LIN28 coordinately promotes nucleolar/ribosomal functions and represses the 2C-like transcriptional program in pluripotent stem cells显示文摘LIN28 is an RNA binding protein with important roles in early embryo development, stem cell differentiation/re-programming,tumorigenesis and metabolism.Previous studies have focused mainly on its role in the cytosol where it interacts with Let-7 microRNA precursors or mRNAs,and few have addressed LIN28's role within the nucleus. Here,we show that LIN28 displays dynamic temporal and spatial expression during murine embryo development.Maternal LIN28 expression drops upon exit from the 2-cell stage, and zygotic LIN28 protein is induced at the forming nucleolus during 4-cell to blas-tocyst stage development,to become dominantly expressed in the cytosol after implantation. In cultured pluripotent stem cells(PSCs),loss of LIN28 led to nucleolar stress and activation of a 2-cell/4-cell-like transcriptional program characterized by the expression of endogenous retrovirus genes.Mechanistically,LIN28 binds to small nucleolar RNAs and rRNA to maintain nucleolar integrity, and its loss leads to nucleolar phase separation defects, ribosomal stress and activation of P53 which in turn binds to and activates 2C transcription factor Dux.LIN28 also resides in a complex containing the nucleolar factor Nucleolin(NCL)and the transcrip-tional repressor TRIM28,and LIN28 loss leads to reduced occupancy of the NCL/TRIM28 complex on the Dux and rDNA loci, and thus de-repressed Dux and reduced rRNA expression.Lin28 knockout cells with nucleolar stress are more likely to assume a slowly cycling, translationally inert and anabolically inactive state, which is a part of previously unappreciated 2C-like transcriptional program. These findings elucidate novel roles for nucleolar LIN28 in PSCs,and a new mechanism linking 2C program and nucleolar functions in PSCs and early embryo development. | Zhen Sun Hua Yu Jing Zhao Tianyu Tan Hongru Pan Yuqing Zhu Lang Chen Cheng Zhang Li Zhang Anhua Lei Yuyan Xu Xianju Bi Xin Huang Bo Gao Longfei Wang Cristina Correia Ming Chen Qiming Sun Yu Feng Li Shen Hao Wu Jianlong Wang Xiaohua Shen George Q.Daley Hu Li Jin Zhang | 2022 | Protein & Cell2022,13,7: | 4
Oxygen permeability and CO_2-tolerance of Ce_(0.8)Gd_(0.2)O_(2-δ)-Ln BaCo_2O_(5+δ) dual-phase membranes显示文摘A series of oxygen permeable dual-phase composite oxides 60 wt% Ce0.8Gd0.2O2-δ-40 wt% Ln Ba Co2O5+δ(CGO-LBCO, Ln = La, Pr, Nd,Sm, Gd and Y) were synthesized through a sol-gel route and effects of the Ln3+cations on their phase structure, oxygen permeability and chemical stability against CO2 were investigated systemically by XRD, SEM, TG-DSC and oxygen permeation experiments. XRD patterns reveal that the larger Ln3+cations(La3+, Pr3+and Nd3+) successfully stabilized the double-layered perovskite structure of sintered LBCO,while the smaller ones(Sm3+, Gd3+, and Y3+) resulted in the partial decomposition of LBCO with some impurities formed. CGO-PBCO yields the highest oxygen permeation flux, reaching 2.8×10-7mol s-1 cm-2at 925?C with 1 mm thickness under air/He gradient. The TG-DSC profiles in 20 mol% CO2/N2 and oxygen permeability experiments with CO2 as sweep gas show that CGO-YBCO demonstrates the best chemical stability against CO2, possibly due to its minimum basicity. The stable oxygen permeation flux of CGO-YBCO under CO2 atmosphere reveals its potential application in the oxy-fuel combustion route for CO2 capture. | Longfei Luo Hongwei Cheng Guangshi Li Xionggang Lu Bo Jiang | 2015 | Journal of Energy Chemistry2015,24,1: | 3

Oxygen permeation and phase structure properties of partially A-site substituted BaCo_(0.7)Fe_(0.225)Ta_(0.075)O_(3-δ) perovskites显示文摘Ba0.9R0.1Co0.7Fe0.225Ta0.075O3-δ(BRCFT, R = Ca, La or Sr) membranes were synthesized by a solid-state reaction. Metal cation Ca2+,La3+or Sr2+doping on A-site partially substituted Ba2+in BaCo0.7Fe0.225Ta0.075O3-δoxides, and its subsequent effects on phase structure stability, oxygen permeability and oxygen desorption were systematically investigated by XRD, TG-DSC, H2-TPR, O2-TPD techniques and oxygen permeation experiments. The partial substitution with Ca2+, La3+or Sr2+, whose ionic radii are smaller than that of Ba2+, succeeded in stabilizing the cubic perovskite structure without formation of impurity phases, as revealed by XRD analysis. Oxygen-involving experiments showed that BRCFT with A-site fully occupied by Ba2+exhibited good oxygen permeation flux under He flow, reaching about 2.3mL min-1 cm-2at 900 ℃ with 1 mm thickness. Of all the membranes, BLCFT membrane showed better chemical stability in CO2, owing to the reduction in alkalinity of the mixed conductor oxide by La doping. In addition, we also found the stability of the perovskite structure under reducing atmospheres was strengthened by increasing the size of A-site cation(Ba2+>La3+>Sr2+>Ca2+). | Bo Jiang Hongwei Cheng Longfei Luo Xionggang Lu Naijun Zhang Jizhong Liu | 2014 | Journal of Energy Chemistry2014,23,2: | 2
