| 2 | Knockdown of OsHox33, a member of the class III homeodomain-leucine zipper gene family, accelerates leaf senescence in rice显示文摘The class III homeodomain-leucine zipper(HD-Zip III)gene family plays important roles in plant growth and development,including regulation of apical embryo patterning,embryonic shoot meristem formation,leaf polarity,vascular development,and meristem function,with a particularly crucial function in leaf development.Although HD-Zip III members are highly conserved in land plants,previous studies,such as genetic analyses based on multiple mutants in Arabidopsis and other plants,suggest that various HD-Zip III family genes have evolved with distinct functions and pleiotropic effects on plant growth and development.In this study,we analyzed a HD-Zip III member,OsHox33,and demonstrated that it plays an important role in age-dependent leaf senescence in rice.We constructed two specific RNAi vectors derived from the 5′-end region and 3′-UTR of OsHox33 to knockdown its expression.Transgenic plants harboring either RNAi construct displayed similar phenotypes of precocious leaf senescence symptoms,suggesting that knockdown of OsHox33 accelerates leaf senescence in rice.pOsHox33::GUS fusion expression and RT-PCR revealed that OsHox33 is highly expressed in young organs,especially in young meristems such as shoot apical meristems,intercalary meristems,and young callus.In addition,real-time PCR indicated that OsHox33 was more highly expressed in young leaves than in old leaves.To further investigate OsHox33 function,we analyzed chloroplast ultrastructure in different-aged leaves of RNAi plants,and found that OsHox33 knockdown accelerated chloroplast degradation,which is consistent with RNAi phenotypes.Finally,real-time PCR studies showed that OsHox33 can regulate the expression of GS1 and GS2,two senescence-associated genes.Taken together,the work presented here provides new insights into the function of HD-Zip III members in plants. | LUAN WeiJiang SHEN Ao JIN ZhiPing SONG SuSheng LI ZhengLong SHA AiHua | 2013 | Science China(Life Sciences)2013,56,12: | 13 |

| 5 | Associations of PGK1 promoter hypomethylation and PGK1-mediated PDHK1 phosphorylation with cancer stage and prognosis:a TCGA pan-cancer analysis显示文摘Background:Cancer cells reprogram metabolism for proliferation.Phosphoglycerate kinase 1(PGK1),as a glycolytic enzyme and newly identified protein kinase,coordinates glycolysis and mitochondrial metabolism.However,the clini-cal significance of PGK1 expression and function in cancer progression is unclear.Here,we investigated the relation-ship between the progression and prognosis of multiple cancer types and PGK1 expression and its function in the mitochondrial metabolism regulation.Methods:We performed pan-cancer analyses of PGK1 mRNA level and DNA methylation in 11,908 tumor tissues and 1582 paired normal tissues across 34 cancer types in The Cancer Genome Atlas datasets.Using specific antibodies against PGK1 S203 and PDHK1 T338 phosphorylation,we performed immunohistochemistry with tissue microarray assay in additional 818 cancer cases with 619 paired normal tissues from five cancer types.Results:The PGK1 mRNA level was significantly elevated with hypomethylation in promotor regions and associated with advanced TNM stage in 15 and four cancer types,respectively.In breast carcinoma,elevated PGK1 mRNA level and promoter hypomethylation were associated with poor prognosis.Positively correlated PGK1 S203 and PDHK1 T338 phosphorylation levels were significantly associated with short overall survival(OS)in cancers of the breast,liver,lung,stomach,and esophagus and with advanced TNM stage in breast and esophageal cancers.PGK1 pS203 and PDHK1 pT338 were also independent predictors of short OS in liver,lung,and stomach cancer.Conclusions:The elevated expression,promoter hypomethylation,and phosphorylation of PGK1 and PDHK1 were related with disease progression and short OS in diverse types of cancer.PGK1 and PDHK1 phosphorylation may be potential prognostic biomarkers. | Fei Shao Xueying Yang Wei Wang Juhong Wang Wei Guo Xiaoli Feng Susheng Shi Qi Xue Shugeng Gao Yibo Gao Zhimin Lu Jie He | 2019 | Cancer Communications2019,39,1: | 4 |
