1Effect of subsoil tillage depth on nutrient accumulation, root distribution, and grain yield in spring maize显示文摘A four-year field experiment was conducted to investigate the effect of subsoiling depth on root morphology, nitrogen(N), phosphorus(P), and potassium(K) uptake, and grain yield of spring maize. The results indicated that subsoil tillage promoted root development,increased nutrient accumulation, and increased yield. Compared with conventional soil management(CK), root length, root surface area, and root dry weight at 0–80 cm soil depth under subsoil tillage to 30 cm(T1) and subsoil tillage to 50 cm(T2) were significantly increased, especially the proportions of roots in deeper soil. Root length, surface area, and dry weight differed significantly among three treatments in the order of T2 > T1 > CK at the12-leaf and early filling stages. The range of variation of root diameter in different soil layers in T2 treatment was the smallest, suggesting that roots were more likely to grow downwards with deeper subsoil tillage in soil. The accumulation of N, P, and K in subsoil tillage treatment was significantly increased, but the proportions of kernel and straw were different. In a comparison of T1 with T2, the grain accumulated more N and P, while K accumulation in kernel and straw varied in different years. Grain yield and biomass were increased by 12.8% and 14.6% on average in subsoil tillage treatments compared to conventional soil treatment. 2Transcriptional regulation of the Herpes Simplex Virus 1α-gene by the viral immediate-early protein ICP22 in association with VP16显示文摘Herpes Simplex Virus 1 (HSV1) is capable of inducing two forms of infection in individuals, and the establishment of which type of infection occurs is linked to the transcriptional activation of viral α genes. One of the HSV1 α genes, ICP22, is known to have multiple functions during virus replication, but its distinct roles are still unclear. This study showed that ICP22 functions as a general repressor for certain viral and cellular promoters, and this transcriptional repression by ICP22 is independent of the specific upstream promoter element, as shown using the CAT enzyme assay system. Further work also found that VP16 interfered with ICP22 mediated transcriptional repression of the viral α4 gene, through interactions with specific elements upstream of the α4 gene promoter. 4Analysis of HSV-I ICP22 effects on HCMV major immediate-early promoter structure显示文摘The human cytomegalovirus (HCMV) major immediate-early (MIE) promoter has strong transcriptional promoting capability. Its cis-acting regulatory elements form a special structure in this region that is repeated multiple times; the biological significance of these elements and their different compositions in the transcriptional promoting process remain unclear. Our results demonstrate that the HSV-I MIE protein ICP22 can generate strong repression of many viral and cellular promoters and enhancers. We further studied the transcriptional effects of ICP22 on structural elements and mutations in various HCMV MIE promoters by using a CAT assay. In spite of different transcriptional effects of all the ele- ments in the presence of ICP22, the transcriptional efficiencies exhibited by mutations generated by different compositions and an entire HCMV promoter, are not the simple sum of the functions of these elements. Furthermore, the transcriptional activities of specific sequences were not affected by the presence of ICP22. 5Immediate-early gene product ICP22 inhibits the trans-transcription activating function of P53-mdm-2显示文摘As a product of HSVI immediate-early gene, ICP22 is capable of interacting with various cellular tran-scriptive and regulatory molecules during viral infection so as to impact the normal cellular molecular mechanism. ICP22 expressed in transfected cells can push the cells’ entering into S phase with binding to mdm-1 promoter region and impact its trans-transcription activating effect by P53. 10Body Size but Not Food Size Determined Head Sexual Dimorphism in Rana kukunoris from the Tibetan Plateau显示文摘Sexual dimorphism between males and females is a common pattern observed in the natural world and has become one of the main focuses of evolutionary biology.Amphibian head size and shape are important factors that may influence food acquisition,but knowledge about the sexual differences between amphibian head size and shape is relatively scant.Here,intersexual differences in the head size and shape of Rana kukunoris,which is a high-elevation frog from the Qinghai-Tibet Plateau,were investigated.Significant differences were found in the head shape but not the head size between sexes.A significant correlation was also observed between prey size and head size.However,these correlations disappeared when individual body size was controlled,suggesting that body size but not food size was the main factor resulting in intersexual differences in head size/shape.The results suggested that food competition between sexes did not result in sexual dimorphism of head shape among these frogs.Hongzhou CHEN Haifen QIN Zhenkun ZHAO Jiahong LIAO Yanfang WU Xueyan LIU Lichun JIANG Buddhi DAYANANDA Wei CHEN 2023Asian Herpetological Research2023,14,2:1
