1Roux-en-Y gastric bypass promotes expression of PDX-1 and regeneration of β-cells in Goto-Kakizaki rats显示文摘AIM:To study the effects of Roux-en-Y gastric bypass(RYGB) on the expression of pancreatic duodenal homeobox-1(PDX-1) and pancreatic β-cell regeneration/neogenesis,and their possible mechanisms in diabetics.METHODS:Three groups of randomly selected nonobese diabetic Goto-Kakizaki(GK) rats were subjected to RYGB,sham-RYGB and sham-operation(sham-op) surgery,respectively.The rats were euthanized at postoperative 1,2,4 and 12 wk.Their pancreases were resected and analyzed using reverse transcription polymerase chain reaction to detect the mRNA of PDX-1.Anti-PDX-1 immunohistochemical(IHC) staining and Western blotting were used to detect the protein of PDX-1.Double IHC staining of anti-Brdu and-insulin was performed to detect regenerated β-cells.The index of double Brdu and insulin positive cells was calculated.RESULTS:In comparison with sham-RYGB and sham-op groups,a significant increase in the expressions of PDX-1 mRNA in RYGB group was observed at all experimental time points(1 wk:0.378 ± 0.013 vs 0.120 ± 0.010,0.100 ± 0.010,F = 727.717,P < 0.001;2 wk:0.318 ± 0.013 vs 0.110 ± 0.010,0.143 ± 0.015,F = 301.509,P < 0.001;4 wk:0.172 ± 0.011 vs 0.107 ± 0.012,0.090 ± 0.010,F = 64.297,P < 0.001;12 wk:0.140 ± 0.007 vs 0.120 ± 0.010,0.097 ± 0.015,F = 16.392,P < 0.001);PDX-1 protein in RYGB group was also increased significantly(1 wk:0.61 ± 0.01 vs 0.21 ± 0.01,0.15 ± 0.01,F = 3031.127,P < 0.001;2 wk:0.55 ± 0.00 vs 0.15 ± 0.01,0.17 ± 0.01,F = 3426.455,P < 0.001;4 wk:0.39 ± 0.01 vs 0.18 ± 0.01,0.22 ± 0.01,F = 882.909,P < 0.001;12 wk:0.41 ± 0.01 vs 0.20 ± 0.01,0.18 ± 0.01,F = 515.833,P < 0.001).PDX-1 mRNA and PDX-1 protein production showed no statistical significance between the two sham groups.Many PDX-1 positive cells could be found in the pancreatic islets of the rats in RYGB group at all time points.In addition,the percentage of Brdu-insulin double staining positive cells was higher in RYGB group than in the other two groups(1 wk:0.22 ± 0.13 vs 0.03 ± 0.06,0.03 ± 0.06,P < 0.05;2 wk:0.28 ± 0.08 vs 0.00 ± 0.00,0.03 ± 0.06,P < 0.05;4 wk:0.24 ± 0.11 vs 0.07 ± 0.06,0.00 ± 0.00,P < 0.001;12 wk:0.20 ± 0.07 vs 0.03 ± 0.06,0.00 ± 0.00,P < 0.05).CONCLUSION:RYGB can increase the expression of pancreatic PDX-1 and induce the regeneration of β-cells in GK rats.The associated regeneration of islet cells may be a possible mechanism that how RYGB could improve type 2 diabetes mellitus.Zhen Li,Dong-Fei Li,Jing-Xing Dai,Yu Bai,Lin Yuan,Southern Medical University,Institute of Basic Medical Anatomy National Key Disciplines,Guangzhou 510515,Guangdong Province,China Hong-Ya Zhang,Lu-Xian Lv,Wen-Qiang Li,Second Affiliated Hospital,Xinxiang Medical College,Henan Province Key Laboratory of Biological Psychiatry,Xinxiang 453002,Henan Province,China Ou Sha,Department of Anatomy,Faculty of Medicine,Shenzhen University,Shenzhen 518060,Guangdong Province,China 2010World Journal of Gastroenterology2010,16,18:14

3Effects of two polybrominated diphenyl ethers(BDE-47,BDE-209) on the swimming behavior, population growth and reproduction of the rotifer Brachionus plicatilis显示文摘Polybrominated diphenyl ethers(PBDEs) are new kinds of persistent organic pollutants(POPs) and their potential threats to the equilibrium and sustainability of marine ecosystems have raised worldwide concerns. Here, two kinds of PBDEs, tetra-BDE(BDE-47)and deca-BDE(BDE-209) were applied, and their toxic effects on the swimming behavior,population growth and reproduction of Brachionus plicatilis were investigated. The results showed that:(1) The actual concentrations of BDE-47 and-209 in the seawater phase measured by GC–MS(Gas Chromatography-Mass Spectrometer) were much lower than their nominal concentrations.(2) In accordance with the 24-hr acute tests, BDE-209 did not show any obvious swimming inhibition to rotifers, but a good correlation did exist between the swimming inhibition rate and BDE-47 concentration suggesting that BDE-47 is more toxic than BDE-209.(3) Both BDE-47 and-209 had a significant influence on the population growth and reproduction parameters of B. plicatilis including the population growth rate, the ratio of ovigerous females/non-ovigerous females(OF/NOF), the ratio of mictic females/amictic females(MF/AF), resting egg production and the mictic rate, which indicate that these parameters in B. plicatilis population were suitable for monitoring and assessing PBDEs. Our results suggest that BDE-47 and-209 are not acute lethal toxicants and may pose a low risk to marine rotifers at environmental concentrations for short-term exposure. They also accumulate differently into rotifers. 