1A Strigolactone Biosynthesis Gene Contributed to the Green Revolution in Rice显示文摘Plant architecture is a complex agronomic trait and a major factor of crop yield,which is affected by several important hormones.Strigolactones(SLs)are identified as a new class hormoneinhibiting branching in many plant species and have been shown to be involved in various developmental processes.Genetical and chemical modulation of the SL pathway is recognized as a promising approach to modify plant architecture.However,whether and how the genes involved in the SL pathway could be utilized in breeding still remain elusive.Here,we demonstrate that a partial loss-of-function allele of the SL biosynthesis gene,HIGH TILLERING AND DWARF 1/DWARF17(HTD1/D17),which encodes CAROTENOID CLEAVAGE DIOXYGENASE 7(CCD7),increases tiller number and improves grain yield in rice.We found that the HTD1 gene had been widely utilized and co-selected with Semidwarf 1(SD1),both contributing to the improvement of plant architecture in modern rice varieties since the Green Revolution in the 1960s.Understanding how phytohormone pathway genes regulate plant architecture and how they have been utilized and selected in breeding will lay the foundation for developing the rational approaches toward improving crop yield.Yuexing Wang Lianguang Shang Hong Yu Longjun Zeng Jiang Hu Shen Ni Yuchun Rao Sanfeng Li Jinfang Chu Xiangbing Meng Lei Wang Ping Hu Jijun Yan Shujing Kang Minghao Qu Hai Lin Tao Wang Quan Wang Xingming Hu Hongqi Chen Bing Wang Zhenyu Gao Longbiao Guo Dali Zeng Xudong Zhu Guosheng Xiong Jiayang Li Qian Qian 2020Molecular Plant2020,13,6:23
2Diel and seasonal variation of methane and carbon dioxide fluxes at Site Guojiaba, the Three Gorges Reservoir显示文摘In order to investigate the CH4and CO2fluxes across the water-gas interface and identify their controlling factors, four diel field campaigns and one monthly sampling campaign during June 2010–May 2011 were carried out at a site near the Three Gorges Dam,China. The averaged CH4and CO2fluxes across the air-water interface from the site were much less than those reported from reservoirs in tropic and temperate regions, and from the natural river channels of the Yangtze River. CH4Fluxes at the site were very low compared to most other reservoirs or natural lakes. One of the most important reasons may be due to the oxidation of CH4in the water column owing to the great water depth and high DO in water in the Three Gorges Reservoir. The averaged monthly CH4and CO2fluxes at the site during the observation year were 0.05 mg/(m2·hr) and 104.43 mg/(m2·hr) respectively with the maximum occurred in July 2010. The monthly CO2fluxes during the observation year were positively correlated to the surface water temperature, and negatively correlated to the air pressure and the surface water pH. The CO2flux showed a positive correlation with DOC to some extent, although not significantly, which indicated that allochthonous organic C was a major source of CO2and biogeochemical processes in this reservoir were C-limited. 3Study on the physiological indices of Pinus sibirica and Pinus koraiensis seedlings under cold stress显示文摘Although Pinus sibirica and Pinus koraiensis are resistant to cold or low temperatures in the cold temperate regions of the northern hemisphere,the former has a stronger cold resistance.Research has been limited to the comparison of physiological responses of the two species to cold stress.In this study,5-year-old seedlings of P.sibirica and P.koraiensis were subjected to six temperature treatments,[20℃(control),0℃,-20℃,-40℃,-60℃,and-80℃],under different stress periods(6,12,24,and 48 h).The results showed that differences in each physiological index were significant between P.sibirica and P.koraiensis,except for the permeability of cell membranes,reactive oxygen species,proline and soluble proteins.An ANOVA test indicated that there were extreme differences among the temperatures for each index,stress time and temperature 9 time for most indices.All indices showed a similar trend for P.sibirica and P.koraiensis with decreasing temperature or the extension of stress time.Soluble sugars and proline increased at 0 to-20℃and then remained unchanged with temperature decline.Other indices showed an increase from 20 to-20℃,stable from-20 to-40℃and a decrease from-40 to-80℃.All the indices increased and then declined along with the prolonged cold stress time,except for the control.From 0 to-40℃,the permeability of cell membranes,relative conductivity,reactive oxygen species and malonaldehyde of P.koraiensis seedlings were higher than in P.sibirica,but superoxide dismutase,peroxidase,catalase activity and soluble sugars,proline,and soluble proteins content emerged as opposite.This study compared the physiological mechanism responses to cold stress between P.sibirica and P.koraiensis to provide the basis for the introduction,distribution,and genetic improvement of these coniferous species.Fang Wang Deyang Liang Xiaona Pei Qinhui Zhang Peng Zhang Jianqiu Zhang Zhimin Lu Yuchun Yang Guifeng Liu Xiyang Zhao 2019Journal of Forestry Research2019,30,4:12
