1Correlation between Expression of DcR3 on Tumor Cells and Sensitivity to FasL显示文摘To investigate the correlation between sensitivity to Fas ligand(FasL)and expression level of decoy receptor 3 (DcR3)on tumor cell surface,Fas/DcR3 mRNA expression was detected by RT-PCR.Anti-DcR3 mAb was used to detect expression level of DcR3 on surface of tumor cells by flow cytometry.Caspase-8,caspase-9,caspase-3,Bcl-2 expressions were analyzed by Western blot,respectively.Sensitivity to apoptosis induced by FasL was determined by Annexin V apoptosis kit.The expressions of DcR3 on the surface of tumor cells from high to low were approximately 35.3% in BGC823 cells,and 21.6% in MCF-7 cells,respectively.The apoptotic rates induced by FasL from low to high were 15.6% in BGC823 cells,and 58.2% in MCF-7 cells,respectively.There was a significant correlation between the expression levels of DcR3 with FasL-inducing apoptosis.Wenzhu Li Changgong Zhang Caixia Chen Guohong Zhuang 2007Cellular & Molecular Immunology2007,4,6:20
2Interactive effect of dissolved organic matter and phenanthrene on soil enzymatic activities显示文摘The investigation of dissolved organic matter(DOM) on urease,catalase and polyphenol oxidase activity in a phenanthrene(Phe)contaminated soil was conducted under laboratory incubation conditions.Values of soil enzymatic activity depended mainly on incubation time.In the initial 16 days,urease activity increased,and was followed by a decrease.In the initial 8 days,catalase activity decreased and then increased.Variation of polyphenol oxidase activity was just the reverse of catalase activity.After 30 days of incubation,no pronounced difference among treatments with Phe,Phe and DOM,and control were detected in urease,catalase and polyphenol oxidase activity.Phe might inhibit urease and catalase,and stimulate polyphenol oxidase.DOM could improve inhibition of Phe in soil urease and catalase activity during the initial period of applying DOM.Nevertheless,DOM had no significant effect on polyphenol oxidase activity in the Phe contaminated soil.There was a negative correlation between catalase and polyphenol oxidase(r =-0.761***),and catalase and urease(r =-0.554**).Additionally,a positive correlation between polyphenol oxidase and urease was also detected(r = 0.701***).It is implied that the formed DOM after application of organic wastes into soils may counteract the inhibition of polycyclic aromatic hydrocarbons in soil enzyme activities.Xinhua Zhan,Wenzhu Wu,Lixiang Zhou,Jianru Liang,Tinghui Jiang College of Resources and Environmental Sciences,Nanjing Agricultural University,Nanjing 210095,China.Greenstar Plant Products Inc.,9430 198 St,Langley,BC,Canada V1M3C8. 2010Journal of Environmental Sciences2010,22,4:10

4Changes in soil carbon stocks and related soil properties along a 50-year grassland-to-cropland conversion chronosequence in an agro-pastoral ecotone of Inner Mongolia,China显示文摘Land use change significantly influences soil properties.There is little information available on the long-term effects of post-reclamation from grassland to cropland on soil properties.We compared soil carbon(C) and nitrogen(N) storage and related soil properties in a 50-year cultivation chronosequence of grassland in the agro-pastoral ecotone of Inner Mongolia.Field surveys on land use changes during the period of 1955-2002 were conducted to build a chronosequence of cropland of different ages since the conversion from grassland.The results showed that soil C and N storage,soil texture,and soil nutrient contents varied with land use types and cropland ages(P<0.01).In the 0-30 cm soil layer,the soil organic carbon(SOC) density was significantly lower in the croplands(3.28 kg C/m2 for C50 soil) than in the grasslands(6.32 kg C/m2).After 5,10,15,20,35,and 50 years of crop planting(years since the onset of cultivation),the SOC losses were 17%,12%,19%,47%,46%,and 48%,respectively,compared with the grasslands.The soil total nitrogen(TN) density of the grasslands was 65 g N/m2,and TN density of the cropland soil was 35 g N/m2 after 50 years