15Clinical progress of nanomedicine-based RNA therapies显示文摘The clinical application of nanoparticles(NPs)to deliver RNA for therapy has progressed rapidly since the FDA approval of Onpattro®in 2018 for the treatment of polyneuropathy associated with hereditary transthyretin amyloidosis.The emergency use authorization or approval and widespread global use of two mRNA-NP based vaccines developed by Moderna Therapeutics Inc.and Pfizer-BioNTech in 2021 has highlighted the translatability of NP technology for RNA delivery.Furthermore,in clinical trials,a wide variety of NP formulations have been found to extend the half-life of RNA molecules such as microRNA,small interfering RNA,and messenger RNA,with limited safety issues.In this review,we discuss the NP formulations that are already used in the clinic to deliver therapeutic RNA and highlight examples of RNA-NPs which are currently under evaluation for human use.We also detail NP formulations that failed to progress through clinical trials,in hopes of guiding future successful translation of nanomedicine-based RNA therapeutics into the clinic.Siyoung A.Lim Alysia Cox Madelynn Tung Eun Ji Chung 2022Bioactive Materials2022,7,6:0

17Understanding Antecedents of Civic Engagement in the Age of Social Media:From the Perspective of Efficacy Beliefs显示文摘This study examines three efficacy beliefs-political self-efficacy,political collective efficacy,and knowledge sharing efficacy-as antecedents of social media use and civic engagement.Employing more than one thousand samples in Singapore,we empirically test(a)a conceptual framework that can provide an understanding of the relationship between the three types of efficacy and civic engagement and(b)the underlying mechanism through which the three types of efficacy beliefs affect civic engagement via social media.The findings suggest that knowledge sharing efficacy was found to play an important role in mediating the relationships between social media use and political self-efficacy,political collective efficacy,respectively,which,in turn,influences the social media use.Siyoung Chung KyuJin Shim 2020Journal of Psychological Research2020,2,3:0
18APOE ε4-dependent effects on the early amyloid pathology in induced neurons of patients with Alzheimer’s disease显示文摘Background: The ε4 allele of apolipoprotein E (APOE ε4) is the strongest known genetic risk factor for late-onset Alzheimer’s disease (AD), associated with amyloid pathogenesis. However, it is not clear how APOE ε4 accelerates amyloid-beta (Aβ) deposition during the seeding stage of amyloid development in AD patient neurons. Methods: AD patient induced neurons (iNs) with an APOE ε4 inducible system were prepared from skin fibroblasts of AD patients. Transcriptome analysis was performed using RNA isolated from the AD patient iNs expressing APOE ε4 at amyloid-seeding and amyloid-aggregation stages. Knockdown of IGFBP3 was applied in the iNs to investigate the role of IGFBP3 in the APOE ε4-mediated amyloidosis. Results: We optimized amyloid seeding stage in the iNs of AD patients that transiently expressed APOE ε4. Remarka-bly, we demonstrated that Aβ pathology was aggravated by the induction of APOE ε4 gene expression at the amyloid early-seeding stage in the iNs of AD patients. Moreover, transcriptome analysis in the early-seeding stage revealed that IGFBP3 was functionally important in the molecular pathology of APOE ε4-associated AD. Conclusions: Our findings suggest that the presence of APOE ε4 at the early Aβ-seeding stage in patient iNs is critical for aggravation of sporadic AD pathology.
