3GOLM1 exacerbates CD8^(+)T cell suppression in hepatocellular carcinoma by promoting exosomal PD-L1 transport into tumor-associated macrophages显示文摘The immunosuppressive microenvironment plays an important role in tumor progression and immunotherapy responses.Golgi membrane protein 1(GOLM1)is correlated to hepatocellular carcinoma(HCC)progression and metastasis.However,little is known about the role of GOLM1 in regulating the immunosuppressive environment and its impact on immunotherapeutic efficacy in HCC.In this study,GOLM1 was positively correlated with infiltrating tumor-associated macrophages(TAMs)expressed high levels of programmed death-ligand 1(PD-L1)and CD8^(+)T cell suppression in HCC tissues.Both gain-and loss-of-function studies determined a close correlation between GOLM1 and immunosuppression.In the mechanism,GOLM1 promoted COP9 signalosome 5-mediated PD-L1 deubiquitination in HCC cells and increased the transport of PD-L1 into exosomes via suppression of Rab27b expression.Furthermore,co-culture with exosomes derived from HCC cells upregulated the expression of PD-L1 on macrophages.Zoledronic acid in combination with anti-PD-L1 therapy reduced PD-L1^(+)TAMs infiltration and alleviated CD8^(+)T cell suppression,resulting in tumor growth inhibition in the mouse HCC model.Together,our study unveils a mechanism by which GOLM1 induces CD8^(+)T cells suppression through promoting PD-L1 stabilization and transporting PD-L1 into TAMs with exosome dependent.Targeting PD-L1^(+)TAM could be a novel strategy to enhance the efficacy of anti-PD-L1 therapy in HCC.Jinhong Chen Zhifei Lin Lu Liu Rui Zhang Yan Geng Minghao Fan Wenwei Zhu Ming Lu Lu Lu Huliang Jia Jubo Zhang Lun-Xiu Qin 2021Signal Transduction and Targeted Therapy2021,6,12:5

5Key technologies for the construction of the Xiluodu high arch dam on the Jinsha River in the development of hydropower in western China显示文摘Hydropower development in China is concentrated in the country's western regions.Among all the rivers in China,the lower course of the Jinsha River contains the richest hydro-energy resource,and therefore,4 mammoth hydropower plants are under construction on this particular section of the river at Wudongde,Baihetan,Xiluodu,and Xiangjiaba.The water-blocking structures of the hydropower facilities at Wudongde,Baihetan and Xiluodu are all arch dams of around 300 m high.In view of changes in the geological conditions at the foundation of the Xiluodu dam on the riverbed after excavation started,the designs of expanding foundation surface excavation and dovetailing the dam body and foundation rock on both upstream and downstream sides were introduced,allowing the arch dam and foundation to fit each other and improving the stress conditions of the dam body and foundation.By dividing the dam body into various concrete sections,the dynamic properties of concrete were adequately adjusted to the distribution of stress in the dam body.In addition,the use of the most optimal concrete material and mixture ratio allowed thermodynamics of concrete to satisfy the requirements of the strength,durability,temperature control and crack prevention of the concrete.Moreover,rigorous temperature control measures were introduced to prevent harmful cracking,thus enhancing the integrity of the arch dam.Furthermore,sophisticated construction machinery,scientific testing methods,and sound construction techniques were employed to ensure the uniformity and reliability of concrete placement.The 'Digital Dam' for the Xiluodu project,which is based on the theory of total life cycle,has supplied strong support for construction process control and decision-making.Lu Youmei Fan Qixiang Zhou Shaowu Li Bingfeng Li Wenwei 2012Engineering Sciences2012,10,2:3
6Glucose metabolism controls humanγδ-cell-mediated tumor immunosurveillance in diabetes显示文摘Patients with type 2 diabetes mellitus(T2DM)have an increased risk of cancer.The effect of glucose metabolism onγδT cells and their impact on tumor surveillance remain unknown.Here,we showed that high glucose induced Warburg effect type of bioenergetic profle in Vy9vδ2 T cells,leading to excessive lactate accumulation,which further inhibited lytic granule secretion by impairing the traffcking of cytolytic machinery to the Vy9vδ2 T-cell-tumor synapse by suppressing AMPK activation and resulted in the loss of antitumor activity in vitro,in vivo and in patients.Strikingly,activating the AMPK pathway through glucose control or metformin treatment reversed the metabolic abnormalities and restored the antitumor activity of Vy9vδ2 T cells.These results suggest that the impaired antitumor activity of Vy9vδ2 T cells induced by dysregulated glucose metabolism may contribute to the increased cancer risk in T2DM patients and that metabolic reprogramming by targeting the AMPK pathway with metformin may improve tumor immunosurveillance.Xiaofeng Mu Zheng Xiang Yan Xu Jing He Jianwen Lu Yuyuan Chen Xiwei Wang Chloe Ran Tu Yanme Wenyue Zhang Zhinan Yin Wing-hang Leung Yu-Lung Lau Yinping Liu Wenwei Tu 2022Cellular & Molecular Immunology2022,19,8:2

