1Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance,essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders.The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells,along with the signaling mechanisms governing stem cell self-renewal and differentiation.In this review,we first summarize the biological characteristics of seven types of dental stem cells,including dental pulp stem cells,stem cells from apical papilla,stem cells from human exfoliated deciduous teeth,dental follicle precursor cells,periodontal ligament stem cells,alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva.We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways.We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications.Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade.Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji 2016Genes & Diseases2016,3,4:16
2Sox9 augments BMP2-induced chondrogenic differentiation by downregulating Smad7 in mesenchymal stem cells(MSCs)显示文摘Cartilage injuries caused by arthritis or trauma pose formidable challenges for effective clinical management due to the limited intrinsic proliferative capability of chondrocytes.Autologous stem cell-based therapies and transgene-enhanced cartilage tissue engineering may open new avenues for the treatment of cartilage injuries.Bone morphogenetic protein 2(BMP2)induces effective chondrogenesis of mesenchymal stem cells(MSCs)and can thus be explored as a potential therapeutic agent for cartilage defect repair.However,BMP2 also induces robust endochondral ossification.Although the precise mechanisms through which BMP2 governs the divergence of chondrogenesis and osteogenesis remain to be fully understood,blocking endochondral ossification during BMP2-induced cartilage formation may have practical significance for cartilage tissue engineering.Here,we investigate the role of Sox9-donwregulated Smad7 in BMP2-induced chondrogenic differentiation of MSCs.We find that overexpression of Sox9 leads to a decrease in BMP2-induced Smad7 expression in MSCs.Sox9 inhibits BMP2-induced expression of osteopontin while enhancing the expression of chondrogenic marker Col2a1 in MSCs.Forced expression of Sox9 in MSCs promotes BMP2-induced chondrogenesis and suppresses BMP2-induced endochondral ossification.Constitutive Smad7 expression inhibits BMP2-induced chondrogenesis in stem cell implantation assay.Mouse limb explant assay reveals that Sox9 expands BMP2-stimulated chondrocyte proliferating zone while Smad7 promotes BMP2-intitated hypertrophic zone of the growth plate.Cell cycle analysis indicates that Smad7 induces significant early apoptosis in BMP2-stimulated MSCs.Taken together,our results strongly suggest that Sox9 may facilitate BMP2-induced chondrogenesis by downregulating Smad7,which can be exploited for effective cartilage tissue engineering.Chen Zhao Wei Jiang Nian Zhou Junyi Liao Mingming Yang Ning Hu Xi Liang Wei Xu Hong Chen Wei Liu Lewis L.Shi Leonardo Oliveira Jennifer Moriatis Wolf Sherwin Ho Aravind Athiviraham H.M.Tsai Tong-Chuan He Wei Huang 2017Genes & Diseases2017,4,4:7
3Adaptive Application Offloading Decision and Transmission Scheduling for Mobile Cloud Computing显示文摘Offloading application to cloud can augment mobile devices' computation capabilities for the emerging resource-hungry mobile application, however it can also consume both much time and energy for mobile device offloading application remotely to cloud. In this paper, we develop a newly adaptive application offloading decision-transmission scheduling scheme which can solve above problem efficiently. Specifically, we first propose an adaptive application offloading model which allows multiple target clouds coexisting. Second, based on Lyapunov optimization theory, a low complexity adaptive offloading decision-transmission scheduling scheme has been proposed. And the performance analysis is also given. 4A genome-wide analysis of SWEET gene family in cotton and their expressions under different stresses显示文摘Background: The SWEET(Sugars will eventually be exported transporters) gene family plays multiple roles in plant physiological activities and development process. It participates in reproductive development and in the process of sugar transport and absorption, plant senescence and stress responses and plant-pathogen interaction. However,thecomprehensive analysis of SWEET genes has not been reported in cotton.Results: In this study, we identified 22, 31, 55 and 60 SWEET genes from the sequenced genomes of Gossypium arboreum, G. raimondii, G. hirsutum and G. barbadense, respectively. Phylogenetic tree analysis showed that the SWEET genes could be divided into four groups, which were further classified into 14 sub-clades. Further analysis of chromosomal location, synteny analysis and gene duplication suggested that the orthologs showed a good collinearity and segmental duplication events played a crucial role in the expansion of the family in cotton. Specific MtN3_slv domains were highly conserved between Arobidopsis and cotton by exon-intron organization and motif analysis. In addition, the expression pattern in different tissues indicated that the duplicated genes in cotton might have acquired new functions as a result of sub-functionalization or neo-funtionalization. The expression pattern of SWEET genes showed that the different genes were induced by diverse stresses. The identification and functional analysis of SWEET genes in cotton may provide more candidate genes for genetic modification.Conclusion: SWEET genes were classified into four clades in cotton. The expression patterns suggested that the duplicated genes might have experienced a functional divergence. 