5Effects of feed form and feed particle size on growth performance,carcass characteristics and digestive tract development of broilers显示文摘This study was conducted to evaluate the effects of feed form(mash and crumble-pellet) and feed particle size(fine, medium and coarse) on growth performance, carcass characteristics and digestive tract development of broilers. A total of 1,152 one day-old Ross 308 mixed-sex broilers were used in a factorial arrangement(2×3) based on a completely randomized design with six replicates of 32 birds each.Higher average daily gain(ADG) and average daily feed intake(ADFI) were observed(P < 0.01) for birds fed the crumble-pellet diets(CPD) than for those fed the mash diets(MD) during starter, grower and the entire experiment period. From d 1 to 40, birds fed CPD had a higher(P < 0.01) body weight(BW) than those fed MD. Birds fed CPD had a lower(P < 0.01) feed:gain ratio(F:G) during the starter phase than those fed MD. Medium or coarse particle size increased(P < 0.01) ADG and ADFI during the starter phase, but birds fed fine particle size diets had lower(P < 0.01) F:G during the grower phase. In MD,medium and coarse particle sizes resulted in higher(P < 0.05) BW, ADG and ADFI than fine particle size during the whole experiment. In CPD, particle size had no significant effect on growth performance, as indicated by a feed form × particle size interaction(P < 0.05). At 41 days of age, ten birds per treatment were randomly selected and killed for slaughter yields and digestive tract characteristics determination.It was shown that particle size and feed form alone had no significant effect on slaughter yields, so changes was the feed form × particle size interaction. The relative empty weight of the gizzard was greater(P < 0.01) and the relative length of the ileum was longer(P < 0.05) in birds fed MD than in those fed CPD. 6Manganese salts function as potent adjuvants显示文摘Aluminum-containing adjuvants have been used for nearly 100 years to enhance immune responses in billions of doses of vaccines.To date,only a few adjuvants have been approved for use in humans,among which aluminum-containing adjuvants are the only ones widely used.However,the medical need for potent and safe adjuvants is currently continuously increasing,especially those triggering cellular immune responses for cytotoxic T lymphocyte activation,which are urgently needed for the development of efficient virus and cancer vaccines.Manganese is an essential micronutrient required for diverse biological activities,but its functions in immunity remain undefined.We previously reported that Mn^(2+) is important in the host defense against cytosolic dsDNA by facilitating cGAS-STING activation and that Mn^(2+)alone directly activates cGAS independent of dsDNA,leading to an unconventional catalytic synthesis of 2′3′-cGAMP.Herein,we found that Mn^(2+) strongly promoted immune responses by facilitating antigen uptake,presentation,and germinal center formation via both cGAS-STING and NLRP3 activation.Accordingly,a colloidal manganese salt(Mn jelly,MnJ)was formulated to act not only as an immune potentiator but also as a delivery system to stimulate humoral and cellular immune responses,inducing antibody production and CD4^(+)/CD8^(+)T-cell proliferation and activation by either intramuscular or intranasal immunization.When administered intranasally,MnJ also worked as a mucosal adjuvant,inducing high levels of secretory IgA.MnJ showed good adjuvant effects for all tested antigens,including T cell-dependent and T cell-independent antigens,such as bacterial capsular polysaccharides,thus indicating that it is a promising adjuvant candidate.Rui Zhang Chenguang Wang Yukun Guan Xiaoming Wei Mengyin Sha Mengran Yi Miao Jing Mengze Lv Wen Guo Jing Xu Yi Wan Xin-Ming Jia Zhengfan Jiang 2021Cellular & Molecular Immunology2021,18,5:7

8Long-term adenosine A1 receptor activation-induced sortilin expression promotes α-synuclein upregulation in dopaminergic neurons显示文摘Prolonged activation of adenosine A1 receptor likely leads to damage of dopaminergic neurons and subsequent development of neurodegenerative diseases.However,the pathogenesis underlying long-term adenosine A1 receptor activation-induced neurodegeneration remains unclear.In this study,rats were intraperitoneally injected with 5 mg/kg of the adenosine A1 receptor agonist N6-cyclopentyladenosine(CPA)for five weeks.The mobility of rats was evaluated by forced swimming test,while their cognitive capabilities were evaluated by Y-maze test.Expression of sortilin,α-synuclein,p-JUN,and c-JUN proteins in the substantia nigra were detected by western blot analysis.In addition,immunofluorescence staining of sortilin andα-synuclein