6Data-driven engineering framework with AI algorithm of Ginkgo Folium tablets manufacturing显示文摘Smart manufacturing still remains critical challenges for pharmaceutical manufacturing.Here,an original data-driven engineering framework was proposed to tackle the challenges.Firstly,from sporadic indicators to five kinds of systematic quality characteristics,nearly 2,000,000 real-world data points were successively characterized from Ginkgo Folium tablet manufacturing.Then,from simplex to the multivariate system,the digital process capability diagnosis strategy was proposed by multivariate C_(pk)integrated Bootstrap-t.The C_(pk)of Ginkgo Folium extracts,granules,and tablets were discovered,which was 0.59,0.42,and 0.78,respectively,indicating a relatively weak process capability,especially in granulating.Furthermore,the quality traceability was discovered from unit to end-to-end analysis,which decreased from 2.17 to 1.73.This further proved that attention should be paid to granulating to improve the quality characteristic.In conclusion,this paper provided a data-driven engineering strategy empowering industrial innovation to face the challenge of smart pharmaceutical manufacturing.Lijuan Ma Jing Zhang Ling Lin Tuanjie Wang Chaofu Ma Xiaomeng Wang Mingshuang Li Yanjiang Qiao Yongxiang Wang Guimin Zhang Zhisheng Wu 2023Acta Pharmaceutica Sinica B2023,13,5:1
7A study on the characteristics of heavy metals in orange ecosystem显示文摘An investigation was conducted to identify the characteristics of heavy metals in orange ecosystem in three areas in the Three Gorge Reservoir Region by using the modified Tessier sequential extraction procedure.The contents of Cu and Pb in different speciation fractions of the soils can be ordered as the Residual(F6)>Reducible(F4)>Oxidable(F5)>Water soluble(F1)>Carbonate(F3)>Exchangeable(F2),and the contents of Zn and Ni follow the order of the Residual(F6)>Oxidable(F5)>Reducible(F4)>Water soluble(F1)>Carbonate(F3)> Exchangeable(F2),and those of Cd follow the order of the Reducible(F4)>Oxidable(F5)>Exchangeable(F2)> Carbonate(F3)>Residual(F6)>Water soluble(F1).The concentrations of Cu in the different organs of oranges follow the increasing sequence of pulp>leaf>pericarp,those of Zn and Ni following the increasing sequence of leaf >pulp>pericarp,and those of Pb and Cd follow the increasing sequence of leaf>pericarp>pulp.Correlation degrees of different forms of same heavy metals in soils are according to the order of Cu>Zn>Cd>Pb>Ni.Correlation degrees of different heavy metals in same forms are according to the order that the forms of Reducible(F4),Oxidable(F5) and Residual(F6) are higher than the forms of Water soluble(F1),Exchangeable(F2) and Carbonate(F3).There are either positive or negative correlations between heavy metals in leaves and in pulp,while there are all positive correlations between heavy mentals in leaves and in pericarp.There are significant correlations between the concentrations of some heavy metals in leaves,pericarp or pulp,and those in speciation fractions in the soils.LI Qilin WEI Chaofu HUANG Yun WANG Liwei WANG Dingyong 2010Chinese Journal Of Geochemistry2010,29,1:1

9Presence of multiple abnormal immunologic markers is an independent prognostic factor of diffuse large B-cell lymphoma显示文摘Autoimmune diseases (ADs) increase the risk of non-Hodgkin's lymphoma and contribute to poor prognosis of patients. However, the association between immunologic markers and clinical outcome has rarely been investigated. This study aims to analyze the prognostic value of pretreatment immunologic markers in newly diagnosed patients with diffuse large B-cell lymphoma (DLBCL). We retrospectively reviewed the data on 502 patients with DLBCL treated in our institution from January 2013 to March 2018. Survival functions were estimated using Kaplan Meier method and Cox regression model. The 3-year progression free survival (PFS) and overall survival (OS) rates were 70.2% and 80.9%, respectively, and the complete remission (CR) rate was 78.1%. Among the patients, those with multiple (≥3) abnormal immunologic markers had significantly shorter 3-year PFS (52.7% vs. 77.3%, P < 0.001) and OS (68.5% vs. 85.8%, P=0.001) than those without multiple abnormal immunologic markers.Multivariate analysis revealed that the presence of multiple abnormal immunologic markers and the elevated serum levels of lactate dehydrogenase were the independent adverse prognostic factors for PFS (P= 0.008, P < 0.001) and OS (P = 0.003, P < 0.001). Meanwhile, advanced Ann Arbor stage was an independent adverse prognostic factor for PFS (P= 0.001) and age > 60 years for OS (P= 0.014). 