Human obstructive(postvasectomy)and nonobstructive azoospermia-Insights from scRNA-Seq and transcriptome analysis显示文摘A substantial number of male infertility is caused by azoospermia. However, the underlying etiology and the molecular basis remain largely unknown.Through single-cell(sc)RNA sequencing,we had analyzed testis biopsy samples from two patients with obstructive azoospermia(OA)and nonobstructive azoospermia(NOA).We found only somatic cells in the NOA samples and explored the transcriptional changes in Sertoli cells in response to a loss of interactions with germ cells. Moreover,we observed a germ cell population discrepancy between an OA(postvasectomy)patient and a healthy individual. We confirmed this observation in a secondary study with two datasets at GSM3526588 and GSE124263 for detailed analysis wherein the regulatory mechanisms at the transcriptional level were identified. These findings thus provide valuable information on huma n spermatoge nesis, and we also ide ntified in sightful info rmation for further research on reproducti on-related diseases. | Shitao Chen Geng An Hanshu Wang Xiaolong Wu Ping Ping Longfei Hu Yunmei Chen Jue Fan C.Yan Cheng Fei Sun | 2022 | Genes & Diseases2022,9,3: | 2
Frequency/time domain modeling of a direct drive point absorber wave energy converter显示文摘Two different methods to model a point absorber wave energy converter(WEC)with direct drive linear power take-off(PTO)are proposed in the present study:the frequency domain(FD)method and the time domain(TD)method. In the FD analysis, the frequency response function(FRF)of the WEC device is obtained via the equation of motion, and the expressions of power capture width in regular and random waves are derived as well.In the TD modeling, based on a state space approximation of the convolution term in the motion equation, both regular wave and random wave simulations are carried out.The regular wave simulation results indicate that the state space approximation is sufficiently accurate and the capture width reaches the maximum in the vicinity of the natural frequency. In the random wave simulations, the effects of buoy size, the PTO damping and wave climate on the power capture width are discussed in detail, which leads to the conclusion that the capture widths are influenced by the natural frequency of the WEC device, peak frequency of the wave spectrum, the amplitude of FRF and PTO damping.Furthermore, the increase of the capture width is at the cost of a relatively large buoy size and PTO damping when control is not included. | CHENG ZhengShun YANG JianMin HU ZhiQiang XIAO LongFei | 2014 | Science China(Physics,Mechanics & Astronomy)2014,57,2: | 2
Oxygen Permeation and Stability of Ce_(0.8)Gd_(0.2)O_(2-δ)-PrBaCo_(2-x)FexO_(5+δ) Dual-phase Composite Membranes显示文摘Dual-phase membranes of 60 wt% Ce_(0.8)Gd_(0.2)O_(2-δ)-40 wt% Pr Ba Co2exFexO3 d(0 x 2) were prepared by a combined citrate and ethylene diamine tetraacetic acid(EDTA) complexing method. X-ray diffraction(XRD)results revealed the good chemical compatibility between ion-conducting phase CGO and electron-conducting phases PBC2 xFxO after sintering in air. The Fe ionic dopant had a significant effect on the phase structure stability and oxygen permeability under CO2 atmosphere, which was confirmed by XRD, thermogravimetrye differential scanning calorimetry(TGeD SC), scanning electron microscopy(SEM) and oxygen permeation experiments. CGOeP BC0.5F1.5O dual-phase membrane demonstrated a stable oxygen permeation flux of2.71x10-7mol cm 2s 1with 50 mol% He/CO2 as the sweep gas at 925 C, and this value was much higher than that of perovskite-type membranes. The rate-limiting step in the oxygen permeation process changed from the bulk diffusion to the surface oxygen exchange when the CGOeP BC0.5F1.5O membrane thickness decreased to 0.8 mm or less. Due to the high oxygen permeation fluxes and the excellent structural stability under CO2 atmosphere, the CGOeP BC0.5F1.5O membrane is a great potential candidate material for separating oxygen from air in the oxy-fuel combustion techniques. | Bo Jiang Hongwei Cheng Longfei Luo Xionggang Lu Zhongfu Zhou | 2014 | Journal of Materials Science & Technology2014,30,12: | 2
