2Single-cell Immune Landscape of Human Recurrent Miscarriage显示文摘t Successful pregnancy in placental mammals substantially depends on the establishment of maternal immune tolerance to the semi-allogenic fetus.Disorders in this process are tightly associated with adverse pregnancy outcomes including recurrent miscarriage(RM).However,an indepth understanding of the systematic and decidual immune environment in RM remains largely lacking.In this study,we utilized single-cell RNA-sequencing(scRNA-seq)to comparably analyze the cellular and molecular signatures of decidual and peripheral leukocytes in normal and unexplained RM pregnancies at the early stage of gestation.Integrative analysis identifies 22 distinct cell clusters in total,and a dramatic difference in leukocyte subsets and molecular properties in RM cases is revealed.Specifically,the cytotoxic properties of CD8^(+)effector T cells,nature killer(NK),and mucosal-associated invariant T(MAIT)cells in peripheral blood indicates apparently enhanced pro-inflammatory status,and the population proportions and ligand–receptor interactions of the decidual leukocyte subsets demonstrate preferential immune activation in RM patients.The molecular features,spatial distribution,and the developmental trajectories of five decidual NK(dNK)subsets have been elaborately illustrated.In RM patients,a dNK subset that supports embryonic growth is diminished in proportion,while the ratio of another dNK subset with cyto toxic and immune-active signature is significantly increased.Notably,a unique pro-inflammatory CD56^(+)CD16^(+)dNK subset substantially accumulates in RM decidua.These findings reveal a comprehensive cellular and molecular atlas of decidual and peripheral leukocytes in human early pregnancy and provide an in-depth insight into the immune pathogenesis for early pregnancy loss.Feiyang Wang Wentong Jia Mengjie Fan Xuan Shao Zhilang Li Yongjie Liu Yeling Ma Yu-Xia Li Rong Li Qiang Tu Yan-Ling Wang 2021Genomics, Proteomics & Bioinformatics2021,19,2:8
3A new algorithm of retrieving a petroleum substances absorption coefficient in sea water based on a remote sensing image显示文摘Establishing the remote sensing algorithm of retrieving the absorption coefficient of seawater petroleum substances is an efficient way to improve the accuracy of retrieving a seawater petroleum concentration using a remote sensing technology. A remote sensing reflectance is a basic physical parameter in water color remote sensing. Apply it to directly retrieve the absorption coefficient of seawater petroleum substances is of potential advantage. The absorption coefficient of waters containing petroleum [ACWCP, a_o(λ)], consists of the absorption coefficient of pure water [ACPW, a_w(λ)], plankton [ACP, a_(ph)(λ)], colored scraps [ACCS, a_(d,g)(λ)], and petroleum substance [ACPS, a_(oil)(λ)]. Among those, ACCS consists of the absorption coefficient of nonalgal particle [ACNP, a_d(λ)] and colored dissolved organic matter [ACCDOM, a_g(λ)]. For waters containing petroleum, the retrieved ACCS using the existing method is a combination absorption coefficient of ACNP,ACCDOM and ACPA [CAC, a_(d,g,oil)(λ)]. Therefore, the principle question is how to extract ACPS from CAC.Through the analysis of the three proportion tests conducted between the year of 2013 and 2015 and the corresponding remote sensing data, an algorithm of retrieving the absorption coefficient of petroleum substances is proposed based on remote sensing reflectance. First of all, ACPS and CAC are retrieved from the reflectance using the quasi-analytical algorithm(QAA), with some parameter modified. Secondly, given the fact that the backscatter coefficient [BC, b_(bp)(555)] of total particles at 555 nm can be obtained completely from the reflectance, the relation between BC and ACNP in petroleum contaminated water can be established. As a result, ACNP can be calculated. Then, combining the remote sensing retrieving algorithm of a_g(440), the method of achieving the spectral slope of the absorption coefficient can be established, from which ACCDOM,can be calculated. Finally, ACPS can be computed as the residual. 