1Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
2High-Resolution Genome-wide Association Study Identifies Genomic Regions and Candidate Genes for Important Agronomic Traits in Wheat显示文摘Wheat(Triticum aestivum)is a major staple food crop worldwide.Genetic dissection of important agronomic traits is essential for continuous improvement of wheat yield to meet the demand of the world's growing population.We conducted a large-scale genome-wide association study(GWAS)using a panel of 768 wheat cultivars that were genotyped with 327609 single-nucleotide polymorphisms generated by genotyping-by-sequencing and detected 395 quantitative trait loci(QTLs)for 12 traits under 7 environments.Among them,273 QTLs were delimited to≤1.0-Mb intervals and 7 of them are either known genes(Rht-D,Vrn-B1,and Vrn-D1)that have been cloned or known QTLs(TaGA2ox8,APO1,TaSus1-7B,and Rht12)that were previously mapped.Eight putative candidate genes were identified for three QTLs that enhance spike seed setting and grain size using gene expression data and were validated in three bi-parental populations.Protein sequence analysis identified 33 putative wheat orthologs that have high identity with rice genes in QTLs affecting similar traits.Large r^2 values for additive effects observed among the QTLs for most traits indicated that the phenotypes of these identified QTLs were highly predictable.Results from this study demonstrated that significantly increasing GWAS population size and marker density greatly improves detection and identification of candidate genes underlying a QTL,solidifying the foundation for large-scale QTL fine mapping,candidate gene validation,and developing functional markers for genomics-based breeding in wheat.Yunlong Pang Chunxia Liu Danfeng Wang Paul St.Amand Amy Bernardo Wenhui Li Fang He Linzhi Li Liming Wang Xiufang Yuan Lei Dong Yu Su Huirui Zhang Meng Zhao Yunlong Liangi Hongze Jia Xitong Shen Yue Lu Hongming Jiang Yuye Wu Anfei Li Honggang Wang Lingrang Kong Guihua Bai Shubing Liu 2020Molecular Plant2020,13,9:8
3Design,methodology,and baseline of whole city‑million scale children and adolescents myopia survey(CAMS)in Wenzhou,China显示文摘Background:Myopia is the most common visual impairment in children and adolescents worldwide.This study described an economical and effective population-based screening pipeline and performed the project of a million scale children and adolescents myopia survey(CAMS),which will shed light on the further study of myopia from the level of epidemiology and precision medicine.Methods:We developed a novel population-based screening pattern,an intelligent screening process and internetbased information transmission and analysis system to carry out the survey consisting of school children in Wenzhou,China.The examination items include unaided distance visual acuity,presenting distance visual acuity,and noncycloplegic autorefraction.Myopia and high myopia were defined as spherical equivalent(SE)≤−1.00 diopters(D)and SE≤−6.00 D,respectively.Next,the reports of the vision checking were automatically sent to parents and the related departments.The CAMS project will be done two to four times annually with the support of the government.An online eyesight status information management system(OESIMS)was developed to construct comprehensive and efficient electronic vision health records(EVHRs)for myopia information inquiry,risk pre-warning,and further study.Results:The CAMS completed the first-round of screening within 30 days for 99.41%of Wenzhou students from districts and counties,in June 2019.A total of 1,060,925 participants were eligible for CAMS and 1,054,251(99.37%participation rate)were selected through data quality control,which comprised 1305 schools,and 580,609,251,050 and 170,967 elementary,middle,and high school students.The mean age of participants was 12.21±3.32 years(6-20 years),the female-to-male ratio was 0.82.The prevalence of myopia in elementary,middle,and high school students was 38.16%,77.52%,and 84.00%,respectively,and the high myopia incidence was 0.95%,6.90%,and 12.98%.Conclusions:The CAMS standardized myopia screening model involves automating large-scale information collection,data transmission,data analysis and early warning,thereby supporting myopia prevention and control.The entire survey reduced 90%of staff,cost,and time consumption compared with previous surveys.This will provide new insights for decision support for public health intervention.Liangde Xu Youyuan Zhuang Guosi Zhang Yunlong Ma Jian Yuan Changseng Tu MiaoMiao Li Wencan Wang Yaru Zhang Xiaoyan Lu Jing Li Xinting Liu Zhengbo Xue Meng Zhou Jie Sun Jinhua Bao Ming Li Fan Lu Hong Wang Jianzhong Su Jia Qu 2021Eye and Vision2021,8,1:7