Probabilistic stability analysis of Bazimen landslide with monitored rainfall data and water level fluctuations in Three Gorges Reservoir, China显示文摘Landslide is a common geological hazard in reservoir areas and may cause great damage to local residents’life and property. It is widely accepted that rainfall and periodic variation of water level are the two main factors triggering reservoir landslides. In this study, the Bazimen landslide located in the Three Gorges Reservoir(TGR)was back-analyzed as a case study.Based on the statistical features of the last 3-year monitored data and field instrumentations, the landslide susceptibility in an annual cycle and four representative periods was investigated via the deterministic and probabilistic analysis, respectively. The results indicate that the fluctuation of the reservoir water level plays a pivotal role in inducing slope failures, for the minimum stability coefficient occurs at the rapid decline period of water level. The probabilistic analysis results reveal that the initial sliding surface is the most important area influencing the occurrence of landslide, compared with other parts in the landslide. The seepage calculations from probabilistic analysis imply that rainfall is a relatively inferior factor affecting slope stability. This study aims to provide preliminary guidance on risk management and early warning in the TGR area. | Wengang ZHANG Libin TANG Hongrui LI Lin WANG Longfei CHENG Tingqiang ZHOU Xiang CHEN | 2020 | Frontiers of Structural and Civil Engineering2020,14,5: | 1
Digital Twin-Assisted Knowledge Distillation Framework for Heterogeneous Federated Learning显示文摘In this paper,to deal with the heterogeneity in federated learning(FL)systems,a knowledge distillation(KD)driven training framework for FL is proposed, where each user can select its neural network model on demand and distill knowledge from a big teacher model using its own private dataset. To overcome the challenge of train the big teacher model in resource limited user devices, the digital twin(DT)is exploit in the way that the teacher model can be trained at DT located in the server with enough computing resources. Then, during model distillation, each user can update the parameters of its model at either the physical entity or the digital agent. The joint problem of model selection and training offloading and resource allocation for users is formulated as a mixed integer programming(MIP)problem. To solve the problem,Q-learning and optimization are jointly used, where Q-learning selects models for users and determines whether to train locally or on the server, and optimization is used to allocate resources for users based on the output of Q-learning.Simulation results show the proposed DT-assisted KD framework and joint optimization method can significantly improve the average accuracy of users while reducing the total delay. | Xiucheng Wang Nan Cheng Longfei Ma Ruijin Sun Rong Chai Ning Lu | 2023 | China Communications2023,20,2: | 0
Environmental selection and evolutionary process jointly shape genomic and functional profiles of mangrove rhizosphere microbiomes显示文摘Mangrove reforestation with introduced species has been an important strategy to restore mangrove ecosystem functioning. However, how such activities affect microbially driven methane(CH4),nitrogen(N),and sulfur(S)cycling of rhizosphere microbiomes remains unclear. To understand the effect of environmental selection and the evolutionary process on microbially driven biogeochemical cycles in native and introduced mangrove rhizospheres,we analyzed key genomic and functional profiles of rhizosphere microbiomes from native and introduced mangrove species by metagenome sequencing technologies.Compared with the native mangrove(Kandelia obovata,KO),the introduced mangrove(Sonneratia apetala,SA)rhizosphere microbiome had significantly(p<0.05)higher average genome size(AGS)(5.8 vs.5.5 Mb),average 16S ribosomal RNA gene copy number(3.5 vs.3.1),relative abundances of mobile genetic elements, and functional diversity in terms of the Shannon index(7.88 vs.7.84)but lower functional