| 2 | Organic carbon accumulation capability of two typical tidal wetland soils in Chongming Dongtan,China显示文摘We measured organic carbon input and content of soil in two wetland areas of Chongming Dongtan(Yangtze River Estuary) to evaluate variability in organic carbon accumulation capability in different wetland soils.Observed differences were investigated based on the microbial activity and environmental factors of the soil at the two sites.Results showed that the organic carbon content of wetland soil vegetated with Phragmites australis(site A) was markedly lower than that with P.australis and Spartina alterniflora(site B).Sites differences were due to higher microbial activity at site A,which led to higher soil respiration intensity and greater carbon outputs.This indicated that the capability of organic carbon accumulation of the site B soils was greater than at site A.In addition,petroleum pollution and soil salinity were different in the two wetland soils.After bio-remediation,the soil petroleum pollution at site B was reduced to a similar level of site A.However,the culturable microbial biomass and enzyme activity in the remediated soils were also lower than at site A.These results indicated that greater petroleum pollution at site B did not markedly inhibit soil microbial activity.Therefore,differences in vegetation type and soil salinity were the primary factors responsible for the variation in microbial activity,organic carbon output and organic carbon accumulation capability between site A and site B. | Shiping Zhang Lei Wang Jiajun Hu Wenquan Zhang Xiaohua Fu Yiquan Le Fangming Jin | 2011 | Journal of Environmental Sciences2011,23,1: | 11 |
| 3 | Internet of Things to network smart devices for ecosystem monitoring显示文摘Smart, real-time, low-cost, and distributed ecosystem monitoring is essential for understanding and managing rapidly changing ecosystems. However, new techniques in the big data era have rarely been introduced into operational ecosystem monitoring, particularly for fragile ecosystems in remote areas.We introduce the Internet of Things(IoT) techniques to establish a prototype ecosystem monitoring system by developing innovative smart devices and using IoT technologies for ecosystem monitoring in isolated environments. The developed smart devices include four categories: large-scale and nonintrusive instruments to measure evapotranspiration and soil moisture, in situ observing systems for CO2 and d13 C associated with soil respiration, portable and distributed devices for monitoring vegetation variables, and Bi-CMOS cameras and pressure trigger sensors for terrestrial vertebrate monitoring. These new devices outperform conventional devices and are connected to each other via wireless communication networks. The breakthroughs in the ecosystem monitoring IoT include new data loggers and longdistance wireless sensor network technology that supports the rapid transmission of data from devices to wireless networks. The applicability of this ecosystem monitoring IoT is verified in three fragile ecosystems, including a karst rocky desertification area, the National Park for Amur Tigers, and the oasis-desert ecotone in China. By integrating these devices and technologies with an ecosystem monitoring information system, a seamless data acquisition, transmission, processing, and application IoT is created. | 4 | Extraction, identification, and antioxidant property evaluation of limonin from pummelo seeds显示文摘Limonin, the main bioactive phytochemical constituent of limonoids with multi-functions, is enriched in citrus fruits and often found at a high concentration in citrus seeds. The present study was attempted to introduce a new and efficient extraction method to isolate limonoids from pummelo seeds, and to evaluate the antioxidant property of the main constituent limonin in Hep G2 cells. Three key single factors were identified for the extraction of limonoids from pummelo seeds using the Box-Behnken experiment design of response surface methodology(RSM), and the optimized extraction parameters were treatment with 89.68 m L of anhydrous acetone for 4.62 h at 78.94C, while the yield of limonoids was 11.52 mg/g. The structure of isolated main constituent of the limonoids was further identified as limonin by Fourier transform infrared(FT-IR) spectrometer