| 7 | Single-cell RNA Sequencing Reveals Sexually Dimorphic Transcriptome and Type 2 Diabetes Genes in Mouse Islet β Cells显示文摘Type 2 diabetes(T2D)is characterized by the malfunction of pancreaticβcells.Susceptibility and pathogenesis of T2D can be affected by multiple factors,including sex differences.However,the mechanisms underlying sex differences in T2D susceptibility and pathogenesis remain unclear.Using single-cell RNA sequencing(scRNA-seq),we demonstrate the presence of sexually dimorphic transcriptomes in mouseβcells.Using a high-fat diet-induced T2D mouse model,we identified sex-dependent T2D altered genes,suggesting sex-based differences in the pathological mechanisms of T2D.Furthermore,based on islet transplantation experiments,we found that compared to mice with sexmatched islet transplants,sex-mismatched islet transplants in healthy mice showed down-regulation of genes involved in the longevity regulating pathway ofβcells.Moreover,the diabetic mice with sex-mismatched islet transplants showed impaired glucose tolerance.These data suggest sexual dimorphism in T2D pathogenicity,indicating that sex should be considered when treating T2D.We hope that our findings could provide new insights for the development of precision medicine in T2D. | Gang Liu Yana Li Tengjiao Zhang Mushan Li Sheng Li Qing He Shuxin Liu Minglu Xu Tinghui Xiao Zhen Shao Weiyang Shi Weida Li | 2021 | Genomics, Proteomics & Bioinformatics2021,19,3: | 1 |
| 8 | Influence of severe neck angulation on hemodynamic and clinical outcomes following endovascular aneurysm repair:a hemodynamic analysis and a retrospective cohort study显示文摘Background: For patients with severe neck angulation (SNA), hemodynamic and clinical outcomes following endovascular aneurysm repair (EVAR) are still unclear. This study aimed to explore the influence of SNA on hemodynamic and clinical outcomes following EVAR.Methods: This study included a hemodynamic analysis and a retrospective cohort study from West China Hospital of Sichuan University between January 2011 and December 2020. The Cox regression model, inverse probability of treatment weighting (IPTW) analysis, sensitivity analysis, and subgroup analysis were applied. Primary outcome was type IA endoleak (T1AEL).Results: In this hemodynamic analysis, nine non-severe neck angulation (nSNA) and 16 SNA idealized models were constructed. We found a significant difference in drag force between SNA and nSNA models (7.016 ± 2.579 Nvs. 4.283 ± 1.460 N,P = 0.008), and proximal neck angles were significantly associated with the magnitude of drag force (F = 0.082 ×α-0.006 ×β + 2.818, α: 95% confidence interval [CI] 0.070-0.094;P = 0.001;β: 95% CI -0.019 to 0.007;P = 0.319). In our cohort study, 514 nSNA patients (71.5 ± 8.5 years;459 males) and 208 SNA patients (72.5 ± 7.8 years;135 males) were included, with a median follow-up duration of 34 months (16-63 months). All baseline characteristics were well balanced after IPTW matching. We found that SNA was associated with a significant risk of adverse limb event (hazard ratio [HR] 2.18, 95% CI 1.09-3.12), yet was not associated with T1AEL, overall survival, or reintervention. In patients without proximal or distal additional procedures (DAP), subgroup analyses suggested a significant risk of T1AEL (Proximal: HR 5.25, 95% CI 1.51-18.23;Distal: HR 5.07, 95% CI 1.60-16.07) and adverse limb event (Proximal: HR 2.27, 95% CI 1.01-5.07;Distal: HR 2.91, 95% CI 1.30-6.54) in SNA patients. However, no noticeable difference was observed in patients with proximal or DAP.Conclusions: SNA has a critical influence on hemodynamic and clinical outcomes following EVAR. | 9 | Analysis of aortic wall stress and morphology in patients with type B aortic dissection显示文摘Risk assessment is critical in preventing aortic dissection(AD).This study aims to evaluate the wall stress(WS)distribution,especially at the locations of proximal tears in patient-specific type-B aortic dissection(TBAD)to explore the pathogenesis of dissection.In addition,the shape of the aortas were assessed and associated with TBAD risk.In this paper,30 three-dimensional models were reconstructed based on patient-specific CT angiography images,and finite element analysis(FEA)was used to analyze the admission blood pressure.In addition,the anatomic variables including the head vessel angles,the aorta diameters,curvatures and torturosity ware measured.Tears occurred at either local high WS or low WS region,and these acute TBAD patients can be classified into three groups based on the location of initial intimal tears.In addition,the WS values at tear sites of each group showed significant difference(P<0.001).Moreover,the angles of the left subclavian and brachiocephalic arteries were statistical different among three groups.Increased wall stress or decreased wall strength both contribute to the pathogenesis of aortic dissections.In addition,abnormal head vessel angles may be monitored as an important risk factor for aortic dissection,and its specific features may further help to determine the potential tear location. | Hongrui Hu Zhan Liu Guoxin Chen Ding Yuan Tinghui Zheng | 2021 | Medicine in Novel Technology and Devices2021,,3: | 0 |

