1Effect of temperature and moisture on soil organic carbon mineralization of predominantly permafrost peatland in the Great Hing'an Mountains,Northeastern China显示文摘Boreal peatlands represent a large global carbon pool.The relationships between carbon mineralization,soil temperature and moisture in the permafrost peatlands of the Great Hing'an Mountains,China,were examined.The CO2 emissions were measured during laboratory incubations of samples from four sites under different temperatures(5,10,15,and 20°C) and moisture contents(0%,30%,60%,100% water holding capacity(WHC) and completely water saturated).Total carbon mineralization ranged from 15.51 to 112.92 mg C under the treatments for all sites.Carbon mineralization rates decreased with soil depth,increased with temperature,and reached the highest at 60% WHC at the same temperature.The calculated temperature coefficient(Q10) values ranged from 1.84 to 2.51 with the soil depths and moisture.However,the values were not significantly affected by soil moisture and depth for all sites due to the different peat properties(P > 0.05).We found that the carbon mineralization could be successfully predicted as a two-compartment function with temperature and moisture(R2 > 0.96) and total carbon mineralization was significantly affected by temperature and moisture(P < 0.05).Thus,temperature and moisture would play important roles in carbon mineralization of permafrost peatlands in the Great Hing'an Mountains,indicating that the permafrost peatlands would be sensitive to the environment change,and the permafrost peatlands would be potentially mineralized under future climate change.Xianwei Wang Xiuzhen Li Yuanman Hu Jiujun Lv Ju Sun Zongmei Li Zhifeng Wu 2010Journal of Environmental Sciences2010,22,7:26
2A Pilot Study of the Therapeutic Efficacy and Mechanism of Artesunate in the MRL/lpr Murine Model of Systemic Lupus Erythematosus显示文摘Recent evidence indicates that artesunate has immunomodulatory properties that might be useful for treating autoimmune disease.In this study,we conducted a pilot study and explored the effect and mechanism of artesunate on the treatment of systemic lupus erythematosus using an MRL/lpr murine model.MRL/lpr mice were divided into control,cyclophosphamide(CTX)and artesunate treatment groups.Blood was collected to measure serum levels of creatinine,antinuclear antibody(ANA)and anti-double-stranded DNA(anti-dsDNA)antibody.Twenty-four-hour urine was collected to measure levels of proteinuria.The concentration of monocyte chemotactic protein-1(MCP-1)in serum and urine was measured.The expression of MCP-1 in kidney was detected by Western blot and immunohistochemistry assay,respectively.The expression of B cell activating factor(BAFF)in spleen was determined by real time-PCR and immunoblotting.We found that artesunate significantly increased the survival rate,body weight and blood leukocyte counts,and reduced the serum levels of ANA and anti-dsDNA antibody titer,24 h urinary protein,and serum creatinine.Our results indicated that artesunate could decrease MCP-1,major pro-inflammation cytokine,in serum,urine and kidney.We also found that the level of BAFF,the major B cell activation factor,was decreased in artesunate treated MRL/lpr mice.Its efficacy was comparable with that of CTX in this study.Taken together,we have demonstrated that artesunate can inhibit the progression of disease and reverse the pathologic lesion of lupus nephritis.Ouyang Jin Huayong Zhang Zhifeng Gu Shengnan Zhao Ting Xu Kangxing Zhou Bo Jiang Jie Wang Xiaofeng Zeng Lingyun Sun 2009Cellular & Molecular Immunology2009,6,6:20

4Metallic nanostructures for light trapping in energy-harvesting devices显示文摘Solar energy is abundant and environmentally friendly.Light trapping in solar-energy-harvesting devices or structures is of critical importance.This article reviews light trapping with metallic nanostructures for thin film solar cells and selective solar absorbers.The metallic nanostructures can either be used in reducing material thickness and device cost or in improving light absorbance and thereby improving conversion efficiency.The metallic nanostructures can contribute to light trapping by scattering and increasing the path length of light,by generating strong electromagnetic field in the active layer,or by multiple reflections/absorptions.We have also discussed the adverse effect of metallic nanostructures and how to solve these problems and take full advantage of the light-trapping effect.Chuan Fei Guo Tianyi Sun Feng Cao Qian Liu Zhifeng Ren 2014Light(Science & Applications)2014,3,1:14
