1The Performance of Whole Genome Amplification Methods and Next-Generation Sequencing for Pre-Implantation Genetic Diagnosis of Chromosomal Abnormalities显示文摘Reliable and accurate pre-implantation genetic diagnosis(PGD) of patient's embryos by next-generation sequencing(NGS) is dependent on efficient whole genome amplification(WGA) of a representative biopsy sample. However, the performance of the current state of the art WGA methods has not been evaluated for sequencing. Using low template DNA(15 pg) and single cells, we showed that the two PCR-based WGA systems Sure Plex and MALBAC are superior to the REPLI-g WGA multiple displacement amplification(MDA) system in terms of consistent and reproducible genome coverage and sequence bias across the 24 chromosomes, allowing better normalization of test to reference sequencing data. When copy number variation sequencing(CNV-Seq) was applied to single cell WGA products derived by either Sure Plex or MALBAC amplification, we showed that known disease CNVs in the range of 3e15 Mb could be reliably and accurately detected at the correct genomic positions. 2Viscous lithospheric structure beneath Sumatra inferred from post-seismic gravity changes detected by GRACE显示文摘Satellite gravity data of the regional rheological structure of the lithosphere in the vicinity of Sumatra is used as evidence to enable a better understanding of the regional geodynamic environment.The data is interpreted using the theory of post-seismic viscoelastic relaxation.Coand post-seismic changes in the gravity field resulting from the 2004 M w 9.3 Sumatra earthquake were calculated from Gravity Recovery and Climate Experiment (GRACE) satellite data.A spatial Gaussian filter,500 km wide,was used in the calculation.The results indicate that there were significant co-seismic jumps in both uplifted and subducted regions.The magnitude of the jump in the subducted zone was ~9 10 8 m/s 2,more significant than the ~2 10 8 m/s 2 jump observed in the uplifted zone.However,a positive gravity change occurred in the uplifted zone very soon after the earthquake.The rheological structure of the lithosphere has a great effect on deformation and its determination is a fundamental part of developing reliable numerical simulations in geodynamics.Based on the temporally-variable gravity field observed by GRACE,the viscous lithospheric structure of the Sumatra area is investigated with a self-gravitating,half space,viscoelastic earth model.The estimated viscosity is of the order of 1.0 10 18 Pa·s and there are differences in the rheological parameters on the two sides of the fault.The factors that affect the viscosity are discussed in connection with the tectonic structure of the Sumatra area.WANG WuXing SHI YaoLin SUN WenKe ZHANG Jing 2011Science China Earth Sciences2011,54,8:9

5Effects of Gaussian filter in processing GRACE data: Gravity rate of change at Lhasa,southern Tibet显示文摘In this paper, the spatial gravity distribution over Tibetan Plateau and the gravity rate of change at Lhasa for different Gaussian filter radii are computed using GRACE data. Results show that the estimate of the gravity rate of change is spatial radius-dependent of the Gaussian filter. The GRACE-estimated gravity rate of change agrees well with the surface measured one. In other words, the GRACE-estimated gravity rate of change has a limited value as that obtained by surface measurement when the spatial filter radius reaches zero. Then numerical simulations are made for different spatial radii of the Gaussian filter to investigate its behaviors when applied to surface signals. Results show that the estimate of a physical signal is filter-radius dependent. If the computing area is equal to or less than the mass area, especially for a uniformly distributed mass, the estimate gives an almost correct result, no matter what filter radius is used. The estimate has large error because of the signal leakage caused by harmonic truncation if the computing area is much bigger than the mass distribution (or inverse for a small mass anomaly). If a mass anomaly is too small, it is difficult to recover it from space observation unless the filter radius is extremely small. If the computing point (or area) is outside the mass distribution, the estimated result is almost zero, particularly for small filter radii. These properties of the Gaussian filter are helpful in applying GRACE data in different geophysical problems with different spatial position and geometrical size. We further discuss physical sources causing the scalar gravity change at Lhasa. Discussions indicate that the gravity rate of change at Lhasa is not caused by the present-day ice melting (PDIM) (or Little Ice Age, LIA) effect because no ice melting occurs in Lhasa city and nearby. The gravity rate of change is attributable mainly to tectonic deformation associated with the Indian Plate collision. 