1High matrix metalloproteinase-9 expression induces angiogenesis and basement membrane degradation in stroke-prone spontaneously hypertensive rats after cerebral infarction显示文摘Basement membrane degradation and blood-brain barrier damage appear after cerebral infarction, severely impacting neuronal and brain functioning; however, the underlying pathogenetic mechanisms remain poorly understood. In this study, we induced cerebral infarction in strokeprone spontaneously hypertensive rats by intragastric administration of high-sodium water(1.3% NaCl) for 7 consecutive weeks. Immunohistochemical and immunofluorescence assays demonstrated that, compared with the non-infarcted contralateral hemisphere, stroke-prone spontaneously hypertensive rats on normal sodium intake and Wistar-Kyoto rats, matrix metalloproteinase-9 expression, the number of blood vessels with discontinuous collagen IV expression and microvessel density were significantly higher, and the number of continuous collagen IV-positive blood vessels was lower in the infarct border zones of stroke-prone spontaneously hypertensive rats given high-sodium water. Linear correlation analysis showed matrix metalloproteinase-9 expression was positively correlated with the number of discontinuously collagen IV-labeled blood vessels and microvessel density in cerebral infarcts of stroke-prone spontaneously hypertensive rats. 2Quantitative calculation for the dissipated energy of fault rock burst based on gradient-dependent plasticity显示文摘The capacity of energy absorption by fault bands after rock burst was calculated quantitatively according to shear stressshear deformation curves considering the interactions and interplaying among microstructures due to the heterogeneity of strain softening rock materials. The post-peak stiffness of rock specimens subjected to direct shear was derived strictly based on gradientdependent plasticity, which can not be obtained from the classical elastoplastic theory. Analytical solutions for the dissipated energy of rock burst were proposed whether the slope of the post-peak shear stress-shear deformation curve is positive or not. The analytical solutions show that shear stress level, confining pressure, shear strength, brittleness, strain rate and heterogeneity of rock materials have important influence on the dissipated energy. The larger value of the dissipated energy means that the capacity of energy dissipation in the form of shear bands is superior and a lower magnitude of rock burst is expected under the condition of the same work done by external shear force. 3Numerical study on the fracturing mechanism of shock wave interactions between two adjacent blast holes in deep rock blasting显示文摘With the application of electronic detonators, millisecond blasting is regarded as a signifi cant promising approach to improve the rock fragmentation in deep rock blasting. Thus, it is necessary to investigate the fracturing mechanisms of short-delay blasting. In this work, a rectangle model with two circle boreholes is modeled as a particles assembly based on the discrete element method to simulate the shock wave interactions induced by millisecond blasting. The rectangle model has a size of 12 × 6 m (L × W) and two blast holes have the same diameter of 12 cm. The shock waves are simplifi ed as time-varying forces applied at the particles of walls of the two boreholes. Among a series of numerical tests in this study, the spacing between two adjacent boreholes and delay time of millisecond blasting are considered as two primary variables, and the decoupling charge with a coeffi cient of 1.5 is taken into account in each case. The results show that stress superposition is not a key factor for improving rock fragmentation (tensile stress interactions rather than compressive stress superposition could aff ect the generation of cracks), whereas collision actions from isolated particles or particles with weakened constraints play a crucial role in creating the fracture network. 4Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. 5Mycorrhizal symbiosis modulates the rhizosphere microbiota to promote rhizobia–legume symbiosis显示文摘Plants establish symbioses with mutualistic fungi,such as arbuscular mycorrhizal(AM)fungi,and bacteria,such as rhizobia,to exchange key nutrients and thrive.Plants and symbionts have coevolved and represent vital components of terrestrial ecosystems.Plants employ an ancestral AM signaling pathway to establish intracellular symbioses,including the legume–rhizobia symbiosis,in their roots.Nevertheless,the relationship between the AM and rhizobial symbioses in native soil is poorly understood.Here,we examined how these distinct symbioses affect root-associated bacterial communities in Medicago truncatula by performing quantitative microbiota profiling(QMP)of 16S rRNA genes.We found that M.truncatula mutants that cannot establish AM or rhizobia symbiosis have an altered microbial load(quantitative abundance)in the rhizosphere and roots,and in particular that AM symbiosis is required to assemble a normal quantitative root-associated microbiota in native soil.Moreover,quantitative microbial co-abundance network analyses revealed