1Mannan-binding lectin directly interacts with Toll-like receptor 4 and suppresses lipopolysaccharide-induced inflammatory cytokine secretion from THP-1 cells显示文摘Mannan-binding lectin(MBL)plays a key role in the lectin pathway of complement activation and can influence cytokine expression.Toll-like receptor 4(TLR4)is expressed extensively and has been demonstrated to be involved in lipopolysaccharide(LPS)-induced signaling.We first sought to determine whether MBL exposure could modulate LPS-induced inflammatory cytokine secretion and nuclear factor-kB(NF-kB)activity by using the monocytoid cell line THP-1.We then investigated the possible mechanisms underlying any observed regulatory effect.Using ELISA and reverse transcriptase polymerase chain reaction(RT-PCR)analysis,we found that at both the protein andmRNAlevels,treatment withMBLsuppresses LPS-induced tumor-necrosis factor(TNF)-a and IL-12 production in THP-1 cells.An electrophoretic mobility shift assay and western blot analysis revealed that MBL treatment can inhibit LPS-induced NF-kB DNA binding and translocation in THP-1 cells.While the binding of MBL to THP-1 cells was evident at physiological calcium concentrations,this binding occurred optimally in response to supraphysiological calcium concentrations.This binding can be partly inhibited by treatment with either a soluble form of recombinant TLR4 extracellular domain or anti-TLR4 monoclonal antibody(HTA125).Activation of THP-1 cells by LPS treatment resulted in increased MBL binding.We also observed that MBL could directly bind to the extracellular domain of TLR4 in a dose-dependent manner,and this interaction could attenuate the binding of LPS to cell surfaces.Taken together,these data suggest that MBL may affect cytokine expression through modulation of LPS-/TLR-signaling pathways.These findings suggest that MBL may play an important role in both immune regulation and the signaling pathways involved in cytokine networks.Mingyong Wang Yue Chen Yani Zhang Liyun Zhang Xiao Lu Zhengliang Chen 2011Cellular & Molecular Immunology2011,8,3:21
2Influences of preload on the friction and wear properties of high-speed instrument angular contact ball bearings显示文摘For starved-oil or solid lubrication of high-speed instrument angular contact ball bearings, friction heating and wear are the main reasons of bearing failures. This paper presents a dynamic wear simulation model to investigate the impacts of different preload methods and the changes of preload caused by wear on bearing wear life. The integral value QV of stress and sliding velocity in the contact ellipses between a ball and the inner and outer races determines friction heating and wear. The changes of QV with the friction coefficient and the wear volume under constantforce preload and fixed-position preload are analyzed. Results show that under the same initial preload, the QV decreases with an increase of the friction coefficient for both preload methods, and the latter is slightly larger. The wear of the ball and the race is equivalent to the ball diameter reduction.The QV of constant-force preload is almost not changed with a decrease of the ball diameter, but for fixed-position preload, the value decreases firstly and then increases substantially due to insufficient preload, and slipping occurs, the ball diameter is reduced by 0.025%, while the preload is reduced by 60.33%. 3Interfacial compatibility issues in rechargeable solid-state lithium metal batteries:a review显示文摘Solid-state lithium metal batteries(SSLBs)contain various kinds of interfaces,among which the solid electrode|solid electrolyte(ED|SE)interface plays a decisive role in the battery's power density and cycling stability.However,it is still lack of comprehensive knowledge and understanding about various interfacial physical/chemical processes so far.Although tremendous efforts have been dedicated to investigate the origin of large interfacial resistance and sluggish charge(electron/ion)transfer process,many scientific and technological challenges still remain to be clarified.In this review,we detach and discuss the critical individual challenge,including charge transfer process,chemical and electrochemical instability,space charge layers,physical contact and mechanical instability.The fundamental concepts,individual effects on the charge transfer and potential solutions are summarized based on material's thermodynamics,electrode kinetics and mechanical effects.It is anticipated that future research should focus on quantitative analysis,modeling analysis and in-situ microstructure characterizations in order to obtain an efficient manipulation about the complex interfacial behaviors in all solid-state Li batteries.Hongchun Wang Jianping Zhu Yu Su Zhengliang Gong Yong Yang 2021Science China Chemistry2021,64,6:3
4Leachability and fractionation of heavy metals in stainless steelmaking plant dusts显示文摘The leachability and fractionation of the selected heavy metals (Cr, Cd, Pb and Zn) in the stainless steelmaking plant dusts were studied by the modified HJ/T299-2007 method, the modified TCLP test and a seven-step sequential extraction procedure. The results show that the stainless steelmaking plant dusts are considered as hazardous materials since at least one of the leachable heavy metals exceeds the corresponding limit levels of State Environmental Protection Administration of China. Pb is the most extractable heavy metals by the sequential extraction procedure, while Cr, Cd and Zn mainly presents in the residual fraction bound to spinel phase and silicates. The mobile heavy metals in the dusts, which present as the species of water soluble, exchangeable species and the species bounded to carbonates and hydroxides, can be estimated as 0.08-1.2% of Cr, 7.6%-31.8% of Cd, 34.1% of Pb, 0.052-0.4% of Zn, respectively.Guojun MA Wei FAN Zhengliang XUE Wei WANG Hui TANG 2010Acta Metallurgica Sinica(English Letters)2010,23,4:2

20Glioma stem cells and neural stem cells respond differently to BMP4 signaling显示文摘Malignant glioma is a highly heterogeneous and invasive primary brain tumor characterized by high recurrence rates,resistance to combined therapy,and dismal prognosis.Glioma stem cells(GSCs)are likely responsible for tumor progression,resistance to therapy,recurrence,and poor prognosis owing to their high self-renewal and tumorigenic potential.As a family member of BMP signaling,bone morphogenetic protein4(BMP4)has been reported to induce the differentiation of GSCs and neural stem cells(NSCs).However,the molecular mechanisms underlying the BMP4-mediated effects in these two cell types are unclear.In this study,we treated hGSCs and hNSCs with BMP4 and com-pared the phenotypic and transcriptional changes between these two cell types.Phenotypically,we found that the growth of hGSCs was greatly inhibited by BMP4,but the same treatment only increased the cell size of hNSCs.While the RNA sequencing results showed that BMP4 treatment evoked significantly transcriptional changes in both hGSCs and hNSCs,the profiles of differentially expressed genes were distinct between the two groups.A gene set that specifically targeted the proliferation and differentiation of hGSCs but not hNSCs was enriched and then validated in hGSC culture.Our results suggested that hGSCs and hNSCs responded differently to BMP4 stimulation.Understanding and investigating different responses between hGSCs and hNSCs will benefit finding partner factors working together with BMP4 to further suppress GSCs proliferation and stemness without disturbing NSCs.Xin-Xin Han Chunhui Cai Li-Ming Yu Min Wang Wenhan Yang Dai-Yu Hu Jie Ren Lu-Ying Zhu Jia-Jia Deng Qing-Qing Chen Hua He Zhengliang Gao 2022Cell Regeneration2022,11,1:1
