1Evaluation of a granulated formula for the nerve root type and vertebral artery type of cervical spondylosis:a multicenter,single-blind,randomized,controlled,phase Ⅲ clinical trial显示文摘OBJECTIVE:To evaluate the effectiveness and safety of cervical spondylosis formula granules in reducing the symptoms of patients with the nerve root type and the vertebral artery type of cervical spondylosis.METHODS:This was a multicenter, single-blind,randomized, controlled trial.From April 2002 to November 2003, 499 patients were randomly assigned to either the treatment or the control group.The treatment group was orally administered granules prepared with a formula for cervical spondylosis, while the control group was given Jingfukang granules.The treatment course was 1 month for both groups.RESULTS:In patients with the nerve root type of cervical spondylosis, the total effect rate in the treatment group(87.21%) was significantly higher than that in the control group(80.70%, P < 0.01).After the treatment period in both groups, the treatment group had a significantly greater rate of resolution of pain, numbness of the upper limbs, muscle strength of the upper limbs, and fatigue than the control group(all P < 0.05).In patients with the vertebral artery type of cervical spondylosis, the total effect rate in the treatment group(82.07%) was similar to that in the control group(71.21%, P >0.05).After the treatment period in both groups,the treatment group had a significantly greater rate of resolution of weakness of the waist and knees than the control group(P < 0.05).CONCLUSION:The cervical spondylosis formula granules significantly improve numbness, muscle strength, and fatigue, and reduce pain in patients with the nerve root type of cervical spondylosis,and improve the weakness of the waist and knees in patients with the vertebral artery type of cervical spondylosis.Luo Xiaodong Wang Peng Li Zhe Liang De Huang Feng Liu Jinwen Chen Xinyu Hu Guoheng Fang Suting Zhang Huajian 2017Journal of Traditional Chinese Medicine2017,37,2:28
2Community Structure and Seasonal Variation of Soil Arthropods in the Forest-Steppe Ecotone of the Mountainous Region in Northern Hebei,China显示文摘Soil fauna have been receiving more and more attention because they play an important role in nutrient cycling.However,there is a lack of information on soil arthropods in the forest-steppe ecotone in the mountainous region of northern Hebei,which makes it difficult to meet the need of protecting biodiversity in this area.Soil arthropod communities were investigated in the forest-steppe ecotone in northern Hebei province to provide basic information on changes in mountain soil fertility,which could promote the development of soil arthropod communities in mountain ecotones.From the preliminary identification,a total of 7994 individual soil arthropods were collected,which belonged to 25 groups,6 classes and 24 orders.Acarina,Hymenoptera and Collembola were the dominant groups in the ecotone.The number of Acarina was higher than Collembola,and this phenomenon was obviously different from other areas in the same climate zone.The increased abundance of rare groups in the Forest zone with the richer vegetation,higher arthropod abundance and more substantial litter depth,could be interpreted as a reaction to the suitable soil environment and food supply.And these rare groups were sensitive to environmental changes,which could be regarded as biotic indicators for evaluating soil quality.The analysis of community diversity showed that the abundance index (d),the Shannon-Wiener index (H'),the evenness index (J) and the density-group index (DG) were significantly higher in the forest zone,lower in the forest-steppe zone,and lowest in the meadow-steppe zone.Seasonal variations in community composition correlated with changes in average air temperature and precipitation in this ecotone.Groups and individuals of soil arthropod communities in the three zones were present in greater numbers in the middle of the rainy season than in the early or late periods of the rainy season as a whole.At the same time,seasonal changes in soil arthropod communities from different plots were also influenced by habitat condition.ZHU Xinyu GAO Baojia YUAN Shengliang HU Yunchuan 2010Journal of Mountain Science2010,7,2:17