that AM symbiosis affects Rhizobiales hubs among plant microbiota and benefits the plant holobiont.Through QMP of rhizobial rpoB and AM fungal SSU rRNA genes,we revealed a new layer of interaction whereby AM symbiosis promotes rhizobia accumulation in the rhizosphere of M.truncatula.We further showed that AM symbiosis-conditioned microbial communities within the M.truncatula rhizosphere could promote nodulation in different legume plants in native soil.Given that the AM and rhizobial symbioses are critical for crop growth,our findings might inform strategies to improve agricultural management.Moreover,our work sheds light on the co-evolution of these intracellular symbioses during plant adaptation to native soil conditions.Xiaolin Wang Huan Feng Yayu Wang Mingxing Wang Xingguang Xie Huizhong Chang Like Wang Jicheng Qu Kai Sun Wei He Chunyan Wang Chuanchao Dai Zhaohui Chu Changfu Tian Nan Yu Xuebin Zhang Huan Liu Ertao Wang 2021Molecular Plant2021,14,3:6
6Progress on design and related R&D activities for the water-cooled breeder blanket for CFETR显示文摘The water-cooled ceramic breeder (WCCB) blanket is one of the blanket candidates for Chinese fusion engineering testing reactor (CFETR) and is being developed at the Institute of Plasma Physics, Chinese Academy of Sciences (ASIPP). This paper reviews design and evolution of the WCCB blanket for CFETR, and presents a new WCCB blanket design according to the latest CFETR core parameters (major and minor radii are R = 7.2 m and a = 2.2 m, respectively) and missions. This new design is expected to satisfy multiple CFETR operation modes of 0.2, 0.5, 1.0, and 1.5 GW fusion power and achieve tritium self-sufficiency. The feasibility of the updated blanket design is evaluated from the aspects of neutronics and thermo-hydraulics. 7Comparative genomic and transcriptomic analyses uncover the molecular basis of high nitrogen-use efficiency in the wheat cultivar Kenong 9204显示文摘Studying the regulatory mechanisms that drive nitrogen-use efficiency(NUE)in crops is important for sustainable agriculture and environmental protection.In this study,we generated a high-quality genome assembly for the high-NUE wheat cultivar Kenong 9204 and systematically analyzed genes related to nitrogen uptake and metabolism.By comparative analyses,we found that the high-affinity nitrate transporter gene family had expanded in Triticeae.Further studies showed that subsequent functional differentiation endowed the expanded family members with saline inducibility,providing a genetic basis for improving the adaptability of wheat to nitrogen deficiency in various habitats.To explore the genetic and molecular mechanisms of high NUE,we compared genomic and transcriptomic data from the high-NUE cultivar Kenong 9204(KN9204)and the low-NUE cultivar Jing 411 and quantified their nitrogen accumulation under high-and low-nitrogen conditions.Compared with Jing 411,KN9204 absorbed significantly more nitrogen at the reproductive stage after shooting and accumulated it in the shoots and seeds.Transcriptome data analysis revealed that nitrogen deficiency clearly suppressed the expression of genes related to cell division in the young spike of Jing 411,whereas this suppression of gene expression was much lower in KN9204.In addition,KN9204 maintained relatively high expression of NPF genes for a longer time than Jing 411 during seed maturity.Physiological and transcriptome data revealed that KN9204 was more tolerant of nitrogen deficiency than Jing 411,especially at the reproductive stage.The high NUE of KN9204 is an integrated effect controlled at different levels.Taken together,our data provide new insights into the molecular mechanisms of NUE and important gene resources for improving wheat cultivars with a higher NUE trait.Xiaoli Shi Fa Cui Xinyin Han Yilin He Long Zhao Na Zhang Hao Zhang Haidong Zhu Zhexin Liu Bin Ma Shusong Zheng Wei Zhang Jiajia Liu Xiaoli Fan Yaoqi Si Shuiquan Tian Jianqing Niu Huilan Wu Xuemei Liu Zhuo Chen Deyuan Meng Xiaoyan Wang Liqiang Song Lijing Sun Jie Han Hui Zhao Jun Ji Zhiguo Wang Xiaoyu He Ruilin Li Xuebin Chi Chengzhi Liang Beifang Niu Jun Xiao Junming Li Hong-Qing Ling 2022Molecular Plant2022,15,9:6
8Evaluation of a laying-hen tracking algorithm based on a hybrid support vector machine显示文摘Background: Behavior is an important indicator reflecting the welfare of animals. Manual analysis of video is the most commonly used method to study animal behavior. However, this approach is tedious and depends on a subjective judgment of the analysts. There is an urgent need for automatic identification of individual animals and automatic tracking is a fundamental part of the solution to this problem.Results: In this study, an algorithm based on a Hybrid Support Vector Machine(HSVM) was developed for the automated tracking of individual laying hens in a layer group. More than 500 h of video was conducted with laying hens raised under a floor system by using an experimental platform. The experimental results demonstrated that the HSVM tracker outperformed the Frag(fragment-based tracking method), the TLD(Tracking-Learning-Detection),the PLS(object tracking via partial least squares analysis), the Mean Shift Algorithm, and the Particle Filter Algorithm based on their overlap rate and the average overlap rate.Conclusions: The experimental results indicate that the HSVM tracker achieved better robustness and state-of-theart performance in its ability to track individual laying hens than the other algorithms tested. 