4A Hybrid Inexact Optimization Model for Land-use Allocation of China显示文摘Land scarcity has become the prominent obstacle on the way to sustainable development for China. Under the constraints of land shortage, how to allocate the finite land resources to the multiple land users in China considering various political, environmental, ecological and economic conditions have become research topics with great significance. In this study, an interval fuzzy national-scale land-use model(IFNLM) was developed for optimizing land systems of China. IFNLM is based on an integration of existing interval linear programming(ILP), and fuzzy flexible programming(FFP) techniques. IFNLM allows uncertainties expressed as discrete interval values and fuzzy sets to be incorporated within a general optimization framework. It can also facilitate national-scale land-use planning under various environmental, ecological, social conditions within a multi-period and multi-option context. Then, IFNLM was applied to a real case study of land-use planning in China. The satisfaction degree of environmental constraints is between 0.69 and 0.97, the system benefit will between 198.25 × 1012 USD and 229.67 × 1012 USD. The results indicated that the hybrid model can help generate desired policies for land-use allocation with a maximized economic benefit and minimized environmental violation risk. 5Effect of interaction between different CeO_(2)plane and platinum nanoparticles on catalytic activity of Pt/CeO_(2)in toluene oxidation显示文摘The plane exposure of support vitally affects the catalytic performance of the catalyst.In this work,CeO_(2)nanorods((110)plane exposure),nano-octahedrons((111)plane exposure)and nano-cubes((100)plane exposure)were prepared as the supports of Pt/CeO_(2)samples to investigate the effect of CeO_(2)plane exposure on total toluene oxidation.Characterizations reveal that the(110)plane of CeO_(2)is more helpful to the dispersion of Pt species,followed by(111)face.The improved dispersion of Pt species can enhance the metal-supports interaction,which promotes the electron transfer of CeO_(2)carrier to Pt nanoparticles and the adsorption-activation of O_(2),thereby facilitating the total oxidation of toluene via the Langmuir-Hinshelwood(L-H)mechanism.Therefore,Pt/CeO_(2)-r(nanorods)sample expresses excellent catalytic performance of toluene oxidation.Finally,the procedure of toluene total oxidation was studied by in-situ diffuse reflectance infrared Fourier transform(DRIFT)spectroscopy.We expect that this work can contribute to the development of an effective sample for the decomposition of volatile organic compounds(VOCs).Shengsheng Chang Yong jia Yiqing Zeng Fuping Qian Lina Guo Shenghua Wu Jinli Lu Yunlong Han 2022Journal of Rare Earths2022,40,11:6
6Progress and challenges in blocked impurity band infrared detectors for space-based astronomy显示文摘Astronomical detection at infrared wavelengths is crucial in astrophysics due to the critical information in this wavelength range.Blocked impurity band(BIB) infrared detectors are desirable for space-based astronomical observation due to their broad response range, low dark currents, high quantum efficiencies, and excellent radiation resistance. In this review, typical BIB device structures and device physics development are first introduced. Subsequently, we discuss progress in Si-based BIB detectors with different doping types and emphasize their applications in space-based infrared detection. Additionally, we discuss recent efforts on pixel performance optimization, response extension, and higher operating temperature devices. 7Operation of the CSNS Penning surface H^- ion source显示文摘The accelerator complex of the China Spallation Neutron Source (CSNS) consists of a H- linear accelerator (linac) and a rapid cycling synchrotron (RCS). The linac contains a Penning surface H- ion source. The designed energy and the beam current of the source are 50 keV and 20 mA respectively, with a normalized root mean square (norm. rms.) emittance of 0.2π mm mrad. The manufactures and tests of the discharge chamber are in great progress. The construction of H- ion source test stand has been completed, and the operation of the source is also in progress. Stable H- ion beams with energy of 50 keV and current up to 50 mA are attained. 9Colonization of endophyte Acremonium sp. D212 in Panax notoginseng and rice mediated by auxin and jasmonic acid显示文摘Endophytic fungi can be beneficial to plant growth. However, the molecular mechanisms under-lying colonization of Acremonium spp. remain unclear. In this study, a novel endophytic Acremonium strain was isolated from the buds of Panax notoginseng and named Acremonium sp. D212. The Acremonium sp. D212 could colonize the roots of P. notoginseng, enhance the resistance of P. notoginseng to root rot disease, and promote root growth and saponin bio-synthesis in P. notoginseng. Acremonium sp. D212 could secrete indole-3-acetic acid (IAA) and jasmonic acid (JA), and inoculation with the fungus increased the endogenous levels of IAA and JA in P. noto-ginseng. Colonization of the Acremonium sp. D212 in the roots of the rice line Nipponbare was dependent on the concentration of methyl jasmonate (MeJA) (2–15μmol/L) and 1-naphthalenacetic acid (NAA) (10–20μmol/L). Moreover, the roots of the JA signaling-defective coi1-18 mutant were colonized by Acremonium sp. D212 to a lesser degree than those of the wild-type Nipponbare and miR393b-overexpressing lines, and the colonization was res-cued by MeJA but not by NAA. It suggests that the cross-talk between JA signaling and the auxin biosynthetic pathway plays a crucial role in the colonization of Acremonium sp. 10Establishment of two-dimensional gel electrophoresis for soybean protein isolate and its