3The HuangZaoSi Maize Genome Provides Insights into Genomic Variation and Improvement History of Maize显示文摘Maize is a globally important crop that was a classic model plant for genetic studies. Here, we report a 2.2 Gb draft genome sequence of an elite maize line, HuangZaoSi (HZS). Hybrids bred from HZS-improved lines (HILs) are planted in more than 60% of maize fields in China. Proteome clustering of six completed sequeneed maize genomes show that 638 proteins fall into 264 HZS-specific gene families with the majority of contributions from tandem duplication events. Resequencing and comparative analysis of 40 HZSrelated lines reveals the breeding history of HILs. More than 60% of identified selective sweeps were clustered in identity.by.descent conserved regions, and yield-related genes/QTLs were enriched in HZS characteristic selected regions. Furthermore, we dem on strated that HZS-specific family genes were not uniformly distributed in the genome but enriched in improvement/function.related genomic regions. 5Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis.However,little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins,including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types,and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels.The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies.Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry,providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations.Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu 2022Cell Research2022,32,1:8
6Tianshanbeilu and the Isotopic Millet Road: reviewing the late Neolithic/Bronze Age radiation of human millet consumption from north China to Europe显示文摘The westward expansion of human millet consumption from north China has important implications for understanding early interactions between the East and West.However,few studies have focused on the Xinjiang Uyghur Autonomous Region,the vast geographical area directly linking the ancient cultures of the Eurasian Steppe and the Gansu Corridor of China.In this study,we present the largest isotopic investigation of Bronze Age China(n=110)on material from the key site of Tianshanbeilu,in eastern Xinjiang.The large range ofδ13C values(–17.6 to–7.2;–15.5±1.2)provides direct evidence of unique dietary diversity and consumption of significant C4 resources(millets).The highδ15N results(10.3 to 16.7;14.7±0.8)likely reflect sheep/goat and wild game consumption and the arid climate of the Taklamakan Desert.Radiocarbon dates from four individuals indicate Tianshanbeilu was in use between 1940 and1215 cal bc.The Tianshanbeilu results are then analysed with respect to 52 Bronze Age sites from across Eurasia,to investigate the spread and chronology of significant human millet consumption and human migration.This isotopic survey finds novel evidence that the second millennium bc was a dynamic period,with significant dietary interconnectivity occurring between north China,Central Asia and Siberia.Further,we argue that this‘Isotopic Millet Road’extended all the way to the Mediterranean and Central Europe,and conclude that these C4 dietary signatures of millet consumption reflect early links(migration and/or resource transfer)between the Bronze Age inhabitants of modern-day China and Europe.Tingting Wang Dong Wei Xien Chang Zhiyong Yu Xinyu Zhang Changsui Wang Yaowu Hu Benjamin T.Fuller 2019National Science Review2019,6,5:8
7Shear creep characteristics and constitutive model of limestone显示文摘The characters of limestone in weak interlayer of a high rocky slope in Xuzhou, China, are studied by shear static test and shear creep test. The results show that limestone specimens have attenuation creep properties and constant rate creep properties, almost have no accelerated creep properties. The exponential type empirical formula is selected to fit creep grading curves by polynomial regression analysis method, and the square sums of the fitting results residual are in the order of 10^(-7). Then grade creep curves at every shear loads are set up. Combining creep rate-time curve, the creep properties of limestone are analyzed. As the physical meaning of component model is clearer, the Poytin–Thomson model is set up. Through the least square method, the optimal parameters of Poytin–Thomson model are obtained,and the sums of squared residuals belong to 10^(-3)order of magnitude, which can meet the accuracy requirements of engineering calculation. 8Growth and characterization of 0.8-μm gate length AlGaN/GaN HEMTs on sapphire substrates显示文摘AlGaN/GaN high electron mobility transistor (HEMT) structures were grown on 2 inch sapphire substrates by MOCVD, and 0.8-μm gate length devices were fabricated and measured. It is shown by resistance mapping that the HEMT structures have an average sheet resistance of approximately 380 ?/sq with a uniformity of more than 96%. The 1-mm gate width devices using the materials yielded a pulsed drain current of 784 mA/mm at Vgs=0.5 V and Vds=7 V with an extrinsic transconductance of 200 mS/mm. A 20-GHz unity current gain cutoff frequency (?T ) and a 28-GHz maximum oscillation frequency (?max) were obtained. 