2Secretory/releasing proteome-based identification of plasma biomarkers in HBV-associated hepatocellular carcinoma显示文摘For successful therapy, hepatocellular carcinoma (HCC) must be detected at an early stage. Herein, we used a proteomic approach to analyze the secretory/releasing proteome of HCC tissues to identify plasma biomarkers. Serum-free conditioned media (CM) were collected from primary cultures of cancerous tissues and surrounding noncancerous tissues. Proteomic analysis of the CM proteins permitted the identification of 1365 proteins. The enriched molecular functions and biological processes of the CM proteins, such as hydrolase activity and catabolic processes, were consistent with the liver being the most important metabolic organ. Moreover, 19% of the proteins were characterized as extracellular or membrane-bound. For validation, secretory proteins involved in transforming growth factor-β signaling pathways were validated in plasma samples. Alphafetoprotein (AFP), metalloproteinase (MMP)1, osteopontin (OPN), and pregnancy-specific beta-1-glycoprotein (PSG)9 were significantly increased in HCC patients. The overall performance of MMP1 and OPN in the diagnosis of HCC remained greater than that of AFP. In addition, this study represents the first report of MMP1 as a biomarker with a higher sensitivity and specificity than AFP. Thus, this study provides a valuable resource of the HCC secretome with the potential to investigate serological biomarkers. 3Heavy mineral assemblage characteristics and the Cenozoic paleogeographic evolution in southwestern Qaidam Basin显示文摘Based on the analysis of heavy mineral assemblages in Cenozoic southwestern Qaidam Basin, we found that different areas have variable heavy mineral assemblage characteristics, which suggested that there were two source areas—the Altyn Mountains and the Qimen Tagh-East Kunlun Mountains. In Ganchaigou-Shizigou-Huatugou(Area A), which was mainly source from the Altyn Mountains, its heavy minerals were mainly composed of zircon, Ti-oxides, and wollastonite in the Paleoceneearly Eocene and mainly of unstable minerals, especially amphibole, in the middle Eocene-Oligene. Since the late OligoceneMiocene, the heavy minerals were still mainly unstable minerals, but the content of epidote increased and the content of amphibole decreased. In Qigequan-Hongliuquan(Area B), which was the mixed source from the Altyn Mountains and the Qimen Tagh-East Kunlun Mountains, its heavy minerals were mainly garnet, epidote, and amphibole. The source of LücaotanDongchaishan-Kunbei(Area C) was mainly from the Qimen Tagh-East Kunlun Mountains, heavy minerals in the sediments in Area C were mainly zircon and Ti-oxides in Paleogene and garnet, epidote, and amphibole in Neogene. In Yuejin-Youshashan(Area D), where the stable minerals and unstable minerals were present simultaneously, the heavy mineral assemblages was controlled by multi-direction source. The variation of heavy mineral assemblages in southwestern Qaidam Basin shows that Altyn Mountains was of low-lying topographic relief in Paleocene-early Eocene, and the rapid uplift of Altyn Mountains started from the middle Eocene. In Paleogene, the Altyn Tagh Fault had a slow strike-slip velocity, but the strike-slip velocity increased greatly since the late Oligocene, leading to a strike-slip displacement above 300 km since Neogene. 4The Structure and Formation Mechanism of a Sea Fog Event over the Yellow Sea显示文摘In this paper, a heavy sea fog event occurring over the Yellow Sea on 11 April 2004 was investigated based upon observational and modeling analyses. From the observational analyses, this sea fog event is a typical advection cooling case. Sea surface temperature(SST) and specific humidity(SH) show strong gradients from south to north, in which warm water is located in the south and consequently, moisture is larger in the south than in the north due to evaporation processes. After fog formation, evaporation process provides more moisture into the air and further contributes to fog evolution. The sea fog event was reproduced by the Regional Atmospheric Modeling System(RAMS) reasonably. The roles of important physical processes such as radiation, turbulence as well as atmospheric stratification in sea fog's structure and its formation mechanisms were analyzed using the model results. The roles of long wave radiation cooling, turbulence as well as atmospheric stratification were analyzed based on the modeling results. It is found that the long wave radiative cooling at the fog top plays an important role in cooling down the fog layer through turbulence mixing. The fog top cooling can overpower warming from the surface. Sea fog develops upward with the aid of turbulence. The buoyancy term, i.e., the unstable layer, contributes to the generation of TKE in the fog region. 