was performed to detect expression in the substantia nigra.The results showed that,compared with adenosine A1 receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine(5 mg/kg)+CPA co-treated rats,motor and memory abilities were reduced,surface expression of sortin andα-synuclein in dopaminergic neurons was reduced,and total sortilin and totalα-synuclein were increased in CPA-treated rats.MN9D cells were incubated with 500 nM CPA alone or in combination with 10μM SP600125(JNK inhibitor)for 48 hours.Quantitative real-time polymerase chain reaction analysis of sortilin andα-synuclein mRNA levels in MN9D cells revealed upregulated sortilin expression in MN9D cells cultured with CPA alone,but the combination of CPA and SP600125 could inhibit this expression.Predictions made using Jasper,PROMO,and Alibaba online databases identified a highly conserved sequence in the sortilin promoter that was predicted to bind JUN in both humans and rodents.A luciferase reporter assay of sortilin promoter plasmid-transfected HEK293T cells confirmed this prediction.After sortilin expression was inhibited by sh-SORT1,expression of p-JUN and c-JUN was detected by western blot analysis.Long-term adenosine A1 receptor activation levels upregulatedα-synuclein expression at the post-transcriptional level by affecting sortilin expression.The online tool Raptor-X-Binding and Discovery Studio 4.5 prediction software predicted that sortilin can bind toα-synuclein.Co-immunoprecipitation revealed an interaction between sortilin andα-synuclein in MN9D cells.Our findings indicate that suppression of prolonged adenosine A1 receptor activation potently inhibited sortilin expression andα-synuclein accumulation,and dramatically improved host cognition and kineticism.This study was approved by the University Committee of Animal Care and Supply at the University of Saskatchewan(approval No.AUP#20070090)in March 2007 and the Animals Ethics Committee of University of South China(approval No.LL0387-USC)in June 2017.Yun-Cheng Lv An-Bo Gao Jing Yang Li-Yuan Zhong Bo Jia Shu-Hui Ouyang Le Gui Tian-Hong Peng Sha Sun Francisco S.Cayabyab 2020Neural Regeneration Research2020,15,4:4

10Sortilin promotes macrophage cholesterol accumulation and aortic atherosclerosis through lysosomal degradation of ATP-binding cassette transporter A1 protein显示文摘Sortilin is closely associated with hyperlipidemia and the risk of atherosclerosis(AS).The role of sortilin and the underlying mechanism in peripheral macrophage are not fully understood.In this study,we investigated the effect of macrophage sortilin on ATP-binding cassette transporter A1(ABCA1)expression,ABCA1-mediated cholesterol efflux,and aortic AS.Macrophage sortilin expression was upregulated by oxidized low-density lipoproteins(ox-LDLs)in both concentration-and time-dependent manners.Its expression reached the peak level when cells were incubated with 50μg/ml ox-LDL for 24 h.Overexpression of sortilin in macrophage reduced cholesterol efflux,leading to an increase in intracellular total cholesterol,free cholesterol,and cholesterol ester.Sortilin was found to bind with ABCA1 protein and suppress macrophage ABCA1 expression,resulting in a decrease in cholesterol efflux from macrophages.The inhibitory effect of sortilin in cholesterol efflux was partially reversed by treatment with chloroquine,a lysosomal inhibitor.On the contrary,the ABCA1 protein level and ABCA1-mediated cholesterol efflux is increased by sortilin short hairpin RNA transfection.The fecal and biliary cholesterol 3H-sterol from cholesterol-laden mouse peritoneal macrophage was reduced by sortilin overexpression through lentivirus vector(LV)-sortilin in low-density lipoprotein receptor knockout mice,which was prevented by co-treatment with chloroquine.Treatment with LV-sortilin reduced plasma high-density lipoprotein and increased plasma ox-LDL levels.Accordingly,aortic lipid deposition and plaque area were exacerbated,and ABCA1 expression was reduced in mice in response to infection with LV-sortilin alone.These effects of LV-sortilin were partially reversed by chloroquine.Sortilin enhances lysosomal degradation of ABCA1 protein and suppresses ABCA1-mediated cholesterol efflux from macrophages,leading to foam cell formation and AS development.Yuncheng Lv Jing Yang Anbo Gao Sha Sun Xilong Zheng Xi Chen Wei Wan Chaoke Tang Wei Xie Suyun Li Dongming Guo Tianhong Peng Guojun Zhao Liyuan Zhong 2019Acta Biochimica et Biophysica Sinica2019,51,5:4
