1Dynamics of Microbial Communities in Bulk and Developing CucumberRhizosphere Soil显示文摘The microbial population dynamics in bulk and developing cucumber rhizospheres were studiedby cultivation and cultivation-independent approach based on directly extracted DNA toprovide baseline data. Soil and rhizosphere samples were taken from tested field 2, 4, 7 and10 weeks after the seeds were planted, which was positively related to the corresponding dateof cucumber growth stages. The plate culture amount showed that total number of bacteria,fungi and actinomyces began to rise when cucumber planted and quickly reached peak at seedlingor blossom period, but decreased slightly later. Bacterial population in rhizosphere washigher by comparison with that of counterpart except for seedling and flowering stages, butthe shift trend of them were quite similar all the time. Nitrogen fixating, nitrobacter andammonifying bacteria showed the same change tendency in population as bacteria and actinomycesdid, however, cellulose-decomposing bacteria had the contrary rhizosphere effect as cucumberdeveloped. Data revealed that positive relevance existed between the dominant rhizospheremicrobe population and cucumber development. PCR was employed to amplify the V3 region of 16SrDNA, then the products were subjected to denaturing gradient gel electrophoresis(DGGE). DGGEprofile indicated that a few microbe species lived stable in farmland soil, but some wereinfluenced by population due to cucumber roots growth. Significant difference was observed inbulk and rhizosphere soils especially for the seedling and flowering samples. Few prominentbands in DGGE patterns, which displayed stronger or less illumination, means the representativebacteria had great population variation in that period. These phenomena indicated thatflowering cucumber heavily affected rhizosphere bacteria, or the bacteria, most probably theuncultured bacteria, functioned specially to cucumber blossom. 2Numerical Simulation of Temperature and Stress Fields of Ti-Alloy Thin-well Barrel during Quenching Process显示文摘The mathematical models have been established to describe the temperature and stress profiles in T16A14V cylinder during quenching. The residual stress and deformation of the workpiece can be predicted precisely based on ANSYS software. The simulated results show that the temperature of the divided element decreases faster at the edge than that at the internal of the workpiece during quenching from 1050 to 20°C. The largest temperature difference and dimension change in diameter are about 90°C and -0.935%, respectively. 3A chromosome-level genome assembly reveals that a bipartite gene cluster formed via an inverted duplication controls monoterpenoid biosynthesis in Schizonepeta tenuifolia显示文摘Biosynthetic gene clusters(BGCs)are regions of a genome where genes involved in a biosynthetic pathway are in proximity.The origin and evolution of plant BGCs as well as their role in specialized metabolism remain largely unclear.In this study,we have assembled a chromosome-scale genome of Japanese catnip(Schizonepeta tenuifolia)and discovered a BGC that contains multiple copies of genes involved in four adjacent steps in the biosynthesis of p-menthane monoterpenoids.This BGC has an unprecedented bipartite structure,with mirrored biosynthetic regions separated by 260 kilobases.This bipartite BGC includes identical copies of a gene encoding an old yellow enzyme,a type of flavin-dependent reductase.In vitro assays and virus-induced gene silencing revealed that this gene encodes the missing isopiperitenone reductase.This enzyme evolved from a completely different enzyme family to isopiperitenone reductase from closely related Mentha spp.,indicating convergent evolution of this pathway step.Phylogenomic analysis revealed that this bipartite BGC has emerged uniquely in the S.tenuifolia lineage and through insertion of pathway genes into a region rich in monoterpene synthases.The cluster gained its bipartite structure via an inverted duplication.The discovered bipartite BGC for p-menthane biosynthesis in S.tenuifolia has similarities to the recently described duplicated p-menthane biosynthesis gene pairs in the Mentha longifolia genome,providing an example of the convergent evolution of gene order.This work expands our understanding of plant BGCs with respect to both form and evolution,and highlights the power of BGCs for gene discovery in plant biosynthetic pathways.Chanchan Liu Samuel J.Smit Jingjie Dang Peina Zhou Grant T.Godden Zheng Jiang Wukun Liu Licheng Liui Wei Lin Jinao Duan Qinan Wu Benjamin R.Lichman 2023Molecular Plant2023,16,3:2
4Toxoplasmosis researches in China显示文摘Toxoplasmosis, caused by Toxoplasma gondii ( T. gondii ), an obligate intracellular parasite, is a globally distributed zoonosis. It is estimated that one third of population has been infected by this parasite worldwide. In some regions of Europe, the serum antibodies against T. gondii were detected in more than 80% of the examined population. In China, T. gondii was isolated for the first time from a rabbit in Fujian Province in 1954, 1 and the first case of human toxoplasmosis was reported in Jiangxi Province in 1964. 2 Later, in 1978, the first epidemic survey on toxoplasmosis was carried out in Guangxi Province. In 1982, the first national survey on toxoplasmosis supported by the National Natural Sciences Foundation of China was carried out and the result was published in 1988. Since that, many surveys on toxoplasmosis were reported in China. These results indicated that T. gondii infection in this country is very common and in some circumstances, the infection causes serious disease. For instance, T. gondii infection in pregnant women causes abortion, stillbirth, abnormal embryo and thus, it has been considered as the most commonly infectious agent of “TORCH syndrome” (TORCH syndrome refers to infection of a developing fetus or newborn by any of a group of infectious agents. “TORCH” is an acronym meaning toxoplasmosis, other agents, rubella, cytomegalovirus, and herpes simples). In immunocompromised patients, toxoplasmosis is also one of the most frequent complications. Because of the medical importance of T. gondii , more and more Chinese researchers are interested in this parasite. Aetiology Infected animals are the most important source for T. gondii infection, although humans can also be taken as the infective source for this disease particularly the vertical transmission from pregnant woman to fetus through the placenta. T. gondii widely distributes in variant animals from mammals to birds. 