9Analysis of insidious fault activation and water inrush from the mining floor显示文摘Based on the stress field distribution rule of the mining floor under abutment pressure, we have established a simplified mechanical model, which contains multiple factors relating to activation and evolution of insidious water-conductive faults. The influence of normal and shear stresses on fault activation and effective shear stress distribution in the fault plane was acquired under mining conditions.Using fracture mechanics theory to calculate the stress intensity factor of an insidious fault front, we have derived the criterion for main fault activation. Results indicate that during the whole working face advance, transpressions are exerted on fault planes twice successively in opposite directions. In most cases, the second transpression is more likely to lead to fault activation. Activation is influenced by many factors, predominant among which are: burial depth of the insidious fault, friction angle of the fault plane, face advance direction and pore water pressure. 10CMHX008,a PPAR γ partial agonist,enhances insulin sensitivity with minor influences on bone loss显示文摘Traditional thiazolidinediones(TZDs),such as rosiglitazone,are peroxisome proliferator-activated receptor g(PPARg)potent agonists that can be used to treat type 2 diabetes but carry unwanted effects,including increased risk for fracture.The present work aimed to compare the insulin-sensitizing efficacies and bone-loss side effects of CMHX008,a novel TZDs-like PPARg partial agonist,with those of rosiglitazone.A TR-FRET PPARg competitive binding assay was used to compare the binding affinity between CMHX008 and rosiglitazone.Mice were administered vehicle,CMHX008 or rosiglitazone for 16 weeks.Mesenchymal stem cells(MSCs)were used to examine differences in differentiation into osteoblasts after compounds treatment.TR-FRET showed lower affinity to PPARg by CMHX008 compared with rosiglitazone.Mice treated with CMHX008 showed insulin sensitization similar to that of mice treated with rosiglitazone,which was related to the significant inhibition of PPARg Ser273 phosphorylation and improved insulin sensitivity by facilitating the phosphorylation of insulin receptor and Akt in adipose tissues.Micro-CT and histomorphometric analyses demonstrated that the degree of trabecular bone loss after treatment with CMHX008 was weaker than that observed with rosiglitazone,as evidenced by consistent changes in BV/TV,Tb.N,Tb.Th,Tb.Sp,and the mineral apposition rate.MSCs treated with CMHX008 showed higher ALP activity and mRNA levels of bone formation markers than did cells treated with rosiglitazone in the osteoblast differentiation test.Thus,CMHX008 showed insulin-sensitizing effects similar to those of rosiglitazone with a lower risk of bone loss,suggesting that PPARg sparing eliminates the skeletal side effects of TZDs while maintaining their insulin-sensitizing properties.Yi Hou Xuemei Cao Xiangnan Hu Xinyu Li Xiaoqin Shi Hongying Wang Chuan Peng Jiayu Li Jibin Li Qifu Li Chaodong Wu Xiaoqiu Xiao 2018Genes & Diseases2018,5,3:6
11The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 13Coriander Genomics Database:a genomic,transcriptomic,and metabolic database for coriander显示文摘Coriander(Coriandrum sativum L.),also known as cilantro,is a globally important vegetable and spice crop.Its genome and that of carrot are models for studying the evolution of the Apiaceae family.Here,we developed the Coriander Genomics Database(CGDB,http://gffzzc8ce0bd092c048a1hp9ubv0xqqp0o60uk.ffgz.tsg.suse.edu.cn/)to collect,store,and integrate the genomic,transcriptomic,metabolic,functional annotation,and repeat sequence data of coriander and carrot to serve as a central online platform for Apiaceae and other related plants.Using these data sets in the CGDB,we intriguingly found that seven transcription factor(TF)families showed significantly greater numbers of members in the coriander genome than in the carrot genome.The highest ratio of the numbers of MADS TFs between coriander and carrot reached 3.15,followed by those for tubby protein(TUB)and heat shock factors.As a demonstration of CGDB applications,we identified 17 TUB family genes and conducted systematic comparative and evolutionary analyses.RNA-seq data deposited in the CGDB also suggest dose compensation effects of gene expression in coriander.CGDB allows bulk downloading,significance searches,genome browser analyses,and BLAST searches for comparisons between coriander and other plants regarding genomics,gene families,gene collinearity,gene expression,and the metabolome.A detailed user manual and contact information are also available to provide support to the scientific research community and address scientific questions.CGDB will be continuously updated,and new data will be integrated for comparative and functional genomic analysis in Apiaceae and other related plants.Xiaoming Song Fulei Nie Wei Chen Xiao Ma Ke Gong Qihang Yang Jinpeng Wang Nan Li Pengchuan Sun Qiaoying Pei Tong Yu Jingjing Hu Xinyu Li Tong Wu Shuyan Feng Xiu-Qing Li Xiyin Wang 2020Horticulture Research2020,7,1:4
