| 3 | Characterization and fine mapping of an early senescence mutant (es-t) in Oryza sativa L.显示文摘An es-t (early senescence-temporary) mutant, produced by ethylene methylsulfonate treatment of strain Nipponbare, was identified in rice. The leaves of es-t appeared yellow at the seedling stage, and had decreased chlorophyll content. Rust spots were found during growth in es-t, especially at the leaf margin and tip. The plants showed a typical early-senescence phenotype at the milky stage. The leaf surface of es-t appeared smoother than wild-type leaves under a scanning electron microscope, because the leaves lack siliceous protuberances around the stoma. Chloroplasts grow abnormally and are filled with many starch grains in es-t. Paraffin section analysis showed that the development of the sclerenchyma cells and vascular bundles were also abnormal in es-t. Genetic analysis indicated that es-t was controlled by a recessive gene, which was finely mapped to a 42-kb interval on chromosome 5. | 4 | Molecular feature and therapeutic perspectives of immune dysregulation, polyendocrinopathy, enteropathy, X-linked syndrome显示文摘Regulatory T(Treg) cells, a subtype of immunosuppressive CD4^+T cells, are vital for maintaining immune homeostasis in healthy people. Forkhead box protein P3(FOXP3), a member of the forkhead-wingedhelix family, is the pivotal transcriptional factor of Treg cells. The expression, post-translational modifications, and protein complex of FOXP3 present a great impact on the functional stability and immune plasticity of Treg cells in vivo. In particular, the mutation of FOXP3 can result in immune dysregulation,polyendocrinopathy, enteropathy, X-linked(IPEX) syndrome, which is a rare genetic disease mostly diagnosed in early childhood and can soon be fatal. IPEX syndrome is related to several manifestations,including dermatitis, enteropathy, type 1 diabetes, thyroiditis, and so on. Here, we summarize some recent findings on FOXP3 regulation and Treg cell function. | 5 | Gap junction-mediated cell-to-cell communication in oral development and oral diseases: a concise review of research progress显示文摘Homoeostasis depends on the close connection and intimate molecular exchange between extracellular, intracellular and intercellular networks. Intercellular communication is largely mediated by gap junctions(GJs), a type of specialized membrane contact composed of variable number of channels that enable direct communication between cells by allowing small molecules to pass directly into the cytoplasm of neighbouring cells. Although considerable evidence indicates that gap junctions contribute to the functions of many organs, such as the bone, intestine, kidney, heart, brain and nerve, less is known about their role in oral development and disease. In this review, the current progress in understanding the background of connexins and the functions of gap junctions in oral development and diseases is discussed. The homoeostasis of tooth and periodontal tissues, normal tooth and maxillofacial development, saliva secretion and the integrity of the oral mucosa depend on the proper function of gap junctions.Knowledge of this pattern of cell–cell communication is required for a better understanding of oral diseases. | 7 | Spatiotemporal evolution of prehistoric Neolithic-Bronze Age settlements and influencing factors in the Guanting Basin, northeast Tibetan Plateau显示文摘For almost two decades, the relationship between prehistoric natural disasters that struck the Guanting Basin in northeast Tibetan Plateau and the destruction of Lajia, an archeological site, has attracted scholarly attention and been widely discussed. Whereas most studies have focused on the impacts of disasters on a single site within the Guanting Basin, few have examined patterns of spatiotemporal evolution of human settlements from the Neolithic to the Bronze Age. Consequently, there is a lack of clarity on the processes and mechanisms underlying the evolution of prehistoric human-land relationships in the Guanting Basin. We therefore examined spatiotemporal variations in settlements in the Guanting Basin during the period, based on the locations, altitudes, and areas of archaeological sites. We found that four sites were located on the third terrace of the Yellow River during the late Yangshao period(5500–5000 cal yr BP) and distributed within a small area. During the period between the Majiayao and Qijia cultures(5300–3600 cal yr BP), the number of sites evidently increased and the scale and distribution of settlements expanded, with settlements generally shifting toward the lower elevation areas of the Guanting Basin.During the Xindian period(3400–2700 cal yr BP), the number and scale of sites showed a downward trend and the distribution of settlements contracted. The Xindian settlement underwent altitude-based spatial differentiation, with some groups moving to areas at higher altitudes and others remained in lower altitude areas. Moreover, we found that the number, scale, and distribution range of Neolithic and Bronze Age sites in the Guanting Basin were closely related to the evolution and distribution patterns of prehistoric cultures in the regions of Gansu and Qinghai, which were further affected by climate change and agricultural development. | 11 | Differential expression of FOXO1 during development and myoblast differentiation of Qinchuan cattle and its association analysis with growth traits显示文摘Our previous work reported a relationship between FOXO1 mutations and growth of Qinchuan(QC) cattle. Here, we performed differential expression analysis of FOXO1 and its association analysis with growth traits in QC cattle. First, we measured the expression of the FOXO1 gene in nine tissues during three developmental stages. The results showed that FOXO1 was abundantly expressed in tissues of calves but was strongly repressed in adulthood, although there was significant transcription in skeletal muscle. FOXO1 expression showed gradual up-regulation during differentiation of primary bovine skeletal muscle cells.We also identified six SNPs of the bovine FOXO1 gene by sequencing DNA pools of samples from 488 individuals, and association analysis indicated that five SNPs were significantly associated with some growth traits in the QC population. We further analyzed four haplotype combinations of the six SNPs and found significant correlation with body length(P<0.01). In conclusion, FOXO1 participates in bovine myocyte differentiation and expression, and may be a strong candidate as a gene that affects growth traits that could be exploited