDM-1 have not been clarified.Here,we report the threedimensional structure of NDM-1 with two catalytic zinc ions in its active site.Biological and mass spectroscopy results revealed that D-captopril can effectively inhibit the enzymatic activity of NDM-1 by binding to its active site with high binding affinity.The unique features concerning the primary sequence and structural conformation of the active site distinguish NDM-1 from other reported metallo-β-lactamases(MBLs)and implicate its role in wide spectrum drug resistance.We also discuss the molecular mechanism of NDM-1 action and its essential role in the pandemic of drug-resistant NDM-1 bacteria.Our results will provide helpful information for future drug discovery targeting drug resistance caused by NDM-1 and related metallo-β-lactamases. | Yu Guo Jing Wang Guojun Niu Wenqing Shui Yuna Sun Honggang Zhou Yaozhou Zhang Cheng Yang Zhiyong Lou Zihe Rao | 2011 | Protein & Cell2011,2,5: | 16 |
| 4 | Chimeric antigen receptor-engineered T-cell therapy for liver cancer显示文摘Background: Chimeric antigen receptor-engineered T-cell(CAR-T) therapy is a newly developed immunotherapy used in the treatment of cancers. Because CAR-T therapy has shown great success in treating CD19-positive hematological malignancies, its application has been explored in the treatment of solid tumors, such as liver cancer. In this review, we discuss the immune characteristics of liver cancer, the obstacles encountered during the application of CAR-T therapy, and preclinical and clinical progress in the use of CAR-T therapy in patients with liver cancer.Data sources: The data on CAR-T therapy related to liver cancers were collected by searching Pub Med and the Web of Science databases prior to December 2017 with the keywords 'chimeric antigen receptor','CAR-T', 'liver cancer', 'hepatocellular carcinoma', and 'solid tumor'. Additional articles were identified by manual search of references found in the primary articles. The data for clinical trials were collected by searching Clinical Trials.gov.Results: The liver has a tolerogenic nature in the intrahepatic milieu and its tumor microenvironment significantly affects tumor progression. The obstacles that reduce the efficacy of CAR-T therapy in solid tumors include a lack of specific tumor antigens, limited trafficking and penetration of CAR-T cells to tumor sites, and an immunosuppressive tumor microenvironment. To overcome these obstacles, several strategies have emerged. In addition, several strategies have been developed to manage the side effects of CAR-T, including enhancing the selectivity of CARs and controlling CAR-T activity. To date, no clinical trials of CAR-T therapy against HCC have been completed. However, preclinical studies in vitro and in vivo have shown potent antitumor efficacy. Glypican-3, mucin-1, epithelial cell adhesion molecule, carcinoembryonic antigen, and other targets are currently being studied.Conclusions: The application of CAR-T therapy for liver cancer is just beginning to be explored and more research is needed. | 5 | Carbon dioxide emissions from cities in China based on high resolution emission gridded data显示文摘Based on the China high resolution emission gridded data(1 km spatial resolution), this article is aimed to create a Chinese city carbon dioxide(CO_2) emission data set using consolidated data sources as well as normalized and standardized data processing methods. Standard methods were used to calculate city CO_2 emissions, including scope 1 and scope 2. Cities with higher CO_2 emissions are mostly in north, northeast, and eastern coastal areas. Cities with lower CO_2 emissions are in the western region. Cites with higher CO_2 emissions are clustered in the Jing-Jin-Ji Region(such as Beijing, Tianjin, and Tangshan), and the Yangtze River Delta region(such as Shanghai and Suzhou).The city per capita CO_2 emission is larger in the north than the south. There are obvious aggregations of cities with high per capita CO_2 emission in the north. Four cities among the top 10 per capita emissions(Erdos, Wuhai, Shizuishan, and Yinchuan) cluster in the main coal production areas of northern China. This indicates the significant impact of coal resources endowment on city industry and CO_2 emissions. The majority(77%) of