5CD4^(+)CD25^(+)but not CD4^(+)Foxp3^(+) T cells as a regulatory subset in primary biliary cirrhosis显示文摘Increasing evidence indicates a role for regulatory T cells(Tregs)in the immune response and in autoimmune diseases,but the role of Tregs and cytokines in autoimmune hepatic diseases remains largely unclear and controversial,especially in patients with primary biliary cirrhosis(PBC).This study was undertaken to investigate Tregs and different cytokines in the liver and peripheral blood of PBC patients.We found that these patients demonstrated a reduction of CD4^(+)CD25^(+) T cells but elevated CD4^(+)Foxp3^(+) T cells in peripheral blood mononuclear cells(PBMCs)and CD41 T cells.The percentage of CD4^(+)CD25^(+) T cells in PBMCs was negatively correlated with elevated plasma interferon(IFN)-c levels.A liver-specific analysis showed that the frequency of Foxp31 Tregs,transforming growth factor(TGF)-b1 and IFN-c were increased in PBC patients.Our findings suggest that an imbalance between CD4^(+)CD25^(+) Tregs and cytotoxic cytokines plays a crucial role in the pathogenesis of PBC while the role of Foxp3 needs further investigation.Dandan Wang Huayong Zhang Jun Liang Zhifeng Gu Qiang Zhou Xiangshan Fan Yayi Hou Lingyun Sun 2010Cellular & Molecular Immunology2010,7,6:9
6Upper crustal velocity and seismogenic environment of the M7.0 Jiuzhaigou earthquake region in Sichuan, China显示文摘On August 8,2017,a magnitude 7.0 earthquake occurred in Jiuzhaigou County,Sichuan Province,China.The deep seismogenic environment and potential seismic risk in the eastern margin of Tibetan Plateau have once again attracted the close attention of seismologists and scholars at home and abroad.The post-earthquake scientific investigation could not identify noticeable surface rupture zones in the affected area;the complex tectonic background and the reason(s)for the frequent seismicity in the Jiuzhaigou earthquake region are unclear.In order to reveal the characteristics of the deep medium and the seismogenic environment of the M7.0 Jiuzhaigou earthquake region,and to interpret the tectonic background and genesis of the seismicity comprehensively,in this paper,we have reviewed all available observation data recorded by the regional digital seismic networks and large-scale,dense mobile seismic array(China Array)for the northern section of the North-South Seismic Belt around Jiuzhaigou earthquake region.Using double-difference seismic tomography method to invert the three-dimensional P-wave velocity structure characteristics of the upper crust around the Jiuzhaigou earthquake region,we have analyzed and discussed such scientific questions as the relationship between the velocity structure characteristics and seismicity in the Jiuzhaigou earthquake region,its deep tectonic environment,and the ongoing seismic risk in this region.We report that:the P-wave velocity structure of the upper crust around the Jiuzhaigoug earthquake region exhibits obvious lateral inhomogeneity;the distribution characteristics of the shallow P-wave velocity structure are closely related to surface geological structure and formation lithology;the M7.0 Jiuzhaigou earthquake sequence is closely related to the velocity structure of the upper crust;the mainshock of the M7.0 earthquake occurred in the upper crust;the inhomogeneous variation of the velocity structure of the Jiuzhaigou earthquake area and its surrounding medium appears to be the deep structural factor controlling the spatial distribution of the mainshock and its sequence.The 3D P-wave velocity structure also suggests that the crustal low-velocity layer of northeastern SGB(Songpan-GarzêBlock)stretches into MSM(Minshan Mountain),and migrates to the northeast,but the tendency to emerge as a shallow layer is impeded by the high-velocity zone of Nanping Nappe tectonics and the Bikou Block.Our results reveal an uneven distribution of high-and low-velocity structures around the Tazang segment of the East Kunlun fault zone.Given that the rupture caused by the Jiuzhaigou earthquake has enhanced the stress fields at both ends of the seismogenic fault,it is very important to stay vigilant to possible seismic hazards in the large seismic gap at the Maqu-Maqên segment of the East Kunlun fault zone.DaHu Li ZhiFeng Ding Yan Zhan PingPing Wu LiJun Chang XiangYu Sun 2021Earth and Planetary Physics2021,5,4:4
7Multiscale modeling of mechanical behavior and failure mechanism of 3D angle-interlock woven aluminum composites subjected to warp/weft directional tension loading显示文摘The mechanical behavior and progressive damage mechanism of novel aluminum matrix composites reinforced with 3D angle-interlock woven carbon fibers were investigated using a multiscale modeling approach.The mechanical properties and failure of yarns were evaluated using a microscale model under different loading scenarios.On this basis,a mesoscale model was developed to analyze the tensile behavior and failure mechanism of the composites.The interfacial decohesion,matrix damage,and failure of fibers and yarns were incorporated into the microscopic and mesoscopic models.The stress–strain curves and fracture modes from simulation show good agreement with the experimental curves and fracture morphology.Local interface and matrix damage initiate first under warp directional tension.Thereafter,interfacial failure,weft yarn cracking,and matrix failure occur successively.Axial fracture of warp yarn,which displays a quasi-ductile fracture characteristic,dominates the ultimate composites failure.Under weft directional tension,interfacial failure and warp yarn rupture occur at the early and middle stages.Matrix failure and weft yarn fracture emerge simultaneously at the final stage,leading to the cata-strophic failure of composites.The weft directional strength and fracture strain are lower than the warp directional ones because of the lower weft density and the more serious brittle fracture of weft yarns.Zhenjun WANG Siyuan YANG Shiping SUN Yingfeng ZHANG Changchun CAI Bowen XIONG Wei YANG Zhifeng XU Huan YU 2021Chinese Journal of Aeronautics2021,34,8:4

