| 15 | Preparation and Pharmacodynamic Evaluation of Kang Zhu Gan Lu Oral Proliposome, a New Hypoglycemic Tibetan Medicine显示文摘[Objectives]To effectively solve the problem of poor oral bioavailability of total flavonoids in Kang Zhu Gan Lu(ZYZH),a new hypoglycemic Tibetan solid preparation,ZYZH oral proliposome was prepared by modern preparation method,and its hypoglycemic effect was evaluated.[Methods]ZYZH proliposome was prepared by carrier deposition method,and its particle morphology and Zeta potential were detected.Taking encapsulation efficacy as indictor,the optimal prescription and preparation process of ZYZH oral proliposome were screened by orthogonal experiment.In addition,the hypoglycemic activity of ZYZH oral liposome in alloxan-induced hyperglycemic mice was studied.[Results]The repose angle,average particle size,encapsulation efficiency,and Zeta potential of ZYZH proliposome are 34.2°,(442.5±3.5)nm,67.7%and(-47.2±3.4)mV,respectively.Compared with the model group,the fasting blood glucose level of the hyperglycemic mice significantly declined(P<0.01),and their body weight changed insignificantly after four weeks of oral administration of ZYZH proliposome(P>0.05).[Conclusions]The preparation process of ZYZH oral proliposome is characterized by simple operation,high encapsulation efficiency and good stability.The new hypoglycemic Tibetan drug ZYZH oral liposome has obvious hypoglycemic effect on alloxan-induced hyperglycemic mice. | Weini DAI Yu MAO Lingmin LIN Xinran MI | 2019 | Medicinal Plant2019,10,5: | 0 |
| 16 | Circadian clock control of MRTF/SRF pathway suppresses beige adipocyte thermogenic recruitment显示文摘The morphological transformation of adipogenic progenitors into mature adipocytes requires dissolution of actin cytoskeleton with loss of myocardin-related transcription factor(MRTF)/serum response factor(SRF)activity.Circadian clock confers temporal control in adipogenic differentiation,while the actin cytoskeleton–MRTF/SRF signaling transduces extracellular physical niche cues.Here,we define a novel circadian transcriptional control involved in actin cytoskeleton–MRTF/SRF signaling cascade that modulates beige fat thermogenic function.Key components of actin dynamic–MRTF/SRF pathway display circadian regulation in beige fat depot.The core clock regulator,brain and muscle arnt-like 1(Bmal1),exerts direct transcriptional control of genes within the actin dynamic–MRTF/SRF cascade that impacts actin cytoskeleton organization and SRF activity.Employing beige fat-selective gene-targeting models together with pharmacological rescues,we further demonstrate that Bmal1 inhibits beige adipogenesis and thermogenic capacity in vivo via the MRTF/SRF pathway.Selective ablation of Bmal1 induces beigeing with improved glucose homeostasis,whereas its targeted overexpression attenuates thermogenic induction resulting in obesity.Collectively,our findings identify the clock–MRTF/SRF regulatory axis as an inhibitory mechanism of beige fat thermogenic recruitment with significant contribution to systemic metabolic homeostasis. | Xuekai Xiong Weini Li Ruya Liu Pradip Saha Vijay Yechoor Ke Ma | 2022 | Journal of Molecular Cell Biology2022,14,12: | 0 |