cities have annual CO_2 emissions below 50 million tons. The mean annual emission, among all cities, is 37 million tons. Emissions from service-based cities, which include the smallest number of cities, are the highest. Industrial cities are the largest category and the emission distribution from these cities is close to the normal distribution. Emissions and degree of dispersion, in the other cities(excluding industrial cities and service-based cities), are in the lowest level. Per capita C02 emissions in these cities are generally below 20 t/person(89%) with a mean value of 11 t/person. The distribution interval of per capita CO_2 emission within industrial cities is the largest among the three city categories. This indicates greater differences among per capita CO_2 emissions of industrial cities. The distribution interval of per capita CO_2 emission of other cities is the lowest, indicating smaller differences of per capita CO_2 emissions among this city category. Three policy suggestions are proposed: first, city CO_2 emission inventory data in China should be increased,especially for prefecture level cities. Second, city responsibility for emission reduction, and partitioning the national goal should be established, using a bottom-up approach based on specific CO_2emission levels and potential for emission reductions in each city. | 8 | Effect of temperature on anoxic metabolism of nitrites to nitrous oxide by polyphosphate accumulating organisms显示文摘Temperature is an important physical factor, which strongly influences biomass and metabolic activity. In this study, the effects of temperature on the anoxic metabolism of nitrite(NO-2) to nitrous oxide(N2O) by polyphosphate accumulating organisms, and the process of the accumulation of N2O(during nitrite reduction), which acts as an electron acceptor, were investigated using 91% ± 4% Candidatus Accumulibacter phosphatis sludge. The results showed that N2O is accumulated when Accumulibacter first utilize nitrite instead of oxygen as the sole electron acceptor during the denitrifying phosphorus removal process. Properties such as nitrite reduction rate, phosphorus uptake rate, N2O reduction rate, and polyhydroxyalkanoate degradation rate were all influenced by temperature variation(over the range from 10 to 30°C reaching maximum values at 25°C). The reduction rate of N2O by N2O reductase was more sensitive to temperature when N2O was utilized as the sole electron acceptor instead of NO2, and the N2O reduction rates, ranging from 0.48 to 3.53 N2O-N/(hr·g VSS), increased to 1.45 to 8.60 mg N2O-N/(hr·g VSS). The kinetics processes for temperature variation of 10 to 30°C were(θ1 = 1.140–1.216 and θ2= 1.139–1.167). In the range of 10°C to 30°C, almost all of the anoxic stoichiometry was sensitive to temperature changes. | 9 | Auranofin mitigates systemic iron overload and induces ferroptosis via distinct mechanisms显示文摘Iron homeostasis is essential for health;moreover,hepcidin-deficiency results in iron overload in both hereditary hemochromatosis and iron-loading anemia.Here,we identified iron modulators by functionally screening hepcidin agonists using a library of 640 FDA-approved drugs in human hepatic Huh7 cells.We validated the results in C57BL/6J mice and a mouse model of hemochromatosis(Hfe^(−/−)mice).Our screen revealed that the anti-rheumatoid arthritis drug auranofin(AUR)potently upregulates hepcidin expression.Interestingly,we found that canonical signaling pathways that regulate iron,including the Bmp/Smad and IL-6/Jak2/Stat3 pathways,play indispensable roles in mediating AUR’s effects.In addition,AUR induces IL-6 via the NF-κB pathway.In C57BL/6J mice,acute treatment with 5 mg/kg AUR activated hepatic IL-6/hepcidin signaling and decreased serum iron and transferrin saturation.Whereas chronically treating male Hfe^(−/−)mice with 5 mg/kg AUR activated hepatic IL-6/hepcidin signaling,decreasing systemic iron overload,but less effective in females.Further analyses revealed that estrogen reduced the ability of AUR to induce IL-6/hepcidin signaling in Huh7 cells,providing a mechanistic explanation for ineffectiveness of AUR in female Hfe^(−/−)mice.Notably,high-dose AUR(25 mg/kg)induces ferroptosis and causes lipid peroxidation through inhibition of thioredoxin