| 2 | Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual. | Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang | 2020 | Protein & Cell2020,11,4: | 44 |

| 4 | Efficacy and safety of Xiangsha Liujunzi granules for functional dyspepsia: A multi-center randomized double-blind placebo-controlled clinical study显示文摘AIM To assess the efficacy and safety of a Chinese herbal medicine(CHM), Xiangsha Liujunzi granules, in the treatment of patients with functional dyspepsia(FD). METHODS We performed a randomized, double-blind, placebocontrolled trial with patients from three centers. Two hundred and sixteen subjects diagnosed with FD according to ROME Ⅲ criteria and confirmed by upper gastrointestinal endoscopy and spleen-deficiency and Qi-stagnation syndrome were selected to receive Xiangsha Liujunzi granules or placebo for 4 wk in a 2:1 ratio by blocked randomization. The subjects also received follow-up after the 4-wk intervention. Herbal or placebo granules were dissolved in 300 m L of water. Participants in both groups were administered 130 m L(45 ℃) three times a day. Participants were evaluated prior to and following 4 wk of the intervention in terms of changes in the postprandial discomfort severity scale(PDSS) score, clinical global impression(CGI) scale score, hospital anxiety and depression scale(HADS) score, traditional Chinese medicine symptoms score(SS), scores of various domains of the 36-item short form health survey(SF-36), gastric emptying(GE) and any observed adverse effects. RESULTS Compared with the placebo group, patients in the CHM group showed significant improvements in the scores of PDSS, HADS, SS, SF-36 and CGI scale(P < 0.05 or P < 0.01). | 5 | Cap-assisted endoscopic sclerotherapy for hemorrhoids: Methods, feasibility and efficacy显示文摘AIM: To evaluate the methodology, feasibility, safety and efficacy of a novel method called cap-assisted endoscopic sclerotherapy(CAES) for internal hemorrhoids.METHODS: A pilot study on CAES for grade Ⅰ to Ⅲ internal hemorrhoids was performed. Colon and terminal ileum examination by colonoscopy was performed for all patients before starting CAES. Polypectomy and excision of anal papilla fibroma were performed if polyps or anal papilla fibroma were found and assessed to be suitable for resection under endoscopy. CAES was performed based on the requirement of the cap, endoscope, disposable endoscopic long injection needle, enough insufflated air and sclerosing agent.RESULTS: A total of 30 patients with grade Ⅰ to Ⅲ internal hemorrhoids was included. The follow-up was more than four weeks. No bleeding was observed after CAES. One(3.33%) patient claimed mild tenesmus within four days after CAES in that an endoscopist performed this procedure for the first time. | 8 | Association between CYP2C19*2/*3Polymorphisms and Coronary Heart Disease显示文摘This study sought to explore the relationship between cytochrome P450 2C19(CYP2C19)*2/*3 polymorphisms and the development of coronary heart disease(CHD),and to evaluate the influence of the single nucleotide polymorphisms(SNPs)on the occurrence of adverse clinical events in CHD patients.A total of 231 consecutive patients candidate for percutaneous coronary intervention genotyped for CYP2C19*2(681G>A)and*3(636G>A)polymorphisms were enrolled.The adverse clinical events were recorded during a follow-up period of 14 months.The incidence of CHD,according to coronary angiography,was significantly higher(P=0.025)in CYP2C19*2 carriers group.Stepwise binary logistic regression analysis revealed that among factors that potentially influenced the presence of CHD(age>60 years,gender,BMI,etc.),CYP2C19*2 carriers(OR 1.94,95%CI:1.08-3.50,P=0.028)and male gender(OR 2.74,95%CI:1.58-4.76,P=0.001)were independent predictors,which were associated with the presence of CHD.The follow-up results showed that the incidence of adverse cardiovascular events within 14 months of discharge was significantly higher in the CYP2C 19*2 carriers than in the non-carriers(21.6%vs.6.3%,P=0.019).The results of the multivariate Cox proportional hazards model showed that CYP2C19*2 loss-of-flinction was the only independent factor which predicted the coronary events during the follow-up period of 14 months(OR=3.65,95%CI:1.09-12.25,P=0.036).The adverse impact of CYP2C19*2 polymorphisms was found not only in the risk of the presence of CHD,but also in the adverse cardiovascular events in CHD patients during the follow-up period of 14 months.However the same influence was not found in CYP2C19*3 mutation in Chinese Han population. | Ying-ying ZHANG Xin ZHOU Wen-jie JI Ting LIU Jing MA Ying ZHANG Yu-ming LI | 2019 | Current Medical Science2019,39,1: | 11 |

