| 1 | Differential gene expression in nontransgenic and transgenic“M.26”apple overexpressing a peach CBF gene during the transition from eco-dormancy to bud break显示文摘The CBF signal pathway is responsible for a significant portion of plant responses to low temperature and freezing.Overexpression of CBF genes in model organisms such as Arabidopsis thaliana enhances abiotic stress tolerance but also reduces growth.In addition to these effects,overexpression of the peach(Prunus persica[L.]Batsch)CBF1 gene in transgenic apple(Malus x domestica Borkh.)line T166 also results in early entry into and late exit from dormancy.Although the regulation of dormancy-induction and dormancy-release occur while the CBF regulon is operative in perennial,woody plants,how overexpression of CBF1 affects these dormancy-related changes in gene expression is incompletely understood.The objective of the present study was to characterize global changes in gene expression in peach CBF1-overexpressing and non-transformed apple bark tissues at different states of dormancy via RNA-seq.RNAseq bioinformatics data was confirmed by RT-qPCR on a number of genes.Results indicate that the greatest number of significantly differentially expressed genes(DEGs)occurred in April when dormancy release and bud break normally occur but are delayed in Line T166.Genes involved in storage and inactivation of auxin,GA,and cytokinin were generally upregulated in T166 in April,while those for biosynthesis,uptake or signal transduction were generally downregulated in T166.Genes for cell division and cambial growth were also downregulated in T166 relative to the non-transformed line.These data suggest that overexpression of the peach CBF1 gene impacts growth hormone homeostasis and as a result the activation of growth in the spring,and most likely growth cessation in the fall as well. | Timothy Artlip Adam McDermaid Qin Ma Michael Wisniewski | 2019 | Horticulture Research2019,6,1: | 6 |

| 7 | Evaluation of the association of C5 with neovascular age-related macular degeneration and polypoidal choroidal vasculopathy显示文摘Background:Neovascular age-related macular degeneration(AMD)and polypoidal choroidal vasculopathy(PCV)are sight-threatening maculopathies with both environmental and genetic risk factors.We have previously shown relative risks posed by genes of the complement pathways to neovascular AMD and PCV.Methods:In this study,we investigated the haplotype-tagging single nucleotide polymorphisms(SNPs)in the complement component 5(C5)gene in 708 unrelated Chinese individuals:200 neovascular AMD patients,233 PCV patients and 275 controls.Six tagging SNPs in C5 were genotyped.Univariate single SNP association analysis,haplotype-based association analysis and gene-gene interaction analysis between C5 and other AMD-associated genes were performed.Results:The results revealed none of the six tagging SNPs of the C5 gene had a significant association with neovascular AMD or PCV(P>0.05).We also found insignificant haplotype-based association,and no significant SNPSNP interaction between C5 and other genes(including C2-CFB-RDBP-SKIV2L,SERPING1,CETP,ABCG1,PGF,ANGPT2,CFH and HTRA1)for neovascular AMD and PCV.Conclusions:This study showed no statistical significance in the genetic association of C5 with neovascular AMD or PCV in a Hong Kong Chinese population.Further studies in large samples from different populations are warranted to elucidate the role of C5 in the genetic susceptibility of AMD and PCV. | Ke Liu Li Ma Timothy Y.Y.Lai Marten E.Brelen Pancy O.S.Tam Clement C.Tham Chi Pui Pang Li Jia Chen | 2019 | Eye and Vision2019,6,1: | 1 |

| 9 | Friction-based riveting technique for AZ31 magnesium alloy显示文摘A new friction-based riveting technique, Rotating Hammer Riveting(RHR), is demonstrated to fully form AZ31 Mg rivet heads in a mere 0.23 s. Heat and pressure generated through severe plastic deformation during the process was sufficient to form the Mg rivet head without the need for a pre-heating operation. Due to preliminary twinning and followed by dynamic recrystallization, AZ31 Mg grains in the rivet head were refined during RHR, which enhance the formability of Mg rivets by triggering grain boundary sliding and reducing plastic anisotropy of Mg. Methods: Immunohistochemical assays and functional analysis on the immune system were performed on RTVP-/- mice and RTVP-1+/+ mice. Protein-protein interaction (PPI) was used to predict the functions of RTVP-1. Results: Abnormal lymphocyte growth kinetics and reduced numbers of CD8+ T cells were revealed in lymph nodes of RTVP-1-/- mice. Expression of phenotypic markers of maturation in bone marrow-derived dendritic cells (DC) was impaired in RTVP-1-/- DC following antigenic stimulation in vitro and RTVP-1-/- DC failed to provide normal CD4+ T cell stimulatory activities in vivo. RTVP-1-/- mice failed to generate normal CTL or antibody responses in vivo after vaccination. In vivo tumor challenge experiments using a mouse cancer cell line demonstrated that the growth rate of subcutaneous tumors in RTVP-1-/- mice was significantly increased and CD8+ T cell infiltration significantly reduced compared to RTVP-1+/+ mice. PPI showed that RTVP-1 protein closely interacted with molecules associated with immune response and cancer metastasis.
