| 1 | Enteric glial cells and their role in gastrointestinal motor abnormalities: Introducing the neuro-gliopathies显示文摘The role of enteric glial cells has somewhat changed from that of mere mechanical support elements, gluing together the various components of the enteric nervous system, to that of active participants in the complex interrelationships of the gut motor and inflammatory events. Due to their multiple functions, spanning from supporting elements in the myenteric plexuses to neurotransmitters, to neuronal homeostasis, to antigen presenting cells, this cell population has probably more intriguing abilities than previously thought. Recently, some evidence has been accumulating that shows how these cells may be involved in the pathophysiological aspects of some diseases. | 9 | From Vietnamese plants to a biflavonoid that relieves inflammation by triggering the lipid mediator class switch to resolution显示文摘Chronic inflammation results from excessive pro-inflammatory signaling and the failure to resolve the inflammatory reaction.Lipid mediators orchestrate both the initiation and resolution of inflammation.Switching from pro-inflammatory to pro-resolving lipid mediator biosynthesis is considered as efficient strategy to relieve chronic inflammation,though drug candidates exhibiting such features are unknown.Starting from a library of Vietnamese medical plant extracts,we identified isomers of the biflavanoid 8-methylsocotrin-4’-ol from Dracaena cambodiana,which limit inflammation by targeting 5-lipoxygenase and switching the lipid mediator profile from leukotrienes to specialized pro-resolving mediators(SPM).Elucidation of the absolute configurations of 8-methylsocotrin-4’-ol revealed the 2 S,γSisomer being most active,and molecular docking studies suggest that the compound binds to an allosteric site between the 5-lipoxygenase subdomains.We identified additional subordinate targets within lipid mediator biosynthesis,including microsomal prostaglandin E2 synthase-1.Leukotriene production is efficiently suppressed in activated human neutrophils,macrophages,and blood,while the induction of SPM biosynthesis is restricted to M2 macrophages.The shift from leukotrienes to SPM was also evident in mouse peritonitis in vivo and accompanied by a substantial decrease in immune cell infiltration.In summary,we disclose a promising drug candidate that combines potent 5-lipoxygenase inhibition with the favorable reprogramming of lipid mediator profiles. | Tran Thi Van Anh Alilou Mostafa Zhigang Rao Simona Pace Stefan Schwaiger Christian Kretzer Veronika Temml Carsten Giesel Paul M.Jordan Rossella Bilancia Christina Weinigel Silke Rummler Birgit Waltenberger Tran Hung Antonietta Rossi Hermann Stuppner Oliver Werz Andreas Koeberle | 2021 | Acta Pharmaceutica Sinica B2021，11，6： | 1 |

| 20 | Hepatitis C risk assessment,testing and referral for treatment in urban primary care:Role of race and ethnicity显示文摘AIM: To determine rates of hepatitis C (HCV) risk factor ascertainment, testing, and referral in urban primary care practices, with particular attention to the effect of race and ethnicity. METHODS: Retrospective chart review from four primary care sites in Philadelphia; two academic primary care practices and two community clinics was performed. Demographics, HCV risk factors, and other risk exposure information were collected. RESULTS: Four thousand four hundred and seven charts were reviewed. Providers documented histories of injection drug use (IDU) and transfusion for less than 20% and 5% of patients, respectively. Only 55% of patients who admitted IDU were tested for HCV. Overall, minorities were more likely to have information regarding a risk factor documented than their white counterparts (79% vs 68%, P < 0.0001). Hispanics were less likely to have a risk factor history documented, compared to blacks and whites (P < 0.0001). Overall, minorities were less likely to be tested for HCV than whites in the presence of a known risk factor (23% vs 35%, P = 0.004). Among patients without documentation of risk factors, blacks and Hispanics were more likely to be tested than whites (20% and 24%, vs 13%, P < 0.005, respectively). CONCLUSION: (1) Documentation of an HCV risk factor history in urban primary care is uncommon, (2)Racial differences exist with respect to HCV risk factor ascertainment and testing, (3) Minority patients, positive for HCV, are less likely to be referred for subspecialty care and treatment.