| 6 | Regulation of Leaf Longevity by DML3-Mediated DNA Demethylation显示文摘Leaf senescence is driven by the expression of senescence-associated genes(SAGs).Development-specific genes often undergo DNA demethylation in their promoter and other regions,which regulates gene expression.Whether and how DNA demethylation regulates the expression of SAGs and thus leaf senescence remain elusive.Whole-genome bisulfite sequencing(WGBS)analyses of wild-type(WT)and demeter-like 3(dml3)Arabidopsis leaves at three developmental stages revealed hypermethylation during leaf senescence in dml3 compared with WT,and 20556 differentially methylated regions(DMRs)were identified by comparing the methylomes of dml3 and WT in the CG,CHG,and CHH contexts.Furthermore,we identified that 335 DMR-associated genes(DMGs),such as NAC016 and SEN1,are upregulated during leaf senescence,and found an inverse correlation between the DNA methylation levels(especially in the promoter regions)and the transcript abundances of the related SAGs in WT.In contrast,in dml3 the promoters of SAGs were hypermethylated and their transcript levels were remarkably reduced,and leaf senescence was significantly delayed.Collectively,our study unraveled a novel epigenetic regulatory mechanism underlying leaf senescence in which DML3 is expressed at the onset of and during senescence to demethylate promoter,gene body or 3'UTR regions to activate a set of SAGs. | Lu Yuan Dan Wang Liwen Cao Ningning Yu Ke Liu Yongfeng Guo Susheng Gan Liping Chen | 2020 | Molecular Plant2020,13,8: | 3 |
| 7 | Arabidopsis ENOR3 regulates RNAi-mediated antiviral defense显示文摘Viruses can infect host plants to cause severe diseases and substantial agricultural loss, while plants have evolved RNA interference(RNAi) strategy to defend against viral infection. Despite enormous efforts, only a few host proteins in RNAi pathway were shown to mediate antiviral defense, including RNA-dependent RNA polymerase 1(RDR1), RDR6, DICER-LIKE 2(DCL2) and DCL4. In this study, we carried out a genetic screen for antiviral factors of RNAi pathway in Arabidopsis rdr6 background via inoculation with a 2 bdeficient Cucumber Mosaic Virus(CMV-D2b). We identified a mutant susceptible to CMV-D2b, referred to as enhancer of rdr6(enor) 3-1 rdr6, and found that ENOR3 encodes a functionally unknown protein with high homology to the mammalian Non Imprinted in Prader-Willi/Angelman(NIPA) magnesium transporters. ENOR3 inhibits accumulation of CMV-D2b and acts additively with RDR1, RDR6, DCL2 and DCL4 in antiviral defense. | 8 | Effects of Fertility and Density on Biomass Production,Translocation and Lodging Resistance of Millet(Setaria italica L.)in North China显示文摘In this study,the plant biomass production,biomass translocation rates across tissues and the lodging resistant-associated traits of millet( Setaria italica L.) in North China were investigated. Among the four summer millet cultivars,Baogu 19 exhibited improved plant biomass( PB) production at flowering and maturity stages,biomass translocation amount( BTA) from vegetative tissues to seeds during filling period,and lodging resistant-associated( LRA) traits compared with other cultivars,including enhanced stem lignin contents,increased anti-broken resistance( ABR),anti-puncturing resistance( APR),and stem diameter( SD) of plants. Compared with treatment regular cultivation( RC),high fertility treatment( HF) increased the plant BP,BTA from vegetative tissue to seed at filling stage,and the plant LRA traits; whereas high density treatment( HD) decreased the plant BP at plant level,plant BTA from vegetative tissues to seeds at filling stage,and the plant LRA traits. Correlation analysis revealed that stem ABR is significantly correlated with the plant lodging resistant-associated traits including APR and SD in the summer millet cultivars examined under various cultivation treatments.