10Structural Insights into the CatalyticMechanism of a Plant Diterpene Glycosyltransferase SrUGT76G1显示文摘Diterpene glycosyltransferase UGT76G1 from Stevia rebaudiana (SrUGT76G1) is key to the generation ofeconomically important steviol glycosides (SGs), a group of natural sweeteners with high-intensity sweetness. SrUGT76G1 accommodates a wide range of steviol-derived substrates and many other small molecules. We report here the crystal structures of SrUGT76G1 in complex with multiple ligands to answer howthis enzyme recognizes diterpenoid aglycones and catalyzes the 1,3-sugar chain branching. A spaciouspocket for sugar-acceptor binding was observed from the determined SrUGT76G1 structures, which canexplain its broad substrate spectrum. Residues Gly87 and Leu204 lining the pocket play key roles in switching between diterpenoid and flavonoid glucosylation. An engineered mutant of SrUGT76G1, T284S, couldcatalyze a selectively increased production of next-generation sweetener rebaudioside M, with diminishedside product of rebaudioside I. 11Can the Grain-for-Green Program Really Ensure a Low Sediment Load on the Chinese Loess Plateau?显示文摘The Chinese Loess Plateau is the most seriously eroded area in the world and contributes the vast majority of the sediment that goes into the Yellow River.Since the 1950s,progressive soil and water conservation measures have been implemented—in particular,large-scale ecological restoration has been ongoing since 1999—resulting in a significant reduction of the sediment load.However,the mechanism of the sediment transport dynamics is not fully understood due to multiple and complicated influencing factors including climate change and human activities(e.g.,ecological restoration).A challenging question,then,arises:Is the current low sediment level a“new normal”in this era and in the future?To address this question,we selected a typical loess hilly region where considerable ecological restoration has been implemented,and which is regarded as the site of the first and most representative Grainfor-Green program in the Loess Plateau.We investigated the evolution of discharge–sediment relationships in the past decades(1960–2010)and their association with the soil and water conservation measures in this area.The results showed that there was a distinct change in the regression parameters of the commonly used annual discharge–sediment regression equation—a continuously increasing trend of parameter b and a decreasing trend of parameter a,accompanying the ecological restoration.The increase in exponent b(i.e.,a steeper slope)implies a potential lower sediment load resulting from low discharge and a potential higher sediment load resulting from large discharge.This finding may question the new normal of a low sediment level and implies the potential risk of a large sediment load during extremely wet years.Pengcheng Sun Yiping Wu Zhifeng Yang Bellie Sivakumar Linjing Qiu Shuguang Liu Yanpeng Cai 2019Engineering2019,5,5:3
12A novel entry point for pedicle screw placement in the thoracic spine显示文摘This study was aimed to introduce a novel entry point for pedicle screw fixation in the thoracic spine and compare it with the traditional entry point. A novel entry point was found with the aim of improving accuracy, safety and stability of pedicle screw technique based on anatomical structures of the spine. A total of 76 pieces of normal thoracic CT images at the transverse plane and the thoracic pedicle anatomy of 6 cadaveric specimens were recruited.Transverse pedicle angle(TPA), screw length, screw placement accuracy rate and axial pullout strength of the two different entry point groups were compared. There were significant differences in the TPA, screw length, and the screw placement accuracy rate between the two groups(P<0.05). The maximum axial pullout strength of the novel entry point group was slightly larger than that of the traditional group. However, the difference was not significant(P > 0.05). 