1Particle size distribution and characteristics of heavy metals in road-deposited sediments from Beijing Olympic Park显示文摘Due to rapid urbanization and industrialization, heavy metals in road-deposited sediments(RDSs) of parks are emitted into the terrestrial, atmospheric, and water environment, and have a severe impact on residents' and tourists' health. To identify the distribution and characteristic of heavy metals in RDS and to assess the road environmental quality in Chinese parks, samples were collected from Beijing Olympic Park in the present study. The results indicated that particles with small grain size(<150 μm) were the dominant fraction. The length of dry period was one of the main factors affecting the particle size distribution, as indicated by the variation of size fraction with the increase of dry days. The amount of heavy metal(i.e., Cu, Zn, Pb and Cd) content was the largest in particles with small size(<150 μm) among all samples. Specifically, the percentage of Cu, Zn, Pb and Cd in these particles was 74.7%, 55.5%, 56.6% and 71.3%, respectively.Heavy metals adsorbed in sediments may mainly be contributed by road traffic emissions. The contamination levels of Pb and Cd were higher than Cu and Zn on the basis of the mean heavy metal contents. Specifically, the geoaccumulation index(I geo) decreased in the order:Cd>Pb>Cu>Zn. This study analyzed the mobility of heavy metals in sediments using partial sequential extraction with the Tessier procedure. 2Finding of high-pressure mafic granulites in the Amdo basement,central Tibet显示文摘High-pressure mafic granulites with a peak mineral assemblage of garnet + clinopyroxene + rutile + quartz were found in the Amdo basement,central Tibet. Two kinds of symplectites were identified that are composed of orthopyroxene + plagioclase ± spinel and hornblende + plagioclase around garnet,which were interpreted to develop during the retrogressing stages in the granulites. P-T estimates suggested that peak metamorphic conditions were about 860-920°C and 1.46-1.56 GPa,which retrogressed from post-peak phase at 820-890°C and 0.88-1.15 GPa to amphibolite facies at 550-670°C and 0.52-0.65 GPa. These three stages define a clockwise P-T path with near-isothermal decompression and cooling following the peak high-pressure metamorphism. This suggests that the Amdo granulites underwent an initial subduction to a deep crustal level of ~50 km and then were rapidly exhumed to a shallow crustal level (~20 km). 3Hydrogen peroxide modulates abscisic acid signaling in root growth and development in Arabidopsis显示文摘Exogenous abscisic acid(ABA)can inhibit root growth and promote formation of more root hairs in the root tip of Arabidopsis.However,the molecular mechanisms that underlie root ABA signaling are largely unknown.We report here that hydrogen peroxide(H2O2)reduces the root growth of wild type, and the phenotype of H2O2 on the root growth is similar to ABA response.Meanwhile ABA-induced changes in the morphology of root system can be partly reversed by ascorbic acid in wild type and abolished in NADPH oxidase defective mutant atrbohF and atrbohC.Further,ABA can induce H2O2 accumulation in the root cells and enhance transcription level of OXI1,which is necessary for many more AOS-dependent processes such as root hair growth in Arabidopsis.Our results suggest that H2O2 as an important signal molecule is required for the ABA-regulated root growth and development in Arabidopsis.BAI Ling ZHOU Yun ZHANG XiaoRan SONG ChunPeng Cao MingQing 2007Chinese Science Bulletin2007,52,8:12

5Olmutinib(HM61713) reversed multidrug resistance by inhibiting the activity of ATPbinding cassette subfamily G member 2 in vitro and in vivo显示文摘Overexpressing of ATP-binding cassette(ABC) transporters is the essential cause of multidrug resistance(MDR), which is a significant hurdle to the success of chemotherapy in many cancers.Therefore, inhibiting the activity of ABC transporters may be a logical approach to circumvent MDR.Olmutinib is an epidermal growth factor receptor(EGFR) tyrosine kinase inhibitor(TKI), which has been approved in South Korea for advanced EGFR T790 M-positive non-small cell lung cancer(NSCLC). Here,we found that olmutinib significantly increased the sensitivity of chemotherapy drug in ABCG2-overexpressing cells. Furthermore, olmutinib could also increase the retention of doxorubicin(DOX) and rhodamine 123(Rho 123) in ABC transporter subfamily G member 2(ABCG2)-overexpressing cells. In addition, olmutinib was found to stimulate ATPase activity and inhibit photolabeling of ABCG2 with [^(125) I]-iodoarylazidoprazosin(IAAP). However, olmutinib neither altered ABCG2 expression at protein and m RNA levels nor blocked EGFR, Her-2 downstream signaling of AKT and ERK. Importantly,olmutinib enhanced the efficacy of topotecan on the inhibition of S1-MI-80 cell xenograft growth. All the results suggest that olmutinib reverses ABCG2-mediated MDR by binding to ATP bind site of ABCG2 and increasing intracellular chemotherapeutic drug accumulation. 