| 11 | Clinical Application of Chromosome Microarray Analysis in Han Chinese Children with Neurodevelopmental Disorders显示文摘Chromosome microarray analysis(CMA) is a cost-effective molecular cytogenetic technique that has been used as a first-line diagnostic test in neurodevelopmental disorders in the USA since 2011. The impact of CMA results on clinical practice in China is not yet well studied, so we aimed to better evaluate this phenomenon.We analyzed the CMA results from 434 patients in our clinic, and characterized their molecular diagnoses, clinical features, and follow-up clinical actions based on these results. The overall diagnostic yield for our patients was 13.6%(59 out of 434). This gave a detection rate of 14.7%for developmental delay/intellectual disability(DD/ID,38/259) and 12% for autism spectrum disorders(ASDs,21/175). Thirty-three recurrent(n≥2) variants were found, distributed at six chromosomal loci involving known chromosome syndromes(such as DiGeorge, Williams Beuren, and Angelman/Prader-Willi syndromes).The spectrum of positive copy number variants in our study was comparable to that reported in Caucasian populations, but with specific characteristics. Parental origin tests indicated an effect involving a significant maternal transmission bias to sons. The majority of patients with positive results(94.9%) had benefits, allowing earlier diagnosis(36/59), prioritized full clinical management(28/59), medication changes(7/59), a changed prognosis(30/59), and prenatal genetic counseling(15/59). Our results provide information on de novo mutations in Chinese children with DD/ID and/or ASDs. Our data showed that microarray testing provides immediate clinical utility for patients. | 12 | Association between Plasma Metal Levels and Diabetes Risk: a Case-control Study in China显示文摘Objective Many metals, some of which have been classified as environmental endocrine disruptors, are used extensively in everyday consumer products and are ubiquitous in our living environment. In the present study, we aimed to explore the associations between the prevalence risk of type 2 diabetes and plasma levels of 20 trace elements as well as those of heavy metals in a Han Chinese population. Methods We conducted a case-control study to investigate the associations between plasma concentrations of 20 metals and diabetes in Jiangsu province. A total of 122 newly diagnosed cases of type 2 diabetes and 429 matched controls were recruited from community physical examinations in Suzhou City of Jiangsu Province. Plasma metal levels were measured by inductively-coupled plasma mass spectrometry. Results After adjusting for confounders, plasma vanadium, chromium, manganese, copper, zinc, arsenic, selenium, strontium, palladium, cadmium, cesium, and barium were associated with diabetes risk(P < 0.05). The adjusted OR increased with increasing concentration of vanadium, manganese, copper, zinc, and cesium. Conclusion Many metals, including manganese, copper, zinc, arsenic, selenium, and cadmium in plasma, are associated with the morbidity of diabetes. | 14 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 17 | An improved method for prediction of tomato photosynthetic rate based on WSN in greenhouse显示文摘In order to improve the efficiency of CO2 fertilizer and promote high quality and yield,it is necessary to precisely control CO2 fertilizer by wireless sensor network based on a model of photosynthetic rate prediction in greenhouse.An experiment was carried out on tomato plants in greenhouse for photosynthetic rate prediction modeling combined rough set and BP neural network.In data acquiring phase,plants growth information and greenhouse environmental information that may have influences on photosynthetic rate,including plant height,stem diameter,the number of leaves and chlorophyll content of functional leaves,air temperature,air humidity,light intensity,CO2 concentration and soil moisture,which were measured.And LI-6400XT photosynthetic rate instrument was used for obtaining net photosynthetic rate of functional leaf.After preliminary processing,135 sets of data were obtained.And twelve of them were used for model test of neural network,while the others were used for modeling.All of the data were normalized before modeling.Two models were built to predict photosynthetic rate based on BP neural network.One had total nine input parameters.The other had six input parameters,chlorophyll content,air temperature,air humidity,light intensity,CO2 concentration,and soil moisture,which were reducted from original nine based on attributes reduction theory of rough set.Both two models have one output parameter,the net photosynthetic rate of single leaf.The genetic algorithm was adopted to reduct attributes.Since continuous data cannot be processed by rough set,the K-mean cluster method was used to discretize the data of nine input parameters before attributes reduction.The prediction results of two models showed that the model with six input parameters had a mean absolute error of 0.6958,an average relative