5Expression of ipt gene driven by tomato fruit specific promoter and its effects on fruit development of tomato显示文摘Fruit specific promoter (2A12) from Lycopersi-com esculentum and cDNA of isopentenyl-transferase (ipt) from Ti plasmid of Agrobacterium tumerfaciens C58 were cloned by PCR procedure respectively. Two plant expression vectors with 2A12/gus or 2A12/ipt were respectively constructed. These two chimeric genes were transferred into tomato by Agrobacterium mediated procedure. The results of Southern hybridization showed that the fusion genes had been integrated into tomatoes. The result of gus histochemi-cal staining showed that 2A12 had high fruit specific expressive capability in transgenic tomato. The ipt expression resulted in accumulation of high level of cytokinins (CTKs) in fruit lead to developmental changes in fruits and seeds. The fruit of ipt transformed tomato had the hyperplastic placenta with very few seeds or even seedless. The shelf life of transgenic fruits elongated for 1-2 weeks. 7Structural Asymmetry of Dorsolateral Prefrontal Cortex Correlates with Depressive Symptoms: Evidence from Healthy Individuals and Patients with Major Depressive Disorder显示文摘In this study,we investigated the role of structural asymmetry of the dorsolateral prefrontal cortex(DLPFC) in the continuum of depression from healthy individuals to patients.Structural magnetic resonance imaging was performed in 70 patients with major depressive disorder(MDD),49 matched controls,and 349 healthy university students to calculate structural asymmetry indexes of the DLPFC.First-episode,treatment-naive MDD patients showed a relatively lower asymmetry index than healthy controls,and their asymmetry index was negatively correlated with the depressive symptoms.This abnormality was normalized by antidepressants in medicated MDD patients.Furthermore,the asymmetry index was negatively correlated with the depressive symptoms in university students;this was replicated at two time points in a subgroup of students,suggesting good test–retest reliability.Our findings are consistent with previous studiesthat support the imbalance hypothesis of MDD and suggest a potential structural basis underlying the functional asymmetry of the DLPFC in depression.In future,the structural index of the DLPFC may become a potential biomarker to evaluate individuals' risk for the onset of MDD.Wei Liu Yu Mao Dongtao Wei Junyi Yang Xue Du Peng Xie Jiang Qiu 2016Neuroscience Bulletin2016,32,3:4
8Shear-responsive boundary-lubricated hydrogels attenuate osteoarthritis显示文摘Lipid-based boundary layers formed on liposome-containing hydrogels can facilitate lubrication.However,these boundary layers can be damaged by shear,resulting in decreased lubrication.Here,a shear-responsive boundary-lubricated drug-loaded hydrogel is created by incorporating celecoxib(CLX)-loaded liposomes within dynamic covalent bond-based hyaluronic acid(HA)hydrogels(CLX@Lipo@HA-gel).The dynamic cross-linked network enables the hydrogel to get restructured in response to shear,and the HA matrix allows the accumulation of internal liposome microreservoirs on the sliding surfaces,which results in the formation of boundary layers to provide stable lubrication.Moreover,hydration shells formed surrounding the hydrogel can retard the degradation process,thus helping in sustaining lubrication.Furthermore,in vitro and in vivo experiments found that CLX@Lipo@HA-gels can maintain anabolic-catabolic balance,alleviate cartilage wear,and attenuate osteoarthritis progression by delivering CLX and shear-responsive boundary lubrication.Overall,CLX@Lipo@HA-gels can serve as shear-responsive boundary lubricants and drug-delivery vehicles to alleviate friction-related diseases like osteoarthritis.Yiting Lei Xingkuan Wang Junyi Liao Jieliang Shen Yuling Li Zhengwei Cai Ning Hu Xiaoji Luo Wenguo Cui Wei Huang 2022Bioactive Materials2022,7,10:4