5Identification of ExpIdentification of Expressed Resistance Gene Analogs from Peanut (Arachis hypogaea L.) Expressed Sequence Tags显示文摘Low genetic diversity makes peanut (Arachis hypogaea L.) very vulnerable to plant pathogens, causing severe yield loss and reduced seed quality. Several hundred partial genomic DNA sequences as nucleotide-binding-site leucine-rich repeat (NBS-LRR) resistance genes (R) have been identified, but a small portion with expressed transcripts has been found. We aimed to identify resistance gene analogs (RGAs) from peanut expressed sequence tags (ESTs) and to develop polymorphic markers. The protein sequences of 54 known R genes were used to identify homologs from peanut ESTs from public databases. A total of 1,053 ESTs corresponding to six different classes of known R genes were recovered, and assembled 156 contigs and 229 singletons as peanut-expressed RGAs. There were 69 that encoded for NBS-LRR proteins, 191 that encoded for protein kinases, 82 that encoded for LRR-PK/transmembrane proteins, 28 that encoded for Toxin reductases, 11 that encoded for LRR-domain containing proteins and four that encoded for TM-domain containing proteins. Twenty-eight simple sequence repeats (SSRs) were identified from 25 peanut expressed RGAs. One SSR polymorphic marker (RGA121) was identified. Two polymerase chain reaction-based markers (Ahsw-1 and Ahsw-2) developed from RGA013 were homologous to the Tomato Spotted Wilt Virus (TSWV) resistance gene. All three markers were mapped on the same linkage group AhIV. 6Multistage Enrichment of the Sawafuqi Uranium Deposit: New Insights into Sandstone-hosted Uranium Deposits in the Intramontane Basins of Tian Shan, China显示文摘Meso-Cenozoic intracontinental orogenic processes in the Tian Shan orogenic belt have significant effect on the sandstone-hosted uranium deposits in the intramontane basins and those adjacent to the orogen. The Sawafuqi uranium deposit, which is located in the South Tian Shan orogenic belt, is investigated to reveal the relationships between uranium mineralization and orogenies. Recent exploration results show that the Sawafuqi uranium deposit has tabular, stratiform, quasi-stratiform, and lens-like orebodies and various geological characteristics different from typical interlayer oxidation zone sandstone-hosted uranium deposits. Systematic studies of ore samples from the Sawafuqi uranium deposit using a variety of techniques, including thin section observation, α-track radiograph, electron microprobe and scanning electron microscope, suggest that uranium mineralization is closely related to pyrite and organic matter. Mineralization-related alterations in the host rocks are mainly silicification and argillation including kaolinite, illite(and illite-smectite mixed layer) and chlorite. Tree stages of mineralization were identified in the Sawafuqi uranium deposit:(i) uranium-bearing detritus and synsedimentary initial pre-enrichment;(ii) interlayer oxidization zone uranium mineralization; and(iii) vein-type uranium mineralization. The synsedimentary uranium pre-enrichment represents an early uranium enrichment in the Sawafuqi uranium deposit, and interlayer oxidation zone uranium mineralization formed the main orebodies, which are superimposed by the vein-type uranium mineralization. Combining the results of this study with previous studies on the Meso-Cenozoic orogenies of South Tian Shan, it is proposed that the synsedimentary uranium pre-enrichment of the Sawafuqi uranium deposit was caused by Triassic Tian Shan uplift, and the interlayer oxidation zone uranium mineralization occurred during the Eocence-Oligocene period, when tectonism was relatively quiet, whereas the vein-type uranium mineralization took place in relation to the strong orogeny of South Tian Shan since Miocene.LIU Zhangyue PENG Suping QIN Mingkuan LIU Hongxun HUANG Shaohua HE Zhongbo GUO Qiang XU Qiang SONG Jiye 2017Acta Geologica Sinica(English Edition)2017,91,6:3
7Mechanism of Silurian Hydrocarbon Pool Formation in the Tarim Basin显示文摘There are three formation stages of Silurian hydrocarbon pools in the Tarim Basin. The widely distributed asphaltic sandstones in the Tazhong (central Tarim) and Tabei (northern Tarim) areas are the results of destruction of hydrocarbon pools formed in the first-stage,and the asphaltic sandstones around the Awati Sag were formed in the second-stage. 9Rapid assessment of DNA damage induced by polystyrene nanosphere suspension using a photoelectrochemical DNA sensor显示文摘Nanomaterials have been used increasingly in a wide variety of applications, and some of them have shown toxic effects on experimental animals and cells. In this study, a previously established photoelectrochemical DNA sensor was employed to rapidly detect DNA damage induced by polystyrene nanosphere (PSNS) suspensions. In the sensor, a double-stranded DNA film was assembled on a semiconductor electrode, and a DNA intercalator, Ru(bpy)2(dppz)2+ (bpy = 2,2'-bipyridine, dppz = dipyrido[3,2-a:2',3'-c]phenazine) was used as the photoelectrochemical signal indicator. After the DNA-modified electrode was exposed to 2.0 mg/mL PSNS suspension, photocurrent of DNA-bound Ru(bpy)2(dppz)2+ decreased by about 20%. The decrease is attributed to the chemical damage of DNA and consequently less binding of Ru(bpy)2(dppz)2+ molecules to the electrode. Gel electrophoresis of DNA samples incubated with PSNS suspension confirmed DNA damage after the chemical exposure. However, in both photoelectrochemical and gel electrophoresis experiments, extensively washed PSNS did not induce any DNA damage, and the supernatant of PSNS suspension exhibited comparable DNA damage as the unwashed PSNS suspension. Furthermore, UV-visible absorption spectrum of the supernatant displayed a pattern very similar to that of styrene oxide (SO), a compound which has been shown to induce DNA damage by forming covalent DNA adducts. It is therefore suggested that styrene oxide and other residual chemicals in the PSNS may be responsible for the observed DNA damage. 