and nuclear magnetic resonance(NMR)spectrum. Moreover, the molecular data in Hep G2 cells revealed that limonin exerted its anti-oxidant property mainly by the activation of nuclear factor(erythroid-2)-like 2(Nrf2)/kelch-like ECHassociated protein 1(Keap1)-antioxidant response element(ARE) pathway in the form of transcriptional regulation of Nrf2 m RNA and posttranscriptional regulation of Nrf2/Keap1 system. | 5 | Variation in cis-regulation of a NAC transcription factor contributes to drought tolerance in wheat显示文摘Drought is a major environmental factor limiting wheat production worldwide,and developing drought-tolerant cultivars is a central challenge for wheat breeders globally.Therefore,it is important to identify genetic components determining drought tolerance in wheat.In this study,we identified a wheat NAC gene(TaNAC071-A)that is tightly associated with drought tolerance by a genome-wide association study.Knockdown of TaNAC071-A in wheat attenuated plant drought tolerance,whereas its overexpression significantly enhanced drought tolerance through improved water-use efficiency and increased expression of stress-responsive genes.This heightened water-saving mechanism mitigated the yield loss caused by water deficit.Further candidate gene association analysis showed that a 108-bp insertion in the promoter of TaNAC071-A alters its expression level and contributes to variation in drought tolerance among wheat accessions.This insertion contains two MYB cis-regulatory elements(CREs)that can be directly bound by the MYB transcription activator,TaMYBL1,thereby leading to increased TaNAC071-A expression and plant drought tolerance.Importantly,introgression of this 108-bp insertion allele,TaNAC071-AIn-693,into drought-sensitive cultivars could improve their drought tolerance,demonstrating that it is a valuable genetic resource for wheat breeding.Taken together,our findings highlight a major breakthrough in determining the genetic basis underlying phenotypic variation in wheat drought tolerance and showcase the potential of exploiting CRE-containing indels for improving important agronomical traits. | Hude Mao Shumin Li Bin Chen Chao Jian Fangming Mei Yifang Zhang Fangfang Li Nan Chen Tian Li Linying Du Li Ding Zhongxue Wang Xinxiu Cheng Xiaojing Wang Zhensheng Kang | 2022 | Molecular Plant2022,15,2: | 9 |

| 7 | Coagulation factors VII, IX and X are effective antibacterial proteins against drug-resistant Gram-negative bacteria显示文摘Infections caused by drug-resistant“superbugs”pose an urgent public health threat due to the lack of effective drugs;however, certain mammalian proteins with intrinsic antibacterial activity might be underappreciated. Here, we reveal an antibacterial property against Gram-negative bacteria for factors VII, IX and X, three proteins with well-established roles in initiation of the coagulation cascade. These factors exert antibacterial function via their light chains (LCs). Unlike many antibacterial agents that target cell metabolism or the cytoplasmic membrane, the LCs act by hydrolyzing the major components of bacterial outer membrane, lipopolysaccharides, which are crucial for the survival of Gram-negative bacteria. The LC of factor VII exhibits in vitro efficacy towards all Gram-negative bacteria tested, including extensively drug-resistant (XDR) pathogens, at nanomolar concentrations. It is also highly effective in combating XDR Pseudomonas aeruginosa and Acinetobacter baumannii infections in vivo. Through decoding a unique mechanism whereby factors VII, IX and X behave as antimicrobial proteins, this study advances our understanding of the coagulation system in host defense, and suggests that these factors may participate in the pathogenesis of coagulation disorder-related diseases such as sepsis via their dual functions in blood coagulation and resistance to infection. | 9 | Genetic analysis and molecular mapping of a novel gene for zebra mutation in rice (Oryza sativa L.)