10Prediction of mechanical ventilation outcome by early abdominal-visceral-blood-flow-and-function score in critically ill patients after cardiopulmonary bypass in the ICU: A prospective observational study显示文摘Background:Abdominal organs are important organs that sense and respond to ischemia and hypoxia,but there are few evaluation methods.We use ultrasonography to evaluate abdominal organ function and blood flow in patients with mechanical ventilation(MV)after cardiopulmonary bypass and to obtain a semiquantitative score for abdominal organ function and blood flow.Methods:Patients with cardiopulmonary bypass in the Critical Care Department of Peking Union Medical College Hospital in China from March to July 2021 were enrolled in this prospective observational study.The correlation of the abdominal-visceral-blood-flow-and-function score(AVBFS)with the duration of MV,number of days spent in the intensive care unit(ICU),acute physiology and chronic health evaluation II(APACHE-II),sequential organ failure assessment(SOFA),lactate,epinephrine,and norepinephrine use was analyzed,and the results were used to assess the predictive value of the receiver operating characteristic curve(ROC)regression analysis score for the duration of MV.Results:Of the 92 patients who underwent cardiopulmonary bypass,41 were finally included.The AVBFS were significantly correlated with the duration of MV,number of days spent in the ICU,APACHE-II score,SOFA score,and norepinephrine use time.The AVBFS in a group of patients using ventilators≥36 h were significantly higher than those obtained for a group of patients using ventilators<36 h(P<0.05).The evaluation results for the AVBFS at 0-12 h after ICU admission were as follows:area under the ROC curve(AUC)=0.876(95%confidence interval[CI]:0.767 to 0.984),cut-off value=2.5,specificity=0.842,and sensitivity=0.773.Conclusions:Abdominal visceral organ function and blood perfusion can be used to evaluate gastrointestinal function.It is related to early and late extubation after cardiac surgery.Chaofu Yue Longxiang Su Jun Wang Na Cui Yuankai Zhou Wei Cheng Bo Tang Xi Rui Huaiwu He Yun Long 2024Journal of Intensive Medicine2024,4,1:0

12TET (Ten-eleven translocation) family proteins: structure, biological functions and applications显示文摘Ten-eleven translocation(TET)family proteins(TETs),specifically,TET1,TET2 and TET3,can modify DNA by oxidizing 5-methylcytosine(5mC)iteratively to yield 5-hydroxymethylcytosine(5hmC),5-formylcytosine(5fC),and 5-carboxycytosine(5caC),and then two of these intermediates(5fC and 5caC)can be excised and return to unmethylated cytosines by thymine-DNA glycosylase(TDG)-mediated base excision repair.Because DNA methylation and demethylation play an important role in numerous biological processes,including zygote formation,embryogenesis,spatial learning and immune homeostasis,the regulation of TETs functions is complicated,and dysregulation of their functions is implicated in many diseases such as myeloid malignancies.In addition,recent studies have demonstrated that TET2 is able to catalyze the hydroxymethylation of RNA to perform post-transcriptional regulation.Notably,catalytic-independent functions of TETs in certain biological contexts have been identified,further highlighting their multifunctional roles.Interestingly,by reactivating the expression of selected target genes,accumulated evidences support the potential therapeutic use of TETs-based DNA methylation editing tools in disorders associated with epigenetic silencing.In this review,we summarize recent key findings in TETs functions,activity regulators at various levels,technological advances in the detection of 5hmC,the main TETs oxidative product,and TETs emerging applications in epigenetic editing.Furthermore,we discuss existing challenges and future directions in this field.Xinchao Zhang Yue Zhang Chaofu Wang Xu Wang 2023Signal Transduction and Targeted Therapy2023,8,9:0