| 13 | Hemodynamics in renal arteries with partial coverage after endovascular aneurysm repair affected by renal orientations显示文摘Endovascular aortic repair(EVAR)has been increasingly used to treat abdominal aortic aneurysm(AAA)due to its lower mortality and shorter recovery time.However,the complications caused by renal artery coverage remained a major concern.In this study,three types of renal orientation and five types of partial renal coverage degree were adopted to numerically investigate the flow pattern,wall shear stress distribution,and blood perfusion during a cardiac cycle.The results show that as for the mild partial coverage(=<30%),the renal artery with a downward orientation suffered fewer unfavorable wall shear stress(WSS)distribution and more blood perfusion.It should be noted that the renal artery with horizontal orientation suffered more unfavorable WSS distribution.In terms of severe coverage(≥50%),all of the renal arteries displayed highly disturbed wall shear distribution,resulting in a significant reduction in blood perfusion It was interesting to note that the artery with an upward orientation had a smaller rate of reduction in blood perfusion.It can be concluded that as for the mild renal coverage after EVAR,renal arteries with downward orientation may have the advantage of reducing the possibility of renal occlusion,while renal arteries with horizontal orientation may increase the unfavorable wall shear distribution.On the other hand,as for serious coverage(≥50%),high risk of renal ischemia remains the main concern for all types of renal arteries due to highly disturbed wall shear distribution would significantly increase regardless of renal orientation. | Jun Wen Xinhui Liu Jianguo Lei Jiarong Wang Xiyang Chen Tinghui Zheng Ding Yuan | 2023 | Medicine in Novel Technology and Devices2023,,2: | 0 |
| 14 | Influence of morphology and hemodynamics on thrombosis in kawasaki disease patients显示文摘Kawasaki Disease(KD)patients with co-occurrent coronary artery aneurysms(CAAs)are prone to thrombosis.This study explores the thrombotic influence of morphology and hemodynamics in KD patients with CAAs.Patient-specific computed tomography angiogram images of 9 patients were used to rebuild coronary models(including 23 aneurysms;10 in thrombosed group,and 13 in non-thrombosed group)and perform computational simulations to obtain the hemodynamic parameters.The thrombosed and non-thrombosed groups were compared in terms of six parameters,namely,the maximum diameter(D_(max)),aspect ratio(R_(l/d)),shape,time-averaged wall shear stress(TAWSS),oscillatory shear index(OSI),and relative residence time(RRT).The results showed that:(1)In thrombosed aneurysms,there were several vortex structures(eddy zone),of which the position changed continuously in a cardiac cycle.In non-thrombosed aneurysms,the streamlines showed a large streaming zone.(2)Abnormal hemodynamic areas of aneurysms with thrombosis tended to appear in the same region(especially the proximal and near myocardial side of aneurysms).(3)In the non-thrombosed group,there was a correlation between the morphological and hemodynamic parameters.In thrombosed aneurysms,the flow pulsatility has a significant impact on the flow pattern.The thrombosed coronary aneurysms exhibited more risk factors and the co-location of hemodynamic abnormalities was consistent with the position of thrombosis.A score of risk factors could provide the thrombotic information of coronary aneurysms,which might be helpful for long-term clinical management of KD patients. | Haoyao Cao Zhuxiang Xiong Zhan Liu Yan Li Huaxia Pu Jing Liu Liqing Peng Tinghui Zheng | 2023 | Medicine in Novel Technology and Devices2023,,2: | 0 |
| 15 | The exploration of flow pattern in the superior vena cava of healthy adults:A 4D flow magnetic resonance imaging study显示文摘Our study sought to investigate the blood flow pattern in the superior vena cava(SVC)of healthy adults and to describe the development and characteristics of the flow pattern using 4D flow magnetic resonance imaging(MRI).4D flow MRI data with full coverage of SVC and brachiocephalic veins(BVs)were acquired with a 3-Testa MRI in thirty healthy adults(age:28.70±9.09).Hemodynamic parameters in the SVC,including velocity,velocity vector,flow visualization(pathlines,streamlines),flow waveform and energy loss,were obtained with specialized commercial post-processing software based on 4D flow MRI data.This study found that:(1)The SVC has a pulsating flow waveform with double peaks.(2)Based on BVs flow of the SVC pathline visualization,flow patterns could be divided into three development types:twining(n=14),untwining(n=)and no helical flow(n=7).(3)With the decreasing blood velocity,helical flow areas tended to gradually extend.(4)There were no significant differences in most hemodynamic parameters among the three types.The exploration of the blood flow characteristics of normal SVC may be the first step for capturing pathogenic features before the patients develop symptoms with a certain positive significance.The potential physiological significance of these phenomena deserves more exploration in future studies. | Huaxia Pu Haoyao Cao Yubo Fan Jinge Zhang Simeng Wang Zhan Liu Xiaoyue Zhou Ning Jin Tinghui Zheng Liqing Peng | 2023 | Medicine in Novel Technology and Devices2023,,2: | 0 |
