The research on method of interlayer modeling based on seismic inversion and petrophysical facies显示文摘Currently, the three-dimensional distribution of interlayer is realized by stochastic modeling.Traditionally, the three-dimensional geological modeling controlled by sedimentary facies models is built on the basis of logging interpretation parameters and geophysical information.Because of shallow gas-cap,the quality of three-dimensional seismic data vertical resolution in research area cannot meet the interlayer research that is below ten meters. Moreover, sedimentary facies cannot commendably reveal interlayer distribution and the well density is very sparse in research area. So,it is difficult for conventional technology to finely describe interlayers. In this document,it uses L1-L2 combined norm constrained inversion to enhance the recognition capability of interlayer in seismic profile and improve the signal to noise ratio, the wave group characteristics and the vertical resolution of three-dimensional data and classifies petrophysical facies of interlayer based on core, sedimentary facies and logging interpretation. The interlayer model which is based on seismic inversion model and petrophysical facies can precisely simulate the distribution of reservoir and interlayer. The results show that the simulation results of this new methodology are consistent with the dynamic production perfectly which provide a better basis for producing and mining remaining oil and a new interlayer modeling method for sparse well density. | Chao Cheng Wengang Yu Xiaojun Bai | 2016 | Petroleum2016,2,1: | 3
Genetically modified CD7-targeting allogeneic CAR-T cell therapy with enhanced efficacy for relapsed/refractory CD7-positive hematological malignancies:a phase I clinical study显示文摘Chimeric antigen receptor(CAR)-T cell therapy against T cell malignancies faces major challenges including fratricide between CAR-T cells and product contamination from the blasts.Allogeneic CAR-T cells, generated from healthy donor T cells, can provide ready-to-use,blast-free therapeutic products, but their application could be complicated by graft-versus-host disease(GvHD)and host rejection. Here we developed healthy donor-derived,CD7-targeting CAR-T cells(RD13-01)with genetic modifications to resist fratricide,GvHD and allogeneic rejection,as well as to potentiate antitumor function.A phase I clinical trial(NCT04538599)was conducted with twelve patients recruited(eleven with T cell leukemia/lymphoma,and one with CD7-expressing acute myeloid leukemia). All patients achieved pre-set end points and eleven proceeded to efficacy evaluation. No dose-limiting toxicity,GvHD,immune effector cell-associated neurotoxicity or severe cytokine release syndrome(grade≥3)were observed.28 days post infusion,81.8%of patients(9/11)showed objective responses and the complete response rate was 63.6%(7/11,including the patient with AML).3 of the responding patients were bridged to allogeneic hematopoietic stem cell transplantation. With a median follow-up of 10.5 months,4 patients remained in complete remission.Cytomegalovirus(CMV)and/or Epstein-Barr virus(EBV)reactivation was observed in several patients, and one died from EBV-associated diffuse large B-cell lymphoma(DLBCL).Expansion of CD7-negative normal T cells was detected post infusion. In summary,we present the first report of a Phase I clinical trial using healthy donor-derived CD7-targeting allogeneic CAR-T cells to treat CD7^(+)hematological malignancies. Our results demonstrated the encouraging safety and efficacy profiles of the RD13-01 allogeneic CAR-T cells for CD7^(+)tumors. | Yongxian Hu Yali Zhou Mingming Zhang Houli Zhao Guoqing Wei Wengang Ge Qu Cui Qitian Mu Gong Chen Lu Han Tingting Guo Jiazhen Cui Xiaoyan Jiang Xiujun Zheng Shuhui Yu Xiaolong Li Xingwang Zhang Mingxi Chen Xiuju Li Ming Gao Kang Wang Cheng Zu Hao Zhang Xiaohong He Yanbin Wang Dongrui Wang Jiangtao Ren He Huang | 2022 | Cell Research2022,32,11: | 2
Targeting histone deacetylases for cancer therapy: Trends and challenges显示文摘Dysregulation of histone deacetylases(HDACs) is closely related to tumor development and progression. As promising anticancer targets, HDACs have gained a great deal of research interests and two decades of effort has led to the approval of five HDAC inhibitors(HDACis). However, currently traditional HDACis, although effective in approved indications, exhibit severe off-target toxicities and low sensitivities against solid tumors, which have urged the development of next-generation of HDACi. This review investigates the biological functions of HDACs, the roles of HDACs in oncogenesis, the structural features of different HDAC isoforms, isoform-selective inhibitors, combination therapies, multitarget agents and HDAC PROTACs. We hope these data could inspire readers with new ideas to develop novel HDACi with good isoform selectivity, efficient anticancer effect, attenuated adverse effect and reduced drug resistance. | Tao Liang Fengli Wang Reham M.Elhassan Yongmei Cheng Xiaolei Tang Wengang Chen Hao Fang Xuben Hou | 2023 | Acta Pharmaceutica Sinica B2023,13,6: | 1