10Understanding tissue-specificity with human tissue-specific regulatory networks显示文摘Tissue-specificity is important for the function of human body. However, it is still not clear how the functional diversity of different tissues is achieved. Here we construct gene regulatory networks in 13 human tissues by integrating large-scale transcription factor(TF)-gene regulations with gene and protein expression data. By comparing these regulatory networks, we find many tissue-specific regulations that are important for tissue identity. In particular, the tissue-specific TFs are found to regulate more genes than those expressed in multiple tissues, and the processes regulated by these tissue-specific TFs are closely related to tissue functions.Moreover, the regulations that are present in certain tissue are found to be enriched in the tissue associated disease genes, and these networks provide the molecular context of disease genes. 13Expression of p73, ER and PR in esophageal carcinoma and precancerous lesions显示文摘Objective:To test the expression of mutant p73,p53,ER and PR proteins in the esophageal normal mucosa,hyperplasia,dysplasia and squamous cell carcinoma,and research the clinically pathological significance and the correlation,for the early diagnosis,prognostic measure and therapy in clinic.Methods:With Immunohistochemistry,it was examined to show these tumor markers’ expression in different epithelial lesions of 40 esophageal squamous cell carcinomas,14 dyspla-sias,14 hyperplasias and 14 normal mucosas.Results:The expression of p73 was 55%,21%,0% and 0% in the esophageal carcinoma,dysplasia,hyperplasia and normal mucosa,respectively.The significant difference in expression of p73(P<0.001)was observed between the esophageal normal mucosia,hyperplasia,dysplasia and esophageal squamous cell carcinoma with Fisher’s exact test.Difference in expression of p73(P<0.05)was observed between the esophageal squamous cell carcinoma and dysplasia with χ2 test.The expression of p73 showed non-correlation with the patient’s age,sex,tumor’s grade,lymph-node metastasis and invasive depth(P>0.05);Similarly,the expression of mutant p53 was 67.5%,35.7%,7% and 0%,respectively;In like manner,the expression of ER was 55%,21.4%,14.2% and 0%,respectively;The expression of PR was 57.5%,14.28%,0% and 0%,respectively.The significant difference in expression of PR(P<0.001)was observed with Fisher’s exact test.Difference in expression of PR(P<0.05)was observed between the esophageal squamous cell carcinoma and dysplasia with χ2 test.The expression of PR(P<0.05)was correlated with lymph-node metastasis,and showed non-correlation with the patient’s age,sex,tumor’s grade,and invasive depth(P>0.05).Moreover,over-expression of mutant p53 and p73 showed significant correlation with ER and PR protein’s positive expression(P<0.05).Conclusion:P73 protein may become a new tumor’s marker to diag-nose esophageal squamous cell carcinoma.Because the expression of p73 protein was closely correlated with ER and PR,they could be simultaneously examined to help to early diagnose,prognosticate and cure esophageal carcinoma.Sheng Fan Jie Ai Jun Wang Jiani Fan Xiaoguang Guo Suping Guo Xiaofeng Liu Jianping Wei 2006The Chinese-German Journal of Clinical Oncology2006,5,6:1

17Status of Stratigraphy Research in China显示文摘Scientific research and productive practice for earth history are inseparable from the accurate stratigraphic framework and time framework. Establishing the globally unified, precise and reliable chronostratigraphic series and geological time series is the major goal of the International Commission on Stratigraphy(ICS). Under the leadership of the ICS, the countries around the world have carried out research on the Global Standard Stratotype-section and Points(GSSPs) for the boundaries of chronostratigraphic systems. In the current International Chronostratigraphic Chart(ICC), 65 GSSPs have been erected in the Phanerozoic Eonothem, and one has yet been erected in the Precambrian Eonothem. Based on the progress of research on stratigraphy especially that from its subcommissions, the ICS is constantly revising the ICC, and will publish a new International Stratigraphic Guide in 2020. After continual efforts and broad international cooperation of Chinese stratigraphers, 10 GSSPs within the Phanerozoic Eonothem have been approved and ratified to erect in China by the ICS and IUGS. To establish the standards for stratigraphic division and correlation of China, with the support from the Ministry of Science and Technology, the National Natural Science Foundation of China