of crop planting.Both the SOC and TN densities could be quantitatively determined by a negative exponential function of cropland age(P<0.0001,R2=0.8528;P<0.0001,R2=0.9637).The dissolved organic carbon(DOC) content,soil available potassium(AK) content,clay content,and pH value were decreased;and the soil bulk density and sand content were increased since the conversion of grassland into cropland during the 50-year period.Our results show soil nutrients were higher in grassland than in cropland.The conversion of grasslands to croplands induced a loss of soil C storage and changes of related soil properties.The reclamation time of cultivated soil(cropland age) had significant effects on soil properties in the study area.Yan JIAO Zhu XU JiaoHong ZHAO WenZhu YANG 2012Journal of Arid Land2012,4,4:8
5Fibrinogen-like protein 2 expression correlates with microthrombosis in rats with type 2 diabetic nephropathy显示文摘Fibrinogen-like protein 2 (fgl2),a novel prothrombinase,is involved in microthrombosis.We examined fgl2 expression in the glomerular and tubulointerstitial capillaries and its correlation with microthromsis in rats with streptozocin-induced type 2 diabetic nephropathy.Our RT-PCR and immunoblotting analysis showed that fgl2 mRNA and protein levels were increased in microvascular endothelial cells of the glomeruli and renal interstitia at week 19 and became significantly elevated with the development of diabetic nephropathy (P < 0.01).Fgl2 was not or only weakly expressed in the renal tissues of normal rats.Furthermore,a direct significant correlation (r=0.543,P < 0.01) was found between fgl2 expression and microthrombotic capillaries in the renal tissues.Enzyme linked immunosorbent assays (ELISA) additionally showed that circulating TNF-α levels in rats with type 2 diabetes were significantly elevated and closely correlated with fgl2 expression (r=0.871,P < 0.01).Our results suggest that fgl2 may activate renal microthrombosis,thus contributing to glomerular hypertension and renal ischemia.Guanhua Su Kun Liu Yan Wang Jue Wang Xiaowei Li Wenzhu Li Yuhua Liao Zhaohui Wang 2011The Journal of Biomedical Research2011,25,2:6
6Identification and molecular mechanism of angiotensin-converting enzyme inhibitory peptides from Larimichthys crocea titin显示文摘This study aimed to identify novel ACEI peptides from Larimichthys crocea titin using in silico approaches and to clarify the molecular interaction mechanism.The hydrolyzed peptides of titin were compared with known ACEI peptides in the AHTPDB and BIOPEP-UWM database.Furthermore,peptides were evaluated for their solubility,ADMET properties,ΔG(kcal/mol)values,and in vitro ACEI activity.Molecular mechanism of ACE-peptide was performed by molecular interactions and binding orientation study.The results revealed that IC50 values of Trp-Ala-Arg(WAR)and Trp-Gln-Arg(WQR)were(31.2±0.8)and(231.33±0.02)mol/L,respectively.The docking interactions result suggested that ACE-WAR and ACEWQR complexes have same binding site,including the residues LYS511,TYR520,TYR523,HIS353,and HIS513.Molecular docking of two tripeptides WAR and WQR with ACE studies predicted their binding site and clarified the interaction between ACE and its inhibitors.The molecular docking data are consistent with the ACE inhibitory activity of the studied peptides.The results showed that Larimichthys crocea titin may be a valuable source for developing nutraceutical food.Yue Fan Zhipeng Yu Wenzhu Zhao Long Ding Fuping Zheng Jianrong Li Jingbo Liu 2020Food Science and Human Wellness2020,9,3:5
