1Co-encapsulation of curcumin and doxorubicin in albumin nanoparticles blocks the adaptive treatment tolerance of cancer cells显示文摘The adaptive treatment tolerance (ATT) of cancer cells is the main encumbrance to cancer chemotherapy. A potential solution to this problem is to treat cancer cells with multiple drugs using nanoparticles (NPs). In this study, we tested the co-administration of curcumin (Cur) and doxorubicin (Dox) to MCF-7 resistant breast cancer cells to block the ATTand elicit efficient cell killing. Drugs were co-administered to cells both sequentially and simultaneously. Sequential drug co-administration was carried out by pre-treating the cells with albumin nanoparticles (ANPs) loaded w让h Cur (Cur@ANPs) followed by treatment with Dox-loaded ANPs (Dox@ANPs). Simultaneous drug co-administration was carried out by treating the cells with ANPs loaded with both the drugs (Cur/Dox@ANPs). We found that the simultaneous drug co-administration led to a greater intra-cellular accumulation of Dox and cell killing with respect to the sequential drug co-administration. However;the simultaneous drug co-administration led to a lower intracellular accumulation of Cur with respect to the sequential drug co-administration. We showed that this result was due to the aggregation and entrapment of Cur in the lysosomes as soon as it was released from Cur@ANPs, a phenomenon called lysosomotropism. In contrast, the simultaneous release of Dox and Cur from Cur/Dox@ANPs into the lysosomes led to lysosomal pH elevation, which, in turn, avoided Cur aggregation, led to lysosome swelling and drug release in the cytosol, and finally provoked efficient cell killing. 8Wind and Photovoltaic Power Time Series Data Aggregation Method Based on an Ensemble Clustering and Markov Chain显示文摘Reducing the input wind and photovoltaic power time series data can improve the efficiency of time sequential simulations.In this paper,a wind and photovoltaic power time series data aggregation method based on an ensemble clustering and Markov chain(ECMC)is proposed.The ECMC method can effectively reduce redundant information in the data.First,the wind and photovoltaic power time series data were divided into scenarios,and ensemble clustering was used to cluster the divided scenarios.At the same time,the Davies-Bouldin Index(DBI)is adopted to select the optimal number of clusters.Then,according to the temporal correlation between wind and photovoltaic scenarios,the wind and photovoltaic clustering results are merged and reduced to form a set of combined typical day scenarios that can reflect the characteristics of historical data within the calculation period.Finally,based on the Markov Chain,the state transition probability matrix of various combined typical day scenarios is constructed,and the aggregation state sequence of random length is generated,and then,the combined typical day scenarios of wind and photovoltaic were sampled in a sequential one-way sequence according to the state sequence and then are built into a representative wind and photovoltaic power time series aggregation sequence.A provincial power grid was chosen as an example to compare the multiple evaluation indexes of different aggregation methods.The results show that the ECMC aggregation method improves the accuracy and efficiency of time sequential simulations.Jingxin Jin Lin Ye Jiachen Li Yongning Zhao Peng Lu Weisheng Wang Xuebin Wang 2022CSEE Journal of Power and Energy Systems2022,8,3:1

12Mo-based perpendicularly magnetized thin films with low damping for fast and low-power consumption magnetic memory显示文摘Perpendicular magnetic anisotropy-based magnetic tunnel junctions(p-MTJs) with low Gilbert damping constant(α) are of particular interest for fast and low-power consumption magnetic random-access memory(MRAM). However, obtaining a faster switching speed and lower power consumption is still a big challenge. Herein, we report a Mo-based perpendicular double free layer structure with a low Gilbert damping constant of 0.02 relative to W-based films, as measured by time-resolved magnetooptical Kerr effect equipment. To show the influence of different film structures on the Gilbert damping constant, we measured the Mo-based single free layer. Thereafter, we deposited the full stacks with the Mo-based double free layer and obtained a high tunneling magnetoresistance of 136.3% and high thermal stability. The results of high-resolution transmission electron microscopy(HR-TEM) and energy-dispersive