显示文摘A novel zebra mutant,zebra-15,derived from the restorer line Jinhui10(Oryza sativa L. ssp. indica) treated by EMS,displayed a distinctive zebra leaf from seedling stage to jointing stage. Its chlorophyll content decreased(55.4%) and the ratio of Chla/Chlb increased(90.2%) significantly in the yellow part of the zebra-15,compared with the wild type. Net photosynthetic rate and fluorescence kinetic parameters showed that the decrease of chlorophyll content significantly influenced the photosynthetic efficiency of the mutant. Genetic analysis of F2 segregation populations derived from the cross of Xinong1A and zebra-15 indicated that the zebra leaf trait is controlled by a single recessive nuclear gene. Ninety-eight out of four hundred and eighty pairs of SSR markers showed the diversity between the Xinong1A and the zebra-15,their F2 population was then used for gene mapping. Zebra-15(Z-15) gene was primarily restricted on the short arm of chromosome 5 by 150 F2 recessive individuals,19.6 cM from marker RM3322 and 6.0 cM from marker RM6082. | 10 | Identification of Rice QTLs for Important Agronomic Traits with Long-Kernel CSSL-Z741 and Three SSSLs显示文摘Rice kernel shape affects kernel quality(appearance) and yield(1000-kernel weight) and therefore is an important agronomic trait, but its inheritance is complicated. We identified a long-kernel rice chromosome segment substitution line(CSSL), Z741, derived from Nipponbare as a recipient and Xihui 18 as a donor parent. Z741 has six substitution segments distributed on rice chromosomes 3, 6, 7, 8 and 12 with an average replacement length of 5.82 Mb. Analysis of a secondary F2 population from a cross between Nipponbare and Z741 identified 20 QTLs for important agronomic traits. The kernel length of Z741 is controlled by a major QTL(qKL3) and a minor QTL(qKL7). Candidate gene prediction and sequencing indicated that qKL3 may be an allele of OsPPKL1, which encodes a protein phosphatase implicated in brassinosteroid signaling, and qKL7 is an unreported QTL. Finally, we validated eight QTLs(qKL3, qKL7, qRLW3-1, qRLW7, qPH3-1, qKWT3, qKWT7 and qNPB6) using three selected singlesegment substitution lines(SSSLs), S1, S2 and S3. Also, we detected five QTLs(qKL6, qKW3, qKW7, qKW6 and qRLW6) in S1, S2 and S3, which were not found in the Nipponbare/Z741 F2 population. However, qNPB3, qNPB7 and qPL3 QTLs were not validated by the three SSSLs in 2019, suggesting that minor QTLs are susceptible to environmental factors. | 14 | Dynamic changes in fibrinogen levels of patients with acute cerebral infarction after taking defibrase显示文摘BACKGROUND: At present, as a therapeutic drug mainly for reducing fibrinogen (FIB) levels, the dynamic influence of defibrase on the FIB levels of patients with acute cerebral infarction has not been clearly ascertained. OBJECTIVE: To observe the dynamic changes in FIB levels of patients with acute cerebral infarction at different time points after taking defibrase. DESIGN, TIME AND SETTING: Randomized controlled clinical trial. The study was conducted in the Department of Neurology, the Second Affiliated Hospital of Jinan University, from June to November 2006. PARTICIPANTS: Sixty patients with acute cerebral infarction, who had been treated by the Neurological Department of the Second Affiliated Hospital of Jinan University from June to November 2006, were selected, including 37 males and 23 females, aged 35–75 years. All cases met the diagnostic criteria formulated by the Fourth National Cerebrovascular Disease Conference within 12 hours of onset. All the patients were confirmed with definite hemiparesis and cerebral infarction without coma, and were randomly divided into two groups: a treatment group (n =40) and a control group (n =20). Patients’ families had the right to be informed and agree with the treatment, which had permission from the Hospital Ethics Committee. METHODS: Patients in the control group were given routine treatment with 30 mL fleabane and 0.75 g cytidine diphosphate added to 500 mL saline solution once a day for 14 consecutive days. Patients in the treatment group were given routine treatment and Haiwang defibrase injection (purchased from Changchu Guoao Bio-Pharmaceutical Co. Ltd., Approval document number H10983237) within 12 hours of infarction. Defibrase doses of 15, 12.5 and 10 U were given over 2 hours according to the patients’ pre-treatment plasma FIB levels of ≥ 4.50 g/L, 3.50-4.49 g/L and 1.00-3.49 g/L, respectively. Plasma FIB levels in the treatment group were measured before, and once every six hours for 48 hours after administration of defibrase. Later, measurements were taken once every 12 hours and FIB levels were kept