potentials involved in CH4 cycling(e.g.,mcrABCDG and pmoABC),N2 fixation(nifHDK),and inorganic S cycling(dsrAB,dsrC,dsrMKJOP,soxB,sqr, and fccAB).Similar results were also observed from the recovered Proteobacterial metagenome-assembled genomes with a higher AGS and distinct functions in the introduced mangrove rhizosphere.Additionally, salinity and ammonium were identified as the main environmental drivers of functional profiles of mangrove rhizosphere microbiomes through deterministic processes. This study advances our understanding of microbially mediated biogeochemical cycling of CH_(4),N,and S in the mangrove rhizosphere and provides novel insights into the influence of environmental selection and evolutionary processes on ecosystem functions, which has important implications for future mangrove reforestation. | Xiaoli Yu Qichao Tu Jihua Liu Yisheng Peng Cheng Wang Fanshu Xiao Yingli Lian Xueqin Yang Ruiwen Hu Huang Yu Lu Qian Daoming Wu Ziying He Longfei Shu Qiang He Yun Tian Faming Wang Shanquan Wang Bo Wu Zhijian Huang Jianguo He Qingyun Yan Zhili He | 2023 | mLife2023,2,3: | 0

Electrochemical Evaluation of Tumor Development via Cellular Interface Supported CRISPR/Cas Trans-Cleavage显示文摘Evaluating tumor development is of great importance for clinic treatment and therapy. It has been known that the amounts of sialic acids on tumor cell membrane surface are closely associated with the degree of cancerization of the cell. So,in this work, cellular interface supported CRISPR/Cas trans-cleavage has been explored for electrochemical simultaneous detection of two types of sialic acids,i.e.,N-glycolylneuraminic acid(Neu5Gc)and N-acetylneuraminic acid(Neu5Ac).Specifically,PbS quantum dot-labeled DNA modified by Neu5Gc antibody is prepared to specifically recognize Neu5Gc on the cell surface, followed by the binding of Neu5Ac through our fabricated CdS quantum dot-labeled DNA modified by Sambucus nigra agglutinin.Subsequently, the activated Cas12a indiscriminately cleaves DNA,resulting in the release of PbS and CdS quantum dots, both of which can be simultaneously detected by anodic stripping voltammetry.Consequently,Neu5Gc and Neu5Ac on cell surface can be quantitatively analyzed with the lowest detection limits of 1.12 cells/mL and 1.25 cells/mL,respectively.Therefore,a ratiometric electrochemical method can be constructed for kinetic study of the expression and hydrolysis of Neu5Gc and Neu5Ac on cell surface, which can be further used as a tool to identify bladder cancer cells at different development stages. Our method to evaluate tumor development is simple and easy to be operated,so it can be potentially applied for the detection of tumor occurrence and development in the future. | Liangfen Cheng Fuhan Yang Longfei Tang Lelin Qian Xu Chen Feng Guan Juan Zhang Genxi Li | 2022 | Research2022,,3: | 0
Evaluation of different primers of the 18S rRNA gene to profile amoeba communities in environmental samples显示文摘Amoeboid protists,an assemblage of organisms belonging to different phylogenetic lineages, have drawn increasing attention due to their crucial ecological roles in various environments and their potential health risks.Currently,18S rRNA gene sequencing is widely applied for the detection of amoebae. However,it is not clear which is the best primer pair for 18S rRNA gene amplification in amoebae. This study compared the four most commonly used primer pairs for revealing the diversity, composition, core species, and community assembly processes of amoebae in water and sediments. We found that the choice of primers artificially influences the detection of community composition of amoebae. We also found that short-read fragments may lead to mismatches in taxonomy and were not suitable for phylogenetic analyses. In contrast,full-length primers could detect the highest number of amoeba lineages and annotate 80%of reads belonging to amoebae to known species.However,full-length primers did not detect as many amoeba species as V4 primers. Moreover,we showed that beta diversity and community assembly determination were largely unaffected by primer choice, but different primers could influence our interpretations of the ecological process underlying stochasticity and determinism. This study indicates that full-length read sequencing and V4 region Illumina sequencing are suitable for profiling amoeba diversity in the environment. | Xiafei Zheng Zhili He Cheng Wang Qingyun Yan Longfei Shu | 2022 | Water Biology and Security2022,1,3: | 0