reductase(TXNRD)activity.We demonstrate the ferroptosis inhibitor ferrostatin significantly protects liver toxicity induced by highdose AUR without comprising its beneficial effect on iron metabolism.In conclusion,our findings provide compelling evidence that TXNRD is a key regulator of ferroptosis,and AUR is a novel activator of hepcidin and ferroptosis via distinct mechanisms,suggesting a promising approach for treating hemochromatosis and hepcidin-deficiency related disorders. | Lei Yang Hao Wang Xiang Yang Qian Wu Peng An Xi Jin Weiwei Liu Xin Huang Yuzhu Li Shiyu Yan Shuying Shen Tingbo Liang Junxia Min Fudi Wang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 5 |

| 16 | Deletion of IFT20 in early stage T lymphocyte differentiation inhibits the development of col agen-induced arthritis显示文摘IFT20 is the smallest member of the intraflagellar transport protein(IFT) complex B. It is involved in cilia formation. Studies of IFT20 have been confined to ciliated cells. Recently, IFT20 was found to be also expressed in non-ciliated T cells and have functions in immune synapse formation and signaling in vitro. However, how IFT20 regulates T-cell development and activation in vivo is still unknown. We deleted the IFT20 gene in early and later stages of T-cell development by crossing IFT20flox/flox(IFT20f/f) mice with Lck-Cre and CD4-Cre transgenic mice, and investigated the role of IFT20 in T-cell maturation and in the development of T cell-mediated collagen-induced arthritis(CIA). We found that both Lck-Cre/IFT20f/f and CD4-Cre/IFT20f/f mice were indistinguishable from their wild-type littermates in body size, as well as in the morphology and weight of the spleen and thymus. However, the number of CD4- and CD8-positive cells was significantly lower in thymus and spleen in Lck-Cre/IFT20f/f mice. Meanwhile, the incidence and severity of CIA symptoms were significantly decreased, and inflammation in the paw was significantly inhibited in Lck-Cre/IFT20f/f mice compared to Lck-Cre/IFT201/1littermates. Deletion IFT20 in more mature T cells of CD4-Cre/IFT20f/f mice had only mild effects on the development of T cells and CIA. The expression of IL-1b, IL-6 and TGF-b1 were significantly downregulated in the paw of Lck-Cre/IFT20f/f mice, but just slight decreased in CD4-Cre/IFT20f/f mice. | 17 | Discovering small molecules as Wnt inhibitors that promote heart regeneration and injury repair显示文摘There are intense interests in discovering pro regenerative medicine leads that can promote cardiac differentiation and regeneration,as well as repair damaged heart tissues.We have combined zebrafish embryo-based screens with cardiomyogenesis assays to discover selective small molecules that modulate heart development and regeneration with minimal adverse effects.Two related compounds with novel structures,named as Cardiomogen 1 and 2(CDMG1 and CDMG2),were identified for their capacity to promote myocardial hyperplasia through expansion of the cardiac progenitor cell population.We find that Cardiomogen acts as a Wnt inhibitor by targeting p-catenin and reducing Tcf/Lef-mediated transcription in cultured cells.CDMG treatment of amputated zebrafish hearts reduces nuclear p-catenin in injured heart tissue,increases cardiomyocyte(CM)proliferation,and expedites wound healing,thus accelerating cardiac muscle regeneration.Importantly,Cardiomogen can alleviate the functional deterioration of mammalian hearts after myocardial infarction.Injured hearts exposed to CDMG1 display increased newly formed CMs and reduced fibrotic scar tissue,which are in part attributable to the^-catenin reduction.Our findings indicate Cardiomogen as a Wnt inhibitor in enhancing injury-induced CM proliferation and heart regeneration,highlighting the values of embryo-based small molecule screens in discovery of effective and safe medicine leads. | Shuying Xie Wenbin Fu Guangju Yu Xueli Hu Kaa Seng Lai Xiangwen Peng Yating Zhou Xuejiao Zhu Ptamen Christov Leah Sawyer Terri T.Ni Gary A.Sulikowski Zhongzhou Yang Ethan Lee Chunyu Zeng Wei E.Wang Tao P.Zhong | 2020 | Journal of Molecular Cell Biology2020,12,1: | 4 |