14Fatigue crack propagation behavior of Ni-based superalloys after overloading at elevated temperatures显示文摘The fatigue crack propagation behavior of three superalloys subjected to a single overloading at elevated temperatures was investigated. The fatigue crack propagation rate FCPR versus stress intensity factor range data da/dN—ΔK were calculated using the two-point secant method. It was found that the crack growth rates of the investigated materials were retarded after overloading with an overload ratio R OL=1.6. The size of the plastic zone in the front of the crack tip and its relation to loading level were discussed. The overload retardation effects are attributed to crack closure. 15Clinical significance of germline copy number variation in susceptibility of human diseases显示文摘Germline copy number variation(CNV) is considered to be an important form of human genetic polymorphisms. Previous studies have identified amounts of CNVs in human genome by advanced technologies, such as comparative genomic hybridization, single nucleotide genotyping, and high-throughput sequencing. CNV is speculated to be derived from multiple mechanisms, such as nonallelic homologous recombination(NAHR) and nonhomologous end-joining(NHEJ). CNVs cover a much larger genome scale than single nucleotide polymorphisms(SNPs), and may alter gene expression levels by means of gene dosage, gene fusion, gene disruption, and long-range regulation effects, thus affecting individual phenotypes and playing crucial roles in human pathogenesis. The number of studies linking CNVs with common complex diseases has increased dramatically in recent years. Here, we provide a comprehensive review of the current understanding of germline CNVs, and summarize the association of germline CNVs with the susceptibility to a wide variety of human diseases that were identified in recent years. 16LncDACH1 promotes mitochondrial oxidative stress of cardiomyocytes by interacting with sirtuin3 and aggravates diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy(DCM)is a common complication in diabetic patients.The molecular mechanisms of DCM remain to be fully elucidated.The intronic long noncoding RNA of DACH1(lnc DACH1)has been demonstrated to be closely associated with heart failure and cardiac regeneration.In this study,we investigated the role of lnc DACH1 in DCM and the underlying molecular mechanisms.The expression of lnc DACH1 was increased in DCM hearts and in high glucose-treated cardiomyocytes.Knockout of lnc DACH1 reduced mitochondrial oxidative stress,cell apoptosis,cardiac fibrosis and hypertrophy,and improved cardiac function in DCM mice.Overexpression of lnc DACH1 exacerbated mitochondria-derived reactive oxygen species(ROS)level and apoptosis,decreased activity of manganese superoxide dismutase(Mn-SOD);while silencing of lnc DACH1 attenuated ROS production,mitochondrial dysfunction,cell apoptosis,and increased the activity of Mn-SOD in cardiomyocytes treated with high glucose.Lnc DACH1 directly bound to sirtuin3(SIRT3)and facilitated its degradation by ubiquitination,therefore promoting mitochondrial oxidative injury and cell apoptosis in mouse hearts.In addition,SIRT3 silencing abrogated the protective effects of lnc DACH1 deficiency in cardiomyocytes.In summary,lnc DACH1 aggravates DCM by promoting mitochondrial oxidative stress and cell apoptosis via increasing ubiquitination-mediated SIRT3 degradation in mouse hearts.Inhibition of lnc DACH1 represents a novel therapeutic strategy for the intervention of diabetic cardiomyopathy.Qi Zhang Danyang Li Xue Dong Xiaowen Zhang Junwu Liu Lili Peng Bo Meng Qi Hua Xinyu Pei Lu Zhao Xiaoxi Hu Yang Zhang Zhenwei Pan Yanjie Lu Baofeng Yang 2022Science China(Life Sciences)2022,65,6:4