11The potential value of microRNA-4463 in the prognosis evaluation in hepatocellular carcinoma显示文摘The purpose of this study is to measure the expression of microRNA-4463 and microRNA-6087 between normal persons and patients with hepatocellular carcinoma(HCC),and to clarify the meaning of them in the prognosis evaluation in HCC.Forty-five samples from healthy people and patients,who had been diagnosed with hepatocellular carcinoma before any treatment,were collected to study respectively.Real-time PCR was used to detect the expression of miRNA-4463 and miRNA-6087 in the serum of control group and hepatocellular carcinoma patients.The expression of miR-4463 in the serum of HCC patients was significantly higher than that in control group(P<0.05),and the expression level was independent of gender,tumor size,cell types,stages,alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL)and HBsAg status(P>0.05).But there was a significant difference of different level of AFP in HCC(P<0.05),and the difference between the group of AFP lower than 400 ug/l and the control group is statistically significant(P<0.05).Besides,the survival time had showed a significant difference at the high and low expression levels(P<0.05).But the expression level of miRNA-6087 was no difference in HCC and control group.The disorder of miRNA-4463 occurred in HCC,even the AFP level doesn’t rises.What’s more,patients who get the high level of miRNA-4463 seem to have a shorter survival time.And it contributes great to the prognostic evaluation.This is the first study to illustrate the potential significance of miRNA-4463 in the prognosis in HCC.Tian Hu Jincheng Li Chuhong Zhang Xiu lv Sai Li Sha He Hanxing Yan Yixi Tan Mingsheng Lei Meiling Wen Jianhong Zuo 2017Genes & Diseases2017,4,2:3

14Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART.Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu 2021Cell Research2021,31,8:2
15In-situ emb e dding zeolitic imidazolate framework derived Co–N–C bifunctional catalysts in carbon nanotube networks for flexible Zn–air batteries显示文摘Recently, the development of high-performance bifunctional oxygen catalysts integrated with flexible conductive scaffolds f or rechargeable metal-air batteries has attracted considerable interest, driving by fastgrowing wearable electronics. Herein, we report a flexible bifunctional oxygen catalyst thin film consisting of Co–N–C bifunctional catalysts embedding in carbon nanotube(CNT) networks. The catalyst is readily prepared by pyrolysis of cobalt-based zeolitic imidazolate frameworks(ZIF-67) that are in-situ synthesized in CNT networks. Such catalyst film demonstrates very high catalytic activities for oxygen reduction(onset potential: 0.91 V, and half-wave potential: 0.87 V vs. RHE) and oxygen evolution(10 m Acm^-2 at 1.58 V) reactions, high methanol tolerance property, and long-term stability(97% current retention). Moreover, our integrated catalyst film shows very good structure flexibility and robustness. Based on the obtained film air electrodes, flexible Zn–air batteries demonstrate low charging and discharging overpotentials(0.82 V at 1 m A cm^-1) and excellent structure stability in the bending tests. 18Concomitant extrahepatic autoimmune diseases do not compromise the long-term outcomes of primary biliary cholangitis显示文摘Background:Primary biliary cholangitis(PBC)patients often have concomitant extrahepatic autoimmune(EHA)diseases including Sjögren’s syndrome(SS),systemic sclerosis(SSc),rheumatoid arthritis(RA),and autoimmune thyroid disease.The present study aimed to describe the prevalence of EHA diseases in PBC and explore the impact of EHA diseases on the long-term outcomes of PBC in Chinese patients.Methods:Medical records of PBC patients diagnosed in our institute were retrospectively reviewed.Pa-tients were followed up by a standardized telephone interview.The endpoints were defined as liver-related death and/or liver transplantation.Results:Totally 247 of the 985(25.1%)PBC patients enrolled in the study had at least one concomi-tant EHA disease.Sjögren’s syndrome(n=140,14.2%)was the most frequent one,followed by rheuma-toid arthritis(RA)(n=56,5.7%)and Hashimoto’s thyroiditis(n=45,4.6%).Patients with EHA dis-eases were more common in females(P<0.001)and in those with a family history of autoimmune disease(P=0.017).Overall,no differences were found between PBC patients with and without EHA dis-eases in terms of biochemical response rates to ursodeoxycholic acid,the incidence of hepatic events,or transplant-free survival.RA and EHA≥2 were protective factors for hepatic events in univariate Cox analysis,but the results became insignificant in multivariate analysis.Conclusions:Concomitant EHA diseases were common in PBC patients but did not compromise the long-term outcomes of PBC.Sha Chen Meng-Qi Li Wei-Jia Duan Bu-Er Li Shu-Xiang Li Ting-Ting Lv Lin Ma Ji-Dong Jia 2022Hepatobiliary & Pancreatic Diseases International2022,21,6:1