6Characterization of flavor fingerprinting of red sufu during fermentation and the comparison of volatiles of typical products显示文摘Traditional Chinese red sufu is a popular condiment,and typical flavor is an important indicator of sensory qualities in commercial products.In this study,the volatile aroma analysis of Wangzhihe red sufu(WRS)from different fermentation stages and four typical red sufu products from different locations was carried out by gas chromatography-mass spectrometry(GC–MS),gas chromatography-mass spectrometry/olfactory(GC–MS/O),electronic nose(E-nose)and sensory evaluation.Results showed that 106 volatile compounds were identified in Wangzhihe red sufu during fermentation process,in which phenolics and alcohols were dominant at molded and salted phetze stages,while esters and alcohols became predominate at post-fermentation stage.The volatile aroma substances varied at each fermentation stage.Furthermore,86 volatiles,including 16 aroma-active compounds,were detected in four typical red sufu products.The multivariate analysis results showed the difference in samples from different fermentation stages and typical red sufu products according to GC–MS and E-nose analysis.Combined with flavor omics,discriminant model was established for effective discrimination of samples from different fermentation stages and locations,as well as references to sufu maturity extent.The study presented a new strategy for quality evaluation of red sufu,which could be the supplement to quality evaluation standards.Peng Wang Xiaoran Ma Wenping Wang Dandan Xu Xin Zhang Jian Zhang Yong Sun 2019Food Science and Human Wellness2019,8,4:7
7A PKS gene, pks-1, is involved in chaetoglobosin biosynthesis, pigmentation and sporulation in Chaetomium globosum显示文摘Chaetomium globosum is one of the most common fungi in nature. It is best known for producing chaetoglobosins; however, the molecular basis of chaetoglobosin biosynthesis is poorly understood in this fungus. In this study, we utilized RNA interference (RNAi) to characterize a polyketide synthase gene, pks-1, in C. globosum that is involved in the production of chaetoglobosin A. When pks-1 was knocked down by RNAi, the production of chaetoglobosin A dramatically decreased. Knock-down mutants also displayed a pigment-deficient phenotype. These results suggest that the two polyketides, melanin and chaetoglobosin, are likely to share common biosynthetic steps. Most importantly, we found that pks-1 also plays a critical role in sporulation. The silenced mutants of pks-1 lost the ability to produce spores. We propose that polyketides may modulate cellular development via an unidentified action. We also suggest that C. globosum pks-1 is unique because of its triple role in melanin formation, chaetoglobosin biosynthesis and sporulation. 8Integrin-interacting protein Kindlin-2 induces mammary tumors in transgenic mice显示文摘Kindlin-2, an integrin-interacting protein, regulates breast cancer progression. However, currently, no animal model to study the role of Kindlin-2 in the carcinogenesis of mammary gland is available. We established a Kindlin-2 transgenic mouse model using a mammary gland-specific promoter, mammary tumor virus(MMTV) long terminal repeat(LTR). Kindlin-2 was overexpressed in the epithelial cells of the transgenic mice. The mammary gland ductal trees were found to grow faster in MMTV-Kindlin-2 transgenic mice than in control mice during puberty. Kindlin-2 promoted mammary gland growth as indicated by more numerous duct branches and larger lumens, and more alveoli were formed in the mammary glands during pregnancy under Kindlin-2 overexpression. Importantly, mammary gland-specific expression of Kindlin-2 induced tumor formation at the age of 55 weeks on average. Additionally, the levels of estrogen receptor and progesterone receptor were decreased, whereas human epidermal growth factor receptor 2 and β-catenin were upregulated in the Kindlin-2-induced mammary tumors. 