7In situ generation of Li_(3)N concentration gradient in 3D carbon-based lithium anodes towards highly-stable lithium metal batteries显示文摘The uncontrolled dendrite growth of lithium metal anodes(LMAs)caused by unstable anode/electrolyte interface and uneven lithium deposition have impeded the practical applications of lithium metal batteries(LMBs).Constructing a robust artificial solid electrolyte interphase(SEI)and regulating the lithium deposition behavior is an effective strategy to address these issues.Herein,a three-dimensional(3D)lithium anode with gradient Li_(3)N has been in-situ fabricated on carbon-based framework by thermal diffusion method(denoted as CC/Li/Li_(3)N).Density functional theory(DFT)calculations reveal that Li_(3)N can effectively promote the transport of Li^(+)due to the low energy barrier of Li^(+)diffusion.As expected,the Li_(3)N-rich conformal artificial SEI film can not only effectively stabilize the interface and avoid parasitic reactions,but also facilitate fast Li^(+)transport across the SEI layer.The anode matrix with uniformly distributed Li3N can enable homogenous deposition of Li,thus preventing Li dendrite propagation.Benefiting from these merits,the CC/Li/Li_(3)N anode achieves ultralong-term cycling for>1000 h at a current density of 2 m A cm^(-2)and dendrite-free Li deposition at an ultrahigh rate of 20 m A cm^(-2).Moreover,the full cells coupled with LiFePO4cathodes show extraordinary cycling stability for>300 cycles in liquidelectrolyte-based batteries and display a high-capacity retention of 96.7%after 100 cycles in solid-state cells,demonstrating the promising prospects for the practical applications of LMBs.Wenzhu Cao Weimin Chen Mi Lu Cheng Zhang Du Tian Liang Wang Faquan Yu 2023Journal of Energy Chemistry2023,,1:4

9Induction of Tumor Cell Apoptosis via Fas/DR5显示文摘The apoptosis inducing effects on tumor cell lines MGC803, BEL7402 and HL60 by Fas ligand and anti-human DR5 monoclonal antibodies (anti-DR5 mAb) and the underlying mechanism was studied. Fas/DR5 mRNA was detected by RT-PCR. Cytotoxicity exerted by FasL/anti-DR5 mAb on tumor cell lines was measured by MTT assay and the induced apoptosis was determined by agarose gel electrophoresis. Flow cytometry was employed to analyze the mode of cell death. The mRNA expression of DR5 in MGC803 and BEL7402 cells after giving anti-DR5 mAb was up-regulated compared with control group, while it was down-regulated in HL60 cells in the same condition. The mRNA expression of Fas in HL60 was higher after giving FasL compared with control group, while it was lower in MGC803 and BEL7402. MGC803 and BEL7402 were sensitive to anti-DR5 mAb but partially to FasL, and HL60 was sensitive to FasL but less sensitive to anti-DR5 mAb. Apoptosis induced by Fas ligand and anti-DR5 mAb vary among tumor cell lines. 13Effect of strain on DRX of 7050 aluminum alloy during extrusion at high temperature显示文摘Extrusion experiments with different filling contents at the temperature of 440 ℃ were conducted for solution treated 7050 aluminum alloy.The microstructure of typical regions with different filler contents were described qualitatively and characterized quantitatively by electron backscattered diffraction technique(EBSD) and TEM.Fraction of deformed structure decrease continuously since ε=0.72.Second phase particles distribute dispersively and interact with dislocations when ε=0.66,which delays the occurrence of DRX.As the strain increases,particles dissolve into the matrix,and their number reduces rapidly.The influence of particles on dislocation accumulation is impaired,so dislocations accumulate on the subgrain boundaries continuously,and lead to the transformation of subgrain boundaries into high angle grain boundaries(HAGBs).The continuous dynamic recrystallization(CDRX) occurrs at the strain of 0.81~0.85.At ε=0.92,CDRX convert rapidly with the increase of HAGBs fraction.But the size of CDRX grains is deduced to be minimum 3.3 μm by theoretical analysis,and it is obtained from experimental result that the maximal size of CDRX is 5.3 μm.ZHU Danyang,ZHEN Liang,HU Wenhao,CHEN Junzhou,CUI Yuexian,and SHAO Wenzhu School of Materials Science and Engineering,Harbin Institute of Technology,Harbin 150001,China 2007Rare Metals2007,26,S1:3