9Effects of capture and captivity on plasma corticosterone and metabolite levels in breeding Eurasian Tree Sparrows显示文摘Background: Bringing free-living animals into captivity subjects them to the stress of both capture and captivity, leading to the alteration of normal physiological processes and behaviors through activation of the hypothalamic– pituitary–adrenal axis. In free-living birds, although elevated plasma corticosterone (CORT) is an important adaptation regulating physiological and behavioral responses during the process of capture and captivity stress, little information is currently available on the effects of such stress on plasma metabolite levels. Methods: We examined the effects of immediate capture and 24-h captivity on body mass, body condition, plasma CORT, and metabolite levels including glucose (Glu), triglyceride (TG), total cholesterol (TC), uric acid (UA), in breeding Eurasian Tree Sparrows (Passer montanus). Results: CORT and Glu levels were increased significantly by the stress of capture, whereas TC and UA levels decreased. Body mass, body condition declined notably after 24 h in captivity, but CORT, Glu, and UA levels increased. Furthermore, male sparrows had lower TG levels after both capture and captivity than those of females. The relationships between plasma CORT and metabolite levels varied between sexes. Conclusions: Our results revealed that the metabolic status of Eurasian Tree Sparrows could be dramatically altered by capture and captivity. 12De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. 14The temporal dynamics of antimicrobial-resistant Salmonella enterica and predominant serovars in China显示文摘Salmonella enterica is one of the most common bacterial pathogens in humans and animals.Systematic studies on the trends and geographical distribution of antimicrobial-resistant Salmonella and dominant serovars have been well studied in European and American countries while not in China.Here,taking the One-Health strategy,we used>35000 Salmonella enterica isolates to explore the temporal and spatial dynamics of dominant serovars in China.We found that Salmonella Typhimurium was the dominant serovar causing human infection in China,which was consistent with Australia but inconsistent with North American and European countries.The proportion of Salmonella serovars Typhimurium,London,Rissen,Corvallis,Meleagridis,Kentucky,and Goldcoast showed an increasing trend during 2006-2019.We randomly selected 1962 isolates for comparative genomics and antimicrobial resistance studies and found that the number of antibiotic resistance genes(ARGs)per isolate increased 1.84 and 2.69 times of human and non-human origins,respectively,spanning 14 years.The proportion of antimicrobial-resistant Salmonella isolates had an increasing trend during 2006-2019,especially beta-lactam,quinolone,tetracycline,and rifampicin resistance.Moreover,we found that higher diversity of sequence types(STs)in S.Typhimurium than in other serovars,ST34 from pig and ST 19 from chicken origin,were mainly associated with isolates causing child and adult gastro-infection,respectively.Our results fill in the data gap on the trends of dominant serovars and antimicrobial resistance of Salmonella enterica in China.These data provide useful information for public health decision-makers prioritizing interventions for foodborne diseases and food safety.Yanan Wang Yue Liu Na Lyu Zhiyuan Li Sufang Ma Demin Cao Yuanlong Pan Yongfei Hu Hua Huang George F.Gao Xuebin Xu Baoli Zhu 2023National Science Review2023,10,3:4
15Characteristics and distribution patterns of reef complexes on the carbonate platform margin in deep water areas:the western South China Sea显示文摘As a potential oil and gas reservoir,reef complexes have been a research focus from petroleum geologists for a long time.There are favorable conditions for the development of reef complexes in the South China Sea; however,their internal structures,evolution and distribution are still poorly understood.Based on 2D and 3D seismic data,the internal structures and evolution patterns of the reef complexes on the carbonate platform margin in the deep water areas over the western South China Sea were studied in detail.The result shows that two types of reef complexes,i.e.,fault controlling platform margin reef complexes and ramp reef complexes have been developed in the study area.The reef complexes have independent or continuous mound or lenticular seismic reflections,with three internal structures(i.e.,aggrading,prograding and retrograding structures).There are different growth rates during the evolution of the reef complexes,resulting in the formation of catch-up reefs,keep-up reefs and quick step reefs.The study also reveals that different platform margin reef complexes have different internal structures and distributions,because of the different platform types.These results may be applied to the exploration and prediction of carbonate platform margin reef complexes in other areas that are similar to the study area.CHEN Ping LI Xushen WANG Yahui LU Yongchao ZHONG Zehong CHEN Lei ZUO Qianmei MA Yiquan WANG Chao DU Xuebin 2015Acta Oceanologica Sinica2015,34,10:3