5The role of gut microbiota and its metabolites short-chain fatty acids in food allergy显示文摘Emerging evidence indicated that the increase in food allergy(FA)over the past few decades was associated with the abnormal compositional and metabolic changes of gut microbiota.Gut microbiota played a vital role in maintaining the homeostasis of the immune system and the dysbiosis of gut microbiota promoted the occurrence of FA.Recent research suggested that short-chain fatty acids(SCFAs),the main metabolites derived from gut microbiota,contributed to FA protection.Herein,we provided a comprehensive review on the relationship between gut microbiota and FA.The multifaceted mechanisms underlymg beneficial effects of gut microbiota composition/metabolites on the regulation of diverse cellular pathways in intestinal epithelial cells,dendritic cells,innate lymphoid cells,T cells,B cells and mast cells in the immune system were discussed systematically.These findings emphasized the positive function of gut microbiota in FA and provided novel ideas for the treatment or prevention of FA in the future.Chen Chen Chenglong Liu Ke Zhang Wentong Xue 2023Food Science and Human Wellness2023,12,3:2
6节段性小肠移植手术中采用自体髂内血管进行血管重建显示文摘背景:本研究旨在评估自体髂内动静脉可否用于异体及自体节段性小肠移植手术的血管重建。方法:2011年1月至2019年1月间34例小肠移植患者纳入研究,其中19例为亲属活体移植,15例为自体移植。收集患者临床资料、血管重建类型及术后并发症进行分析。结果:全组男性20例,女性14例,中位年龄35岁。在34例小肠移植中,22例(64.7%,其中异体移植和自体移植各11例)采用自体髂内动脉脉进行血管重建,12例(35.3%)直接行血管吻合。血管重建耗时(21˘6)min。与直接吻合组相比,血管重建组患者手术总时长(530˘226 vs 440˘116 min,P=0.208)和冷缺血时间(159˘49 vs 125˘66 min,P=0.097)均有延长的趋势,但差异并未达到统计学意义。直接吻合组术后血栓形成发生率较血管重建组有增高的趋势(16.7%vs 0%,P=0.118)。中位随访36.9个月,无一例出现静脉狭窄或假性动脉瘤。19例异体移植患者中,4例(21.1%)发生急性排斥反应,1例(5.2%)出现慢性排斥反应。结论:本组数据显示,采用自体髂内动静脉进行血管重建可有力促进小肠移植的开展,同时有望降低术后血管并发症的风险。Guosheng Wu Yinglun Wu Mian Wang Wentong Zhang Chaoxu Liu Tingbo Liang 2021Gastroenterology Report2021,9,4:2
7Effective nanotherapeutic approach for metastatic breast cancer treatment by supplemental oxygenation and imaging-guided phototherapy显示文摘Metastasis remains the primary cause for mortality of breast cancer.Despite advances in current therapeutic agents,patients with metastatic breast cancer still have poor prognoses.Tumor hypoxia,a key microenvironment factor,is emerging as an attractive target to prevent metastasis and is also involved with resistance to phototherapy.Here,we show an effective nanotherapeutic approach based on manganese dioxide-coated polydopamine nanocarriers to trigger robust anti-tumor and anti-metastasis responses against metastatic breast cancer by supplemental oxygenation and multimodal imaging-guided phototherapies.In cancer cells,the produced oxygen by the developed nanoplatform decreases the expression of hypoxia-inducible factors 1 a to inhibit tumor metastasis,and enhances the efficacy of photodynamic therapy.This nanotherapeutic approach enables the combined photodynamic/photothermal treatments with great inhibition on cell migration and invasion in vitro.Moreover,the nanotherapeutics effectively suppresses primary tumor progress and inhibits lung metastasis in v ivo in a breast cancer mouse model with satisfying biosafety.This study suggests that the tumor hypoxia-targeting nanotherapeutics have great potential for preventing and treating metastatic cancers.Jialing Hu Fuan Wang Feng Liu Wentong Sun Qunying Jiang Yahua Liu Yun Zhao Xiaoqing Liu 2020Nano Research2020,13,4:1

12Changes in wheat protein digestibility and allergenicity: role of Pediococcus acidilactici XZ31 and yeast during dough fermentation显示文摘Wheat flour,as the most important source of food globally,is one of the most common causative agents of food allergy.This study aimed to investigate the effects of fermentation on wheat protein digestibility and allergenicity.Protein digestibility were evaluated using the sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis and enzyme-linked immunosorbent assay.The effect of protein on intestinal permeability was investigated by Caco-2 cell monolayers.Co-culture fermentation with Pediococcus acidilactici XZ31 and yeast leads to improvement in digestibility of wheat protein compared to single strain fermentation.Fermentation leads to a decrease in albumin/globulin antigenicity and an increase in gluten R5 reactivity,with the most significant changes in the co-culture group.Digestion strengthen the decrease of protein antigenicity and counteracts the difference in antigenicity induced by fermentation between