application显示文摘To optimize the conditions for the establishment of two-dimensional gel electrophoresis(2-DE)of soybean protein isolate(SPI),we investigated Ampholine mixture,anodic and cathodic electrolytes,loading amount of sample,acrylamide concentration,p H gradient and gel staining method in twodimensional gel electrophoresis to optimize the protein imaging conditions in two-dimensional gel.The results of mixed-level design experiments showed that Ampholine,loading amount and gel staining method had significant effect(P<0.05)on 2-DE of SPI.The optimal conditions were Ampholine mixture(pH 3–10+pH 5–7 or pH 4–6+pH 5–7),loading amount of 2 mg sample and silver staining.Although the acrylamide concentration of the gel,the p H gradient,the anodic and cathodic electrolyte solutions had significant statistical effects on the protein separation degree,the complexity of the protein composition and the visibility of the gel images were more inclined to the 12%gel,the 3–10 pH gradient and the H3PO4/NaOH electrolyte.According to the established conditions,the hydrolyzed products of SPI emulsion were determined by 2-DE,and the dynamic changes of protein in the process of enzymatic hydrolysis were described.Xinkai Lu Yaoming Cui Junjun Guan Xue Liu Hao Zhu Xuyang Ji Jianzhang Zheng Yunlong Cheng Xiaofei Fu 2020Grain & Oil Science and Technology2020,3,3:2

12CFD–DEM simulation of particle deposition characteristics of pleated air filter media based on porous media model显示文摘In this study, the three-dimensional physical model of pleated air filtration media was simplified to porous media model, and the calculation parameters of porous media were obtained based on experimental data. The model of V-shaped pleated air filter media is constructed, the height of the media pleat is 50 mm and the pleat thickness is 4 mm, the pleat angle is 3.7°. The Hertz-Mindlin contact model was modified by Johnson Kendall Roberts (JKR) adhesion contact model. The deposition process of particles in media was simulated based on computational fluid dynamics (CFD) theory and discrete element method (DEM). Results show that the CFD–DEM coupling method can be effectively applied to the macro research of pleated air filter media. The particles will form dust layer and dendrite structure on the fiber surface, and the dust layer will affect the subsequent air flow organization, and the dendrite structure will eventually form a “particle wall”. The formation of the “particle wall” will prevent the particles from moving further in the fluid domain, which makes area of pleated angle become the “low efficiency” part about the particle deposition. 15Guided Wave Based Damage Detection Method for Aircraft Composite Structures under Varying Temperatures显示文摘Guided waves based damage detection methods using base signals offer the advantages of simplicity of signal generation and reception,sensitivity to damage,and large area coverage;however,applications of the technology are limited by the sensitivity to environmental temperature variations.In this paper,a Spearman Damage Index-based damage diagnosis method for structural health condition monitoring under varying temperatures is presented.First,a PZT sensor-based Guided wave propagation model is proposed and employed to analyze the temperature effect.The result of the analysis shows the wave speed of the Guided wave signal has higher temperature sensitivity than the signal fluctuation features.Then,a Spearman rank correlation coefficient-based damage index is presented to identify damage of the structure under varying temperatures.Finally,a damage detection test on a composite plate is conducted to verify the effectiveness of the Spearman Damage Index-based damage diagnosis method.Experimental results show that the proposed damage diagnosis method is capable of detecting the existence of the damage and identify its location under varying temperatures.Dongyue Gao Yunlong Ma Zhanjun Wu Yuebin Zheng Hongbo Lu 2021Structural Durability & Health Monitoring2021,15,1:1

17All-optical attosecond time domain interferometry显示文摘Interferometry,a key technique in modern precision measurements,has been used for length measurement in engineering metrology and astronomy.An analogous time-domain interferometric technique would represent a significant complement to spatial domain applications and require the manipulation of interference on extreme time and energy scales.Here,we report an all-optical interferometer using laser-driven high order harmonics as attosecond temporal slits.By controlling the phase of the temporal slits with an external field,a time domain interferometer that preserves both attosecond temporal resolution and hundreds of me V energy resolution is implemented.We apply this exceptional temporal resolution to reconstruct the waveform of an arbitrarily polarized optical pulse,and utilize the provided energy resolution to interrogate the abnormal character of the transition dipole near the Cooper minimum in argon.This novel attosecond interferometry paves the way for high precision measurements in the time-energy domain using all-optical approaches.Zhen Yang Wei Cao Yunlong Mo Huiyao Xu Kang Mi Pengfei Lan Qingbin Zhang Peixiang Lu 2021National Science Review2021,8,10:1