16Adiabatic shear localization evolution for steel based on the Johnson-Cook model and gradient-dependent plasticity显示文摘Gradient-dependent plasticity is introduced into the phenomenological Johnson-Cook model to study the effects of strain-hardening, strain rate sensitivity, thermal-softening, and microstructure. The microstructural effect (interactions and interplay among microstructures) due to heterogeneity of texture plays an important role in the process of development or evolution of an adiabatic shear band with a certain thickness depending on the grain diameter. The distributed plastic shear strain and deformation in the shear band are derived and depend on the critical plastic shear strain corresponding to the peak flow shear stress, the coordinate or position, the internal length parameter, and the average plastic shear strain or the flow shear stress. The critical plastic shear strain, the distributed plastic shear strain, and deformation in the shear band are numerically predicted for a kind of steel deformed at a constant shear strain rate. Beyond the peak shear stress, the local plastic shear strain in the shear band is highly nonuniform and the local plastic shear deformation in the band is highly nonlinear. Shear localization is more apparent with the increase of the average plastic shear strain. 17Comparison of the defluoridation efficiency of calcium phosphate and chitin in the exoskeleton of Antarctic krill显示文摘Calcium(Ca),phosphorus(P),and chitin are the main components of the exoskeleton of krill.Defluoridation of a solution of sodium fluoride(NaF) using calcium phosphate(Ca3(PO4)2) and chitin as defluoridation agents was studied.Orthogonal experiments were designed to find the optimum reaction conditions for defluoridation,to obtain the maximum defluoridation efficiency and fluoride removal capacity of calcium phosphate and chitin.At the same time,a comparison of the capacity of the two defluoridation agents was made.The results suggest that calcium phosphate has a far greater capability than chitin for the removal of fluoride(F) from water under similar reaction conditions.It is also suggested that Antarctic krill is likely to adsorb fluoride via compounds such as calcium phosphate,hydroxyapatite,and other compounds of Ca and P with the general form(Ca,X) x(PO4,HPO4,Y) y(OH,Z) z,in addition to chitin.WANG Zhangmin YIN Xuebin 2012Advances in Polar Science2012,23,3:3
18Transforming growth factor β1 is a differentially expressed candidate protein of congestive heart failure with Qi-deficiency-blood-stasis syndrome显示文摘OBJECTIVE: To investigate the role of tongue coating fluid protein in regulation of congestive heart failure(CHF) in Qi-deficiency-blood-stasis syndrome.METHODS: We studied patients with CHF(3 patients with Qi-deficiency-blood-stasis syndrome and 3 without Qi-deficiency-blood-stasis syndrome) to investigate differentially expressed proteins. We also included a control group. A biotin label-based antibody array was used for testing tongue coating fluid samples from patients. Net-work analysis of these differentially expressed proteins was conducted using the STRING database,which can predict the relations between differentially expressed proteins and CHF with Qi-deficiency-blood-stasis syndrome.RESULTS: A total of seven differentially expressed proteins were identified, and among these, transforming growth factor β1(TGF-β1) gets a particular attention for us has drawn specific attention.Network analysis showed a homologous relationship of TGF-β1 with bone morphogenetic protein15, which is associated with myocardial fibrosis.CONCLUSION: Occurrence and development of CHF may result from certain DE-proteins and associated signaling pathways. 19Characteristics and Driving Forces of Spatial Expansion of Oasis Cities and Towns in Hexi Corridor, Gansu Province, China显示文摘This paper presents an integrated study of urban spatial expansion in the Hexi Corridor, Gansu Province, China based on TM, ETM remote sensing data in 1987, 1990, 1995, 2000, 2006 and 2011. The study explores the characteristics of urban spatial expansion and dynamic mechanism by using expansion speed index, expansion intensity index, compact index, fractal dimension, and extended flexibility index. We built the index system of influencing factors of urban spatial expansion by using the grey incidence model. The results showed that urban spatial expansion rate in the Hexi Corridor has been on the upward trend since 1987. Expansion intensity showed an obvious upward trend, however, the upward trend varied in different urban areas. In addition, the urban structure was loose relatively, but the urban compactness was more obvious. The urban spatial form tended to be simple, and the urban land use tended to become more intensive. Urban spatial expansion experienced several stages: padding internally, external expansion and padding internally.