groups.However,pretreatment with P.acidilactici XZ31 reduces the amount of allergens across Caco-2 monolayer and attenuates the gluten-induced increase in permeability of Caco-2 cell monolayer by reducing actin polymerization and villous atrophy.Co-culture fermentation reduces gluten-induced cell monolayer damage to a greater extent than P.acidilactici XZ31 monoculture.These results gives valuable insight into the effects of P.acidilactici XZ31 fermentation on the allergenicity and toxicity of wheat proteins,which contribute to promoting the application of multi-strain leavening agent in hypoallergenic and gluten-free wheat products.Wenhui Fu Chen Chen Chenglong Liu Sha Tao Wentong Xue 2023Food Science and Human Wellness2023,12,6:0
13Specific knockout of Sox2 in astrocytes greatly reduces reactive astrocyte formation and promotes recovery after early postnatal TBI in mouse cortex显示文摘Reactive astrocytes are a double edged sword for traumatic brain injury(TBD),whether their overall effect on injury is beneficial or harmful depends on the balance between the loss and gain of function.However,the mechanism of reactive astrocytes after TBI is poorly understood.Recently more and more studies have been conducted to regulate the gene expression of astrocytes as a target to treat the disease.Here,we generated early postnatal TBI in the mouse cortex,indicating typical reactive astrocytes and glial scar were observed around damaged area;We next deleted Sox2 in astrocytes to observe its effects on reactive astrocytes and tissue repair after postnatal injury in the mouse cortex.As a result,specific deletion of Sox2 in astrocytes greatly inhibited the formation of reactive astrocytes and glial scars,and significantly promoted nerve regeneration in the damaged area and post injured repair.Together,our study indicate that Sox2 plays an important role in reactive astrocytes after early postnatal TBI in the mouse cortex.Therefore,Sox2 in astrocytes may be a putative target to develop new therapeutic tools for TBI treatment.Liu Yitong Hong Wentong Gong Pifang Pan Xinjie Qi Guibo Qin Song 2021解剖学杂志2021,44,S01:0
14Spatiotemporal variability of rain-on-snow events in the arid region of Northwest China显示文摘Rain-on-snow(ROS)events involve rainfall on snow surfaces,and the occurrence of ROS events can exacerbate water scarcity and ecosystem vulnerability in the arid region of Northwest China(ARNC).In this study,using daily snow depth data and daily meteorological data from 68 meteorological stations provided by the China Meteorological Administration National Meteorological Information Centre,we investigated the spatiotemporal variability of ROS events in the ARNC from 1978 to 2015 and examined the factors affecting these events and possible changes of future ROS events in the ARNC.The results showed that ROS events in the ARNC mainly occurred from October to May of the following year and were largely distributed in the Qilian Mountains,Tianshan Mountains,Ili River Valley,Tacheng Prefecture,and Altay Prefecture,with the Ili River Valley,Tacheng City,and Altay Mountains exhibiting the most occurrences.Based on the intensity of ROS events,the areas with the highest risk of flooding resulting from ROS events in the ARNC were the Tianshan Mountains,Ili River Valley,Tacheng City,and Altay Mountains.The number and intensity of ROS events in the ARNC largely increased from 1978 to 2015,mainly influenced by air temperature and the number of rainfall days.However,due to the snowpack abundance in areas experiencing frequent ROS events in the ARNC,snowpack changes exerted slight impact on ROS events,which is a temporary phenomenon.Furthermore,elevation imposed lesser impact on ROS events in the ARNC than other factors.In the ARNC,the start time of rainfall and the end time of snowpack gradually advanced from the spring of the current year to the winter of the previous year,while the end time of rainfall and the start time of snowpack gradually delayed from autumn to winter.This may lead to more ROS events in winter in the future.These results could provide a sound basis for managing water resources and mitigating related disasters caused by ROS events in the ARNC.YANG Zhiwei CHEN Rensheng LIU Zhangwen ZHAO Yanni LIU Yiwen WU Wentong 2024Journal of Arid Land2024,16,4:0