4Transcriptional analysis and histochemistry reveal that hypersensitive cell death and H2O2 have crucial roles in the resistance of tea plant (Camellia sinensis (L.) O. Kuntze) to anthracnose显示文摘Anthracnose causes severe losses of tea production in China.Although genes and biological processes involved in anthracnose resistance have been reported in other plants,the molecular response to anthracnose in tea plant is unknown.We used the susceptible tea cultivar Longjing 43 and the resistant cultivar Zhongcha 108 as materials and compared transcriptome changes in the leaves of both cultivars following Colletotrichum fructicola inoculation.In all,9015 and 8624 genes were differentially expressed between the resistant and susceptible cultivars and their controls(0 h),respectively.In both cultivars,the differentially expressed genes(DEGs)were enriched in 215 pathways,including responses to sugar metabolism,phytohormones,reactive oxygen species(ROS),biotic stimuli and signalling,transmembrane transporter activity,protease activity and signalling receptor activity,but DEG expression levels were higher in Zhongcha 108 than in Longjing 43.Moreover,functional enrichment analysis of the DEGs showed that hydrogen peroxide(H2O2)metabolism,cell death,secondary metabolism,and carbohydrate metabolism are involved in the defence of Zhongcha 108,and 88 key genes were identified.Protein–protein interaction(PPI)network demonstrated that putative mitogen-activated protein kinase(MAPK)cascades are activated by resistance(R)genes and mediate downstream defence responses.Histochemical analysis subsequently validated the strong hypersensitive response(HR)and H2O2 accumulation that occurred around the hyphal infection sites in Zhongcha 108.Overall,our results indicate that the HR and H2O2 are critical mechanisms in tea plant defence against anthracnose and may be activated by R genes via MAPK cascades.Yuchun Wang Xinyuan Hao Qinhua Lu Lu Wang Wenjun Qian Nana Li Changqing Ding Xinchao Wang Yajun Yang 2018Horticulture Research2018,5,1:11
5Secondary Crystallization of Na_2CO_3-Modified HZSM-5 Zeolites with Tetrapropylammonium Hydroxide and Their Catalytic Performance in Thiophene Alkylation Reaction显示文摘The Na2CO3-modified HZSM-5 zeolites were further treated by tetrapropylammonium hydroxide(TPAOH) solution. The effect of TPAOH concentration on the secondary crystallization process was investigated. The resulting samples were characterized by a complementary combination of X-ray diffraction, N2 adsorption/desorption, scanning electron microscopy, X-ray fluorescence spectroscopy, XPS, 27 Al and 29 Si magic-angle spinning nuclear magnetic resonance spectroscopy, BET and temperature-programmed desorption techniques. The results showed that the secondary crystallization of the HZSM-5 zeolite could result in migration of non-framework species from the internal channels to the zeolite surface and their transformation into framework species. The catalytic activity of these modified samples for thiophene alkylation was evaluated. 6Influences of drip and flood irrigation on soil carbon dioxide emission and soil carbon sequestration of maize cropland in the North China Plain显示文摘The need is pressing to investigate soil CO_2(carbon dioxide) emissions and soil organic carbon dynamics under water-saving irrigation practices in agricultural systems for exploring the potentials of soil carbon sequestration. A field experiment was conducted to compare the influences of drip irrigation(DI) and flood irrigation(FI) on soil organic carbon dynamics and the spatial and temporal variations in CO_2 emissions during the summer maize growing season in the North China Plain using the static closed chamber method. The mean CO_2 efflux over the growing season was larger under DI than that under FI. The cumulative CO_2 emissions at the field scale were 1959.10 and 1759.12 g/m^2 under DI and FI, respectively. The cumulative CO_2 emission on plant rows(OR) was larger than that between plant rows(BR) under FI, and the cumulative CO2 emission on the irrigation pipes(OP) was larger than that between irrigation pipes(BP) under DI. The cumulative CO_2 emissions of OP, BP and bare area(BA) under DI were larger than those of OR, BR and BA