10Combined peripheral natural killer cell and circulating tumor cell enumeration enhance prognostic efficiency in patients with metastatic triple-negative breast cancer显示文摘Objective:Triple-negative breast cancer(TNBC)is a heterogeneous disease with poor prognosis.Circulating tumor cells(CTCs)are a promising predictor for breast cancer prognoses but their reliability regarding progression-free survival(PFS)is controversial.We aim to verify their predictive value in TNBC.Methods:In present prospective cohort study,we used the Pep@MNPs method to enumerate CTCs in baseline blood samples from 75 patients with TNBC(taken at inclusion in this study)and analyzed correlations between CTC numbers and outcomes and other clinical parameters.Results:Median PFS was 6.0(range:1.0–25.0)months for the entire cohort,in whom we found no correlations between baseline CTC status and initial tumor stage(P=0.167),tumor grade(P=0.783)or histological type(P=0.084).However,among those getting first-line treatment,baseline CTC status was positively correlated with ratio of peripheral natural killer(NK)cells(P=0.032),presence of lung metastasis(P=0.034)and number of visceral metastatic site(P=0.037).Baseline CTC status was predictive for PFS in first-line TNBC(P=0.033),but not for the cohort as a whole(P=0.118).This prognostic limitation of CTC could be ameliorated by combining CTC and NK cell enumeration(P=0.049).Conclusions:Baseline CTC status was predictive of lung metastasis,peripheral NK cell ratio and PFS in TNBC patients undergoing first-line treatment.We have developed a combined CTC-NK enumeration strategy that allows us to predict PFS in TNBC without any preconditions.Xiaoran Liu Ran Ran Bin Shao Hope S.Rugo Yanlian Yang Zhiyuan Hu Zewen Wei Fengling Wan Weiyao Kong Guohong Song Hanfang Jiang Xu Liang Ruyan Zhang Ying Yan Guobing Xu Huiping Li 2018Chinese Journal of Cancer Research2018,30,3:4
11PhaseⅠdose-escalation and expansion study of PARP inhibitor,fluzoparib(SHR3162),in patients with advanced solid tumors显示文摘Objective:Fluzoparib(SHR3162)is a novel,potent poly(ADP-ribose)polymerases(PARP)1,2 inhibitor that showed anti-tumor activity in xenograft models.We conducted a phaseⅠ,first-in-human,dose-escalation and expansion(D-Esc and D-Ex)trial in patients with advanced solid cancer.Methods:This was a 3+3 phaseⅠD-Esc trial with a 3-level D-Ex at 5 hospitals in China.Eligible patients for DEsc had advanced solid tumors refractory to standard therapies,and D-Ex enrolled patients with ovarian cancer(OC).Fluzoparib was administered orally once or twice daily(bid)at 11 dose levels from 10 to 400 mg/d.Endpoints included dose-finding,safety,pharmacokinetics,and antitumor activity.Results:Seventy-nine patients were enrolled from March,2015 to January,2018[OC(47,59.5%);breast cancer(BC)(16,20.3%);colorectal cancer(8,10.1%),other tumors(8,10.1%)];48 patients were treated in the D-Esc arm and 31 in the D-Ex arm.The maximum tolerated dose(MTD)was 150 mg bid,with a half-life of 9.14 h.Grade 3/4 adverse events included anemia(7.6%)and neutropenia(5.1%).The objective response rate(ORR)was 30%(3/10)in patients with platinum-sensitive OC and 7.7%(1/13)in patients with BC.Among patients treated with fluzoparib≥120 mg/d,median progression-free survival(m PFS)was 7.2[95%confidence interval(95%CI),1.8-9.3]months in OC,9.3(95%CI,7.2-9.3)months in platinum-sensitive OC,and 3.5(range,2.0-28.0)months in BC.In patients with germline BC susceptibility gene mutation(g BRCAMut)(11/43 OC;2/16 BC),m PFS was 8.9 months for OC(range,1.0-23.2;95%CI,1.0-16.8)and 14 and 28 months for BC(those two patients both also had somatic BRCAMut).Conclusions:The MTD of fluzoparib was 150 mg bid in advanced solid malignancies.Fluzoparib demonstrated single-agent antitumor activity in BC and OC,particularly in BRCAMut and platinum-sensitive OC.Huiping Li Rongrui Liu Bin Shao Ran Ran Guohong Song Ke Wang Yehui Shi Jihong Liu Wenjing Hu Fu Chen Xiaoran Liu Gairong Zhang Chuanhua Zhao Ru Jia Quanren Wang Hope S.Rugo Yifan Zhang Guangze Li Jianming Xu 2020Chinese Journal of Cancer Research2020,32,3:4