9Prevalence and clinical significance of pathogenic germline BRCA1/2 mutations in Chinese non-small cell lung cancer patients显示文摘Objective: Germline alterations in the breast cancer susceptibility genes type 1 and 2, BRCA1 and BRCA2, predispose individuals to hereditary cancers, including breast, ovarian, prostate, pancreatic, and stomach cancers.Accumulating evidence suggests inherited genetic susceptibility to lung cancer.The present study aimed to survey the prevalence of pathogenic germline BRCA mutations(gBRCAm) and explore the potential association between gBRCAm and disease onset in Chinese advanced non-small cell lung cancer(NSCLC) patients.Methods: A total of 6,220 NSCLC patients were screened using capture-based ultra-deep targeted sequencing to identify patients harboring germline BRCA1/2 mutations.Results: Out of the 6,220 patients screened, 1.03%(64/6,220) of the patients harbored the pathogenic gB RCAm, with BRCA2 mutations being the most predominant mutations(49/64, 76.5%).Patients who developed NSCLC before 50 years of age were more likely to carry gBRCAm(P = 0.036).Among the patients harboring classic lung cancer driver mutations, those with concurrent gBRCAm were significantly younger than those harboring the wild-type gBRCA(P = 0.029).By contrast, the age of patients with or without concurrent gBRCAm was comparable to those of patients without the driver mutations(P = 0.972).In addition, we identified EGFR-mutant patients with concurrent gBRCAm who showed comparable progression-free survival but significantly longer overall survival(P = 0.002) compared to EGFR-mutant patients with wild-type germline BRCA.Conclusions: Overall, our study is the largest survey of the prevalence of pathogenic gBRCAm in advanced Chinese NSCLC patients.Results suggested a lack of association between germline BRCA status and treatment outcome of EGFR-TKI.In addition,results showed a positive correlation between pathogenic gB RCAm and an early onset of NSCLC.Xingsheng Hu Dongyong Yang Yalun Li Li Li Yan Wang Peng Chen Song Xu Xingxiang Pu Wei Zhu Pengbo Deng Junyi Ye Hanhan Zhang Analyn Lizaso Hao Liu Xinru Mao Hai Huang Qian Chu Chengping Hu 2019Cancer Biology & Medicine2019,16,3:4
10Generalized cubature quadrature Kalman filters:derivations and extensions显示文摘A new Gaussian approximation nonlinear filter called generalized cubature quadrature Kalman filter(GCQKF) is introduced for nonlinear dynamic systems. Based on standard GCQKF,two extensions are developed, namely square root generalized cubature quadrature Kalman filter(SR-GCQKF) and iterated generalized cubature quadrature Kalman filter(I-GCQKF). In SR-GCQKF,the QR decomposition is exploited to alter the Cholesky decomposition and both predicted and filtered error covariances have been propagated in square root format to make sure the numerical stability. In I-GCQKF, the measurement update step is executed iteratively to make full use of the latest measurement and a new terminal criterion is adopted to guarantee the increase of likelihood. 13Differential SW16.1 allelic effects and genetic backgrounds contributed to increased seed weight after soybean domestication显示文摘Although seed weight has increased following domestication from wild soybean(Glycine soja) to cultivated soybean(Glycine max), the genetic basis underlying this change is unclear. Using mapping populations derived from chromosome segment substitution lines of wild soybean, we identified SW16.1 as the causative gene underlying a major quantitative trait locus controlling seed weight.SW16.1 encodes a nucleus-localized LIM domaincontaining protein. Importantly, the GsSW16.1 allele from wild soybean accession N24852 had a negative effect on seed weight, whereas the GmSW16.1 allele from cultivar NN1138-2 had a positive effect. Gene expression network analysis,reverse-transcription quantitative polymerase chain reaction, and promoter-luciferase reporter transient expression assays suggested that SW16.1 regulates the transcription of MT4, a positive regulator of seed weight. The natural variations in SW16.1 and other known seed weight genes were analyzed in soybean germplasm. The SW16.1 polymorphism was associated with seed weight in 247 soybean accessions, showing much higher frequency of positive-effect alleles in cultivated soybean than in wild soybean. Interestingly,gene allele matrix analysis of the known seed weight genes revealed that G. max has lost 38.5%of the G. soja alleles and that most of the lost alleles had negative effects on seed weight. 15Seed filling dynamic traits of oil flax in response to nitrogen and phosphorus显示文摘Crop yield is primarily seed-filling-limited in production system under field conditions.This study was aimed to determine whether seed filling traits of oil flax(Linum usitatissimum L.)could be controlled by phosphorus(P),nitrogen(N),and phosphorus and nitrogen(NP)supply.Effects on seed filling traits were investigated in 2 years including capsule diameter,capsule height,capsule dry matter(DM),seed DM per capsule,pericarp DM per capsule,protein content and oil content.DM translocation from pericarps to seed,translocation efficiency,and contribution of photoassimilates during seed filling period were also detected.In a randomized complete block design,cultivar'Longyaza 1'was grown under P(33 kg P/ha),N(75 kg N/ha),and NP(33 kg P/ha and 75 kg N/ha)along with a zerofertilizer(CK)treatment in 2013 and 2014.Results suggested that DM translocation efficiency and contribution efficiency increased to different extent due to P,N or NP application.At 42 DAA(days after anthesis),seed DM per capsule reached the greatest,while protein content and pericarp DM obtained the least level.However,the highest oil content was detected at 35 DAA.A significant positive linear relationship was observed between seed DM,capsule DM and DM translocation in both years.Protein content showed inconsistent relation with oil content.The results indicated that appropriate N and P management could be an effective approach to increase oil flax production.Yaping Xie Bin Wang Limin Wang Junyi Niu Wei Zhao Bin Yan Li Zhao Zhao Dang Wenjuan Li Yanni Qi Jianping Zhang 2019Oil Crop Science2019,4,3:1