under FI, respectively. Additionally, DI promoted root respiration more effectively than FI did. The average proportion of root respiration contributing to the soil CO_2 emissions of OP under DI was larger than that of OR under FI. A general conclusion drawn from this study is that soil CO_2 emission was significantly influenced by the soil water content, soil temperature and air temperature under both DI and FI. Larger concentrations of dissolved organic carbon(DOC), microbial biomass carbon(MBC) and total organic carbon(TOC) were observed under FI than those under DI. The observed high concentrations(DOC, MBC, and TOC) under FI might be resulted from the irrigation-associated soil saturation that in turn inhibited microbial activity and lowered decomposition rate of soil organic matter. However, DI increased the soil organic matter quality(the ratio of MBC to TOC) at the depth of 10–20 cm compared with FI. 7Effects and mechanism of freeze-thawing cycles on the soil N2O fluxes in the temperate semi-arid steppe显示文摘High nitrous oxide(N2O) emissions during freeze-thawing period(FTP) have been observed in many different ecosystems. However, the knowledge about the dynamic of soil N_2O emissions and its main driving mechanism during the freeze-thawing processes in grassland ecosystem is still limited. An in-situ experiment was conducted during the FTP on the sites with 0 and 15% surplus of the average rainfall and two levels of N addition(0,10 g N/(m^2·year)) during growing season(marked as W0N0, W15N0, W0N10, W15N10, respectively) to explore the effects of water and N background on soil N_2O emissions during FTPs and the relationship between soil N_2O emissions and environmental factors. The results indicated that water and N treatments conducted during growing season did not show significant effect on the N_2O effluxes of FTP, but the soil mineral N contents of W0N10 treatment were significantly higher than those of W0N0, W15N0, W15N10treatments(p < 0.05). The soil PLFA concentrations of microbial groups monitored during 2015 spring freeze-thawing period(2015S-FTP) were lower than those during winter freeze-thawing period of 2014(2014W-FTP), while cumulative soil N_2O emissions of 2015S-FTP were higher than those of 2014W-FTP. 8A SPH simulation on large-amplitude sloshing for fluids in a two-dimensional tank显示文摘Smoothed particle hydrodynamics(SPH) is a mesh-free adaptive Lagrangian particle method with attractive features for dealing with the free surface flow.This paper applies the SPH method to simulate the large-amplitude lateral sloshing both with and without a floating body,and the vertical parametrically-excited sloshing in a two-dimensional tank.The numerical results show that the SPH approach has an obvious advantage over conventional mesh-based methods in handling nonlinear sloshing problems such as violent fluid-solid interaction,and flow separation and wave-breaking on the free fluid surface.The SPH method provides a new alternative and an effective way to solve these special strong nonlinear sloshing problems.Wang Lishi Wang Zhuang Li Yuchun 2013Earthquake Engineering and Engineering Vibration2013,12,1:3
9Evaluation of Antitumor,Immunomodulatory and Free Radical Scavenging Effects of A New Herbal Prescription Seaweed Complex Preparation显示文摘Seaweed Complex Preparation(SCP) is a clinical traditional Chinese medicine preparation which is composed of seven traditional Chinese herbs,and it has been used for treatment of lung cancer,liver cancer and digestive cancer.However,little information is available about the pharmacodynamic basis.The antitumor,immunomodulatory and free radical scavenging effects of SCP were evaluated in this study.Transplanted tumor in vivo method was used to determine the antitumor effect.The effects on splenocyte proliferation and phagocytosis of macrophages in tumor-bearing mice were measured by the MTT method and the phagocytizing cock red blood cell(CRBC) method respectively.The scavenging activities of SCP on DPPH and hydroxyl radicals in vitro were investigated.It was found that the medium-dose and high-dose of SCP could significantly inhibit the growth of transplanted hepatic tumor of murine hepatocarcinoma cell line H22,and promote proliferation of splenocytes and phagocytosis of macrophages.SCP possessed noticeable scavenging activities on DPPH and hydroxyl radicals.The antitumor effects of SCP might be achieved by improving immune system and scavenging free radicals,which is in accordance with the viewpoint of traditional Chinese medicine in promoting the body resistance and eliminating pathogenic factors for cancer treatment.LIU Xin SHAO Changlun KONG Wenwen FANG Yuchun WANG Changyun 2013Journal of Ocean University of China2013,12,3:3