12Insights into Convective-scale Predictability in East China: Error Growth Dynamics and Associated Impact on Precipitation of Warm-Season Convective Events显示文摘This study investigated the regime-dependent predictability using convective-scale ensemble forecasts initialized with different initial condition perturbations in the Yangtze and Huai River basin(YHRB)of East China.The scale-dependent error growth(ensemble variability)and associated impact on precipitation forecasts(precipitation uncertainties)were quantitatively explored for 13 warm-season convective events that were categorized in terms of strong forcing and weak forcing.The forecast error growth in the strong-forcing regime shows a stepwise increase with increasing spatial scale,while the error growth shows a larger temporal variability with an afternoon peak appearing at smaller scales under weak forcing.This leads to the dissimilarity of precipitation uncertainty and shows a strong correlation between error growth and precipitation across spatial scales.The lateral boundary condition errors exert a quasi-linear increase on error growth with time at the larger scale,suggesting that the large-scale flow could govern the magnitude of error growth and associated precipitation uncertainties,especially for the strong-forcing regime.Further comparisons between scale-based initial error sensitivity experiments show evident scale interaction including upscale transfer of small-scale errors and downscale cascade of larger-scale errors.Specifically,small-scale errors are found to be more sensitive in the weak-forcing regime than those under strong forcing.Meanwhile,larger-scale initial errors are responsible for the error growth after 4 h and produce the precipitation uncertainties at the meso-β-scale.Consequently,these results can be used to explain underdispersion issues in convective-scale ensemble forecasts and provide feedback for ensemble design over the YHRB.Xiaoran ZHUANG Jinzhong MIN Liu ZHANG Shizhang WANG Naigeng WU Haonan ZHU 2020Advances in Atmospheric Sciences2020,37,8:3
13Os isotopic evidence for a carbonaceous chondritic mantle source for the Nagqu ophiolite from Tibet and its implications显示文摘Early-crystallizing chromian spinel(Cr-spinel) in the Nagqu ophiolite has high Os and low Re contents,and it is resistant to alteration during serpentinization,weathering and metamorphism.The chemical composition of primitive magma is preserved in Cr-spinel,which makes it suitable for determining the initial Os-isotope composition of the mantle source.This study presents Cr-spinel Os isotopes and zircon U-Pb ages for cumulate dunite and gabbro,respectively,in the same cumulate section of the ophiolite at Nagqu in Tibet.The results shed light on the formation and evolution of lithospheric mantle.The Nagqu ophiolite is located in the central part of the Bangong-Nujiang suture zone.It is a remnant of the Neotethyan oceanic crust,and contains cumulate dunite and gabbro.Zircon from the gabbro yielded a weighted mean 206 Pb/238 U age of 183.7±1 Ma.Cr-spinel exhibits Os values of 0.2 to 0.3,suggesting that the mantle source for the dunite is similar to that of carbonaceous chondrites.Thus,the Tibetan lithosphere is primarily a relic of Tethyan oceanic lithosphere,which has formed by the transformation of the normal asthenospheric mantle in the Mesozoic.This is the first study to combine the spinel Os isotopes with accurate zircon U-Pb ages to constrain the geochemical characteristics of the mantle source for the ophiolite.HUANG QiShuai SHI RenDeng LIU DeLiang ZHANG XiaoRan FAN ShuaiQuan DING Lin 2013Chinese Science Bulletin2013,58,1:3
14Ages and Hf isotopes of detrital zircons from the Permian strata in the engbatu area(Inner Mongolia) and tectonic implications显示文摘The Central Asian Orogenic Belt(CAOB) was built up through protracted accretion and collision of a variety of terranes/micro-continents during Neoproterozoice Mesozoic time. To understand potential links among Paleozoic subduction and accretionary processes that were operative during the development of the southeastern CAOB, we conducted a combined U-Pb and Hf-isotope analysis of detrital zircons from previously defined Devonian, Carboniferous and Early Permian strata in the Bengbatu area,Inner Mongolia. Detrital zircons from(meta-) sandstones in these strata commonly yield major Paleozoic age populations at ca. 300-261 Ma, 351-300 Ma and 517-419 Ma, and also give several Precambrian ages that range from 2687 Ma to 544 Ma. The youngest ages redefine the deposition of all these strata to be in the Middle Permian(Wordiane Capitanian) or later, much younger than previously considered.These ages, coupled with regional magmatic records, support an interpretation of most surrounding areas as possible detritus sources, including the Mongolian arcs to the north, the Northern Accretionary Orogen to the south, and the intervening Erenhote Hegenshan Ophiolite Belt. Zircons with magmatic ages of ca. 500-350 Ma and ca. 300-261 Ma display a large range of εHf(t) values(-13.97 to +15.31),whereas ca. 350-300 Ma zircons are dominated by positive εHf(t) values(+0.14 to +16.00). These results support the occurrence of two significant shifts of the zircon εHf(t) values, which has tectonic implications for the understanding of the Carboniferouse Permian evolution of the southeastern CAOB. A marked shift from mixed to positive zircon εHf(t) values at 350 -330 Ma likely manifests the incipient opening of the Hegenshan Ocean, due to the slab rollback of the subducting Paleo-Asian Oceanic lithosphere. 