in a QC cattle breeding program. | 13 | Molecular cloning and functional identification of sterol C24-methyltransferase gene from Tripterygium wilfordii显示文摘Sterol C24-methyltransferase(SMT) plays multiple important roles in plant growth and development. SMT1, which belongs to the family of transferases and transforms cycloartenol into 24-methylene cycloartenol, is involved in the biosynthesis of 24-methyl sterols. Here, we report the cloning and characterization of a cDNA encoding a sterol C24-methyltransferase from Tripterygium wilfordii(Tw SMT1). Tw SMT1(Gen Bank access number KU885950) is a 1530 bp cDNA with a 1041 bp open reading frame predicted to encode a 346-amino acid, 38.62 k Da protein. The polypeptide encoded by the SMT1 cDNA was expressed and purified as a recombinant protein from Escherichia coli(E. coli) and showed SMT activity. The expression of Tw SMT1 was highly up-regulated in T. wilfordii cell suspension cultures treated with methyl jasmonate(Me JA). Tissue expression pattern analysis showed higher expression in the phellem layer compared to the other four organs(leaf, stem, xylem and phloem), which is about ten times that of the lowest expression in leaf. | 14 | Identification and characterization of soybean dreg soluble dietary fibre by combination of extrusion pre-treatment and enzymatic modification显示文摘OBJECTIVES:Soybean dreg is a by-product of soy milk processing,which contains high levels of soluble dietary fibre(SDF).In this study,we aimed to provide comprehensive processes of pre-treated extrusion for the improving structure and properties of soybean dreg soluble dietary fibre(SDSDF),which would be a valuable approach to enhance physiological activity.METHODS:Here,we characteristic the functional role of SDSDF employing to extrusion pretreatment.Soybean dregs were pre-treated using the twin screw extrusion method followed by enzymatic modification using neutral protease,α-amylase,glucoamylase,and cellulose to produce SDSDF.The physical properties and antioxidant activity of SDSDF were investigated.RESULTS:The morphology and crystal structure of SDSDF were observed that,through extrusion processing and enzymatic modification,the SDSDF yield increased by 106.28%.Moreover,the surface structure showed block-shaped or reticular formations in the extruded SDSDF,and the size of block-shaped cells was about 10μm.Infrared spectroscopic analysis showed that a characteristic absorption peak of polysaccharide appeared at 1631 cm−1 during extrusion processing.However,after extrusion processing,decreased absorption peaks were observed for the extruded SDSDF.Furthermore,XRD analysis showed that the 2θdiffraction peak changed at 24.16°for the extruded SDSDF.CONCLUSIONS:The overall findings suggest that the water holding capacity(WHC),oil holding capacity(OHC),expansibility,and the water solubility were significantly decreased in extruded SDSDF.In addition,the scavenging ability of 1,1-diphenyl-2-picrylhydrazyl(DPPH),-OH,O_(2)^(-),and the total reducing power were significantly improved,indicating that beneficial changes had taken place in the crystal structure of cellulose or hemicellulose to improve the physiological activity in extruded SDSDF. | Min Qu Yujia Liu Guang Zhang Sophia Jun Xue Hongchen Fan Yan Wang Shiyou Yu | 2017 | Food Quality and Safety2017,1,2: | 3 |

| 17 | Design and Analysis of an Active Helical Drive Downhole Tractor显示文摘During oil-gas well drilling and completion,downhole tools and apparatus should be conveyed to the destination to complete a series of downhole works.Downhole tractors have been used to convey tools in complex wellbores, however a very large tractive force is needed to carry more downhole tools to accomplish works with high efficiency. A novel serial active helical drive downhole tractor which has significantly improved performance compared with previous work is proposed. All previously reported helical drive downhole tractors need stators to balance the torque generated by the rotator. By contrast, the proposed serial downhole tractor does not need a stator; several rotator-driven units should only be connected to one another to achieve a tractive force multifold higher than that was previously reported. As a result,the length of a single unit is shortened, and the motion flexibility of the downhole tractor is increased. The major performance indicators, namely, gear ratio, velocity, and tractive force, are analyzed. Experimental results show that the maximum tractive force of a single-unit prototype with a length of 900 mm is 165.3 kg or 1620 N. | 18 | Anomaly detection of hot components in gas turbine based on frequent pattern extraction显示文摘Hot components operate in a high-temperature and high-pressure environment. The occurrence of a fault in hot components leads to high economic losses. In general, exhaust gas temperature(EGT) is used to monitor the performance of hot components.However, during the early stages of a failure, the fault information is weak, and is simultaneously affected by various types of interference, such as the complex working conditions, ambient conditions, gradual performance degradation of the compressors and turbines, and noise. Additionally, inadequate effective information of the gas turbine also restricts the establishment of the detection model. To solve the above problems, this paper proposes an anomaly detection method based on frequent pattern extraction. A frequent pattern model(FPM) is applied to indicate the inherent regularity of change in EGT occurring from different types of interference. In this study, based on a genetic algorithm and support vector machine regression, the relationship model between the EGT and interference was tentatively built. The modeling accuracy was then further improved through the selection of the kernel function and training data. Experiments indicate that the optimal kernel function is linear and that the optimal training data should be balanced in addition to covering the appropriate range of operating conditions and ambient temperature. Furthermore, the thresholds based on the Pauta criterion that is automatically obtained during the modeling process, are used to determine whether hot components are operating abnormally. Moreover, the FPM is compared with the similarity theory, which demonstrates that the FPM can better suppress the effect of the component performance degradation and fuel heat value fluctuation. Finally, the effectiveness of the proposed method is validated on seven months of actual data obtained from a Titan130 gas turbine on an offshore oil platform.