10MULTI-TILLERING DWARF1, a new allele of BRITTLE CULM 12, affects plant height and tiller in rice显示文摘Plant height and tillering are crucial factors determining rice plant architecture and influencing rice grain production. In this study, multi-tillering dwarf1(mtd1), a stable multi-tiller and dwarf mutant, was screened from the ethylmethane sulfonate-treated japonica rice variety Wuyunging7. Compared with the wild type, mtd1 mutant exhibited pleiotropic phenotypes, including dwarfism, more tillers, brittle culms and delayed heading date.By employing map-based cloning strategy, the gene MTD1 was finally mapped to an approximately 66-kb region on the short arm of chromosome 9. Sequencing results showed that the gene LOC_Os09g02650(BC12) in mtd1 mutant had a single nucleotide substitution(G to A), which generated a premature translation stop. Over-expressing MTD1/BC12 coding sequence rescued all the phenotypes of mtd1 mutants including plant height and tillers, which confirms that BC12 is the mutated gene in mtd1 mutant.Quantitative reverse transcription-PCR analysis showed that MTD1/BC12 could negatively regulate the expression of MONOCULM 1, IDEAL PLANT ARCHITECTURE1 and Tillering and Dwarf 1, and control rice tillering. Remarkably, a-amylase activity analysis and gibberellic acid(GA)treatment showed that the dwarf phenotype of mtd1 mutant was dependent on GA biosynthesis pathway. 12Associations between gene polymorphisms in two crucial metabolic pathways and growth traits in pigs显示文摘Propanoate metabolism and fatty acid metabolism are crucial metabolic pathways in vivo that have been studied extensively and have shown a significant influence on the growth and diseases of animals.In order to identify the essential genes significantly associated with porcine growth traits at the pathway level,we selected 14 candidate genes with 58 SNPs,including 24 tag SNPs and 34 predicted SNPs,in the two essential pathways to test using the SNaPshot mini-sequencing method.Association analysis was performed using both static and dynamic phenotypic records.The results showed that four SNPs,respectively in SUCLA2,SUCLG2,ACADS,and ALDH1B1,were significantly associated with growth in pigs.In addition,an exciting finding was that the change of transcription factor binding site that resulted from alteration of the bases in SUCLG2 brought out a transcription factor binding site for POU1F1a,which is the product of a notable marker gene for growth of animals.YANG Fan WANG QiongPing WANG MingHui HE Kan PAN YuChun 2012Chinese Science Bulletin2012,57,21:3
13Three Gorges Dam: friend or foe of riverine greenhouse gases?显示文摘Dams are often regarded as greenhouse gas(GHG) emitters. However, our study indicated that the world’s largest dam, the Three Gorges Dam(TGD), has caused significant drops in annual average emissions of CO, CHand NO over 4300 km along the Yangtze River, accompanied by remarkable reductions in the annual export of CO(79%), CH(50%) and NO(9%) to the sea. Since the commencement of its operation in 2003, the TGD has altered the carbonate equilibrium in the reservoir area, enhanced methanogenesis in the upstream, and restrained methanogenesis and denitrification via modifying anoxic habitats through long-distance scouring in the downstream. 14Microsatellite DNA analysis proves nucleus of interspecies reconstructed blastocyst coming from that of donor giant panda显示文摘A method for DNA isolation from early development of blastocyst and further analysis of nuclear and mitochondrial DNA was developed in present study. Total DNA was prepared from interspecies reconstructed blastocyst and a giant panda specific microsatellite locus g010 was successfully amplified. DNA sequencing of the PCR product showed that two sequences of reconstructed blastocysts are the same as that of positive control giant panda. 15Epidemic status of Angiostrongylus cantonensis in Hainan island,China显示文摘Objective:To understand the current epidemiological status of Angiostrongylus cantonensis in Hainan island and provide the scientific evidences for prevention and control of the disease.Methods:The investigation on nature infection condition of intermediate hosts and final hosts of Angiostrongylus cantonensis were carried out in five counties,Hainan Province, and enzyme linked immunosorbent assay(ELISA) was employed to detect antibodies of Angiostrongylus cantonensis in human hosts.Results:A total of 1 612 molluscs were examined, 21.3%of which harbored L3 of Angiostrongylus cantonensis.Among them,the infection rates of Pomacea canaliculata,Achatina fulica,Cepaea and Phlegm bilineatus were 12.3696(64/518), 