15Nitrogen doped hollow carbon nanospheres as efficient polysulfide restricted layer on commercial separators for high-performance lithium-sulfur batteries显示文摘The polysulfide shuttle limits the development of lithium-sulfur(Li-S) batteries with high energy density and long lifespan. Herein, nitrogen doped hollow carbon nanospheres(NHCS) derived from polymerization of dopamine on SiO_(2)nanospheres are employed to modify the commercial polypropylene/polyethylene/polypropylene tri-layer separators(PP/PE/PP@NHCS). The abundant nitrogen heteroatoms in NHCS exhibit strong chemical adsorption toward polysulfides, which can effectively suppress the lithium polysulfides shuttle and further enhance the utilization of active sulfur. Lithium-sulfur batteries employing the PP/PE/PP@NHCS deliver an initial discharge capacity of 1355 mAh/g and retain high capacity of 921 mAh/g after 100 cycles at 0.2 C. At a high rate of 2 C, the lithium-sulfur batteries exhibit capacity of 461 mAh/g after 1000 cycles with a capacity fading rate of 0.049% per cycle. 16Unusual local electric field concentration in multilayer ceramic capacitors显示文摘Local electric-field around multitype pores(dielectric pore,interface pore,electrode pore)in multilayer ceramic capacitors(MLCCs)was investigated using Kelvin probe force microscopy combined with the finite element simulation to understand the effect of pores on the electric reliability of MLCCs.Electricfield is found to be concentrated significantly in the vicinity of these pores and the strength of the local electric-field is 1.5e5.0 times of the nominal strength.Unexpectedly,the concentration degree of the pores in the inner electrode is much higher than that in the dielectrics and dielectric-electrode interfaces.Meanwhile,geometry orientations are found to have a remarkable influence on the local electric field strength.The pores act as an insulation degradation precursor via local electric,thermal center,and oxygen vacancies accumulation center.Such unusual local electric field concentration of multitype pores can provide new insights into the understanding of insulation degradation evolution,processing tailoring and design optimization for MLCCs.Wentong Du Huarong Zeng Weiwei Yang Kunyu Zhao Faqiang Zhang Guorong Li Yongxiang Li Zhifu Liu 2023Journal of Materiomics2023,9,2:0
17Temporal-spatial Generation of Astrocytes in the Developing Diencephalon显示文摘tAstrocytes are the largest glial population in the mammalian brain.However,we have a minimal understanding of astrocyte development,especially fate specification in different regions of the brain.Through lineage tracing of the progenitors of the third ventricle(3V)wall via in-utero electroporation in the embryonic mouse brain,we show the fate specification and migration pattern of astrocytes derived from radial glia along the 3V wall.Unexpectedly,radial glia located in different regions along the 3V wall of the diencephalon produce distinct cell types:radial glia in the upper region produce astrocytes and those in the lower region produce neurons in the diencephalon.With genetic fate mapping analysis,we reveal that the first population of astrocytes appears along the zona incerta in the diencephalon.Astrogenesis occurs at an early time point in the dorsal region relative to that in the ventral region of the developing diencephalon.With transcriptomic analysis of the region-specific 3V wall and lateral ventricle(LV)wall,we identified cohorts of differentially-expressed genes in the dorsal 3V wall compared to the ventral 3V wall and LV wall that may regulate astrogenesis in the dorsal diencephalon.Together,these results demonstrate that the generation of astrocytes shows a spatiotemporal pattern in the developing mouse diencephalon.Wentong Hong Pifang Gong Xinjie Pan Zhonggan Ren Yitong Liu Guibo Qi Jun-Liszt Li Wenzhi Sun Woo-Ping Ge Chun-Li Zhang Shumin Duan Song Qin 2024Neuroscience Bulletin2024,40,1:0