| 18 | On the critical particle size of soil with clogging potential in shield tunneling显示文摘Shield tunneling is easily obstructed by clogging in clayey strata with small soil particles.However,soil clogging rarely occurs in strata with coarse-grained soils.Theoretically,a critical particle size of soils should exist,below which there is a high risk of soil clogging in shield tunneling.To determine the critical particle size,a series of laboratory tests was carried out with a large-scale rotary shear apparatus to measure the tangential adhesion strength of soils with different particle sizes and water contents.It was found that the tangential adhesion strength at the soilesteel interface gradually increased linearly with applied normal pressure.When the particle size of the soil specimen was less than 0.15 mm,the interfacial adhesion force first increased and then decreased as the water content gradually increased;otherwise,the soil specimens did not manifest any interfacial adhesion force.The amount of soil mass adhering to the steel disc was positively correlated with the interfacial adhesion force,thus the interfacial adhesion force was adopted to characterize the soil clogging risk in shield tunneling.The critical particle size of soils causing clogging was determined to be 0.15 mm.Finally,the generation mechanism of interfacial adhesion force was explored for soils with different particle sizes to explain the critical particle size of soil with clogging risk in shield tunneling. | Shuying Wang Zihao Zhou Pengfei Liu Zhao Yang Qiujing Pan Weizhong Chen | 2023 | Journal of Rock Mechanics and Geotechnical Engineering2023,15,2: | 3 |

| 20 | Investigation and control of a Plasmodium falciparum malaria outbreak in Shan Special Region II of Myanmar along the China-Myanmar Border from June to December 2014显示文摘Background:From 2007 to 2013,intensive control measures reduced malaria burden by 90%along the China-Myanmar border.However,despite these measures a P.falciparum malaria outbreak was reported in the Shan Special Region II of Myanmar in June of 2014.Methods:Epidemiological,parasitological and entomological investigations were performed.Dihydroartemisinin piperaquine(DAPQ)was immediately administered to treat parasite positive individuals.Long lasting insecticidal nets(LLIN),indoor residual spraying(IRS)with insecticides and behavior change communication(BCC)were also provided for outbreak control.An embedded efficacy study was conducted evaluating DP.Molecular genotyping via polymerase chain reaction(PCR)was performed on the Kelch gene on chromosome 13.Results:All infections were identified as Plasmodium falciparum by RDT and microscopy.Two fatalities resulted from the outbreak.The attack rate was 72.8%(67/92)and the incidence density rate was 14.2 per 100 person-weeks.The positive rate of rapid diagnostic test(RDT)was 72.2%(65/90)and microscopically-determine parasite rate 42.2%(38/90).Adjusted odds ratio(OR)of multivariate logistic regression analysis for aged<15 years,15–45 years,inappropriate treatment from a private healer and lack of bed nets were 13.51(95%confidence interval,2.21–105.89),7.75(1.48–44.97),3.78(1.30–46.18)and 3.21(1.21–15.19)respectively.In the six surrounding communities of the outbreak site,positive RDT rate was 1.2%(4/328)and microscopically-determine parasite rate 0.6%(2/328).Two light traps collected a total of 110 anopheline mosquitoes including local vectors,An.minimus,An.sinensis and An.maculates.After intensive control,the detection of malaria attacks,parasites and antigen were reduced to zero between July 1 and December 1,2014.The cure rate of P.falciparum patients at day 42 was 94.3%(95%CI,80.8–99.3%).The PCR did not detect K13-propeller mutations.Conclusion:Imported P.falciparum caused the outbreak.Age,seeking inappropriate treatment and lack of bed nets were risk factors for infection during the outbreak.P.falciparum was sensitive to treatment with DAPQ.The integrated measures controlled the outbreak and prevented the spread of P.falciparum effectively.The results of this study indicate that malaria control on the China-Myanmar border,especially among special populations,needs further collaboration between China,Myanmar and international societies. | Hui Liu Jian-Wei Xu Heng-Lin Yang Mei Li Cheng-De Sun Yi-Jie Yin Zhi-Liang Zheng Guang-Yun Zhang Ai-Shui Yu Yong-Hui Yang Chun-Hui Li Shui Ai | 2016 | Infectious Diseases of Poverty2016,5,1: | 3 |