14Effects of brewers' spent grain protein hydrolysates on gas production, ruminal fermentation characteristics, microbial protein synthesis and microbial community in an artificial rumen fed a high grain diet显示文摘Background: Brewers' spent grain(BSG) typically contains 20% – 29% crude protein(CP) with high concentrations of glutamine, proline and hydrophobic and non-polar amino acid, making it an ideal material for producing valueadded products like bioactive peptides which have antioxidant properties. For this study, protein was extracted from BSG, hydrolyzed with 1% alcalase and flavourzyme, with the generated protein hydrolysates(AlcH and FlaH)showing antioxidant activities. This study evaluated the effects of AlcH and FlaH on gas production, ruminal fermentation characteristics, nutrient disappearance, microbial protein synthesis and microbial community using an artificial rumen system(RUSITEC) fed a high-grain diet.Results: As compared to the control of grain only, supplementation of FlaH decreased(P < 0.01) disappearances of dry matter(DM), organic matter(OM), CP and starch, without affecting fibre disappearances;while AlcH had no effect on nutrient disappearance. Neither AlcH nor FlaH affected gas production or VFA profiles, however they increased(P < 0.01) NH_3-N and decreased(P < 0.01) H_2 production. Supplementation of FlaH decreased(P < 0.01)the percentage of CH_4 in total gas and dissolved-CH_4(dCH_4) in dissolved gas. Addition of monensin reduced(P < 0.01) disappearance of nutrients, improved fermentation efficiency and reduced CH_4 and H_2 emissions.Total microbial nitrogen production was decreased(P < 0.05) but the proportion of feed particle associated(FPA) bacteria was increased with FlaH and monensin supplementation. Numbers of OTUs and Shannon diversity indices of FPA microbial community were unaffected by AlcH and FlaH;whereas both indices were reduced(P < 0.05) by monensin. Taxonomic analysis revealed no effect of AlcH and FlaH on the relative abundance(RA) of bacteria at phylum level, whereas monensin reduced(P < 0.05) the RA of Firmicutes and Bacteroidetes and enhanced Proteobacteria. Supplementation of FlaH enhanced(P < 0.05) the RA of genus Prevotella, reduced Selenomonas, Shuttleworthia, Bifidobacterium and Dialister as compared to control;monensin reduced(P < 0.05) RA of genus Prevotella but enhaced Succinivibrio.Conclusions: The supplementation of FlaH in high-grain diets may potentially protect CP and starch from ruminal degradation, without adversely affecting fibre degradation and VFA profiles. It also showed promising effects on reducing CH_4 production by suppressing H_2 production. 15Effect of commercial slow-release urea product on in vitro rumen fermentation and ruminal microbial community using RUSITEC technique显示文摘Background:The objectives of this study were to determine the effect of commercial slow-release urea(SRU)on in vitro fermentation characteristics,nutrient digestibility,gas production,microbial protein synthesis and bacterial community using a rumen simulation technique(RUSITEC).The experiment was a completely randomized design with four treatments and four replications of each treatment.Treatments were:control diet(no SRU addition),control diet plus 0.28%SRU(U28),or plus 0.56%SRU(U56),and control diet that was modified substituting a part of soybean meal equivalent to 0.35%SRU(MU35;dry matter[DM]basis).The experiment consisted of 8 d of adaptation and 7 d of data and sample collection.Rumen inoculum was obtained from three ruminally fistulated Angus cows fed the same diet to the substrate incubated.Results:Digestibility of DM,organic matter(OM),crude protein(CP),fibre and starch was not affected,but daily production of gas(P<0.07)and methane(P<0.05)was quadratically increased with increasing SRU supplementation.The increase of SRU addition did not affect fermentation pH and total volatile fatty acid(VFA)production,whereas linearly(P<0.01)decreased proportion of propionate,and linearly(P<0.01)increased acetate to propionate ratio and ammonia nitrogen(N)concentration.The microbial N efficiency was also linearly(P<0.03)improved with increasing supplementation of SRU.In comparison with control diet,the dietary substitution of SRU for part of soybean meal increased(P<0.05)the digestibility of DM,OM and CP and decreased(P<0.02)the total gas production.The total VFA production and acetate to