| 1 | 粪菌移植应用于慢传输型便秘的临床疗效及疗效影响因素:一项前瞻性研究的长期随访结果显示文摘背景和目的肠道菌群参与结肠动力的调控,菌群失调是便秘是病因之一。粪菌移植被证实可以恢复肠道平衡。本研究旨在评估粪菌移植治疗慢传输型便秘(STC)的临床疗效和预后影响因素。方法:52例STC患者接受标准的粪菌移植入组研究,对其进行至少6个月的随访。记录入组患者的排便习惯、结肠传输时间、便秘相关的症状(PAC-SYM评分)、生活质量(PAC-QOL评分)、治疗满意度和不良反应。主要疗效指标是平均每周完全自主排便≥3次的患者比例。结果:治疗后34周、912周、2124周,每周完全自主排便≥3次患者的比例分别为50.0%、38.5%和32.7%,均较治疗前显著提高(均P<0.01)。其他排便情况、结肠传输时间、便秘相关症状和生活质量方面,治疗后亦有显著改善;但这些短期改善在治疗后12周和24周逐渐减小。治疗前排便不尽是唯一的疗效影响因素。治疗和随访期间未发现任何治疗相关的不良反应。结论:本研究显示粪菌移植治疗STC安全有效,但远期疗效有所降低。治疗前排便不尽感较少甚至没有的患者接受粪菌移植的治疗效果更佳。 | Chao Ding Wenting Fan Lili Gu Hongliang Tian Xiaolong Ge Jianfeng Gong Yongzhan Nie Ning Li | 2018 | Gastroenterology Report2018,6,2: | 7 |
| 2 | Characterization of the soybean Gm ALMT family genes and the function of Gm ALMT5 in response to phosphate starvation显示文摘A potential mechanism to enhance utilization of sparingly soluble forms of phosphorus(P) is the root secretion of malate, which is mainly mediated by the ALMT gene family in plants. In this study, a total of 34 Gm ALMT genes were identified in the soybean genome. Expression patterns diverged considerably among Gm ALMTs in response to phosphate(Pi) starvation in leaves, roots and flowers, with expression altered by P availability in 26 of the 34 Gm ALMTs. One root-specific Gm ALMT whose expression was significantly enhanced by Pi-starvation,Gm ALMT_5, was studied in more detail to determine its possible role in soybean P nutrition. Analysis of Gm ALMT_5 tissue expression patterns, subcellular localization, and malate exudation from transgenic soybean hairy rootsoverexpressing Gm ALMT_5, demonstrated that Gm ALMT_5 is a plasma membrane protein that mediates malate efflux from roots. Furthermore, both growth and P content of transgenic Arabidopsis overexpressing Gm ALMT_5 were significantly increased when sparingly soluble Ca-P was used as the external P source. Taken together, these results indicate that members of the soybean Gm ALMT gene family exhibit diverse responses to Pi starvation. New abilities allow in vivo cancer imaging with sensitivity and accuracy that are unprecedented in conventional imaging approaches. The visualization of cellular and molecular behaviors and events within tumors in living subjects is improving our deeper understanding of tumors at a systems level. These advances are being rapidly used to acquire tumor-to-tumor molecular heterogeneity, both dynamically and quantitatively, as well as to achieve more effective therapeutic interventions with the assistance of real-time imaging. | 6 | Assessing the conservation impact of Chinese indigenous chicken populations between ex-situ and in-situ using genome-wide SNPs显示文摘Conservation programs require rigorous evaluation to ensure the preservation of genetic diversity and viability of conservation populations. In this study, we conducted a comparative analysis of two indigenous Chinese chicken breeds, Gushi and Xichuan black-bone, using whole-genome SNPs to understand their genetic diversity, track changes over time and population structure. The breeds were divided into five conservation populations(GS1, 2010, ex-situ;GS2, 2019, ex-situ;GS3, 2019, in-situ;XB1, 2010, in-situ;and XB2, 2019, in-situ) based on conservation methods and generations. The genetic diversity indices of three conservation populations of Gushi chicken showed consistent trends, with the GS3 population under in-situ strategy having the highest diversity and GS2 under ex-situ strategy having the lowest. The degree of inbreeding of GS2 was higher than that of GS1 and GS3. Conserved populations of Xichuan black-bone chicken showed no obvious changes in genetic diversity between XB1 and XB2. In terms of population structure, the GS3 population were stratified relative to GS1 and GS2. According to the conservation priority, GS3 had the highest contribution to the total gene and allelic diversity in GS breed, whereas the contribution of XB1 and XB2 were similar. We also observed that the genetic diversity of GS2 was lower than GS3, which were from the same generation but under different conservation programs(in-situ and ex-situ). While XB1 and XB2 had similar levels of genetic diversity. | 10 | Penalized M-Estimation Based on Standard Error Adjusted Adaptive Elastic-Net显示文摘When there are outliers or heavy-tailed distributions in the data, the traditional least squares with penalty function is no longer applicable. In addition, with the rapid development of science and technology, a lot of data, enjoying high dimension, strong correlation and redundancy, has been generated in real life. So it is necessary to find an effective variable selection method for dealing with collinearity based on the robust method. This paper proposes a penalized M-estimation method based on standard error adjusted adaptive elastic-net, which uses M-estimators and the corresponding standard errors as weights. The consistency and asymptotic normality of this method are proved theoretically. For the regularization in high-dimensional space, the authors use the multi-step adaptive elastic-net to reduce the dimension to a relatively large scale which is less than the sample size, and then use the proposed method to select variables and estimate parameters. Finally, the authors carry out simulation studies and two real data analysis to examine the finite sample performance of the proposed method.