15Electropolymerization of cobalt porphyrins and corroles for the oxygen evolution reaction显示文摘Developing large-scale electrocatalysts using molecular complexes for the oxygen evolution reaction(OER)is of great importance. Herein, four cobalt porphyrins and corroles are deposited on electrode substrates using a simple and fast electropolymerization method. Our results showed that Co-1-P@CC, formed by electropolymerizing Co tetrakis(p-N-pyrrolylphenyl)porphyrin(Co-1-P) on carbon cloth(CC), is the most active OER catalyst in the examined Co porphyrins and corroles in alkaline aqueous solutions by displaying an onset overpotential of 380 m V. 17Recent advances on interface engineering of perovskite solar cells显示文摘Lead halide perovskite solar cells(PSCs)have been rapidly developed in the past decade.Owing to its excellent power conversion efficiency with robust and low-cost fabrication,perovskite quickly becomes one of the most promising candidates for the next-generation photovoltaic technology.With the development of PSCs,the interface engineering has witnessed its increasingly critical role in maximizing the device performance as well as the long-term stability,because the interfaces in PSCs are closely correlated with the defect management,carrier dynamics and surface passivation.This review focuses on interfacial modification between the perovskite active layer and the charge transport layer,as well as the recent advances on high-efficiency and stable PSCs driven by interface engineering strategies.The contributing roles of interface engineering in terms of defect passivation,inhibiting ion migration,optimization of energy band alignment and morphological control are discussed.Finally,based on the latest progress and advances,strategies and opportunities for the future research on interface engineering for PSCs are proposed to promote the development of perovskite photovoltaic technology.Wenjing Yu Xiaoran Sun Mu Xiao Tian Hou Xu Liu Bolin Zheng Hua Yu Meng Zhang Yuelong Huang Xiaojing Hao 2022Nano Research2022,15,1:2
18Biopsy-free in vivo virtual histology of skin using deep learning显示文摘An invasive biopsy followed by histological staining is the benchmark for pathological diagnosis of skin tumors.The process is cumbersome and time-consuming,often leading to unnecessary biopsies and scars.Emerging noninvasive optical technologies such as reflectance confocal microscopy(RCM)can provide label-free,cellular-level resolution,in vivo images of skin without performing a biopsy.Although RCM is a useful diagnostic tool,it requires specialized training because the acquired images are grayscale,lack nuclear features,and are difficult to correlate with tissue pathology.Here,we present a deep learning-based framework that uses a convolutional neural network to rapidly transform in vivo RCM images of unstained skin into virtually-stained hematoxylin and eosin-like images with microscopic resolution,enabling visualization of the epidermis,dermal-epidermal junction,and superficial dermis layers.The network was trained under an adversarial learning scheme,which takes ex vivo RCM images of excised unstained/label-free tissue as inputs and uses the microscopic images of the same tissue labeled with acetic acid nuclear contrast staining as the ground truth.We show that this trained neural network can be used to rapidly perform virtual histology of in vivo,label-free RCM images of normal skin structure,basal cell carcinoma,and melanocytic nevi with pigmented melanocytes,demonstrating similar histological features to traditional histology from the same excised tissue.This application of deep learning-based virtual staining to noninvasive imaging technologies may permit more rapid diagnoses of malignant skin neoplasms and reduce invasive skin biopsies.Jingxi Li Jason Garfinkel Xiaoran Zhang Di Wu Yijie Zhang Kevin de Haan Hongda Wang Tairan Liu Bijie Bai Yair Rivenson Gennady Rubinstein Philip O.Scumpia Aydogan Ozcan 2021Light(Science & Applications)2021,10,12:1
