1Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients.Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin 2021Cellular & Molecular Immunology2021,18,2:8
2Mitochondria-targeted antioxidant peptide SS-31 mediates neuroprotection in a rat experimental glaucoma model显示文摘To investigate the neuroprotective effects of the mitochondria-targeted antioxidant Szeto-Schiller peptide 31 (SS-31) in a rat experimental glaucoma model, SS-31 was intraperitoneally (IP) injected into Sprague-Dawley rats, followed by intracameral injection of polystyrene microspheres to induce elevated intraocular pressure (IOP). After 6 weeks, electroretinography (ERG) and flash visual-evoked potentials (F-VEPs) were recorded to assess retinal function. Hematoxylin-eosin staining was performed on retinal cross-sections to measure ganglion cell complex (GCC) thickness. Apoptotic retinal cells were assessed by TUNEL staining. Brn3a-positive retinal ganglion cells (RGCs) were counted in retinal flat mounts via immunofluorescence. The retinal total SOD, SOD2, and MDA expression levels were assessed in retinal tissue homogenates. The cyt c, Baxz and Bcl-2 protein levels in rat retinas were detected by western blot analysis. Bax and Bcl-2 expressions were also evaluated using immunohistochemistry in paraffinized sections. Our results showed that the rats that received microsphere injection developed elevated IOP. SS-31 ameliorated the reductions in the a- and b-wave amplitudes on ERG and the F-VEP amplitude in glaucomatous eyes. GCC thickness was preserved, TUNEL-positive cells were decreased in the retina, and Brn3a-positive RGCs were in creased in the SS-31-treated glaucoma group compared with those in the non-treated glaucoma group. SS-31 significantly reduced MDA levels and increased SOD2 levels after glaucoma induction. Significant suppression of cyt c release, upregulation of Bcl-2, and downregulation of Bax were observed following SS-31 administration. 5Simulation of Heat Transfer Characteristics in Channel with Square Rib Under Uniform and Turbulent Inlet Conditions显示文摘The detailed flow structures and closely-related heat transfer characteristics are investigated along the wall of a cooling channel with rib tabulator by computation.Three typical Reynolds numbers defined by the rib height are set at 200,500,1300,and the Mach numbers is 0.2,respectively.Two inlet boundary conditions,including the uniform and the fully-developed turbulent conditions,are used to study the turbulence effects on the characteristics of heat transfer in the vicinity of rib and wall.Results show that the local Nusselt number increases when the Reynolds number rises from 200 to 1300.At lower Reynolds number,the turbulent inlet condition generates more tangible heat transfer enhancement.At higher Reynolds number,however,the uniform inlet condition contributes more to the convective heat transfer effects.The paper discovers that the high Nusselt number has a consistent correlation with the positive and negative sign alteration of the shear layer on the wall,which satisfactorily explains the mechanisms of heat transfer enhancement due to the flow.LI Heng YU Ting WANG Duo ZU Yingqing XU Hongyi 2019Transactions of Nanjing University of Aeronautics and Astronautics2019,36,5:1

7Sphingolipid synthesis inhibitor fumonisin B1 causes verticillium wilt in cotton显示文摘Verticillium wilt caused by Verticillium dahliae is a major disease of cotton. Acidic protein–lipopolysaccharide complexes are thought to be the toxins responsible for its symptoms. Here,we determined that the sphingolipid biosynthesis inhibitor fumonisin B1(FB1) acts as a toxin and phenocopies the symptoms induced by V. dahliae. Knocking out genes required for FB1 biosynthesis reduced V. dahliae pathogenicity. Moreover, we showed that overexpression of a FB1 and V. dahliae both downregulated gene, Gh IQD10, enhanced verticillium wilt resistance by promoting the expression of brassinosteroid and anti-pathogen genes. 10Shadow geometry maps for alias-free shadows显示文摘Shadow maps sample scene visibility in the light source space and offer an efficient solution to generate hard shadows.However,they suffer from aliasing artifacts because of discretization errors,inadequate resolution and projection distortion.In this paper,we propose the shadow geometry map method,where a shadow depth map is augmented by storing geometry information about scenes.This leads to a new shadowrendering algorithm that combines a supersampling filter,a geometry-aware reconstruction kernel and an irregular sampling filter.Our method produces high quality alias-free and subpixel supersampling shadow rendering and retains the simplicity and the efficiency of shadow maps.We show that the algorithm pipeline is efficiently parallelized using current programmable graphics hardware and that our method is capable of generating high quality hard shadows.WANG Rui WU YingQing PAN MingHao CHEN Wei HUA Wei 2013Science China(Information Sciences)2013,56,6:1
11Compounding effects of human activities and climatic changes on coexistence of oasis-desert ecosystems:Prioritizing resilient decision-making for a riskier world显示文摘Water-salt balance is critical for the stable coexistence of salt-affected and groundwater-fed oasis-desert ecosystems. Yet, a comprehensive investigation of how soil salinization and groundwater degradation threaten the coexistence of oasis-desert ecosystems is still scarce, especially under the compounding effects of human activities and climatic changes. Here, we assessed the impacts of irrigated agriculture on hydrological regimes in oasisdesert systems, investigated the spatio-temporal variations of soil salinization in irrigated cropland, and evaluated the implications of the interplays of soil salinization and groundwater degradation on the coexistence of oasis-desert ecosystems in northwestern China, based on meaningful modelling approaches and comprehensive measurements over 1995–2020. The results showed that the irrigation return flow coefficient decreased sharply from 0.21 ± 0.09 in the traditional irrigation period to 0.09 ± 0.01 in the water-saving irrigation period. The continuous drop in groundwater tables and significant degradation of groundwater quality are occurring throughout this watershed. The eco-environmental flows are reaching to their limit with watershed closures(i.e.,the drainage from the oasis region into the desert region is being weakened or even eliminated), although these progressions were largely hidden by regional precipitation and streamflow variability. The process of salt migration and accumulation across different landscapes in oasis-desert system is being reshaped, and soil salinization in water-saving agricultural irrigated lands is accelerating with a regional average annual growth rate of18%. The vegetation in this watershed is degrading, and anthropogenic disturbance accelerates this trend. Our results highlight that environmental stress adaptation strategies must account for resilience maintenance to avoid accelerating catastrophic transitions in oasis-desert ecosystems.