error of 7.28%,a root-mean-square error of 0.7428,and a correlation coefficient of 0.9964,while the other model respectively had 0.4026,4.53%,0.3245 and 0.9965,which proved that the model with minimum attributes had higher prediction accuracy.On the other hand,the number of iterations was used to represent the neural network train speed.The result showed that the model with six input parameters had an iteration of 544,while the other had 1038.Hence,the reduction model was applied to controlling CO2 concentration.The net photosynthetic rates at different CO2 concentrations were predicted at a certain condition.The results had the same curve trend with theory analysis,and a high prediction accuracy,which proved that the model was useful for CO2 concentration control. | Ji Yuhan Jiang Yiqiong Li Ting Zhang Man Sha Sha Li Minzan | 2016 | International Journal of Agricultural and Biological Engineering2016,9,1: | 6 |

| 19 | MdMYB8 is associated with flavonol biosynthesis via the activation of the MdFLS promoter in the fruits of Malus crabapple显示文摘Flavonols are polyphenolic compounds that play important roles in plant stress resistance and development.They are also valuable components of the human diet.The Malus crabapple cultivar‘Flame’provides an excellent model for studying flavonol biosynthesis due to the high flavonol content of its fruit peel.To obtain a more detailed understanding of the flavonol regulatory network involved in fruit development,the transcriptomes of the fruit of the Malus cv.‘Flame’from five continuous developmental stages were analyzed using RNA sequencing.A flavonol-related gene module was identified through weighted gene coexpression network analysis(WGCNA),and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis indicated that phytohormones are involved in regulating flavonol biosynthesis during fruit development.A putative transcription factor,MdMYB8,was selected for further study through hub gene correlation network analysis and yeast one-hybrid assays.Stable overexpression or RNAi knockdown of MdMYB8 in transgenic‘Orin’apple calli resulted in a higher or lower flavonol content,respectively,suggesting that MdMYB8 is a regulator of flavonol biosynthesis.This transcriptome analysis provides valuable data for future studies of flavonol synthesis and regulation. | Hua Li Yu Li Jiaxuan Yu Ting Wu Jie Zhang Ji Tian Yuncong Yao | 2020 | Horticulture Research2020,7,1: | 6 |
| 20 | Chang'an Ⅱ Decoction(肠安Ⅱ号方)-Containing Serum Ameliorates Tumor Necrosis Factor-α-Induced Intestinal Epithelial Barrier Dysfunction via MLCK-MLC Signaling Pathway in Rats显示文摘Objective To investigate the effect of Chang’an Ⅱ Decoction(肠安Ⅱ号方))-containing serum on intestinal epithelial barrier dysfunction in rats.Methods Tumor necrosis factor(TNF)-α-induced injury of Caco-2 monolayers were established as an inflammatory model of human intestinal epithelium.Caco-2 monolayers were treated with blank serum and Chang’an Ⅱ Decoction-containing serum that obtained from the rats which were treated with distilled water and Chang’an Ⅱ Decoction intragastrically at doses of 0.49,0.98,1.96 g/(kg·d)for 1 week,respectively.After preparation of containing serum,cells were divided into the normal group,the model group,the Chang’an Ⅱ-H,M,and L groups(treated with 30 ng/mL TNF-αand medium plus 10%high,middle-,and low-doses Chang’an Ⅱ serum,respectively).Epithelial barrier function was assessed by transepithelial electrical resistance(TER)and permeability of fluorescein isothiocyanate(FITC)-labeled dextran.Transmission electron microscopy was used to observe the ultrastructure of tight junctions(TJs).Immunofluorescence of zonula occludens-1(ZO-1),claudin-1 and nuclear transcription factor-kappa p65(NF-κBp65)were measured to determine the protein distribution.The mRNA expression of myosin light chain kinase(MLCK)was measured by real-time polymerase chain reaction.The expression levels of MLCK,myosin light chain(MLC)and p-MLC were determined by Western blot.Results Chang’an Ⅱ Decoction-containing serum significantly attenuated the TER and paracellular permeability induced by TNF-α.It alleviated TNF-α-induced morphological alterations in TJ proteins.The increases in MLCK mRNA and MLCK,MLC and p-MLC protein expressions induced by TNF-αwere significantly inhibited in the Chang’an Ⅱ-H group.Additionally,Chang’an Ⅱ Decoction significantly attenuated translocation of NF-κBp65 into the nucleus.Conclusion High-dose Chang’an Ⅱ-containing serum attenuates TNF-α-induced intestinal barrier dysfunction.The underlying mechanism may be involved in inhibiting the MLCK-MLC phosphorylation signaling pathway mediated by NF-κBp65. | CHEN Ting YIN Xiao-Ian KANG Nan WANG Xiao-ge LI Bao-shuang JI Hai-jie ZHANG Yin-qiang BIAN Li-qun ZHANG Bei-hua WANG Feng-yun TANG Xu-dong | 2020 | Chinese Journal of Integrative Medicine2020,26,10: | 5 |