in a range of 0.5–1.3 g/L for one week. When the FIB level increased to over 1.3 g/L, a 5 U dose of defibrase was given again over two hours. FIB levels of the control group were measured once before treatment and once after one week of treatment MAIN OUTCOME MEASURES: (1) Dynamic changes of FIB levels in the defibrase treatment group. (2) Comparison of dynamic changes of FIB levels in the treatment group and control group before and after treatment for one week. RESULTS: All 60 patients were included in the final analysis. The treatment group’s FIB levels quickly decreased to 0.5–1.3 g/L within 12 hours of taking the first dose of defibrase and reached a minimum in 24 hours. Later, they began to rise slowly into the therapeutic range (0.5–1.3 g/L) in 48 hours. The FIB levels of the treatment group increased slowly to over 1.3 g/L in 60 hours after the first dose of defibrase and then quickly decreased to the therapeutic range after the second dose of 5 U defibrase with a minimum level higher than after the first dose, and higher again after the third dose. After treatment, patients’ FIB levels could be kept in the therapeutic range for about one week. The FIB levels in patients in the treatment group were significantly lower after taking defibrase for one week in comparison with the levels before treatment (P < 0.01). There was no difference in the control group between the levels pre-treatment and one week after treatment (P > 0.05). CONCLUSIONS: FIB levels in patients with acute cerebral infarction quickly decrease after taking the first dose of defibrase and reach a minimum in 24 hours. Later, they begin to rise slowly into the therapeutic range (0.5–1.3 g/L) in 48 hours. The FIB levels increase slowly to over 1.3 g/L in 60 hours. The rice CSSL Z1364 with increased kernel number was identified in a BC3F8 population derived from a cross of Nipponbare as the recipient with Xihui 18 as the donor parent. Z1364 carried three substitution segments distributed on chromosomes 1, 6, and 8. The mean substitution length was 1.19 Mb. Of 17 QTL identified on the substitution segments, qSP1 for spikelets per panicle, qSSD1 for seed-set density, and qNSB1 for number of secondary branches explained respectively 57.34%, 87.7%, and 49.44% of the corresponding phenotypic variance and were all linked to RM6777. Chi-square analysis showed that the increased kernel number in Z1364 was inherited recessively by a single gene. By fine mapping, qSP1 was delimited to a 50-kb region on the short arm of chromosome 1. Based on DNA sequence, a previously uncharacterized rice homolog of Arabidopsis thaliana AT4G32551 was identified as a candidate gene for qSP1 in which mutation increases the number of spikelets and kernels in Z1364. qSP1 was expressed in all tissues, but particularly in 1-cm panicles. The expression levels of OsMADS22, GN1A, and DST were upregulated and those of LAX2, GNP1, and GHD7 were downregulated in Nipponbare. | 16 | Online forecasting model of tundish nozzle clogging显示文摘A nozzle clogging online forecasting model based on hydrodynamics engineering was developed, in which the actual flow rate was calculated from the mold width, thickness, and casting speed. There is a linear relationship between the theoretical flow rate and the slide gate opening ratio as the molten steel level, argon flow rate, and the top slag weight are kept constant, and the relationship can be obtained by regression of the data collected at the beginning of the first heat in each casting sequence when the nozzle clogging does not occur. Then, during the casting, the theoretical flow rate can be calculated at intervals of one second. Comparing the theoretical flow rate with the actual flow rate, the online nozzle clogging ratio can be obtained at intervals of one second. A cold-adapted gram-negative bacterium, hydrocarbon-degrading Pseudoalteromonas sp. NJ289, was isolated from sea-ice of the Antarctica region, and sequenced the whole genome through the next generation sequencing platform. The assembly yielded three contigs representing two chromosomes and one plasmid with the sizes of 3.2 Mb, 636 kb and 1.8 kb, respectively. The G+C contents of genome were 40.83% and included 3 589 ORFs. Functional annotation indicated some potential roles in enzymatic activity and environmental adaptability.