16Research on Open-circuit Fault Tolerant Control of Six-phase Permanent Magnet Synchronous Machine Based on Fifth Harmonic Current Injection显示文摘This paper proposes a novel control approach for fault-tolerant control of dual three-phase permanent magnet synchronous motor(PMSM) under one-phase open-circuit fault.A modified six-phase static coordinate transformation matrix and an extended rotating coordinate transformation matrix are investigated considering the influence of the fifth harmonic space on fault-tolerant control. These mathematical models are further analyzed in the fundamental space and the fifth harmonic space after the fault and to eliminate the coupling between the d-q axis voltage equation in the fundamental wave space and the d-q axis voltage equation in the fifth harmonic space, a secondary rotation coordinate transformation matrix is proposed. To achieve the purpose of reducing torque ripple, the fault-tolerant control method proposed in this paper not only takes the minimum copper loss as the constraint condition, but also injects the fifth harmonic current. 17Rifampicin inhibits apoptosis in rotenone-induced differentiated PC12 cells by ameliorating mitochondrial oxidative stress显示文摘BACKGROUND:Previous studies have shown that rifampicin exhibits neuroprotective effects,but the precise mechanisms remain unclear.Rifampicin is thought to exert the neuroprotective effect as a hydroxyl free radical scavenger.OBJECTIVE:To investigate the protective effects of rifampicin pretreatment on rotenone-induced mitochondrial oxidative stress in differentiated PC12 cells.DESIGN,TIME AND SETTING:A repeated measure,cell-based study was performed at the Department of Neurology,Second Affiliated Hospital,Sun Yat-sen University,China between December 2007 and November 2008.MATERIALS:PC12 cells were a kind gift from the Physiology Laboratory of Zhongshan Medical School,Sun Yat-sen University,China.Rotenone and rifampicin were purchased from Sigma,USA.METHODS:PC12 cells were differentiated by culturing with 100 ng/mL 7S nerve growth factor for 9 days in Dulbecco's modified Eagle's medium/Nutrient Mix F12(DMEM/F12) supplemented with 10% fetal bovine serum.The cells were assigned to six groups according to various treatment conditions:control,cultured with normal media;rifampicin group,treated with 300 μmol/L rotenone for 26 hours;rotenone group,treated with 2.5 μmol/L rotenone for 24 hours;rifampicin pretreatment groups,pretreated with 100,200,and 300 μmol/L rifampicin for 2 hours,respectively,followed by 2.5 μmol/L rotenone for 24 hours.MAIN OUTCOME MEASURES:Mitochondrial membrane potential was measured by fluorescence microscopy and flow cytometry,respectively,using rhodamine123 staining.Intracellular reactive oxygen species formation was analyzed by flow cytometry using 2',7'-dichlorofluorescin-diacetate staining,and intracellular reduced glutathione was measured with a microplate reader.Cell viability was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay.Cell apoptosis was detected by Hoechst 33342 staining and flow cytometry.RESULTS:Increased apoptosis in rotenone-induced,differentiated,PC12 cells was accompanied by the loss of mitochondrial transmembrane potential,the formation of reactive oxygen species,and reduced glutathione depletion(P<0.01).Rotenone-induced mitochondrial dysfunction was blocked in a dose-dependent manner by rifampicin(P<0.05 or P<0.01).CONCLUSION:Pretreatment of differentiated PC12 cells with rifampicin blocked rotenone-induced apoptosis by ameliorating mitochondrial dysfunction and oxidative stress.Shiwen Chen Yuanlin Sun Zhifeng Zeng Enxiang Tao 2010Neural Regeneration Research2010,5,4:2

19Microvascular endothelial cells derived from spinal cord promote spinal cord injury repair显示文摘Neural regeneration after spinal cord injury (SCI) closely relates to the microvascular endothelial cell (MEC)- mediated neurovascular unit formation. However, the effects of central nerve system-derived MECs on neovascularization and neurogenesis, and potential signaling involved therein, are unclear. Here, we established a primary spinal cord-derived MECs (SCMECs) isolation with high cell yield and purity to describe the differences with brain-derived MECs (BMECs) and their therapeutic effects on SCI. Transcriptomics and proteomics revealed differentially expressed genes and proteins in SCMECs were involved in angiogenesis, immunity, metabolism, and cell adhesion molecular signaling was the only signaling pathway enriched of top 10 in differentially expressed genes and proteins KEGG analysis. SCMECs and BMECs could be induced angiogenesis by different stiffness stimulation of PEG hydrogels with elastic modulus 50-1650 Pa for SCMECs and 50-300 Pa for BMECs, respectively. Moreover, SCMECs and BMECs promoted spinal cord or brain-derived NSC (SNSC/BNSC) proliferation, migration, and differentiation at different levels. At certain dose, SCMECs in combination with the NeuroRegen scaffold, showed higher effectiveness in the promotion of vascular reconstruction. The potential underlying mechanism of this phenomenon may through VEGF/AKT/eNOS- signaling pathway, and consequently accelerated neuronal regeneration and functional recovery of SCI rats compared to BMECs.