11Economic Critical Resources for the Industrial Exploitation of Natural Gas Hydrate显示文摘Since the implementation of several pilot production tests were in natural gas hydrate(NGH) reservoirs in terrestrial and marine settings, the study of NGH has entered a new stage of technological development for industrial exploitation. Prior to the industrial exploitation of any given NGH reservoir, the economic feasibility should be examined. The first step of economic evaluation of a NGH reservoir is to know whether its resource amount meets the requirement for industrial exploitation. Unfortunately, few relevant studies have been conducted in this regard. In this study, the net present value(NPV) method is employed to estimate the economic critical resources required for the industrial exploitation of NGHs under different production scenarios. Sensitivity analysis is also performed in order to specify the effects of key factors, such as the number of production wells, gas price, technological improvement and tax incentive, on the economic critical resources. The results indicate that China requires the lowest economic critical resource for a NGH reservoir to be industrially exploited, ranging from 3.62 to 24.02 billion m3 methane. 14Morphological and Sulfur-Isotopic Characteristics of Pyrites in the Deep Sediments from Xisha Trough,South China Sea显示文摘Pyrite is one of the common authigenic minerals in marine sediments.Previous studies have shown that the morphological and isotopic characteristics of pyrite are closely related to the geochemical environment where it is formed.To better understand the for-mation mechanism of authigenic pyrite,we analyzed the isotopic composition,morphology,and distribution of pyrite in the sediment at 500m below the seafloor from Xisha Trough,South China Sea.Mineral morphologies were observed by scanning electron micros-copy and Raman spectrography.X-Ray computed tomography was applied to measure the particle size of pyrite.The size of pyrite crystals in the matrix sediment mainly ranged between 25 and 65µm(av.ca.40µm),although crystals were larger(av.ca.50μm)in the veins.The pyrites had a fine-grained truncated octahedral shape with occasionally well-developed growth steps,which implies the low growth rate and weak anaerobic oxidation of methane-sulfate reduction when pyrite was formed.Theδ^(34)S values of pyrites ranged from+20.8‰Vienna-defined Canyon Diablo Troilite(V-CDT)to+33.2‰V-CDT and from+44.8‰V-CDT to+48.9‰,which suggest two growth stages.In the first stage,with the continuous low methane flux,the pyrite possibly formed in an environment with good access to seawater.In the second stage,the pyrites mainly developed in sediment fractures and appeared in veins,probably due to the limited availability of sulfate.The less exposure of pyrite to the environment in the second stage was probably caused by sediment accumulation or perturbation.In this study,an episodic pyritization process was identified,and the paleoenvironment was reconstructed for the sediment investigated.CHANG Jingyi LIU Yujia LU Hailong LU Jing’an SU Xin YE Jianliang XIE Wenwei 2024Journal of Ocean University of China2024,23,1:0
15Promotion mechanism of self-transmissible degradative plasmid transfer in maize rhizosphere and its application in naphthalene degradation in soil显示文摘Rhizospheres can promote self-transmissible plasmid transfer,however,the corresponding mechanism has not received much attention.Plant-microbe remediation is an effective way to promote pollutant biodegradation;however,some pollutants,such as naphthalene,are harmful to plants and result in inefficient plant-microbe remediation.In this study,trans-fer of a TOL-like plasmid,a self-transmissible plasmid loaded with genetic determinants for pollutant degradation,among different bacteria was examined in bulk and rhizosphere soils as well as addition of maize root exudate and its artificial root exudate(ARE).The results showed that the numbers of transconjugants and recipients as well as bacterial metabolic activities,such as xylE mRNA expression levels and catechol 2,3-dioxygenase(C23O)activ-ities of bacteria,remained high in rhizosphere soils,when compared with bulk soils.The number of transconjugants and bacterial metabolic activities increased with the increasing exudate and ARE concentrations,whereas the populations of donor and recipient bacteria were substantially unaltered at all concentrations.All the experiments consistently showed that a certain number of bacteria is required for self-transmissible plasmid transfer,and that the increased plasmid transfer might predominantly be owing to bacterial metabolic activ-ity stimulated by root exudates and ARE.Furthermore,ARE addition increased naphthalene degradation by transconjugants in both culture medium and soil.Thus,the combined action of a wide variety of components in ARE might contribute to the increased plasmid transfer and naphthalene degradation.These findings suggest that ARE could be an effectively al-ternative for plant-microbe remediation of pollutants in environments where plants cannot survive.Qiuyan Jiang Wenwei Lu Lei Zhang Yeqing Jin Yujing Wang Jun Chen Ziyi Ye Ming Xiao 2022Journal of Environmental Sciences2022,34,5:0

19Experimental study on sand production and coupling response of silty hydrate reservoir with different contents of fine clay during depressurization显示文摘To further understand the characteristics of clay and sand production(hereafter collectively referred to as sand production)and to provide optimization designs of sand control schemes are critical for gas production from clayey silt natural gas hydrate reservoirs in the South China Sea.Thus,gas-water-sand production behavoirs and coupling reservoir subsidence characteristics before,during,and after hydrate dissociation of the clayey silt hydrate reservoirs with different clay contents(5%,10%,15%,20%,25%,and 30%)have been studied through a self-developed experimental system.The results show that with the increase of clay content,the total mass of sand production first increases and then decreases,and it reaches maximum when the clayey content is 20%.The sand production is the lowest before hydrate dissociation and increases significantly during hydrate dissociation,which mainly occurs in the high-speed gas and water production stage at the beginning of hydrate dissociation.After hydrate dissociation,the sand production decreases significantly.During the whole depressurization process,the clay and free sand particles generally move to the sand outlet due to the fluid driving force and overlying stress extrusion.However,for conditions of high clay contents,those particles fail to pass through the sand control screen and gradually accumulate and block the screen by forming a mud cake,which greatly reduce the permeability of the screen and limite sand production as well as gas and water production.Our research lays a foundation for sand production prediction and sand control scheme selection during gas recovery from clayey silty hydrate reservoirs that greatly need to consider a balance between sand control and gas productivity.Xiangyu Fang Dianheng Yang Fulong Ning Linjie Wang Zhichao Liu Yanjiang Yu Wenwei Xie Hongfeng Lu Yanlong Li Meng Xu 2023Petroleum2023,9,1:0