7Weakened anisotropy of mechanical properties in rolled ZK60 magnesium alloy sheets with elevated deformation temperature显示文摘The rolling direction(RD) and the transverse direction(TD) samples were obtained from an as-rolled ZK60 magnesium alloy sheet with strong anisotropy of initial texture and their mechanical properties were tested at various deformation temperatures. Meanwhile, the microstructure and texture of these samples after fracture were investigated. Results revealed that a higher flow stress along the RD than that along the TD at room temperature were ascribed to the strong anisotropy of transitional texture, and this texture effect was remarkably weakened with the increase of deformation temperature. Deformation structure was dominant at 100?C, and was replaced by dynamic recrystallization structure when the deformation temperature increased to 200?C and 300?C. The texture presented a strong texture(transitional texture in the RD sample and basal texture in the TD sample) at 100?C, but its intensity visibly decreased and texture components became more disperse at 200?C and 300?C. 8Cardiac fibroblast heat shock protein 47 aggravates cardiac fibrosis post myocardial ischemia-reperfusion injury by encouraging ubiquitin specific peptidase 10 dependent Smad4 deubiquitination显示文摘Despite complications were significantly reduced due to the popularity of percutaneous coronary intervention(PCI) in clinical trials, reperfusion injury and chronic cardiac remodeling significantly contribute to poor prognosis and rehabilitation in AMI patients. We revealed the effects of HSP47 on myocardial ischemia-reperfusion injury(IRI) and shed light on the underlying molecular mechanism.We generated adult mice with lentivirus-mediated or miRNA(mi1/133TS)-aided cardiac fibroblastselective HSP47 overexpression. Myocardial IRI was induced by 45-min occlusion of the left anterior descending(LAD) artery followed by 24 h reperfusion in mice, while ischemia-mediated cardiac remodeling was induced by four weeks of reperfusion. Also, the role of HSP47 in fibrogenesis was evaluated in cardiac fibroblasts following hypoxia-reoxygenation(HR). Extensive HSP47 was observed in murine infarcted hearts, human ischemic hearts, and cardiac fibroblasts and accelerated oxidative stress and apoptosis after myocardial IRI. Cardiac fibroblast-selective HSP47 overexpression exacerbated cardiac dysfunction caused by chronic myocardial IRI and presented deteriorative fibrosis and cell proliferation.HSP47 upregulation in cardiac fibroblasts promoted TGFβ1-Smad4 pathway activation and Smad4 deubiquitination by recruiting ubiquitin-specific peptidase 10(USP10) in fibroblasts. However, cardiac fibroblast specific USP10 deficiency abolished HSP47-mediated fibrogenesis in hearts. Moreover, blockage of HSP47 with Col003 disturbed fibrogenesis in fibroblasts following HR. 10Characterization on the formation of porosity and tensile properties prediction in die casting Mg alloys显示文摘The 3D visualization of the porosity in high-pressure die casting(HPDC)Mg alloys AZ91D and Mg4Ce2Al0.5Mn(EA42)was investigated by X-ray computed tomography.It was demonstrated that the volumetric porosity at the near-gate location for alloy EA42 was significantly higher than that far from the gate location.This difference resulted from the low valid time during intensified casting pressure conditions.Specimens of alloy EA42 exhibited a narrow pore distribution in the side view(~0.5 mm)compared to the wide distribution(~1.8 mm)of alloy AZ91D,which was mainly attributed to the formation mechanism of the defect band.The formation of microporosity in the defect band of alloy EA42 was inhibited because of the significant latent heat released by a large amount of the Al11Ce3phase segregated in