11High Altitude Frogs (Rana kukonoris) Adopt a Diversified Bethedging Strategy in the Face of Environmental Unpredictability显示文摘Environmental unpredictability can influence strategies of maternal investment among eggs within a clutch. Models predict that breeding females should adopt a diversified bet-hedging strategy in unpredictable environments, but empirical field evidence from Asia is scarce. Here we tested this hypothesis by exploring spatial patterns in egg size along an altitudinal gradient in a frog species(Rana kukunoris) inhabiting the Tibetan Plateau. Within-clutch variability in egg size increased as the environment became variable(e.g., lower mean monthly temperature and mean monthly rainfall at higher altitudes), and populations in environments with more unpredictable rainfall produced eggs that were smaller and more variable in size. We provide support for a diversified bet-hedging strategy in high-altitude environments, which experience dynamic weather patterns and therefore are of unpredictable environmental quality. This strategy may be an adaptive response to lower environmental quality and higher unpredictable environmental variance. Such a strategy should increase the likelihood of breeding success and maximize maternal lifetime fitness by producing offspring that are adapted to current environmental conditions. We speculate that in high-altitude environments prone to physical disturbance, breeding females are unable to consistently produce the optimal egg size due to physiological constraints imposed by environmental conditions(e.g., duration of the active season, food availability). 14Differential-equation Filtering in Seismic Data Processing显示文摘Noise attenuation is necessary before and after stacking in the seismic data processing. We describe the filtering method based on differential equations, which transforms the filtering equations in the frequency domain or in the f-k domain back into the time or time-space domains, and use the finite-difference algorithm to solve the equations. The method does not require the seismic data being stationary; and the filtering parameters can vary at any temporal and spatial point. 19STIM1 expression is associated with osteosarcoma cell survival显示文摘Objective: To examine the role of store-operated calcium entry(SOCE) and stromal interaction molecule 1(STIM1) in survival and migration of osteosarcoma cells and investigate what blockade of store-operated Ca^(2+)contributes to the regulation of osteosarcoma cells.Methods: First, we examined the expression levels of STIM1 in osteosarcoma cell lines by Western analysis and in tissue specimens by immunohistochemistry. Second, we investigated the effect of SOCE and STIM1 on osteosarcoma cell viability using MTS assays and on cell proliferation using colony formation. Third, we investigated the role of SOCE and STIM1 in cell migration using wound healing assays and Boyden chamber assays. Finally, we studied the effect of SOCE on the nuclear factor of activated T-cells cytoplasmic 1(NFATc1)activity by luciferase assays.Results: STIM1 was overexpressed in osteosarcoma cell lines and tissue specimens and was associated with poor survival of osteosarcoma patients. Also, inhibition of SOCE and STIM1 decreased the cell viability and migration of osteosarcoma cells. 20Identification and characterization of multipotential stem cells in immortalized normal ovarian surface epithelial cells显示文摘The ovarian surface epithelium(OsE)is a single layer of squamous-to-cuboidal epithelial cells that experience repetitive ovulatory rupture and subsequent repair.However,the characteristics of human immortalized ovarian surface epithelial cells(IOSE80)remain elusive.This study aims to determine whether IOSE80 cells have the characteristics of stem cell proliferation and multilineage differentiation and their application in regenerative medicine.IOsE80 cells are sequenced by high-throughput transcriptome analysis,and 5 sets of public data are used to compare the differences between IosE8o cells and bone marrow mesenchymal stem cells,pluripotent stem cells,and oocytes in transcriptome profiling.The IOsE80 cells present a cobblestone-like monolayer and express the epithelial cell marker KRT18;the stem cell markers IFITM3,ALDH1A1,and VIM;lowly express stem cell marker LGR5 and germ cell markers DDX4 and DAZL.In addition,the GO terms'regulation of stem cell proliferation','epithelial cell proliferation',etc.,are significantly enriched(P<0.05).IOSE80 cells have the potential to act as mesenchymal stem cells to differentiate into adipocytes with lipid droplets,osteoblasts,and chondroblasts in vitro.IOSE80 cells express pluripotent stem cell markers,including OCT4,SSEA4,TRA-1-60,and TRA-1-81,and they can be induced into three germ layers in vitro.IOSE80 cells also form oocyte-like cells in vitro and in vivo.In addition,IOSE80 cells exhibit robust proliferation,migration,and ovarian repair functions after in vivo transplantation.This study de-monstrates that losE8o cells have the characteristics of pluripotent/multipotent stem cells,indicating their im-portant role in tissue engineering and regenerative medicine.Lin Hou Hanqing Hong Wenjiao Cao Liutong Wei Lichun Weng Shuang Yuan Chengqi Xiao Qiuwan Zhang Qian Wang Dongmei Lai 2024Acta Biochimica et Biophysica Sinica2024,56,2:0