and the China Geological Survey, Chinese stratigraphers have carried out research on the establishment of Stages in China. A total of 102 stages have been defined in the 'Regional Chronostratigraphic Chart of China(geologic time)', in which 59 stages were studied in depth. In 2014, the 'Stratigraphic Chart of China' was compiled, with the essential contents as follows: the correlation between international chronostratigraphy and regional chronostratigraphy of China(geologic time), the distributive status of lithostratigraphy, the characteristics of geological ages, the biostratigraphic sequence, the magnetostratigraphy, the geological events and eustatic sea-level change during every geological stage. The 'Stratigraphical Guide of China and its Explanation(2014)' was also published. Chinese stratigraphers have paid much attention to stratigraphic research in south China, northeast China, north China and northwest China and they have made great achievements in special research on stratigraphy, based on the 1:1000000, 1:250000, 1:200000 and 1:50000 regional geological survey projects. Manifold new stratigraphic units were discovered and established by the regional geological surveys, which are helpful to improve the regional chronostratigraphic series of China. On the strength of the investigation in coastal and offshore areas, the status of marine strata in China has been expounded. According to the developing situation of international stratigraphy and the characteristics of Chinese stratigraphic work, the contrast relation between regional stratigraphic units of China and GSSPs will be established in the future, which will improve the application value of GSSPs and the standard of regional stratigraphic division and correlation. In addition, the study of stratigraphy of the Precambrian, terrestrial basins and orogenic belts will be strengthened, the Stratigraphic Chart of China will be improved, the typical stratigraphic sections in China will be protected and the applied study of stratigraphy in the fields of oil and gas, solid minerals, etc. will be promoted. 19A Dual Endogenous Stimuli-Responsive Framework Nucleic Acid Nanodevice for MicroRNA Imaging显示文摘Framework nucleic acids(FNAs)have emerged as intelligent sensing systems for the detection of tumor-related biomarkers in living cells.However,orthogonally controlled manipulation of FNAs-based nanodevices for on-site imaging of microRNAs(miRNAs)remains an intractable challenge.Herein,we report a dual endogenous stimuli-responsive FNA nanodevice that can perform miRNA sensing and imaging in a tumor-specificmanner.The sensing function of the nanodevice is silent(OFF)by rationally incorporating adenosine 5′-triphosphate(ATP)aptamer and an abasic site,which can also be activated(ON)with ATP and human apurinic/apyrimidinic endonuclease 1(APE1)in tumor cells,enabling on-site and efficient miRNA imaging with improved tumor specificity.Furthermore,we demonstrate the capability of the nanodevice for specificmiRNAimagingboth in living cells and in vivo based on the“dual keys”(overexpressed ATP and APE1 in tumors)priming mode.Therefore,this work illustrates a simple biosensing methodology with great potential for precise imaging of tumor biomarkers in clinical diagnosis and therapeutic evaluation.Lie Li Haiyan Chen Xiaohong Wen Suping Li Mei Yang Qiuping Guo Kemin Wang 2023CCS Chemistry2023,5,10:0
20Extracellular ATP-activated hybridization chain reaction for accurate and sensitive detection of cancer cells显示文摘Accurate and sensitive detection of caner cells is of significant importance for early diagnosis and treat-ment of cancer.Here,we developed an extracellular ATP-dctivated hybridization chain reaction(HCR)amplification strategy to meet this purpose.This strategy relies on three DNA probes,Apt-trigger,H1-AТP aptamer duplex and hairpin H2.The Apt-trigger probe consists of two com sequence for specific recognition of the target cells.and a trigger sequence for the HCR assembly.Theроnents:an aptamer duplex structure of H1-ATP aptamer causes the tochold in hairpin H1 to be hidden,preventing the strand-ent displacement reaction between haipin H1 and Apt-trigger.Upon activation with ATP the ATP aptamer will blnd to ATP to dissoci iate from hairpin H1,thus leading to an Apt-trigger-induced strand-displacement reaction and subsequent HCR with hairpin H2 on the target cell surface.Benefiting from aptamer recogni-tion and ATP-activated HCR amplification,this strategy can not only perform sensitive quantitative anal-ysis with a detection limit of 25 cells in 200 ul.of binding buffer,but also show desirable specificity and accuracy for identifylng target cells from control cells and mixed cell samples,Imporantly,this method retains stable and good perfor mance for target cell detection in 10%fetal bovine ser rum,den onstrating great potential for clinical diagnosis in complex biological matrices.Furthermore,this strategy can be adapted to detect various types of cancer cells by changing the corresponding aptamer sequence.Lie Li Suping Li Jie Wang Xiaohong Wen Mei Yang Haiyan Chen Qiuping Guo Kemin Wang 2023Chinese Chemical Letters2023,34,12:0