18Optimization of the Mars ionospheric radio occultation retrieval显示文摘Electron density is a key parameter to characterize Martian ionospheric structure and dynamics. Based on the ephemeris and auxiliary information derived from the Spacecraft, Planet, Instruments, C-matrix, and Events(SPICE) toolkit, we calculated the bending angle of signal path from the frequency residuals measured by the Mars Express Radio Science Experiment(Ma RS) of the Mars Express(MEX) mission under the assumption of a spherically symmetric ionosphere. We stratified the ionosphere into layers and assumed a linear change of bending angle between layers, to derive profiles in radial distance of refractivity with the optimized parameters of upper integral limit of 4890 km and baseline correction boundary of 3690 km. Meanwhile, we also compared the retrieved electron density profiles between the frequency residuals of the single-frequency and differential Doppler of the dual-frequency. In total, ~640 electron density profiles of Martian ionosphere between April 2004 and April 2015 were retrieved successfully. There are 24 profiles identified manually that exhibit an additional sporadic layer occurrence below the normal two-layers. 19Glycolytic potential enhanced by blockade of pyruvate influx into mitochondria sensitizes prostate cancer to detection and radiotherapy显示文摘Objective:This study aimed to evaluate the effects of mitochondrial pyruvate carrier(MPC)blockade on the sensitivity of detection and radiotherapy of prostate cancer(PCa).Methods:We investigated glycolysis reprogramming and MPC changes in patients with PCa by using metabolic profiling,RNASeq,and tissue microarrays.Transient blockade of pyruvate influx into mitochondria was observed in cellular studies to detect its different effects on prostate carcinoma cells and benign prostate cells.Xenograft mouse models were injected with an MPC inhibitor to evaluate the sensitivity of 18F-fluorodeoxyglucose positron emission tomography with computed tomography and radiotherapy of PCa.Furthermore,the molecular mechanism of this different effect of transient blockage towards benign prostate cells and prostate cancer cells was studied in vitro.Results:MPC was elevated in PCa tissue compared with benign prostate tissue,but decreased during cancer progression.The transient blockade increased PCa cell proliferation while decreasing benign prostate cell proliferation,thus increasing the sensitivity of PCa cells to 18F-PET/CT(SUVavg,P=0.016;SUVmax,P=0.03)and radiotherapy(P<0.01).This differential effect of MPC on PCa and benign prostate cells was dependent on regulation by a VDAC1-MPC-mitochondrial homeostasis-glycolysis pathway.Conclusions:Blockade of pyruvate influx into mitochondria increased glycolysis levels in PCa but not in non-carcinoma prostate tissue.This transient blockage sensitized PCa to both detection and radiotherapy,thus indicating that glycolytic potential is a novel mechanism underlying PCa progression.The change in the mitochondrial pyruvate influx caused by transient MPC blockade provides a critical target for PCa diagnosis and treatment.Huan Xu Junyi Chen Zhi Cao Xi Chen Caihong Huang Jin Ji Yalong Xu Junfeng Jiang Yue Wang Guowang Xu Lina Zhou Jingyi He Xuedong Wei Jason Boyang Wu Zhong Wang Shancheng Ren Fubo Wang 2022Cancer Biology & Medicine2022,19,9:1
20Frequency and Offset Online Estimation of Sinusoidal Signals Based on High-Order Nonlinear Continuous Differentiator显示文摘Real-time parameter identification of sinusoidal signals is an essential research topic due to its broad utilization in both theoretical studies and engineering practice.A tracking differentiator based online estimation framework has been proposed to simultaneously identify frequencies and offset of given multi-sinusoidal signal.Tracking differentiator is exploited in presented framework to track the time derivatives of measurements which are then utilized to estimate the frequencies and offset.We introduce a tracking differentiator called high-order nonlinear continuous differentiator into the framework,giving birth to a new estimation algorithm.Comparative experiments on both single and two sinusoidal signal are simulated,indicating the superiority of proposed method on both convergent speed and estimation accuracy.WANG Hongwei ZHANG Wei ZUO Junyi WANG Heping 2017Chinese Journal of Electronics2017,26,3:1