18Heavy metal gradients from rural to urban lakes in central China显示文摘Background:Limited information is available on heavy metal patterns in lakes under rapid watershed urbanization,especially considering a large spatial gradient with a long linear distance and great variations in topographic relief.To fill this gap,we studied concentrations of a series of heavy metals in both water and sediments from 20 lakes along a rural to urban gradient in central China,and we aimed to understand the effects of urban processes on heavy metal dynamics in lake ecosystems.Studied lakes were divided into five groups:A(rural reservoir group),B(rural commercial fishing group),C(urban park group),D(urban recreational fishing group),and E(urban commercial fishing group).An inductively coupled plasma optical emission spectrometer(ICP-OES)and an inductively coupled plasma-mass spectrometer(ICP-MS)were used to analyze the heavy metals in water and sediments.Results:An increasing trend of most heavy metals in water from rural to urban lakes was observed.Concentrations of cadmium(Cd),cobalt(Co),lead(Pb),chromium(Cr),arsenic(As),nickel(Ni),magnesium(Mn),iron(Fe),and aluminum(Al)in water were significantly lower in rural group A than those in other groups.Arsenic in sediments of rural group A was lower than those in other groups.No other heavy metal element in sediments was significantly different among groups.The enrichment factor analysis of selected heavy metals showed there were different degrees of enrichments of heavy metals in sediments.The potential ecological risk index showed a low level for heavy metals in sediments of all studied lakes.Conclusions:Results indicated that urban processes could have an impact on heavy metals in lake water.The sources of heavy metals in sediments were more likely from anthropogenic activities.These results could enhance our understanding of metal dynamics in lake ecosystems under urbanization and could help prevent heavy metal pollutions and promote sustainable management of urban ecosystems.Wentong Xia Rui Wang Bin Zhu Lars G.Rudstam Yinglong Liu Yanxue Xu Wei Xin Yushun Chen 2020Ecological Processes2020,9,1:0
19Mid1ip1b modulates apical reorientation of non-centrosomal microtubule organizing center in epithelial cells显示文摘In most kinds of animal cells,the centrosome serves as the main microtubule organizing center(MTOC)that nucleates microtubule arrays throughout the cytoplasm to maintain cell structure,cell division and intracellular transport.Whereas in epithelial cells,non-centrosomal MTOCs are established in the apical domain for generating asymmetric microtubule fibers and cilia in epithelial cells for the organ morphogenesis during embryonic development.However,the mechanism by which MTOCs localize to the apical domain in epithelial cells remains largely unknown.Here,we show that Mid1ip1b has a close interaction with g-tubulin protein,the central component of MTOC,and modulates lumen opening of the neural tube,gut,intestine,and kidney of zebrafish.Knockdown or dominant negative effect of Mid1ip1b resulted in failure of lumen formation of the organs as aforementioned.Moreover,the non-centrosomal MTOCs were unable to orientate to the apical domain in Mid1ip1b knockdown epithelial cells,and the centrosomal MTOCs were inaccurately placed in the apical domain,resulting in defective formation of asymmetric microtubules and misplacement of cilia in the apical domain.These data uncover a molecule that controls the proper localization of MTOCs in the apical domain in epithelial cells for organ morphogenesis during embryonic development.Xin Zhou Chun Xiao Yu Li Yanna Shang Dongqin Yin Siying Li Bo Xiang Ran Lu Yi Ji Yang Wu Wentong Meng Hongyan Zhu Jin Liu Huozhen Hu Xianming Mo Hong Xu 2018Journal of Genetics and Genomics2018,45,8:0
20Detection of Serum Antibodies of Porcine Pseudorabies Virus(PRV)in Binzhou City in 2014显示文摘To understand immunization effect of pseudorabies vaccine and infection status of porcine pseudorabies( PR) in pig farms and guide prevention and control against PR,453 copies of blood samples collected from 43 different scale pig farms in four counties( districts) of Binzhou City were detected with ELISA to investigate PRV g B antibody and g E antibody. Detection results showed that the g B antibody positive rate of sows was 75. 58%,and that of fattening pigs was68. 67%; the pig farms with positive rate higher than 70% accounted for 74. 42% of total survey pig farms. The PRV g E antibody positive rates of sows and fattening pigs were 25. 41% and 26. 67%,and the positive rates of pig farms were 46. 51% and 44. 33%,respectively.