X-ray spectroscopy(EDS) showed that the Mo-based films had better crystallinity,sharper interfaces, and weaker diffusion than the W-based films and thus produced a weaker external contribution of the Gilbert damping constant. As a result of the weak spin-orbit coupling in the Mo-based structure, the intrinsic contribution of the Gilbert damping constant was also weak, thereby leading to the small Gilbert damping constant of the Mo-based stacks. In addition, the macro-spin simulation results demonstrated that the magnetization switching by the spin transfer torque of the Mo-based MTJs was faster than that of the W-based MTJs. 14Osteopontin targeted theranostic nanoprobes for laser-induced synergistic regression of vulnerable atherosclerotic plaques显示文摘Vulnerable atherosclerotic plaque(VASPs)is the major pathological cause of acute cardiovascular event.Early detection and precise intervention of VASP hold great clinical significance,yet remain a major challenge.Photodynamic therapy(PDT)realizes potent ablation efficacy under precise manipulation of laser irradiation.In this study,we constructed theranostic nanoprobes(NPs),which could precisely regress VASPs through a cascade of synergistic events triggered by local irradiation of lasers under the guidance of fluorescence/MR imaging.The NPs were formulated from human serum albumin(HSA)conjugated with a high affinity-peptide targeting osteopontin(OPN)and encapsulated with photosensitizer IR780 and hypoxia-activatable tirapazamine(TPZ).After intravenous injection into atherosclerotic mice,the OPN-targeted NPs demonstrated high specific accumulation in VASPs due to the overexpression of OPN in activated foamy macrophages in the carotid artery.Under the visible guidance of fluorescence and MR dual-model imaging,the precise near-infrared(NIR)laser irradiation generated massive reactive oxygen species(ROS),which resulted in efficient plaque ablation and amplified hypoxia within VASPs.In response to the elevated hypoxia,the initially inactive TPZ was successively boosted to present potent biological suppression of foamy macrophages.After therapeutic administration of the NPs for 2 weeks,the plaque area and the degree of carotid artery stenosis were markedly reduced.Furthermore,the formulated NPs displayed excellent biocompatibility.In conclusion,the developed HSA-based NPs demonstrated appreciable specific identification ability of VASPs and realized precise synergistic regression of atherosclerosis.Mengqi Xu Cong Mao Haoting Chen Lu Liu Yabin Wang Abid Hussain Sulei Li Xu Zhang Ruslan G.Tuguntaev Xing-Jie Liang Weisheng Guo Feng Cao 2022Acta Pharmaceutica Sinica B2022,12,4:0

18Blockchain-based smart contract for smart payment in construction: A focus on the payment freezing and disbursement cycle显示文摘Late payment,and indeed no payment,is a rampant and chronic problem that has plagued the global construction industry for too long.Recent development in blockchain technology,particularly its smart contract,seems to provide a new opportunity to improve this old problem.However,this opportunity is largely unexploited.This study aims to develop a blockchain-based smart contract(BBSC)system for smart payment in the construction industry by focusing on the fundamental cycle of payment freezing(sometimes also synonymously called payment guarantees)and disbursement application.Firstly,a BBSC framework,containing three processes of(a)initiation and configuration,(b)payment freezing,and(c)disbursement application,is developed.Next,based on the framework,the system architecture of the BBSC system,containing three layers of(1)Infrastructure as a Service(IaaS),(2)Blockchain as a Service(BaaS),and(3)Software as a Service(SaaS)is proposed and elaborated.Finally,based on the system architecture,a BBSC prototype system is developed using a real-life modular construction project as a case study.It was found that the prototype system can improve the certainty and efficiency of the progress payment,thereby enabling smart payment in construction transactions.Without advocating radical changes(e.g.,the contractual relationships or the intermediate role of banks in modem construction projects),the prototype can be developed into a real-life BBSC system that can work compatibly with current advancements in the field.Future works are recommended to fine-tune the findings and translate and implement them in real-life applications.Liupengfei WU Weisheng LU Jinying XU 2022Frontiers of Engineering Management2022,9,2:0