5Analysis of Valence Electron Structure in Compound Layer of Steel 42CrMo Plasma Nitrided with Rare Earth Addition显示文摘The valence electron structure (VES) in compound layer of steel plasma-nitrided at 560°C with rare earth (RE) addition was calculated based on the empirical electron theory (EET) of solids and molecules and BLD method. 8Micro-interfaces modulation by UV-ozone substrate treatment for MPEA vapor fluorescence detection显示文摘Drug abuse directly endangers human health and social security,hence its sensitive and rapid detection is vitally important.In recent years,organic film-based fluorescent sensing technology has attracted more and more attention in the detection of drugs and explosives due to its advantages of simple operation and rapid detection.For film-based fluorescent sensors,in addition to sensitive materials,the surface morphology of the film is also an important factor affecting the performance.In previous studies,the regulation of surface morphology mainly depends on concentration changes or complex templates.Here,a novel fluorescent polymer probe was designed and synthesized,and a simple and efficient ultraviolet(UV)-ozone substrate treatment method is used to adjust their surface morphology.The results show that film has an excellent fluorescence enhancement effect upon exposure to methylphenethylamine(MPEA,a simulant of methamphetamine)vapor.The sensing effect of the film is significantly improved after UV-ozone substrate treatment,and the limit of detection was decreased by 10.4 times from 2.59 to 0.25 ppm.Further experiments show that the sensing performance of other fluorescent probe can also be improved by the UV-ozone substrate treatment.This convenient and general method may become a very effective approach to improve the performance of film-based fluorescent sensors.Bin Li Keke Li Wei Xu Mingzhu Yan Jianhao Zhao Wukun Zhang Mingshuai Yuan Yanyan Fu Qingguo He Jiangong Cheng 2023Nano Research2023,16,3:0
9Numerical Prediction of Compound Layer Growth of Steel En40B during Plasma Nitriding显示文摘This paper deals with the kinetics of compound layer growth of steel En40B plasmas nitrided at 520°C for different time and establishes the corresponding mathematical model based on the thermodynamics and kinetics of nitrogen diffusion process. The results show that the compound layer consists of dual phases e (Fe2-3N) and / (Fe4N) and its thickness increases with increasing nitriding time. The calculated data indicate that the critical concentration between compound layer and diffusion one is about 0.82wt%. 10Fatigue Behavior of a Dissimilar Aluminum Alloy Welding Joint With and Without Natural Defect显示文摘In order to investigate the influence of natural defect on the fatigue behavior of 5A06/7A05 dissimilar aluminum alloys welding joint,fatigue tests of two types of specimens with and without defects were carried out systematically under stress amplitude control conditions (stress ratio R=0.1) at normal temperature in laboratory air condition.Furthermore,a new parameter,i e,fatigue defect effect factor (FDEF) was introduced to assess the effect of defect on fatigue strength.The fatigue failure analysis was conducted as well to compare the fatigue and fracture behavior of the two types of specimens.The results show that:(1) natural defects have a strong effect on the fatigue lives of welding joint,and the differences between the specimens with and without defects can reach 80 times under a same theoretical net sectional stress;(2) the FDEF parameter introduced is effective to deal with the defect effect,and the FDEF decreases along with the increase of fatigue life.The mean relative error between the experimental data and predicted fatigue strength based on the FDEF is 10.2%;(3) the macro fracture of both types of specimens have three typical zones,i e,fatigue source zone,crack propagation zone and final fracture zone,while there are more than one fatigue sources for specimens with natural defects.The overall pattern of crack propagation zone and fracture zone are quite similar,but the morphologies are different in details.石亮 WANG Chiquan LIU Zhiyi WANG Juntao WANG Wukun 2022Journal of Wuhan University of Technology(Materials Science)2022,37,5:0

12A Combined Experimental and Numerical Study of Shotcrete Jets and Related Wet Spray Nozzles显示文摘In this research,the dynamics of wet spray nozzles with different geometries,used to accelerate shotcrete,are investigated on the basis of a suitable three-dimensional mathematical model and related numerical method.Simulations have been conducted in the frame of the SIMPLEC algorithm.The k-εturbulence model has been used to account for turbulent effects.The study shows that when the angle of the convergent section is less than 3°,it has a scarce effect on the dynamics of the jet of shotcrete;with the increase of the convergence angle,the shotcrete jet velocity decreases and the nozzle wear increases;when this angle is greater than 6°,the concrete outlet jet velocity is very small and the nozzle can easily be blocked.Experimental results are in good agreement with the outcomes of the numerical simulations,which indicates that the used approach is reliable.Chang Su Qiangqiang Zheng Wukun Zhao 2020Fluid Dynamics & Materials Processing2020,16,5:0