Dual-sized hollow particle incorporated fibroin thermal insulating coatings on catheter for cerebral therapeutic hypothermia显示文摘Selective endovascular hypothermia has been used to provide cooling-induced cerebral neuroprotection, but current catheters do not support thermally-insulated transfer of cold infusate, which results in an increased exit temperature, causes hemodilution, and limits its cooling efficiency.Herein,air-sprayed fibroin/silica-based coatings combined with chemical vapor deposited parylene-C capping film was prepared on catheter. This coating features in dual-sized-hollow-microparticle incorporated structures with low thermal conductivity. The infusate exit temperature is tunable by adjusting the coating thickness and infusion rate.No peeling or cracking was observed on the coatings under bending and rotational scenarios in the vascular models. Its efficiency was verified in a swine model, and the outlet temperature of coated catheter(75μm thickness)was 1.8-2.0◦C lower than that of the uncoated one. This pioneering work on catheter thermal insulation coatings may facilitate the clinical translation of selective endovascular hypothermia for neuroprotection in patients with acute ischemic stroke. | Ming Li Yuan Gao Miaowen Jiang Hongkang Zhang Yang Zhang Yan Wu Wenhao Zhou Di Wu Chuanjie Wu Longfei Wu Luzi Bao Xiaoxiao Ge Zhengfei Qi Ming Wei Ang Li Yuchuan Ding Jicheng Zhang Guangzhen Pan Yu Wu Yan Cheng Yufeng Zheng Xunming Ji | 2023 | Bioactive Materials2023,,8: | 0
CRMP2 promotesthe trafficking of phosphorylated AMPA receptor GluA2 subunit显示文摘CRMP2 is one of the well-studied members of the CRMPs family, which has been demonstrated not only play the roles in neurite extension, dendrites and dendritic spine development, but also in the AMPA receptor trafficking. However, the role of CRMP2 on the trafficking of AMPA receptor GluA2 subunit especially phosphorylated GluA2 is unknown.GST-Pull down and C-IP assays, immunofluorescence staining and whole cell patch clamp recording technique were performed in this study. The results showed that CRMP2 interacted with wild-type GluA2 and promoted the surface expression of wild-type GluA2.No matter GluA2 S880 was exogenously phosphorylated by phorbol myristate acetate(TPA)or endogenously phosphorylated by PKC5,CRMP2 can promote the surface expression of phosphorylated GluA2 and enhance its amplitude and frequency of mEPSCs,increasing the slope of the I-V curve. | Wu Jiaming Cheng Longfei Li Jiong Zhang Jifeng Guo Guoqing Chen Keen | 2021 | 解剖学杂志2021,44,S01: | 0
Touchable cell biophysics property recognition platforms enable multifunctional blood smart health care显示文摘As a crucial biophysical property, red blood cell(RBC)deformability is pathologically altered in numerous disease states, and biochemical and structural changes occur over time in stored samples of otherwise normal RBCs.However, there is still a gap in applying it further to point-of-care blood devices due to the large external equipment(high-resolution microscope and microfluidic pump),associated operational difficulties, and professional analysis.Herein,we revolutionarily propose a smart optofluidic system to provide a differential diagnosis for blood testing via precise cell biophysics property recognition both mechanically and morphologically.Deformation of the RBC population is caused by pressing the hydrogel via an integrated mechanical transfer device. The biophysical properties of the cell population are obtained by the designed smartphone algorithm.Artificial intelligence-based modeling of cell biophysics properties related to blood diseases and quality was developed for online testing. We currently achieve 100%diagnostic accuracy for five typical clinical blood diseases(90 megaloblastic anemia,78 myelofibrosis,84 iron deficiency anemia,48 thrombotic thrombocytopenic purpura, and 48 thalassemias)via real-world prospective implementation;furthermore, personalized blood quality(for transfusion in cardiac surgery)monitoring is achieved with an accuracy of 96.9%.This work suggests a potential basis for next-generation blood smart health care devices. | Longfei Chen Yantong Liu Hongshan Xu Linlu Ma Yifan Wang Le Yu Fang Wang Jiaomeng Zhu Xuejia Hu Kezhen Yi Yi Yang Hui Shen Fuling Zhou Xiaoqi Gao Yanxiang Cheng Long Bai Yongwei Duan Fubing Wang Yimin Zhu | 2021 | Microsystems & Nanoengineering2021,7,6: | 0
