| 1 | Contribution of oxidative stress to pulmonary arterial hypertension显示文摘Recent data implicate oxidative stress as a mediator of pulmonary hypertension (PH) and of the associated pathological changes to the pulmonary vasculature and right ventricle (RV). Increases in reactive oxygen species (ROS), altered redox state, and elevated oxidant stress have been demonstrated in the lungs and RV of several animal models of PH, including chronic hypoxia, monocrotaline toxicity, caveolin-1 knock-out mouse, and the transgenic Ren2 rat which overexpresses the mouse renin gene. Generation of ROS in these models is derived mostly from the activities of the nicotinamide adenine dinucleotide phosphate oxidases, xanthine oxidase, and uncoupled endothelial nitric oxide synthase. As disease progresses circulating monocytes and bone marrow-derived monocytic progenitor cells are attracted to and accumulate in the pulmonary vasculature. Once established, these inflammatory cells generate ROS and secrete mitogenic and fibrogenic cytokines that induce cell proliferation and fibrosis in the vascular wall resulting in progressive vascular remodeling. Deficiencies in antioxidant enzymes also contribute to pulmonary hypertensive states. Current therapies were developed to improve endothelial function, reduce pulmonary artery pressure, and slow the progression of vascular remodeling in the pulmonary vasculature by targeting deficiencies in either NO (PDE-type 5 inhibition) or PGI 2 (prostacyclin analogs), or excessive synthesis of ET-1 (ET receptor blockers) with the intent to improve patient clinical status and survival. New therapies may slow disease progression to some extent, but long term management has not been achieved and mortality is still high. Although little is known concerning the effects of current pulmonary arterial hypertension treatments on RV structure and function, interest in this area is increasing. | 2 | Role of CD56-expressing immature biliary epithelial cells in biliary atresia显示文摘AIM: To analyze the clinical and pathological parameters and expression of the neural cell adhesion molecule(CD56) in patients with biliary atresia(BA).METHODS: Established clinical laboratory markers of hepatic function, including enzyme activity, protein synthesis, and bilirubin metabolism, were evaluated in patients with BA and compared with those in patients with choledochal cysts and neonatal hepatitis. Pathological changes in tissue morphology and fibrosis were examined by histological and tissue collagen staining. Immunohistochemical staining for the biliary epithelial cell markers CD56 and CK19 together with the Notch signaling related molecules Notch1 and Notch2 was performed in the context of alterations in the structure of intrahepatic biliary ducts.RESULTS: Differences in some clinical laboratoryparameters among the three diseases examined were observed, but they did not correlate with the pathological classification of fibrosis in BA. Immunohistochemical staining showed the presence of CD56-positive immature bile ducts in most patients(74.5%) with BA but not in patients with choledochal cysts or neonatal hepatitis. The number of CD56-expressing cells correlated with disease severity, with more positive cells present in the later stages of liver damage(81.8% vs 18.2%). Furthermore, bile plugs were mainly found in CD56-positive immature biliary ducts. Notch signaling was a key regulatory pathway in biliary duct formation and played a role in tissue fibrosis. Notch1 was co-expressed in CD56-positive cells, whereas Notch2 was found exclusively in blood vessels in the portal area of patients with BA. | 3 | Near-infrared fluorescence imaging of a solitary fibrous tumor of the pancreas using methylene blue显示文摘A 67-year-old female presented with unexplained abdominal pain. A contrast-enhanced computed tomography scan of the abdomen incidentally revealed a mass in the uncinate process of the pancreas. This mass was resected and based on histopathological findings, diagnosed as a solitary fibrous tumor (SFT) of the pancreas. A SFT is an extremely rare benign mesenchymal tumor that in 65% of cases affects the visceral pleura but can also affect extra-pleural sites. The intraoperative demarcation of pancreatic tumors, such as SFTs, can bechallenging. | 4 | Downregulation of Hes1 expression in experimental biliary atresia and its effects on bile duct structure显示文摘AIM To analyze the expression and function of the notchsignaling target gene Hes1 in a rhesus rotavirusinduced mouse biliary atresia model. METHODS The morphologies of biliary epithelial cells in biliary atresia patients and in a mouse model were examined by immunohistochemical staining. Then, the differential expression of Notch signaling pathway-related molecules was investigated. Further, the effects of the si RNAmediated inhibition of Hes1 expression were examined using a biliary epithelial cell 3 D culture system.RESULTS Both immature(Ep CAM+) and mature(CK19+) biliary epithelial cells were detected in the livers of biliary atresia patients without a ductile structure and in the mouse model with a distorted bile duct structure. The hepatic expression of transcripts for most Notch signaling molecules were significantly reduced on day 7 but recovered to normal levels by day 14, except for the target molecule Hes1, which still exhibited lower m RNA and protein levels. Expression of the Hes1 transcriptional co-regulator, RBP-Jκ was also reduced. A 3 D gel culture system promoted the maturation of immature biliary epithelial cells, with increased expression of CK19+ cells and the formation of a duct-like structure. The administration of Hes1 si RNA blocked this process. | 6 | Mosquito larvicidal properties of Orthosiphon thymiflorus(Roth)Sleesen.