propionate ratio did not differ between control and MU35,whereas the proportion of butyrate was lower(P<0.05)and that of branched-chain VFA was greater(P<0.05)with MU35 than control diet.Total and liquid-associated microbial N production as well as ammonia N concentration were greater(P<0.03)with MU35 than control diet.Observed operational taxonomic units(OTUs),Shannon diversity index,and beta diversity of the microbial community did not differ among treatments.Taxonomic analysis revealed no effect of adding SRU on the relative abundance of bacteria at the phylum level,while at the genus level,the beneficial impact of SRU on relative abundance of Rikenellaceae and Prevotellaceae in feed particleassociated bacteria,and the abundance of Roseburia in liquid associate bacteria was greater(P<0.05)with MU35.Conclusions:Supplementation of a dairy cow diet with SRU showed potential of increase in ammonia N concentration and microbial protein production,and change fermentation pattern to more acetate production.Adding SRU in dairy cow diet also showed beneficial effect on improving digestibility of OM and fibre.The results suggest that SRU can partially substitute soybean meal in dairy cow diet to increase microbial protein production without impairing rumen fermentation.Yongmei Guo Ling Xiao Long Jin Sumei Yan Dongyan Niu Wenzhu Yang 2022Journal of Animal Science and Biotechnology2022,13,5:3
16Biological evaluation and interaction mechanism of beta-site APP cleaving enzyme 1 inhibitory pentapeptide from egg albumin显示文摘Inhibition of beta-site APP cleaving enzyme1(BACE1)is one of the most promising therapeutic approaches for Alzheimer’s disease.To find natural products for the treatment of Alzheimer’s disease,absorption,distribution,metabolism,excretion and toxicity(ADMET)properties and in vitro BACE1 inhibitory activity of the peptides isolated from egg albumin were evaluated.Then,molecular docking and molecular dynamics simulation were used to explain the molecular mechanism of the interactions between BACE1 and peptides.The IC50 value of peptide KLPGF,with satisfactory ADMET properties,against BACE1 was(8.30±0.56)mmol/L.Molecular docking revealed that KLPGF contacted with the residues of BACE1’s active sites through twelve hydrogen bonds interactions,two hydrophobic interactions,one electrostatic interaction,and two Pi-cation interactions.The 5 ns molecular dynamics simulations confirmed that the structure of KLPGF with BACE1 was stable.Peptide KLPGF contacted the residues Lys321,Asp228,and Asn233 with stable hydrogen bonds.KLPGF may be a potential anti-BACE1 candidate.Zhipeng Yu Sijia Wu Wenzhu Zhao Long Ding David Shiuan Fuping Zheng Jianrong Li Jingbo Liu 2020Food Science and Human Wellness2020,9,2:2
17Ages and Hf isotopes of detrital zircons from the Permian strata in the engbatu area(Inner Mongolia) and tectonic implications显示文摘The Central Asian Orogenic Belt(CAOB) was built up through protracted accretion and collision of a variety of terranes/micro-continents during Neoproterozoice Mesozoic time. To understand potential links among Paleozoic subduction and accretionary processes that were operative during the development of the southeastern CAOB, we conducted a combined U-Pb and Hf-isotope analysis of detrital zircons from previously defined Devonian, Carboniferous and Early Permian strata in the Bengbatu area,Inner Mongolia. Detrital zircons from(meta-) sandstones in these strata commonly yield major Paleozoic age populations at ca. 300-261 Ma, 351-300 Ma and 517-419 Ma, and also give several Precambrian ages that range from 2687 Ma to 544 Ma. The youngest ages redefine the deposition of all these strata to be in the Middle Permian(Wordiane Capitanian) or later, much younger than previously considered.These ages, coupled with regional magmatic records, support an interpretation of most surrounding areas as possible detritus sources, including the Mongolian arcs to the north, the Northern Accretionary Orogen to the south, and the intervening Erenhote Hegenshan Ophiolite Belt. Zircons with magmatic ages of