the defect band during solidification.Additionally,an effective estimator(Z-Propagation)was introduced,which is proposed to predict the projected area fraction of the porosity(f)involved during tensile failure with better effectiveness compared with traditional methods based on the actual fractured surface.By coupling the Z-Propagation method with the critical local strain model,the logarithmic fracture strain and true fracture stress of the specimens were predicted within 3.03%and 1.65%of the absolute value of the average relative error(AARE),respectively.Zixin Li Dejiang Li Wenke Zhou Bo Hu Xingfeng Zhao Jingya Wang Ming Qin Jiangkun Xu Xiaoqin Zeng 2022Journal of Magnesium and Alloys2022,10,7:2
11Variability of streambed vertical hydraulic conductivity with depth along the Elkhorn River,Nebraska,USA显示文摘Determination of streambed hydraulic conductivity is of great importance in the analysis of stream-aquifer interactions and stream ecosystems.In this paper,in situ falling-head standpipe permeameter tests were conducted to determine streambed vertical hydraulic conductivity (Kv) of two connected layers of sediments at 60 test locations from eight sites in the Elkhorn River,Nebraska.Our results show that the Kv values for the upper layer of sediments are generally larger than those in their respective lower layer of sediments.The individual Kv value for the upper layer of sediments from all test locations of the eight sites range from 2.7 to 104.9 m/d,and the average value is 26.6 m/d.The individual Kv value ranges from 0.4 to 73.4 m/d and the average value is 16.1 m/d for the lower layer of sediments.We believe that hyporheic processes can enhance larger streambed Kv in the upper layer of sediments.Inflow and outflow through hyporheic zone produce more porous and permeable sediments and further result in an increasing of streambed Kv.Moreover,we observe that bioturbation activities can destroy the clogging layer of the exposed sediments and create pore spaces in the sediments,which also further increases streambed Kv.SONG JinXi CHEN XunHong CHENG Cheng WANG DeMing WANG WenKe 2010Chinese Science Bulletin2010,55,10:2
12Spatial distribution characteristics and mechanism of nonhydrological time-variable gravity in China's Mainland显示文摘The purpose of this study is to explore nonhydrological mass transfer in China's Mainland.For this purpose,gravity recovery and climate experiment(GRACE)data were obtained to study the spatial distribution of time variant gravity signals in China's Mainland.Then,from auxiliary hydrological data processed according to the current hydrological model,a new more comprehensive hydrological model of China's Mainland was constructed.Finally,the time variant signals of this new hydrological model were removed from the time variant gravity field computed from GRACE data,thus obtaining a description of the nonhydrological mass transfer of China's Mainland.The physical sources and mechanisms of the resulting mass transfer are then discussed.The improved,more realistic,hydrological model used here was created by selecting the hydrological components with the best correlations in existing hydrological models,by use of correlation calculation,analysis,and comparison.This improved model includes water in soils and deeper strata,in the vegetation canopy,in lakes,snow,and glaciers,and in other water components(mainly reservoir storage,swamps,and rivers).The spatial distribution of the transfer signals due to nonhydrological mass in China's Mainland was obtained by subtracting the combined hydrological model from the GRACE time-variable gravity field.The results show that the nonhydrological signals in China's Mainland collected in GRACE data were mainly positive signals,and were distributed in the Bohai Rim and the northern and eastern parts of the Tibetan Plateau.The above nonhydrological mass transfer signals have been studied further and are discussed.The results show that the nonhydrological mass migration signals in the Bohai Rim region originate primarily from sea level change and marine sediment accumulation.The mass accumulation from Indian plate collision in the Tibetan Plateau appears to be the main reason for the increase in the residual gravity field in that region.Yue Shen QiuYu Wang WeiLong Rao WenKe Sun 2022Earth and Planetary Physics2022,6,1:2