17Highly sensitive flexible capacitive pressure sensor with a broad linear response range and finite element analysis of micro-array electrode显示文摘With the continuous development of wearable electronics,health care and smart terminals,highly performance flexible pressure sensors present a huge application prospect.In this study,by introducing the micro-array structured electrodes and dielectric layers with high dielectric constant,capacitive pressure sensor fabricated with a brand new preparation strategy and highly sensitive is proposed.The prepared micro-array structure is the basis for sensors with high sensitivity.Besides,the contact area between the two electrodes changes from linear to planar with the increased loading,which result in a wider linear responding range.In addition,by introducing ceramic dielectric material-barium titanate(BT)fillers into the dielectric layer to increase it’s the dielectric constant,the sensitivity of the sensor shows two-fold increase.Moreover,the sensitivity gradients can be tuned by changing the loading contents of BT particles.Hence,compared with parallel board capacitive sensors with ordinary dielectric layer,these sensors exhibit excellent performance as follow,high sensitivity(up to 4.9 kPa^(-1))under low pressure range(0-2500 Pa),low detection limit(<1.7 Pa),short response time(<50 ms),a stable response over 5000 loading-unloading cycles,bending stability and an adjustable sensitivity.Further,the flexible pressure sensor can detect the pressure of the water droplets and monitor human movement behavior.With the facile design and excellent comprehensive properties,the flexible pressure sensor provides a new approach to improve the sensitivity and shows a broad application prospects in the wearable electronics,health care and smart terminals.Longquan Ma Xuecheng Yu Yuanyuan Yang Yougen Hu Xinyu Zhang Huayuan Li Xing Ouyang Pengli Zhu Rong Sun Ching-ping Wong 2020Journal of Materiomics2020,6,2:4
18Galectin-3-centered paracrine network mediates cardiac inflammation and fibrosis upon β-adrenergic insult显示文摘Rapid over-activation of β-adrenergic receptors (β-AR) following acute stress initiates cardiac inflammation and injury by activating interleukin-18 (IL-18),however,the process of inflammation cascades has not been fully illustrated.The present study aimed to determine the mechanisms of cardiac inflammatory amplification following acute sympathetic activation.With bioinformatics analysis,galectin-3 was identified as a potential key downstream effector of β-AR and IL-18 activation.The serum level of galectin-3 was positively correlated with norepinephrine or IL-18 in patients with chest pain.In the heart of mice treated with β-AR agonist isoproterenol (ISO,5 mg kg^(-1)),galectin-3 expression was upregulated markedly later than IL-18 activation,and Nlrp3^(-/-)and Il18^(-/-)mice did not show ISO-induced galectin-3 upregulation.It was further revealed that cardiomyocyte-derived IL-18 induced galectin-3 expression in macrophages following ISO treatment.Moreover,galectin-3deficiency suppressed ISO-induced cardiac inflammation and fibrosis without blocking ISO-induced IL-18 increase.Treatment with a galectin-3 inhibitor,but not a β-blocker,one day after ISO treatment effectively attenuated cardiac inflammation and injury.In conclusion,galectin-3 is upregulated to exaggerate cardiac inflammation and injury following acute β-AR activation,a galectin-3 inhibitor effectively blocks cardiac injury one day after β-AR insult.Guomin Hu Jimin Wu Huijun Gu Xiangning Deng Wenli Xu Shan Feng Shuaixing Wang Yao Song Zhengda Pang Xiuling Deng Aleksandr E.Vendrov Nageswara R.Madamanchi Marschall S.Runge Xinyu Wang Youyi Zhang Han Xiao Erdan Dong 2023Science China(Life Sciences)2023,66,5:3
19mTOR signaling promotes stem cell activation via counterbalancing BMP-mediated suppression during hair regeneration显示文摘Hair follicles(HFs)undergo cycles of degeneration(catagen),rest(telogen),and regeneration(anagen)phases.Anagen begins when the hair follicle stem cells(HFSCs)obtain sufficient activation cues to overcome suppressive signals,mainly the BMP pathway,from their niche cells.Here,we unveil that mTOR complex 1(mTORC1)signaling is activated in HFSCs,which coincides with the HFSC activation at the telogen-to-anagen transition.By using both an inducible conditional gene targeting strategy and a pharmacological inhibition method to ablate or inhibit mTOR signaling in adult skin epithelium before anagen initiation,we demonstrate that HFs that cannot respond to mTOR signaling display significantly delayed HFSC activation and extended telogen.Unexpectedly,BMP signaling activity is dramatically prolonged in mTOR signaling-deficient HFs.Through both gain-and loss-of-function studies in vitro,we show that mTORC1 signaling negatively affects BMP signaling,which serves as a main mechanism whereby mTORC1 signaling facilitates HFSC activation.Indeed,in vivo suppression of BMP by its antagonist Noggin rescues the HFSC activation defect in mTORC1-null skin.Our findings reveal a critical role for mTOR signaling in regulating stem cell activation through counterbalancing BMP-mediated repression during hair regeneration.Zhili Deng Xiaohua Lei Xudong Zhang Huishan Zhang Shuang Liu Qi Chen Huimin Hu Xinyue Wang Lina Ning Yujing Cao Tongbiao Zhao Jiaxi Zhou Ting Chen Enkui Duan 2015Journal of Molecular Cell Biology2015,7,1:3