22.66%(121/534),20.93%(9/43),and 28.24%(146/517),respectively.Of the 118 rats trapped,13 Rattus norvegicus were found to be infected with Angiostrongylus cantonensis.A total of 459 serum samples were collected and tested.92 serum samples were Angiostrongylus cantonensis antibody-positive.Conclusions:The survey revealed a wide distribution of Angiostrongylus cantonensis in Hainan island.Pomacea canaUculata and Achatina fulica are main intermediate hosts of Angiostrongylus cantonensis.Rattus novegicus is a nature definitive host,indicating that a considerable number of people are at risk of angiostrongyliasis.Health education,rigorous food inspection and surveillance are all needed to prevent angiostrongyliasis outbreaks in future.Ximin Hu Jianwei Du Chongjin Tong Shanqing Wang Jian Liu Yuchun Li Changhua He 2011Asian Pacific Journal of Tropical Medicine2011,4,4:2

17The establishment of polypeptide PSMA-targeted chimericantigen receptor-engineered natural killer cells forcastration-resistant prostate cancer and the induction of ferroptosis-related cell death显示文摘Background:The mortality of castration-resistant prostate cancer(CRPC)is high due to lack of an effective treatment.Chimeric antigen receptor(CAR)-based therapy is a promising immunotherapeutic strategy.Here,we aimed to design a novel CAR-natural killer(NK)cells with a clinically significant tumoricidal effect on CRPC.Methods:We constructed novel CAR-NK92MI cells with a CD244-based recombinant lentiviral vector.Different intracellular segments(CD244,NKG2D,or CD3ζ)were screened to identify the best candidate according to cell lysis assay and CD107a expression levels.To enhance the affinity of the CAR to the tumor antigen,we compared an antibody specific for prostate-specific membrane antigen(anti-PSMA)with PSMA-targeted polypeptide(p-PSMA),which was screened by phage display combinatorial library.Then,CAR-NK92MI cells with both a high affinity for PSMA and a strong tumoricidal capacity were generated.In addition,we verified their tumor-killing effect in vitro and in vivo.The release of cytokine by NK92MI cells was compared with that by CAR-NK92MI cells through flow cytometry and enzyme-linked immunosorbent assay.Moreover,ferroptosis-related cell death was explored as a possible underlying mechanism.Results:Three different CAR intracellular regions CAR1(CD244),CAR2(CD244,NKG2D)and CAR3(CD244,NKG2D,and CD3ζ)were constructed.CAR2 was chosen to confer a stronger tumoricidal ability on CAR-NK92MI cells.Compared with anti-PSMA,p-PSMA exhibited enhanced affinity for the tumor antigen.Thus,p-PSMA-CAR-NK92MI cells,which expressed CAR with a polypeptide-based antigen-binding region,an intracellular CD244 and a NKG2D costimulatory domain,were generated.They could selectively and successfully kill PSMA+target cells and exhibited specific lysis rate of 73.19%for PSMA-positive C4-2 cells and 33.04%for PSMA-negative PC3 cells.Additionally,p-PSMA-CAR-NK92MI cells had significantly higher concentrations of IFN-γ,TNF-αand granzyme B than NK92MI cells.In a CRPC cancer xenograft model,p-PSMA-CAR-NK92MI cells significantly inhibited tumor growth and exerted a more consistent killing effect than NK92MI cells.Moreover,ferroptosis is a potential mechanism through which CAR-NK92MI cells attack cancer cells,and is triggered by IFN-γ.Conclusions:p-PSMA-CAR-NK92MI cells can effectively kill CRPCPSMA+cells in vitro and in vivo.This strategy may provide additional treatment options for patients with CRPC.Liyuan Wu Fei Liu Le Yin Fangming Wang Hui Shi Qinxin Zhao Feiya Yang Dong Chen Xiying Dong Yuchun Gu Nianzeng Xing 2022Cancer Communications2022,42,8:2

19A rice XANTHINE DEHYDROGENASE gene regulates leaf senescence and response to abiotic stresses显示文摘Xanthine dehydrogenase, a member of the molybdenum enzyme family, participates in purine metabolism and catalyzes the generation of ureides from xanthine and hypoxanthine. However, the mechanisms by which xanthine dehydrogenase affects rice growth and development are poorly understood. In the present study, we identified a mutant with early leaf senescence and reduced tillering that we named early senescence and less-tillering 1(esl1). Map-based cloning revealed that ESL1 encodes a xanthine dehydrogenase, and it was expressed in all tissues. Chlorophyll content was reduced and chloroplast maldevelopment was severe in the esl1 mutant. Mutation of ESL1 led to decreases in allantoin, allantoate, and ABA contents. Further analysis revealed that the accumulation of reactive oxygen species in esl1 resulted in decreased photosynthesis and impaired chloroplast development, along with increased sensitivity to abscisic acid and abiotic stresses.