(Family:Labiatae)against mosquito vectors,Anopheles stephensi,Culex quinquefasciatus and Aedes aegypti(Diptera:Culicidae)显示文摘Objective:To determine the mosquito larvicidal activities of hexane,chloroform,ethyl acetate, acetone and methanol leaf extract of Orthosiphon thymiflorus(O.thymiflorus) against Anopheles stephensi(An.stephensi),Culex quinquefasciatus(Cx.quinquefasciatus) and Aedes aegypti (Ae.aegypti).Methods:The larvicidal activity was assayed against three mosquito species at various concentrations ranging from(50-450 ppm) under the laboratory conditions.The LC50 and LC90value of the O.thymiflorus leaf extract was determined by Probit analysis.Results:The LC50 values of hexane,chloroform,ethyl acetate,acetone and methanol extract of O.thymiflorus third instar larvae of An.stephensi were LC50= 201.39,178.76,158.06,139.22 and 118.74 ppm; Cx.quinquefasciatus were LC50=228.13,209.72.183.35,163.55 and 149.96 ppm and Ae.aegypti were LC50=215.65,197.91,175.05,154.80 and 137.26 ppm,respectively.Maximum larvicidal activity was observed in the methanolic extract followed by acetone,ethyl acetate chloroform and hexane extract.The larval mortality was observed after 24 h exposure.No mortality was observed in control.Conclusions:The present results suggest that the effective plant crude extracts have potential to be used as an ideal eco-friendly approach for the control of mosquito vectors.This study provides the first report on the larvicidal activity of this plant crude solvent extract of against An.stephensi,Cx.quinquefasciatus and Ae.aegypti mosquitoes. | K Kovendan K Murugan S Vincent Donald R Barnard | 2012 | Asian Pacific Journal of Tropical Medicine2012,5,4: | 2 |
| 7 | Transposon mouse models to elucidate the genetic mechanisms of hepatitis B viral induced hepatocellular carcinoma显示文摘The major type of human liver cancer is hepatocellular carcinoma(HCC), and there are currently many risk factors that contribute to this deadly disease. The majority of HCC occurrences are associated with chronic hepatitis viral infection, and hepatitis B viral(HBV) infection is currently a major health problem in Eastern Asia. Elucidating the genetic mechanisms associated with HBV-induced HCC has been difficult due to the heterogeneity and genetic complexity associated with this disease. A repertoire of animal models has been broadly used to study the pathophysiology and to develop potential treatment regimens for HBVassociated HCC. The use of these animal models has provided valuable genetic information and has been an important contributor to uncovering the factors involved in liver malignant transformation, invasion and metastasis. Recently, transposon-based mouse models are becoming more widely used in liver cancer research to interrogate the genome by forward genetics and also used to validate genes rapidly in a reverse genetic manner. Importantly, these transposon-based rapid reverse genetic mouse models could become crucial in testing potential therapeutic agents before proceeding to clinical trials in human. | 12 | Persistence and growth of faecal culturable bacterial indicators in water column and sediments of Vidy Bay,Lake Geneva,Switzerland显示文摘The aims of this study was to investigate the persistence and the growth of culturable bacterial indicators(CBI) including total coliforms(TC) and faecal coliforms represented by Escherichia coli,enterococcus(ENT) ,and aerobic mesophilic bacteria(AMB) in the surface sediments and the water column of Vidy Bay(Lake Geneva,City of Lausanne,Switzerland) . The study was carried out for 60 d using microcosms containing Sewage Treatment Plant(STP) effuent and nonsterile water without CBI,as well as contaminated and non-contaminated sediments. The effects of water temperature and of organic matter associated with sediments on the survival of CBI in the sediments and the water column were observed. The number of CBI colonies in the contaminated sediments of Vidy Bay and in the STP effuent was almost identical in the order of 105-107,104-106,103-105,and 104-107 CFU/100 g sediment or /100 mL water for TC,E. coli,ENT,and AMB respectively. A degradation of CBI was observed in the sediments where organic mater content was low and in the water column at a temperature of 10°C after 5 d of experimentation. In addition,a growth of CBI was observed in the sediment which is rich in organic matter at 20°C. The results of this study indicate:(1) the higher concentrations of the CBI observed in dierent points in the water column of Vidy Bay may not be explained only by the recent contribution of the three potential sources of the Bay contamination including STP and the Chamberonne and Flon Rivers,but also by the persistence,removal from sediment and multiplication of CBI in the sediment and water column;(2) the sediment of Vidy Bay constitute a reservoir of CBI and can even support their growth;and(3) the CBI not only survive in sediments,but also can be remobilized and increased in the water column,therefore,it become a permanent microbiological pollution in Vidy Bay. | POTE John HALLER Laurence KOTTELAT Régis SASTRE Vincent ARPAGAUS Philippe WILDI Walter | 2009 | Journal of Environmental Sciences2009,21,1: | 2 |