ca. 500-350 Ma and ca. 300-261 Ma display a large range of εHf(t) values(-13.97 to +15.31),whereas ca. 350-300 Ma zircons are dominated by positive εHf(t) values(+0.14 to +16.00). These results support the occurrence of two significant shifts of the zircon εHf(t) values, which has tectonic implications for the understanding of the Carboniferouse Permian evolution of the southeastern CAOB. A marked shift from mixed to positive zircon εHf(t) values at 350 -330 Ma likely manifests the incipient opening of the Hegenshan Ocean, due to the slab rollback of the subducting Paleo-Asian Oceanic lithosphere. Another shift from positive to mixed zircon εHf(t) values at ca. 300 Ma likely corresponds to a tectonic switch from syn-orogenic subduction-related to post-orogenic extensional setting, genetically related to the tectonic collapse of a formerly overthickened crust.Bing Xu Guochun Zhao Jianhua Li Dongxing Liu Bo Wang Yigui Han Paul R. 19RGD-FasL Induces Apoptosis of Pituitary Adenoma Cells显示文摘This study was to investigate the cytotoxic effects on pituitary adenoma cell lines GH3/MMQ/AtT20 induced by RGD-FasL and the underlying mechanism. Fas/DcR3 mRNAs were detected by RT-PCR and their surface expressions were measured by flow cytometry. Cytotoxicity exerted by RGD-FasL on tumor cells was measured with MTT assay and the induced apoptosis was determined by agarose gel electrophoresis. The cell cycle and apoptosis was assessed by flow cytometry with PI staining. The expressions of caspase8/9/3,Bcl-2,RANKL and JNK2 were detected by Western blotting. Approximately 13.7% of GH3 cells,25.5% of MMQ cells,22.2% of AtT20 cells express Fas,while 23.9% of GH3 cells,24.1% of MMQ cells,4.6% of AtT20 cells express DcR3. The cytotoxic effects of FasL/RGD-FasL on tumor cells were all taken in a dose-dependent manner. Cell lines MMQ/AtT20 showed the same sensitivity to RGD-FasL as to FasL,while cell line GH3 was less sensitive to RGD-FasL. The cell cycle analysis indicated that RGD-FasL could inhibit cells in G0/G1 phase and G2/M phase. In MMQ and AtT20 cells treated with RGD-FasL,the AI was not significantly different from that treated with FasL,while in GH3 cells treated with RGD-FasL,the AI was lower than that treated with FasL. The expressions of caspase-8/9/3,RANKL and JNK2 were increased while that of Bcl-2 was decreased after treatment with RGD-FasL,suggesting that RGD-FasL induces apoptosis through caspase activation. 20Improvement of cardiac function by hepatocyte growth factor via intracoronary transfection in the swine myocardial infarction model显示文摘Objective:To study the amelioration effect of Adenovirus5-mediated human hepatocyte growth factor(Ad5-HGF)on postinfarction heart failure in the swine myocardial infarction model.Methods:Twelve SuZhong young swine were randomly divided into 2 groups with 6 swine in each group:Ad5-HGF-treated group and null-Ad5 group.Four weeks after ligation at left anterior descending coronary artery in swine hearts,Ad5-HGF was transferred to the swine myocardium.Simultaneously,Gated myocardial perfusion imaging was performed to evaluate cardiac perfusion and heart function.After three weeks,Gated myocardial perfusion imaging was performed again,then the hearts were harvested and sectioned to examine the expression of HGF through ELISA.Results:High expression of human HGF was observed in the myocardium of Ad5-HGF-treated group.From 4 weeks to 7 weeks after operation,Left ventricular ejection fraction was increased in Ad5-HGF-treated group.The improvement in LVEF was greater in Ad5-HGF-treated group than that in null-Ad5 group at 7 weeks after operation.Cardiac perfusion was significantly improved in the Ad5-HGF-treated group.Conclusion:High expression of human HGF was observed in the myocardium through intracoronary transfection,which suggests that HGF can ameliorate heart function in swine with postinfarction heart failure.Wei Wang Zhijian Yang Dongchao Ma Shunlin Xu Yourong Zhang Yuqing Zhang Liansheng Wang Bo Chen Kejiang Cao Wenzhu Ma 2006Journal of Nanjing Medical University2006,20,4:2