Probabilistic stability analysis of Bazimen landslide with monitored rainfall data and water level fluctuations in Three Gorges Reservoir, China显示文摘Landslide is a common geological hazard in reservoir areas and may cause great damage to local residents’life and property. It is widely accepted that rainfall and periodic variation of water level are the two main factors triggering reservoir landslides. In this study, the Bazimen landslide located in the Three Gorges Reservoir(TGR)was back-analyzed as a case study.Based on the statistical features of the last 3-year monitored data and field instrumentations, the landslide susceptibility in an annual cycle and four representative periods was investigated via the deterministic and probabilistic analysis, respectively. The results indicate that the fluctuation of the reservoir water level plays a pivotal role in inducing slope failures, for the minimum stability coefficient occurs at the rapid decline period of water level. The probabilistic analysis results reveal that the initial sliding surface is the most important area influencing the occurrence of landslide, compared with other parts in the landslide. The seepage calculations from probabilistic analysis imply that rainfall is a relatively inferior factor affecting slope stability. This study aims to provide preliminary guidance on risk management and early warning in the TGR area. | Wengang ZHANG Libin TANG Hongrui LI Lin WANG Longfei CHENG Tingqiang ZHOU Xiang CHEN | 2020 | Frontiers of Structural and Civil Engineering2020,14,5: | 1

Control Effect of UAV Low Volume Spray Technology on Thrips and Tomato Spotted Wilt Virus in Tobacco Field显示文摘[Objectives]The paper was to explore the influence of unmanned aerial vehicle(UAV)low volume spray technology on the control effect of viruliferous thrips and tomato spotted wilt virus(TSWV)under different pesticide concentrations, and to explore the effective control nodes and methods of thrips and TSWV.[Methods]According to the occurrence characteristics of thrips and TSWV,the field control effects of 4 pesticides on thrips and TSWV were tested by UAV with the thrips number and disease index as indicators, following the principle of regional and periodic control trials.[Results]In the groups of UAV and artificial prevention and control,4 agents were used alternatively, including 1%emamectin benzoate ME,1.7%abamectin imidacloprid ME,5%emamectin benzoate WDG,and 30%pyriproxyfen dinotefuran SL.There was no significant difference in the control effect of thrips and TSWV among treatment 1(recommended dosage of UAV agent),treatment 2(reduction of recommended dosage of UAV agent by 25%)and treatment 4(recommended dosage of artificial control agent),and the control effects on thrips were more than 83.16%in the 3 treatments. The disease index of TSWV in the 3 treatments decreased from 8.64±1.37 in the blank control group to less than 3,which reached the prevention and control threshold. But treatment 3(reduction of recommended dosage of UAV agent by 50%)did not reach the prevention and control threshold. The disease index of TSWV in the blank control area increased with the increase of the number of live nymphs of thrips, and there was a significant positive correlation, with good fitness.[Conclusions]UAV has a significant impact on the control effect of thrips and TSWV under different pesticide concentrations. In the actual flight control application,it is recommended that the amount of the pesticides is about 3/4 and not less than 1/2 of that of the artificial application. The control area should be extended to 100 m of the ridge of the tobacco field, and UAV is used periodically to control thrips and TSWV for 5 times from pre-transplanting stage to pre-squaring stage of flue-cured tobacco. The control effect is significantly different from the traditional artificial application. | Zefan LIU Dean XIE Jiabao HAN Yanfen XIE Li MA Siyuan GAO Ruiqi PENG Yuting ZHAO Xiangze FU Wengang CHENG Yonghong FENG Yan ZHANG Huaping ZHANG Ying HUANG Leijie DAI Lanfeng WANG | 2023 | Plant Diseases and Pests2023,14,4: | 0
